The effects of nitric oxide donors on the sporulation of Eimeria tenella oocysts.

Nitric oxide (NO) is involved in many physiological processes including regulation of blood pressure, immune response, and neural communication. The effects of NO donors on the sporogony of Eimeria tenella oocysts and the blocking effects of NO scavenger, hemoglobin (Hb), against NO donors were investigated in this study. The results showed that the sporulation of oocysts could be completely inhibited by acidic sodium nitrite (NaNO2), S-nitroso-glutathione (GSNO) or S-nitroso-N-acetyl-dl-penicillamine (SNAP) in profound dose-dependent and time-dependent manners, and the inhibiting process was

Nucleic acid stains as indicators of Cryptosporidium parvum oocyst viability.

We developed nucleic acid dye staining methodology for untreated, heat-treated and chemically inactivated C. parvum oocysts. The nucleic acid staining was compared to in vitro excystation and animal infectivity using split samples of oocysts. Among the nucleic acid stains tested, SYTO-9, hexidium and SYTO-59 stained the oocysts consistently, and the staining was related to the infectivity of the oocysts to neonatal CD-1 mice but not to in vitro excystation. The nucleic acid viability assay was used to determine log-inactivations of the oocysts after treatment with ozone, chlorine, chlorine dio

Pattern of Cryptosporidium parvum oocyst excretion by experimentally infected dogs.

Six 6-week-old Beagle dogs were fed Cryptosporidium parvum oocysts of calf origin. All 6 dogs shed oocysts in faeces. Greater numbers of oocysts were detected with a Weber concentration technique (formalin-ethyl acetate extraction and NaCl centrifugal flotation) stained with either fluorescent antibody or modified Ziehl-Neelsen than with other formalin-ether or -ethyl acetate extraction methods. Oocyst numbers g-1 of faeces rose from days 3 to 5 to a first and highest peak lasting to days 7-9, and 5 of the 6 dogs passed oocysts for at least 80 days. However, the numbers of oocysts detected in

Effect of pasteurization on infectivity of Cryptosporidium parvum oocysts in water and milk.

Cryptosporidium parvum is a major cause of diarrheal disease in humans and has been identified in 78 other species of mammals. The oocyst stage, excreted in feces of infected humans and animals, has been responsible for recent waterborne outbreaks of human cryptosporidiosis. High temperature and long exposure time have been shown to render oocysts (suspended in water) noninfectious, but for practical purposes, it is important to know if high-temperature--short-time conditions (71.7 degrees C for 15 s) used in commercial pasteurization are sufficient to destroy infectivity of oocysts. In this s

Detection of Cryptosporidium oocysts in extra-intestinal tissues of sheep and pigs.

Extra-intestinal infections by Cryptosporidium parvum have been detected in pigs and sheep. Detection was carried out by imprints of the mucosa of different organs and viscera in 55 sheep and 57 pigs slaughtered at three abattoirs in Zaragoza (northeast Spain). Imprints were stained by using a modified Ziehl-Neelsen technique. In addition to intestinal infections, cryptosporidial oocysts were found in the gall-bladders of two pigs which were 2 months old, and in some organs of sheep aged 5 days or more, including the gall-bladder (5), mesenteric lymph nodes (2), trachea (7), lung (3) and the u

Oocyst production and immunogenicity of Cryptosporidium muris (strain MCR) in mice.

Three-week-old ICR SPF mice were orally inoculated with one of 5 doses ranging from 2 x 10(2) to 2 x 10(6) oocysts of Cryptosporidium muris (strain MCR) per mouse. Oocyst inoculation was directly proportional to the amount of oocysts shed and was inversely proportional to the period required for peak oocyst production and to the prepatent period. Peak oocyst production occurred between fifteen and thirty-one days with a patent period from 61 to 64 days. Three days after all mice stopped shedding oocysts, they were orally challenged with a single dose of 2 x 10(6) oocysts of the same species. M

Techniques for the recovery and identification of Cryptosporidium oocysts from stool specimens.

Due to increasing numbers of patients with documented infections with Cryptosporidium and other coccidia, it is important for the physician and clinical laboratory to be aware of the appropriate diagnostic techniques necessary for organism recovery and identification. Although Cryptosporidium is found in the gastrointestinal tract, tissue biopsies may be insufficient for organism recovery; the examination of stool specimens is a noninvasive procedure and will provide better overall opportunities for organism recovery. Human clinical specimens were examined from 45 patients with confirmed crypt

Development of a molecular assay for the determination of Eimeria tenella oocyst viability.

Coccidiosis is caused by apicomplexan parasites of the genus Eimeria, which infect epithelial cells of the intestinal tract causing diarrhea and negatively impacting production in the poultry industry. The self-limiting and highly immunogenic nature of infection by Eimeria spp. make live vaccination an effective means of coccidiosis control. Paramount to vaccine efficacy is the ability to administer precise numbers of viable oocysts. Unfortunately, no rapid and accurate method for determination of oocyst viability is available presently. This study presents the development of a qPCR-based assa

Evaluation of a modified method for the detection of Cryptosporidium oocysts on spinach leaves.

Despite the infection risk associated with the consumption of contaminated food, techniques for recovering and detecting Cryptosporidium oocysts from fruit and vegetables are generally inadequate due to the variable recovery efficiencies and high reagent costs, such as those presented by ISO 18744:2016 "Microbiology of the food chain -Detection and enumeration of Cryptosporidium and Giardia in fresh leafy green vegetables and berry fruits". Although an improved method for recovering these parasites from Iceberg lettuce, which reported increased recovery efficiency as well as lower costs, has b

Evaluation of novel oocyst wall protein candidates of Toxoplasma gondii.

Oocyst stage of Toxoplasma gondii is characterized by a durable wall that confers a strong protection to this protozoan parasite in face of harsh environmental conditions. Thus, it is considered the key for transmission of T. gondii. Analysis of oocyst wall composition is mandatory therefore; the aim of this study was to identify novel T. gondii oocyst wall proteins and test their use in detection of these oocysts in environmental samples. Five candidates of novel T. gondii oocyst wall proteins (TgOWPs) were identified and named TgOWP8 through TgOWP12. Recombinant protein of TgOWP8 was express

Toxoplasma gondii: A study of oocyst re-shedding in domestic cats.

The aim of the present study was to evaluate the re-shedding of T. gondii oocysts in cats fed tissue cysts of homologous and heterologous strains 12, 24 and 36 months after the first infection. Thirteen cats were used in the present study and were divided into four groups: G1 (n=2), G2 (n=3), G3 (n=5), and G4 (n=3). G1, G3 and G4 cats were infected with brain cysts of ME49 and G2 with TgDoveBr8, both genotype II strains of T. gondii. The G1 and G2 cats were re-infected after twelve months with brain cysts of VEG strain (genotype III), and G3 cats were re-infected with TgDoveBr1 (genotype II).

Comparison of diagnostic techniques for the detection of Cryptosporidium oocysts in animal samples.

While a large number of laboratory methods for the detection of Cryptosporidium oocysts in faecal samples are now available, their efficacy for identifying asymptomatic cases of cryptosporidiosis is poorly understood. This study was carried out to determine a reliable screening test for epidemiological studies in livestock. In addition, three molecular tests were compared to identify Cryptosporidium species responsible for the infection in cattle, sheep and horses. A variety of diagnostic tests including microscopic (Kinyoun's staining), immunological (Direct Fluorescence Antibody tests or DFA

The detection of a novel type of Cryptosporidium andersoni oocyst in cattle in Japan.

Cryptosporidium muris, found in rodents and cattle, has been recognized as a valid species. However, this organism from cattle was recently separated from C. muris infecting rodents based on molecular data and a transmission study. As a consequence, it has been proposed as a new species, C. andersoni. More recently, C. andersoni, which has infectivity to rodents, was detected in cattle in Japan, where it has been designated as a novel type. However, isolates from cattle in Japan have not been analyzed genetically, and therefore it remains unclear whether a novel type of C. andersoni is distrib

Occurrence of Cryptosporidium sp. oocysts in fecal and water samples in Austria.

Oocysts of Cryptosporidium spp. were detected and differentiated by a modular arranged gene amplification procedure in various samples, mostly human stool, feces of herpetotaxa, and water, in different locations of South and Eastern Austria. Cryptosporidium parvum was found in stool samples of immunocompromised persons, in reptile feces, and in water samples. The presence of Cryptosporidium in an area is probably associated with high human population densities since water from protected sources in sparsely inhabited areas is rarely contaminated.

[Cryptosporidium oocysts and epidemic safety of drinking water in the Russian Federation].

The paper provides evidence that Cryptosporidium oocysts should be necessarily included as an index of the epidemic safety of drinking water in our country. This is evidenced by: the wide spread of cryptosporidiosis among animals and human beings; that of the pathogen of this disease in the environment; large-scale outbreaks of cryptosporidiosis in the world's population; no sanitary and parasitological drinking water monitoring in our country; the use of this parasitological index to evaluate drinking water quality in some countries (USA, UK, Ukraine, etc.).

beta-tubulin mRNA as a marker of Cryptosporidium parvum oocyst viability.

Determining the viability of waterborne Cryptosporidium parvum oocysts remains a technical challenge. rRNA and mRNA were evaluated in a reverse transcription (RT)-PCR assay as potential markers of oocyst viability. The rationale for this approach is the rapid turnover and postmortem decay of cellular RNA. The beta-tubulin mRNA and an anonymous mRNA transcript were chosen as potential markers because they are the only mRNA species in C. parvum known to possess introns. This feature facilitated the distinction between genuine RT-PCR products and PCR products originating from copurifying DNA. Pro
