Lectin histochemical study of olfactory neurons in the eel.

Lectin histochemical studies were performed on paraffin embedded sections of the olfactory system of the eel to identify specific glycoconjugates on the surface of primary olfactory neurons. The olfactory receptors, the olfactory nerve fibres and their terminals in the bulbs were labelled with the lectins (SBA, BSA-I, BSA-I-B4 and DBA) HRP-conjugated or biotinylated. The lectin staining patterns indicate that the membrane of olfactory neurons of the eel had oligosaccharides with alpha-galactose and alpha-N-acetyl-D-galactosamine residues. These findings represent the demonstration of a molecul

Integrating olfactory assessments in consciousness disorders: methodologies and therapeutic potential.

INTRODUCTION: Current clinical assessments for patients with prolonged disorders of consciousness (pDOC) primarily depend on visual and auditory approaches, which often result in misdiagnosis. The olfactory system possesses a distinct anatomical pathway that projects directly to the cortex without thalamic relay, which may retain responsiveness in pDOC patients. Preliminary evidence suggests a link between smelling and consciousness processes, yet there is a gap in applying these findings clinically. METHODS: This review combines insights from neuroanatomy, behavioral neuroscience, and neurom

Development of the terminal nerve system in the shark Scyliorhinus canicula.

The nervus terminalis (or terminal nerve) system was discovered in an elasmobranch species more than a century ago. Over the past century, it has also been recognized in other vertebrate groups, from agnathans to mammals. However, its origin, functions or relationship with the olfactory system are still under debate. Despite the abundant literature about the nervus terminalis system in adult elasmobranchs, its development has been overlooked. Studies in other vertebrates have reported newly differentiated neurons of the terminal nerve system migrating from the olfactory epithelium to the telen

Organizational Principles of the Centrifugal Projections to the Olfactory Bulb.

Centrifugal projections in the olfactory system are critical to both olfactory processing and behavior. The olfactory bulb (OB), the first relay station in odor processing, receives a substantial number of centrifugal inputs from the central brain regions. However, the anatomical organization of these centrifugal connections has not been fully elucidated, especially for the excitatory projection neurons of the OB, the mitral/tufted cells (M/TCs). Using rabies virus-mediated retrograde monosynaptic tracing in Thy1-Cre mice, we identified that the three most prominent inputs of the M/TCs came fr

Histological Properties of Main and Accessory Olfactory Bulbs in the Common Hippopotamus.

The olfactory system of mammals comprises a main olfactory system that detects hundreds of odorants and a vomeronasal system that detects specific chemicals such as pheromones. The main (MOB) and accessory (AOB) olfactory bulbs are the respective primary centers of the main olfactory and vomeronasal systems. Most mammals including artiodactyls possess a large MOB and a comparatively small AOB, whereas most cetaceans lack olfactory bulbs. The common hippopotamus (Hippopotamus amphibius) is semiaquatic and belongs to the order Cetartiodactyla, family Hippopotamidae, which seems to be the closest

Olfactory cilia: our direct neuronal connection to the external world.

An organism's awareness of its surroundings is dependent on sensory function. As antennas to our external environment, cilia are involved in fundamental biological processes such as olfaction, photoreception, and touch. The olfactory system has adapted this organelle for its unique sensory function and optimized it for detection of external stimuli. The elongated and tapering structure of olfactory cilia and their organization into an overlapping meshwork bathed by the nasal mucosa is optimized to enhance odor absorption and detection. As many as 15-30 nonmotile, sensory cilia on dendritic end

Forebrain organization in elasmobranchs.

It has long been known that many elasmobranch fishes have relatively large brains. The telencephalon, in particular, has increased in size in several groups, and as a percent of total brain weight, it is as large as in some mammals. Little is known, however, about the organization, connections, and functions of the telencephalon in elasmobranchs. Early experimental studies indicated that olfaction does not dominate the telencephalon and that other sensory modalities are represented, particularly in the pallium. We have investigated the intrinsic and extrinsic connections of the telencephalon i

Electrophysiology of Olfactory and Optic Nerve in Outpatient and Intraoperative Settings.

Evoked potentials are time-locked electrophysiologic potentials recorded in response to standardized stimuli using scalp electrodes. These responses provide good temporal resolution and have been used in various clinical and intraoperative settings. Olfactory evoked potentials (OEPs) may be used as an adjunct tool in identifying patients of Parkinson disease and Alzheimer dementia. In clinical practice, visual evoked potentials (VEPs) are particularly useful in identifying subclinical cases of optic neuritis and in treatment surveillance. In recent times, pattern electroretinograms and photopi

State-dependent representations of mixtures by the olfactory bulb.

Sensory systems are often tasked to analyse complex signals from the environment, separating relevant from irrelevant parts. This process of decomposing signals is challenging when a mixture of signals does not equal the sum of its parts, leading to an unpredictable corruption of signal patterns. In olfaction, nonlinear summation is prevalent at various stages of sensory processing. Here, we investigate how the olfactory system deals with binary mixtures of odours under different brain states by two-photon imaging of olfactory bulb (OB) output neurons. Unlike previous studies using anaesthetis

MRI tractography reveals the human olfactory nerve map connecting the olfactory epithelium and olfactory bulb.

The olfactory nerve map describes the topographical neural connections between the olfactory epithelium in the nasal cavity and the olfactory bulb. Previous studies have constructed the olfactory nerve maps of rodents using histological analyses or transgenic animal models to investigate olfactory nerve pathways. However, the human olfactory nerve map remains unknown. Here, we demonstrate that high-field magnetic resonance imaging and diffusion tensor tractography can be used to visualize olfactory sensory neurons while maintaining their three-dimensional structures. This technique allowed us

[How is the sense of smell connected? Cellular and molecular mechanisms guiding the development of the synaptic connections from the nose to the cortex (I)].

The physiological particularities that occur during the development of the olfactory system make it one of the most fascinating parts of the central nervous system and one of models that has been most widely studied in order to understand the mechanisms related with axonal growth and guidance towards the right targets. A variety of mechanisms are known, some mediated by contact (laminins, cell adhesion molecules, ephrins, etc.) and others that are secreted (semaphorins, slits, growth factors, etc.), to play diverse roles in establishing the synaptic interactions among the olfactory epithelium,

[Surface of the olfactory region in man observed by scanning electron microscope].

This report describes the surface of the regio olfactoria of a 25-year-old man as observed in the scanning electron-microscope. Corresponding to the distal ends of the receptor cells, the surface displays various planes: (a) Upper-most is a film of mucus of varying thickness. (b) The olfactory hairs, which are arranged closely together, mostly in a parallel fashion in layers, dip into this mucus film. The long, distal parts of the olfactory hairs are of a rather uniform shape and form, together with the mucus film, a single functional unit. (c) Now follows a layer consisting of the short parts

Olfaction in Lamprey Pallium Revisited-Dual Projections of Mitral and Tufted Cells.

The presence of two separate afferent channels from the olfactory glomeruli to different targets in the brain is unravelled in the lamprey. The mitral-like cells send axonal projections directly to the piriform cortex in the ventral part of pallium, whereas the smaller tufted-like cells project separately and exclusively to a relay nucleus called the dorsomedial telencephalic nucleus (dmtn). This nucleus, located at the interface between the olfactory bulb and pallium, in turn projects to a circumscribed area in the anteromedial, ventral part of pallium. The tufted-like cells are activated wit

The connections between the olfactory bulb and the brain in the goldfish.

We have investigated olfactory bulb connections in the goldfish by using both horseradish peroxidase applied to olfactory tract lesions and wheat germ agglutinin-conjugated peroxidase administered to the olfactory bulb. The projections from the bulb pass to the brain in two major bundles: the lateral and the medial olfactory tracts. The lateral tract innervates the posterior terminal field in the area dorsalis. The medial tract divides into two rami. The dorsolateral ramus is the most substantial olfactory bundle in the brain and innervates several targets in both the area dorsalis and the are
