Improving noninvasive diagnosis of cutaneous and gastrointestinal forms of Pythium insidiosum infections in dogs: complementary roles of serum-based ELISA and polymerase chain reaction.

OBJECTIVE: To evaluate the diagnostic performance of ELISA and PCR for detecting pathogenic oomycete infections and to explore associations with clinical presentation and treatment status. METHODS: Canine sera (n = 796) submitted to Pythium Serology Diagnostic Laboratory (from 2023 through 2025) were analyzed by ELISA to detect anti-Pythium antibodies and a PCR targeting Pythium insidiosum, Lagenidium spp, and Paralagenidium spp. Clinical records were reviewed for a subset of submissions. Analyses included χ2 tests, a linear mixed-effects model, and Cohen κ for ELISA-PCR agreement. RESULTS:

Loss of function of a DMR6 ortholog in tomato confers broad-spectrum disease resistance.

Plant diseases are among the major causes of crop yield losses around the world. To confer disease resistance, conventional breeding relies on the deployment of single resistance (R) genes. However, this strategy has been easily overcome by constantly evolving pathogens. Disabling susceptibility (S) genes is a promising alternative to R genes in breeding programs, as it usually offers durable and broad-spectrum disease resistance. In Arabidopsis, the S gene DMR6 (AtDMR6) encodes an enzyme identified as a susceptibility factor to bacterial and oomycete pathogens. Here, we present a model-to-cro

Enhanced resistance to Cucumber mosaic virus in the Arabidopsis thaliana ssi2 mutant is mediated via an SA-independent mechanism.

The Arabidopsis thaliana SSI2 gene encodes a plastid-localized stearoyl-ACP desaturase. The recessive ssi2 mutant allele confers constitutive accumulation of the pathogenesis-related-1 (PR-1) gene transcript and salicylic acid (SA), and enhanced resistance to bacterial and oomycete pathogens. In addition, the ssi2 mutant is a dwarf and spontaneously develops lesions containing dead cells. Here, we show that the ssi2 mutant also confers enhanced resistance to Cucumber mosaic virus (CMV). Compared with the wild-type plant, viral multiplication and systemic spread were diminished in the ssi2 muta

Use of Basic Substances and Other Natural Compounds as Alternatives to Synthetic Fungicides for the Control of Grapevine Downy and Powdery Mildews.

Consumer demand, retailer involvement, policies to reduce pesticide application, and the growth of organic agriculture are steering research toward safer alternatives like basic substances: compounds used as food, food ingredients, or in medicine with a potential secondary application in plant protection. Downy mildew (DM) and powdery mildew (PM) are major grapevine diseases, and many fungicide applications are required for their management. In this 2-year field trial, chitosan, whey, sweet orange essential oil (EO), Equisetum arvense, Urtica spp., and Ampelomyces quisqualis were tested agains

Identification of the crayfish plague fungus Aphanomyces astaci by polymerase chain reaction and restriction enzyme analysis.

To characterise the DNA of the crayfish plague fungus Aphanomyces astaci, Saprolegniales (Oomycetes), primers were developed to amplify a 1050bp segment of the 28S rDNA region. Restriction enzymes were applied to the amplicon obtained, to distinguish A. astaci from 12 fungal species belonging also to the Saprolegniales and five more distantly related fungi. Most of the fungal species included in the study are either known parasites of freshwater crayfish cuticle or can be found in their natural environment. A. astaci DNA was distinguishable from the DNA of other fungal species tested by using

Different pathotypes of the sunflower downy mildew pathogen Plasmopara halstedii all contain isometric virionsdagger.

Eight pathotypes of Plasmopara halstedii were screened to investigate the occurrence of virions and the potential viral influence on the pathogenicity of the sunflower downy mildew pathogen. In 23 of 26 P. halstedii isolates derived from eight countries in Europe, North America and South America, virions were detected by transmission electron microscopy. By contrast, there were no ultrastructural indications of virus-like particles in eight other related Oomycetes. The virions of representative P. halstedii isolates were morphologically and biochemically characterized and compared among each o

Elucidating the ancestral role of Class I HD-Zip transcription factors in land plants.

Class I homeodomain-leucine zipper (C1HDZ) transcription factors regulate diverse developmental and stress-related processes in land plants. While extensively studied in angiosperms, their functions in other lineages are poorly understood. In the liverwort Marchantia polymorpha, the sole C1HDZ orthologue (MpC1HDZ) controls herbivore defense by regulating oil body cell formation. This suggests that the function of C1HDZ genes may have diverged during the evolution of land plants. Here, we further investigated the function of MpC1HDZ in M. polymorpha and its integration into Arabidopsis (Arabido

Dissecting phosphite-induced priming in Arabidopsis infected with Hyaloperonospora arabidopsidis.

Phosphite (Phi), a phloem-mobile oxyanion of phosphorous acid (H(3)PO(3)), protects plants against diseases caused by oomycetes. Its mode of action is unclear, as evidence indicates both direct antibiotic effects on pathogens as well as inhibition through enhanced plant defense responses, and its target(s) in the plants is unknown. Here, we demonstrate that the biotrophic oomycete Hyaloperonospora arabidopsidis (Hpa) exhibits an unusual biphasic dose-dependent response to Phi after inoculation of Arabidopsis (Arabidopsis thaliana), with characteristics of indirect activity at low doses (10 mm

Spiny-cheek crayfish Orconectes limosus carry a novel genotype of the crayfish plague pathogen Aphanomyces astaci.

The oomycete Aphanomyces astaci causes mass mortalities of European crayfish. Different species of North American crayfish, original hosts of this parasite, seem to carry different strains of A. astaci. So far, four distinct genotype groups have been recognised using Random Amplification of Polymorphic DNA (RAPD-PCR). We succeeded in isolating A. astaci from the spiny-cheek crayfish Orconectes limosus, a widespread invader in Europe, and confirmed that this species carries a novel A. astaci genotype. Improving knowledge on the diversity of this parasite may facilitate identification of genotyp

Genome editing of susceptibility and resistance genes for engineering disease-resistant crops: recent advances, challenges and future prospects.

Plant diseases caused by bacterial, fungal, oomycete, and viral pathogens remain a major constraint to crop productivity worldwide. Recent advances in genome-editing technologies have enabled precise manipulation of genes involved in plant-pathogen interactions, providing new opportunities for developing disease-resistant crops. In particular, CRISPR/Cas-based systems have facilitated functional dissection and engineering of susceptibility genes, resistance genes, and immune regulatory networks across diverse crop species. In this review, we initially discuss emerging genome-engineering platfo

The elicitin β-cryptogein's activity in tomato is mediated by jasmonic acid and ethylene signalling pathways independently of elicitin-sterol interactions.

The level of resistance induced in different tomato genotypes after β-CRY treatment correlated with the upregulation of defence genes, but not sterol binding and involved ethylene and jasmonic acid signalling. Elicitins, a family of small proteins secreted by Phytophthora and Pythium spp., are the most well-known microbe-associated molecular patterns of oomycetes, a lineage of fungus-like organisms that include many economically significant crop pathogens. The responses of tomato plants to elicitin INF1 produced by Phytophthora infestans have been studied extensively. Here, we present studies

Unconventionally secreted effectors of two filamentous pathogens target plant salicylate biosynthesis.

Plant diseases caused by fungi and oomycetes pose an increasing threat to food security and ecosystem health worldwide. These filamentous pathogens, while taxonomically distinct, modulate host defense responses by secreting effectors, which are typically identified based on the presence of signal peptides. Here we show that Phytophthora sojae and Verticillium dahliae secrete isochorismatases (PsIsc1 and VdIsc1, respectively) that are required for full pathogenesis. PsIsc1 and VdIsc1 can suppress salicylate-mediated innate immunity in planta and hydrolyse isochorismate in vitro. A conserved tri

First confirmed case of nasal pythiosis in a horse in Thailand.

INTRODUCTION: Pythiosis is caused by Pythium insidiosum, a fungus-like organism in the class Oomycetes. It can infect humans and a variety of animal species in tropical, subtropical and some temperate regions. Cases of animal pythiosis have occurred predominantly in horses in the skin and subcutaneous tissue at the limbs and in the ventral portion of thoracoabdominal wall - lesions in the nasal region are rarely reported. Moreover, although many human pythiosis cases have been reported in Thailand, no cases of animal pythiosis in Thailand have been reported. CASE PRESENTATION: We report a cas

Antibacterial to antifungal conversion of neamine aminoglycosides through alkyl modification. Strategy for reviving old drugs into agrofungicides.

Many Actinomycetes aminoglycosides are widely used antibiotics. Although mainly antibacterials, a few known aminoglycosides also inhibit yeasts, protozoans and important crop pathogenic fungal oomycetes. Here we show that attachment of a C8 alkyl chain to ring III of a neamine-based aminoglycoside specifically at the 4″-o position yields a broad-spectrum fungicide (FG08) without the antibacterial properties typical for aminoglycosides. Leaf infection assays and greenhouse studies show that FG08 is capable of suppressing wheat fungal infections by Fusarium graminearum-the causative agent of Fus

Efficacy of Organic Fungicides for Downy Mildew in Field-Grown Sweet Basil.

Downy mildew is a common, widespread disease affecting basil leaves. No tolerance for disease symptoms, especially on leaves for fresh consumption, necessitates management. Six replicated experiments were conducted between 2010 and 2016 with field-grown basil of a susceptible cultivar exposed to naturally occurring, wind-dispersed sporangiospores of Peronospora belbahrii to evaluate fungicides approved for use on organically produced crops and products in development. Products tested currently registered for use on basil in the U.S. and labeled for downy mildew were Actinovate (Streptomyces ly

Coumarin biosynthesis genes are required after foliar pathogen infection for the creation of a microbial soil-borne legacy that primes plants for SA-dependent defenses.

Plants deposit photosynthetically-fixed carbon in the rhizosphere, the thin soil layer directly around the root, thereby creating a hospitable environment for microbes. To manage the inhabitants of this nutrient-rich environment, plant roots exude and dynamically adjust microbe-attracting and -repelling compounds to stimulate specific members of the microbiome. Previously, we demonstrated that foliar infection of Arabidopsis thaliana by the biotrophic downy mildew pathogen Hyaloperonospora arabidopsidis (Hpa) leads to a disease-induced modification of the rhizosphere microbiome. Soil condition

Geographical spread of bacterial and fungal diseases of crustaceans.

The author discusses the way in which bacterial and fungal diseases of crustaceans can be and have been transmitted via commercial movements of stocks. Two classic examples of such diseases are presented, namely gaffkaemia (a bacterial disease of marine lobsters) and crayfish plague (a fungal disease of freshwater crayfish). In both cases, there is ample evidence that much of the disease spread can be linked to the commercial movement of infected animals caught from infected wild stocks and transported over large distances for wholesale and retail. In the case of crayfish plague, much of the r

Automated air-flow cytometry enables real-time monitoring of Plasmopara viticola sporangia in vineyards.

UNLABELLED: Fungal diseases such as downy mildew (Plasmopara viticola) can severely affect grapevine yield and quality, making precise fungicide applications critical. Decision Support Systems (DSS) in precision viticulture guide treatment timing, but most rely primarily on weather data and overlook biological indicators, such as real-time spore dynamics. To address this, we evaluated an automatic air-flow cytometer (SwisensPoleno Jupiter) for real-time detection and quantification of P. viticola sporangia. The instrument integrates holographic imaging, fluorescence spectroscopy, and AI-based
