Clinical and Genetic Characterization of a Novel RYR1 Variant (p.Gln474His) in Malignant Hyperthermia Susceptibility.

BACKGROUND/OBJECTIVES: Malignant hyperthermia (MH) is a life-threatening pharmacogenetic disorder of skeletal muscle calcium regulation and commonly associated with pathogenic variants in the RYR1 gene. Interpretation of rare RYR1 variants remains challenging, particularly when classified as variants of uncertain significance (VUS). This study describes the clinical, functional, and genetic evaluation of a patient with suspected MH susceptibility carrying a rare RYR1 mutation. METHODS: We report a retrospective case evaluation of a 32-year-old female referred for MH assessment following a pri

A novel evolutionary method for spine detection in ultrasound samples of spina bifida cases.

BACKGROUND AND OBJECTIVES: Spina bifida is a fetal spine defect observed during pregnancy. The defect is caused by unfinished closure of the embryonic neural column. Common diagnosis of the defect is still based on manual examination which aims to detect any deformation on spinal axis. This study proposes a novel evolutionary method for locating spinal axis on sonograms of spina bifida pathology. METHODS: The method involves a meta-heuristic evolutionary approach, where the sonogram is automatically divided into columns and bone regions belonging to the spine are classified. Accordingly, a sp

The future of beef production in South America.

South American beef production varies due to diverse climates, environmental conditions, animal breeds (Bos indicus, Bos taurus and crossbreeds), management strategies, and nutritional sources. Applying technology in the South American beef production system can significantly enhance efficiency, sustainability, and profitability. Reproductive efficiency is a significant challenge, especially in cow-calf operation systems conducted under adverse conditions. Consequently, implementing effective assisted reproduction technologies (ART) can make a significant contribution. In the last two decades,

Case report: Mesothelioma and BAP1 tumor predisposition syndrome: Implications for public health.

BRCA-1 associated protein 1 (BAP1) tumour predisposition syndrome (TPDS) is a hereditary condition characterised by germline mutation of the tumour suppressor BAP1. This disorder is associated with the development of various benign and malignant tumours, mainly involving the skin, eyes, kidneys, and mesothelium. In this article, we report the case of a man recruited through the Apulia (Southern Italy) Mesothelioma Regional Operational Centre of the National Register of Mesotheliomas, who suffered from uveal melanoma, renal cancer, and mesothelioma, and a familial cluster of BAP1 germline mutat

Laboratory surveillance for wild and vaccine-derived polioviruses--worldwide, January 2006-June 2007.

The Global Polio Laboratory Network (GPLN) was established after announcement of the 1988 World Health Assembly resolution to eradicate poliomyelitis. Operating in all six World Health Organization (WHO) regions, the network currently has 146 laboratories that test stool specimens from acute flaccid paralysis (AFP) patients for polioviruses. The virologic data provided by GPLN underpin the global polio eradication initiative, guiding decisions regarding where targeted immunization activities should be conducted, based on confirmed wild or vaccine-derived poliovirus circulation. The data also a

Biological, prognostic, and therapeutic impact of the epigenome in CLL.

Chronic lymphocytic leukemia (CLL) is characterized by widespread alterations in the genetic and epigenetic landscapes which seem to underlie the variable clinical manifestations observed in patients. Over the last decade, epigenomic studies have described the whole-genome maps of DNA methylation and chromatin features of CLL and normal B cells, identifying distinct epigenetic mechanisms operating in tumoral cells. DNA methylation analyses have identified that the CLL methylome contains imprints of the cell of origin, as well as of the proliferative history of the tumor cells, with both being

A Constitutional Mismatch Repair Deficiency Syndrome Presented With an Advanced Rectal Cancer in a Juvenile Female: A Case Report and Literature Review.

The constitutional mismatch repair deficiency (CMMRD) syndrome is a rare and challenging condition with a poor prognosis. It results from biallelic mismatch repair gene mutations and leads to multiorgan cancers. Therefore, we report the first case of advanced juvenile rectal cancer related to CMMRD syndrome in the Gulf region. She is a 13-year-old female, born to non-consanguineous parents with a positive family history of malignancy, presented with an eight-month history of a retractable bulging anal mass associated with diarrhea mixed with blood and constitutional symptoms. She was cachectic

The clinical, imaging, pathological and genetic landscape of bottom-of-sulcus dysplasia.

Bottom-of-sulcus dysplasia (BOSD) is increasingly recognized as a cause of drug-resistant, surgically-remediable, focal epilepsy, often in seemingly MRI-negative patients. We describe the clinical manifestations, morphological features, localization patterns and genetics of BOSD, with the aims of improving management and understanding pathogenesis. We studied 85 patients with BOSD diagnosed between 2005-2022. Presenting seizure and EEG characteristics, clinical course, genetic findings and treatment response were obtained from medical records. MRI (3 T) and 18F-FDG-PET scans were reviewed syst

[Local recurrence of pheochromocytoma associated with von Hippel-Lindau disease 26 years after bilateral adrenalectomy : a case report].

A 60-year-old man who had undergone left adrenalectomy and right partial adrenalectomy for bilateral pheochromocytoma 26 years ago was found to have an elevated serum noradrenaline (NA) and dopamine (DA) during a long-term follow-up. At the time of right partial adrenalectomy, the normal part of the right adrenal gland was preserved. His cousin and second cousin had pheochromocytoma associated with von Hippel-Lindau (VHL) disease. His eldest daughter had cerebellar hemangioblastoma. Computed tomography and magnetic resonance imaging revealed a tumor which was 17 mm in diameter with contrast en

Genetic Determinants of Tau-PET Imaging in Alzheimer's Disease: A Narrative Review of Genome-Wide and Candidate-Gene Studies.

Alzheimer's disease (AD) is characterized by extracellular β-amyloid (Aβ) plaques and intracellular hyperphosphorylated tau neurofibrillary tangles, with tau pathology correlating more strongly than amyloid pathology with cognitive decline and neuronal loss. While traditional genome-wide association studies (GWAS) have identified loci associated with tau-related phenotypes in cerebrospinal fluid, these approaches cannot capture the regional brain patterns of tau deposition that may underlie disease heterogeneity. Recent GWAS incorporating tau-PET imaging have begun to address this gap, identif

CREB-Binding Protein Regulates Cocaine- and Amphetamine-Regulated Transcript Peptide Expression in the Lateral Hypothalamus: Implication in Reward and Reinforcement.

Neuropeptide cocaine- and amphetamine-regulated transcript peptide (CARTp) is known to play an important role in reward processing. The rats conditioned to intra-cranial self-stimulation (ICSS) showed massive upregulation of CART protein and mRNA in the vicinity of the electrode implanted to deliver the electric current directly at the lateral hypothalamus (LH)-medial forebrain bundle (MFB) area. However, the underlying mechanisms leading to the upregulation of CART in ICSS animals remain elusive. We tested the putative role of CREB-binding protein (CBP), an epigenetic enzyme with intrinsic hi

Genetic parameters and genomic regions associated with horn fly resistance in organic Holstein cattle.

Horn flies (Haematobia irritans [L.]) contribute to major economic losses of pastured cattle operations, particularly in organic herds because of limitations on control methods that can be used. The objectives of this research were to determine if resistance to horn flies is a heritable trait in organic Holstein cattle, determine associations with yield traits, and to detect genomic regions associated with fly infestation. Observations of fly load were recorded from 1,667 pastured Holstein cows, of which 640 were genotyped, on 13 organic dairies across the United States. Fly load score was det

The African Polio Laboratory Network: Advancing Regional Poliovirus Detection, Genomic Surveillance, and Eradication.

BACKGROUND: Sustaining progress toward polio eradication requires not only effective vaccination but also a resilient laboratory network capable of timely poliovirus detection, confirmation, and genetic characterization. In the WHO African region, the African polio laboratory network provides essential virological support for eradication efforts; however, persistent circulating Vaccine-derived Polioviruses (cVDPVs) outbreaks and increasing demands of genomic surveillance continue to challenge laboratory capacity. Despite its critical role, evidence describing the operational performance, adapt

Multiple sclerosis.

Differences in the risk of multiple sclerosis depending on racial background, and the high clinical concordance rates in monozygotic compared with dizygotic twins, have stimulated attempts to identify and locate genes that confer susceptibility to the disease. The risk of multiple sclerosis is increased from 1 in 800 in northern European Caucasians to 1 in 3 in the monozygotic co-twins of affected individuals, with intermediate rates for siblings, offspring and more distant relatives. Concordance rates in monozygotic and dizygotic co-twins of affected individuals rise to 35% and 15%, respectiv

Transcriptional regulation of the genes encoding cytochromes P450BM-1 and P450BM-3 in Bacillus megaterium by the binding of Bm3R1 repressor to Barbie box elements and operator sites.

We previously reported (Liang, Q., He, J.-S., and Fulco, A.J. (1995) J. Biol. Chem. 270, 4438-4450) that Bm3R1, a repressor regulating the expression of P450BM-3 in Bacillus megaterium, could bind to Barbie box sequences in the 5'-flanking regions of barbiturate-inducible genes. We've now shown that pentobarbital does not inhibit in vitro binding of Bm3R1 to the P450BM-3 and P450BM-1 Barbie boxes (BB3 and BB1), although the palindromic operator sequence (OIII) of P450BM-3 did have a strong competitive effect on such binding. G39E-Bm3R1, a mutant of Bm3R1, did not bind to either Barbie box. In

[Genes, cognition, and language: some notes regarding velocardiofacial syndrome].

INTRODUCTION: Identification of different syndromes caused by chromosomic microdeletions and their exhaustive characterization in molecular and clinical terms is decisively contributing to elucidate real connections between genes and cognition. DEVELOPMENT: The velocardiofacial syndrome is a disorder caused by a hemizygous deletion of chromosome 22q11.2, which encompasses about 40 genes. Although the hemizygosis of that region gives rise to different structural and functional alterations in difference zones of the body, those having the greatest relevance are those observed in the central ner
