Intranasal chemosensory function of the trigeminal nerve and aspects of its relation to olfaction.

OBJECTIVES: The review outlines characteristics of the intranasal trigeminal chemosensory system. In addition, it provides selective comparisons of the trigeminal and olfactory systems, the two of which interact at multiple levels. RESULTS AND CONCLUSIONS: This interaction between the trigeminal and olfactory systems is an important determinant of sensations of odor. Further, it appears to change as a result of aging and disease. Thus, the interaction between the olfactory and trigeminal systems is not straightforward and may be difficult to predict, but it has a powerful influence on the per

Investigating the Link between Eating Attitudes, Taste and Odour Preferences and the Chemical Senses.

OBJECTIVES: To examine if higher degrees of pathological eating attitudes in a non-clinical sample are associated with odour and taste perception and preferences based on psychophysical ratings. PARTICIPANTS AND METHODS: A total of 80 female university students completed the eating attitudes test (EAT-26), followed by four chemosensory measures including olfactory and gustatory perception plus perceptual ratings and preferences for food odours and tastes. RESULTS: There were no significant correlations between EAT-26 scores and measures of olfactory and gustatory perception. However, a signi

Diseases of the nasal cavity.

Despite garnering minimal attention from the medical community overall, olfaction is indisputably critical in the manner in which we as humans interact with our surrounding environment. As the initial anatomical structure in the olfactory pathway, the nasal airway plays a crucial role in the transmission and perception of olfactory stimuli. The goal of this chapter is to provide a comprehensive overview of olfactory disturbances as it pertains to the sinonasal airway. This comprises an in-depth discussion of clinically relevant nasal olfactory anatomy and physiology, classification systems of

Identifying molecular pathways of olfactory dysfunction in Parkinson's disease through a systems biology framework.

The sense of smell is essential for human perception. Olfactory function declines with increasing age, affecting a substantial portion of the elderly population, and this decline is more pronounced in men. This reduction can be attributed to anatomical and degenerative changes in the brain and olfactory receptors. There is robust clinical evidence indicating an association between olfactory perception decline/deficit (OPD) and major neurodegenerative diseases, with severe deficits observed in Alzheimer's and Parkinson's disease and milder effects noted in other conditions. However, its molecul

Blindness, But Not HMHA Anosmia, Predicts Loneliness: A Psychophysical Study.

Olfactory deficits can play a detrimental role in everyday social functioning. Perception of 3-hydroxy-3-methylhexanoic acid (HMHA)-a body odor component-could also be linked to this research area. However, no study so far has addressed the problem of HMHA perception in the context of the previously reported relationship between olfactory abilities and social difficulties. Here, we tested whether HMHA-specific anosmia predicted loneliness understood both as a cognitive evaluation of social participation and as one's social isolation, and we additionally analyzed the effects and correlates of H

Sensory loss enhances multisensory integration performance.

Auditory and visual sensory loss has repeatedly been shown to alter abilities in remaining sensory modalities. It is, however, unclear whether sensory loss also impacts multisensory integration; an ability that is fundamental for the perception of the world around us. We determined effects of olfactory sensory deprivation on multisensory perception by assessing temporal as well as semantic aspects of audio-visual integration in 37 individuals with anosmia (complete olfactory sensory loss) and 37 healthy, matched controls. Participants performed a simultaneity judgement task to determine the te

Osmotexia and the somatosensory dimension of smell: toward a fourth axis in chemosensory evaluation.

BACKGROUND: The evaluation of olfaction is classically based on three psychophysical axes: threshold, discrimination, and identification. However, these do not fully reflect the multisensory nature of odour perception. It is well established that numerous odours can activate the trigeminal system, thereby generating somatosensory sensations such as texture, density, or temperature. This is a dimension that is currently absent from clinical evaluation models. AIMS/OBJECTIVES: The present study introduces and conceptualises osmotexia as the ability to experience odours in terms of internal text

Target-dependent routing of sensory information in the olfactory system.

Parallel processing, the ability to split the information content of sensory stimuli and handle them simultaneously, is essential for sensory perception in many systems. Mitral (MCs) and tufted cells (TCs) in the olfactory bulb (OB), with their separate but overlapping projection areas, provide an ideal anatomical basis for parallel processing. Whether they convey different information to distinct areas of the olfactory cortex remains unclear. Here, we separated mouse OB output neurons by innervation area using retrograde tracing from the anterior olfactory nucleus (AON) or the anterior pirifo

Effects of Endoscopic Sinus Surgery on Olfactory Function.

PURPOSE OF REVIEW: To review the effects of endoscopic sinus surgery and endonasal approaches to the skull base on olfaction. RECENT FINDINGS: Advancements in endonasal endoscopic approaches to the sinuses and skull base allow for direct treatment of a variety of sinonasal and skull base diseases. However, these extended approaches will often require manipulation of normal anatomical structures and the olfactory neuroepithelium. Depending on the planned procedure and extent of disease, the prognosis of olfactory perception can vary significantly among patients. Endoscopic sinonasal surgical p

Genome-wide association meta-analysis of human olfactory identification discovers sex-specific and sex-differential genetic variants.

Smelling is a human sense, expressing strong sexual dimorphisms. We aim to improve the knowledge of the genetics of human olfactory perception by performing an exploratory genome-wide association meta-analysis of up to 21,495 individuals of European ancestry. By sex-stratified and overall analysis of the identification of twelve odours and an identification score, we discovered ten independent loci, seven of them novel, with trait-wise genome-wide significance (p < 5 × 10-8) involving five odours. Seven of these loci, including four novel ones, are also significant using a stricter study-wide

Non-neuronal expression of SARS-CoV-2 entry genes in the olfactory system suggests mechanisms underlying COVID-19-associated anosmia.

Altered olfactory function is a common symptom of COVID-19, but its etiology is unknown. A key question is whether SARS-CoV-2 (CoV-2) - the causal agent in COVID-19 - affects olfaction directly, by infecting olfactory sensory neurons or their targets in the olfactory bulb, or indirectly, through perturbation of supporting cells. Here we identify cell types in the olfactory epithelium and olfactory bulb that express SARS-CoV-2 cell entry molecules. Bulk sequencing demonstrated that mouse, non-human primate and human olfactory mucosa expresses two key genes involved in CoV-2 entry, ACE2 and TMPR

Characterization of pathological changes in the olfactory system of mice exposed to methylmercury.

Methylmercury (MeHg) is a well-known environmental neurotoxicant that causes severe brain disorders such as Minamata disease. Although some patients with Minamata disease develop olfactory dysfunction, the underlying pathomechanism is largely unknown. We examined the effects of MeHg on the olfactory system using a model of MeHg poisoning in which mice were administered 30 ppm MeHg in drinking water for 8 weeks. Mice exposed to MeHg displayed significant mercury accumulation in the olfactory pathway, including the nasal mucosa, olfactory bulb, and olfactory cortex. The olfactory epithelium was

Hyperbaric oxygen therapy for treatment of COVID-19-related parosmia: a case report.

Parosmia is a qualitative olfactory dysfunction characterized by distortion of odor perception. Traditional treatments for parosmia include olfactory training and steroids. Some patients infected with COVID-19 have developed chronic parosmia as a result of their infection. Here, we present the case of a patient who developed parosmia after a COVID-19 infection that was not improved by traditional treatments but found significant improvement after hyperbaric oxygen therapy[A1].

Dynamics of odor sampling strategies in mice.

Mammalian olfactory receptor neurons in the nasal cavity are stimulated by odorants carried by the inhaled air and their activation is therefore tied to and driven by the breathing or sniffing frequency. Sniffing frequency can be deliberately modulated to alter how odorants stimulate olfactory receptor neurons, giving the animal control over the frequency of odorant exposure to potentially aid odorant detection and discrimination. We monitored sniffing behaviors and odorant discrimination ability of freely-moving mice while they sampled either decreasing concentrations of target odorants or sa

Flavor perception test: evaluation in patients with Kallmann syndrome.

In Kallmann syndrome (KS), congenital hypogonadism is associated with olfactory impairment. To evaluate flavor perception-related disability in KS patients, 30 patients with KS, 12 with normosmic hypogonadism (nIHH), 24 with acquired anosmia (AA), and 58 healthy controls entered the study. All participants completed questionnaires concerning dietary habits, olfaction-related quality of life (QoL), and self-determined olfactory, flavor, and taste abilities prior to undergoing standardized olfactometry and gustometry. Each subject underwent flavor testing, using orally administered aqueous aroma

Dietary habits and olfactory function in individuals with Parkinson's disease: an underexplored association.

Parkinson's disease (PD) is a progressive neurological disorder characterized by motor and non-motor symptoms, including olfactory dysfunction, which may precede motor manifestations by up to 10-20 years. Given the role of olfaction in flavor perception and dietary behavior, olfactory impairment may significantly affect nutritional status and quality of life in individuals with PD. This opinion article discusses the relationship between olfactory dysfunction and dietary habits in PD, highlighting the scarcity and inconsistency of the available evidence. While some studies suggest that lower di

The Sense of Smell Impacts Metabolic Health and Obesity.

Olfactory inputs help coordinate food appreciation and selection, but their role in systemic physiology and energy balance is poorly understood. Here we demonstrate that mice upon conditional ablation of mature olfactory sensory neurons (OSNs) are resistant to diet-induced obesity accompanied by increased thermogenesis in brown and inguinal fat depots. Acute loss of smell perception after obesity onset not only abrogated further weight gain but also improved fat mass and insulin resistance. Reduced olfactory input stimulates sympathetic nerve activity, resulting in activation of β-adrenergic r

Proof-of-concept: SCENTinel 1.1 rapidly discriminates COVID-19 related olfactory disorders.

It is estimated that 20-67% of those with COVID-19 develop olfactory disorders, depending on the SARS-CoV-2 variant. However, there is an absence of quick, population-wide olfactory tests to screen for olfactory disorders. The purpose of this study was to provide a proof-of-concept that SCENTinel 1.1, a rapid, inexpensive, population-wide olfactory test, can discriminate between anosmia (total smell loss), hyposmia (reduced sense of smell), parosmia (distorted odor perception), and phantosmia (odor sensation without a source). Participants were mailed a SCENTinel 1.1 test, which measures odor

The excisional, plication and internal drainage techniques: a comparison of the results for idiopathic hydrocele.

The pathogenesis of Crohn's disease (CD) is still unknown, but the involvement of the olfactory system in CD appears possible. No study to date has systematically assessed the olfactory function in CD patients. We investigated the olfactory function in CD patients in active (n = 31) and inactive disease (n = 27) and in a control group of age- and sex-matched healthy subjects (n = 35). Subjective olfactory testing was applied using the Sniffin' Sticks test. Impaired brain response to odors in patients with varied severity of olfactory loss after traumatic brain injury.

Traumatic brain injury (TBI) as a major public health problem may lead to olfactory dysfunction. However, little is known about brain responses to odors in TBI olfactory loss patients. Nineteen healthy controls and forty TBI olfactory dysfunctional patients (19 with hyposmia and 21 with anosmia) underwent a functional magnetic resonance imaging scan when two odors (peach and coffee) were presented intranasally using a computerized olfactometer. Olfactory performance was measured using the "Sniffin' Sticks" test. TBI patients with hyposmia or anosmia showed decreased odor-induced brain activati
