Effects of L-thyroxine and ovine growth hormone on smoltification of amago salmon (Oncorhynchus rhodurus).

In a study of the hormonal control of salmon smoltification, L-thyroxine (T4; 200 ppm in diet), ovine growth hormone (oGH; 2 micrograms/g intramuscular injection weekly), or a combination of these hormones was administered to underyearling amago salmon parr for 72 days. Administration of both T4 + oGH remarkably stimulated growth of the fish. All hormone treatments caused body silvering of the fish. The body silvering was the greatest in the group treated with T4 + oGH. Both T4 + oGH and oGH increased survival rate of the fish in 32% seawater, and kept plasma osmolarity and plasma sodium conce

Transcriptome-and Metabolome-Based Mechanisms of High-Temperature Adaptation in Triploid Rainbow Trout (Oncorhynchus mykiss).

High-temperature stress poses a critical challenge to cold-water aquaculture; however, the size-associated molecular mechanisms underlying thermal adaptation in triploid rainbow trout (Oncorhynchus mykiss) remain poorly understood. Here, we integrated transcriptomic and metabolomic profiling of liver tissues, which plays central role in energy metabolism and stress integration, from three body-weight classes-small (0.8 kg ± 0.18 kg), medium (1.5 kg ± 0.22 kg), and large (2.5 kg ± 0.31 kg)-sampled under peak summer heat stress (20.5 °C). Transcriptomic analysis identified 974, 570, and 862 grou

Immune effects of lipids isolated from Oncorhynchus mykiss eggs on cyclophosphamide-induced immunosuppression in mice.

Rainbow trout (Oncorhynchus mykiss) eggs are a rich source of polyunsaturated fatty acids (PUFAs), with levels exceeding those found in fish meat, and are also a notable source of bioactive omega-3 fatty acids, primarily eicosapentaenoic acid (EPA)and docosahexaenoic acid (DHA). However, despite their nutritional advantages, their in vivo immunomodulatory properties remain poorly understood. This study explored the immunomodulatory potential of the lipid extract from O. mykiss eggs combined with PEG6000 (OM-PEG) in a cyclophosphamide (CY)-induced immunosuppressive BALB/c mouse model. Mice were

11-Deoxycorticosterone (DOC)'s Action on the Gill Osmoregulation of Juvenile Rainbow Trout (Oncorhynchus mykiss).

In aquaculture, stress can negatively affect fish growth. For years, the cortisol hormone has been thought to play both glucocorticoid and mineralocorticoid functions. Nevertheless, recent research has suggested that 11-deoxycorticosterone (DOC) released during stress could contribute to cortisol actions, though this process is still misunderstood. Here, we evaluated the DOC effects on physiological and early transcriptional responses by RNA-seq. Juvenile rainbow trout were treated with DOC and/or glucocorticoids (mifepristone) or mineralocorticoid (eplerenone) receptor antagonists. Subsequent

Diploid and triploid Chinook salmon (Oncorhynchus tshawytscha) exhibit differential immunological responses to acute thermal stress.

Exposure to temperatures outside of a fish's optimal range results in suppression of the immune system, ultimately leaving aquaculture stocks susceptible to disease outbreaks. This effect is exacerbated in triploid fishes, which demonstrate greater susceptibility to stress than their diploid counterparts. This study investigates the impacts of acute heat stress on the abundance of immune transcripts and proteins in diploid and triploid Chinook salmon (Oncorhynchus tshawytscha), an important finfish crop. This study also demonstrates that acute heat stress induces significant increases in the a

Thiamine Supplementation Improves Survival and Body Condition of Hatchery-Reared Steelhead (Oncorhynchus mykiss) in Oregon.

Early rearing of steelhead (Oncorhynchus mykiss) in Oregon hatcheries is often problematic; fry can become emaciated and die during the period between hatch and first feed. Thiamine (vitamin B1) deficiency has caused early mortality in salmonids; however, the thiamine status of Oregon's steelhead populations is unknown, to date. Of the 26 egg samples from three Oregon hatcheries in 2019, 20 (77%) had thiamine levels < 10 nmol/g, and 13 of those samples (50%) had levels <6.5 nmol/g, suggesting the thiamine deficiency of adult, female steelhead. To investigate if thiamine deficiency was causally

Functional and Molecular Immune Response of Rainbow Trout (Oncorhynchus mykiss) Following Challenge with Yersinia ruckeri.

Currently, aquaculture production of rainbow trout (Oncorhynchus mykiss) is a multibillion dollar industry; nevertheless, the development of this sector has not been exempt from pitfalls related to the recurrent presence of pathogens of bacterial origin. This is the case of Yersinia ruckeri, the etiologic agent of the infectious pathology known as Enteric Red Mouth Disease (ERM), causing serious economic losses that can be as high as 30-70% of production. Although several studies have been performed regarding pathogen features and virulence factors, more information is needed about the host de

Time-course transcriptome analyses of spleen in rainbow trout (Oncorhynchus mykiss) post-Flavobacterium psychrophilum infection.

Flavobacterium psychrophilum, the etiological agent of bacterial coldwater disease and rainbow trout fry syndrome, causes considerable losses in salmonid aquaculture globally. Systemic F. psychrophilum infections in rainbow trout (Oncorhynchus mykiss) lead to a range of clinical signs, including ulcerative lesions in the skin and muscle and splenitis. Previous studies offered an integrative analysis of the skeletal muscle response to F. psychrophilum infection in rainbow trout. However, little is known about the molecular mechanism of immune response in the spleen, which is an important immune

Clinical and histopathological changes in rainbow trout Oncorhynchus mykiss experimentally infected with fungus Bradymyces oncorhynchi.

Bradymyces oncorhynchi is a poorly known melanised fungal species that has been isolated only from a hyperaemic focus near the enlarged spleen of a rainbow trout. Although the pathogenicity of this species to fish is suspected, it has not been fully confirmed. Four laboratory experiments were conducted to test the effect of the fungus on the health of rainbow trout fingerlings. Mycelia were cultivated under different conditions to increase inoculum variability and to test the impact of the conditions on the pathogenicity. The inoculum was subsequently administered by the intraperitoneal route.

Erythromycin and florfenicol treatment of rainbow trout Oncorhynchus mykiss (Walbaum) experimentally infected with Flavobacterium psychrophilum.

Flavobacterium psychrophilum is responsible for significant economic losses in rainbow trout aquaculture. Antimicrobial treatment remains the primary means of control; however, there are limited choices available for use. The objectives of the study were therefore to determine the minimum inhibitory concentrations for erythromycin and florfenicol in selected F. psychrophilum isolates and to evaluate their clinical treatment efficacy in experimentally infected rainbow trout. All isolates tested had moderate susceptibility to florfenicol and erythromycin except one isolate, which had low suscept

Ploidy Has Minimal Effect on Hypoxia Tolerance at High Temperature in Rainbow Trout (Oncorhynchus mykiss).

Polyploidy is an important driver of evolutionary change (generally via tetraploidy) and also serves a practical role in aquaculture and fisheries management (via triploidy). Fundamental changes in cell size and number that accompany polyploidy are predicted to affect cellular and whole-animal physiology due to constraints placed on surface-mediated processes at the cellular level, potentially altering environmental tolerances and optima. The aim of this study was to determine whether the documented reduction in thermal tolerance of aquatic polyploids is a result of their being less hypoxia to

Borax Supplementation Alleviates Hematotoxicity and DNA Damage in Rainbow Trout (Oncorhynchus mykiss) Exposed to Copper.

Heavy metals have harmful effects on health of both ecosystems and organisms to their accumulation ability. Copper (Cu) is an essential element for organism survival, but EPA considers Cu as a priority pollutant. On the other hand, boron has well-defined biological effects in living organisms including cytoprotection and genoprotection, although borax (BX) metabolism is poorly described in fish. Moreover, the effects of boron supplementation against Cu-induced hematotoxicity and DNA damage in aquatic organisms are still undetermined. Therefore, the main aim of this study was to provide an over

Effect of follicle cell autophagy on gonadal development of triploid female rainbow trout (Oncorhynchus mykiss).

Autophagy is a cellular process which occurs in eukaryotic cells. To study the mechanism regulating polyploid fish growth and development is of significance in genetic, because of its growth advantages and economic values. This study focused on triploid female rainbow trout (RBT) which discusses the effects of autophagy on gonadal development of polyploid fish. Autophagy-related genes of RBT lc3b, atg12, atg4b, gabarap1, and bcl2 were cloned, and autophagy gene expressions in gonads were analyzed at different developmental period. Gonadal ultrastructures were observed under transmission electr

Detection and identification of Diphyllobothrium nihonkaiense plerocercoids from wild Pacific salmon (Oncorhynchus spp.) in Japan.

We investigated the risk of diphyllobothriasis from ingestion of wild Pacific salmon in Japan by surveying Diphyllobothrium plerocercoids in 182 salmon samples obtained from Japan. The plerocercoids were not detected in chum salmon (Oncorhynchus keta) (0/26), called Akizake in Japan, caught between September and November. However, the detection rate of plerocercoids in chum salmon, called Tokishirazu in Japan, caught between early April and June, was 51.1% (24/47) with an average of two plerocercoid larvae per fish. The detection rates of cherry salmon (Oncorhynchus masou) and pink salmon (Onc

Susceptibility of sexually mature rainbow trout, Oncorhynchus mykiss to experimental cryptobiosis caused by Cryptobia salmositica.

Sexually mature rainbow trout, Oncorhynchus mykiss, were highly susceptible to cryptobiosis caused by Cryptobia salmositica. Spawning female trout were more susceptible (higher parasitaemia and mortality) than sexually mature males. Most infected female trout (seven of nine) with eggs died before or shortly after spawning; however, none of the nine infected sexually matured males or the uninfected fish died. There was no significant difference in the severity of the anaemia between infected male and female trout. All infected males developed exophthalmia, while this clinical sign was not seen

Evaluation of immune functions of rainbow trout (Oncorhynchus mykiss)--how can environmental influences be detected?

In fish, the first line of defense against infectious microorganisms is based on a broad range of nonspecific humoral and cellular immune mechanisms ("innate immunity") which without prior specific activation can act in forming a more static barrier (Fish Shellfish Immunol. 10 (2000) 243; Dev. Comp. Immunol. 25 (2001) 827). This natural resistance is normally effective enough to protect fish from infectious diseases until specific immune responses are being induced (Fig. 1; Dev. Comp. Immunol. 25 (2001) 841). Healthy fish exhibit both nonspecific and specific immune responses depending directl

Type II iodothyronine deiodinase is preferentially expressed in rainbow trout (Oncorhynchus mykiss) liver and gonads.

It is well admitted that thyroid hormones (TH) play a role in the development of vertebrates. The major secretory product of the thyroid is a pro-hormone, T(4), which is activated in peripheral tissues by outer ring deiodination to T(3). We have isolated from rainbow trout testis, a full length cDNA encoding type II iodothyronine deiodinase (rtD2). The cDNA was 2410 nucleotides long and coded for a polypeptide of 264 amino acids including a selenocysteine residue. The predicted molecular weight of rtD2 was 29.3 kDa and the isoelectric point 8.71. The deduced amino acids sequence showed 80% ide

[Parasitic helminths infection in Coho salmón, Oncorhynchus kisutch, during their return to Simpson river, Chile].

Between may and july 1994, 17 adult returning salmons, Oncorhynchus kisutch, were collected in the River Simpson, Chile. All fishes showed infection by plerocercoids of Diphyllobothrium sp. in different locations: stomach, spleen, liver, mesenteries and gonads. Infection with larval cestodes of an unidentified species of Phillobothriidae was determined in the intestine of seven (41.2%) salmons and its prevalences of infection showed significant differences between female and male salmons. The 94.4% of total plerocercoids of Diphyllobothrium were isolated from the stomach wall. Prevalence and m
