Identification of reovirus reassortants with enhanced oncolytic activity in human fibrosarcoma cells.

Fibrosarcoma is a rare, clinically aggressive soft-tissue sarcoma that can be difficult to treat once advanced, highlighting the need for new therapeutic strategies. Oncolytic virotherapy is an emerging approach that uses replication-competent viruses to selectively infect and lyse tumor cells while stimulating antitumor immune responses. Mammalian orthoreovirus (reovirus) is an attractive candidate due to its generally benign clinical profile and capacity to replicate in many cancer cells. Reoviruses exhibit substantial genetic diversity arising from mutations and reassortment of their segmen

Pathogenicity and molecular evolutionary analysis of novel Avian orthreovirus genotypes IV and V in Upper Egypt.

Avian orthoreovirus (ARV) imposes substantial economic losses on the global poultry industry, primarily through runting-stunting syndrome and viral arthritis. Despite its global prevalence, the molecular epidemiology and pathogenic landscape of ARV strains circulating in Upper Egypt remain largely unexplored. This study investigated the molecular characteristics, phylogenetic relationships, and pathogenic potential of ARV in broiler flocks across Assiut, Sohag, and New Valley governorates. Out of 140 clinical samples from chickens showing stunted growth and tendon swelling, 42 (30%) were posit

The σNS protein of NDRV antagonizes TRIM59-mediated antiviral innate immune response of Cherry Valley duck.

In recent years, outbreak of the novel duck reovirus (NDRV) disease has occurred frequently in duck populations. Due to its rapid spreading, absence of effective control methods, and high treatment costs, the NDRV disease has caused huge losses to waterfowl breeding in China. As reported, four non-structural (NS) proteins are encoded by the NDRV genome, among which the σNS protein is an RNA-binding protein that can improve the stability of bound RNA by forming oligomers (Adams and Cory, 1998). Nevertheless, the mechanism by which it facilitates reovirus replication remains ambiguous. According

Diverse bat organoids provide pathophysiological models for zoonotic viruses.

Bats are important reservoirs of zoonotic pathogens, but suitable model systems for comprehensively exploring host-pathogen interactions and assessing spillover risks remain limited. To address this gap, we developed a collection of bat organoid models spanning five species and four organ types. This multispecies, multiorgan organoid panel showed species- and tissue-specific replication patterns for several viruses, offering robust pathophysiological models for studying respiratory, renal, and enteric zoonotic viruses. Using this platform, we successfully isolated and characterized bat-borne m

A TaqMan-based real-time PCR assay for specific detection of novel duck reovirus in China.

BACKGROUND: In China, Newly emerging duck reovirus (NDRV) variants have been causing major disease problems in cherry valley ducks. NDRV has the potential to cause high morbidity and 5-50% mortality rates. Severe hemorrhagic-necrosis in the liver and spleen were commonly seen in NDRV affected ducks. The availability of upgraded methods for rapid diagnosis of newly emerging DRV variants is crucial for successful DRV infection control and prevention. RESULTS: In this study, we present a TaqMan-based real-time PCR assay (RT-qPCR) for the detection of NDRV infection. Using the conserved regions w

Development of a live attenuated vaccine against Muscovy duck reovirus infection.

The Muscovy duck reovirus (MDRV) is a highly pathogenic virus that causes substantial economic losses in the Muscovy duck industry. While MDRV poses a significant threat to Muscovy ducklings, no vaccine candidates are available to date to alleviate MDRV infection throughout the world. The present study presents efforts toward establishing an attenuated vaccine for MDRV. For this purpose, a live attenuated vaccine strain named CA was obtained via alternate propagation of the MDRV isolate MW9710 in both Muscovy duck embryo fibroblasts (MDEFs) and chicken embryo fibroblasts (CEFs) for 90 passages

A single-tube dual-channel CRISPR platform for genotyping of waterfowl reoviruses.

Waterfowl reovirus (WRVs) includes two genotypes, Genotype I (Muscovy duck reovirus, MDRV) and Genotype II (novel duck reovirus, NDRV). They have the same clinical manifestations, but different pathogenicity, and can co-infect individual hosts, resulting in diagnostic ambiguity that cannot be solved by single-target detection. Here, we report a single-tube dual-channel platform coupling recombinase polymerase amplification (RPA) with orthogonal CRISPR effectors: LbCas12a targeting the conserved S4 gene of MDRV via a ROX-labeled ssDNA reporter, and LwaCas13a targeting the S1 gene of NDRV via a

Age-based host response to Turkey arthritis reovirus in commercial Turkeys in the presence of maternally derived antibodies.

BACKGROUND: Turkey arthritis reovirus (TARV) causes arthritic lameness in market-age turkeys. Since 2011, highly pathogenic TARV strains have caused significant economic losses in the turkey industry due to increased culling, reduced market weights, and decreased carcass quality, necessitating more effective control measures. Autogenous vaccine prevention strategies have been inefficacious partly due to a limited understanding of age-related susceptibility of turkeys to TARV. This study investigated age-related host and gut microbiota responses to TARV infection in commercial turkeys derived f

Transcriptomic analysis revealed ferroptosis in ducklings with splenic necrosis induced by NDRV infection.

Infection with novel duck reovirus (NDRV) results in severe splenic necrosis, leading to immunosuppression, secondary infections with other pathogens, and impairment of the immune effect of the vaccine. However, little is known about NDRV-induced spleen injury and its antagonistic mechanism on the host immune response. In this study, we conducted pathological and comparative transcriptomic analyses of NDRV-infected duck spleens. Our findings elucidated the histopathological progression of splenic necrotic foci formation following NDRV infection and identified splenic macrophages as the primary

Development of a reverse transcription loop mediated isothermal amplification assay for the detection of Mouse reovirus type 3 in laboratory mice.

Mouse reovirus type 3 (Reo-3) infection is a viral disease that is harmful for laboratory mice. No rapid and accurate detection methods are currently available for this infection. In this study, we describe a rapid, simple, closed-tube, one step, reverse transcription-loop-mediated isothermal amplification (RT-LAMP) assay for Reo-3 and compare our assay with indirect enzyme-linked immunosorbent assay (ELISA). Three sets of RT-LAMP primers were designed by sequence analysis of a specific conserved sequence of the Reo-3 S1 gene. Using RS2 primer set, the RT-LAMP assay required 60 min at 65 °C to

Combination therapy with oncolytic viruses and immune checkpoint inhibitors.

Introduction: Immune checkpoint inhibitors (ICI) have dramatically improved the outcome for cancer patients across multiple tumor types. However the response rates to ICI monotherapy remain relatively low, in part due to some tumors cultivating an inherently 'cold' immune microenvironment. Oncolytic viruses (OV) have the capability to promote a 'hotter' immune microenvironment which can improve the efficacy of ICI.Areas covered: In this article we conducted a literature search through Pubmed/Medline to identify relevant articles in both the pre-clinical and clinical settings for combining OVs

In vitro study on role of σB protein in avian reovirus pathogenesis.

Avian reoviruses, members of Orthoreovirus genus was known to cause diseases like tenosynovitis, runting-stunting syndrome in chickens. Among eight structural proteins, the proteins of S-class are mainly associated with viral arthritis but the significance of σB protein in arthritis is not established till date. In this infection pathological condition together with infection of joints often leads to arthritis because joints consists of cartilage which forms lubricating surface between two bones, and has limited metabolic, replicative and repair capacity. To establish the role of σB protein in

Complete sequence of a reovirus associated with necrotic focus formation in the liver and spleen of Muscovy ducklings.

The complete sequence of a reovirus, strain 815-12 associated with necrotic focus formation in the liver and spleen of Muscovy ducklings in China, was determined and compared with sequences of other duck-, goose-, and chicken-origin reoviruses. The 815-12 genome comprised 22,969 bp with 10 dsRNA segments ranging from 1125 bp (S4) to 3958 bp (L1), all of which (except S4) were almost identical in length to the cognate segments of other waterfowl and chicken isolates. Detailed analyses revealed that 815-12 and other waterfowl isolates contained the conserved 3'-terminal pentanucleotide sequence

Reviewing the risk of feed as a vehicle for swine pathogen transmission.

BACKGROUND: While porcine biological hazards have had the potential to be transmitted through feed and feed mills for decades, the emerging threat of foreign animal disease has elevated the concern that these may enter or be transmitted throughout the domestic swine herd via a feed vehicle. OBJECTIVE: The goal of this review was to describe the current classification for emerging porcine biological pathogen transmission through the feed supply chain so resources can be best directed towards those of highest risk. METHODS: By assessing the pathogen severity to pigs and the probability of path

Immunogenic cross-reactivity between different serotypes novel duck reoviruses: Evaluation of cross-protection provided by mono or bivalent vaccine.

To investigate the immunogenic cross-reactivity among different novel duck reovirus isolates, animals immunized with monovalent inactivated duck reovirus were challenged with homologous and heterologous viruses, and serum samples were collected after immunization for cross-neutralization test. The results showed that the virus neutralizing titer against homologous virus was significantly higher than that against heterologous virus(p < 0.000001), which indicated that there was an important difference in antigenicity between DE13 and WL01 strains, which affected the specificity of protective vir

Anatomopathological and molecular characterization of avian reovirus in broilers condemned due to arthritis in Southern Brazil.

Condemnations in slaughterhouses are a recurring cause of losses for the poultry industry. This study aimed to evaluate the involvement of avian reovirus (ARV) in arthritis-related condemnations in a broiler slaughterhouse under Brazilian Federal Inspection. A total of 150 pelvic limbs condemned for arthritis were collected from the inspection line at the time of condemnation, originating from 30 flocks not vaccinated against ARV. For the detection of ARV, real-time polymerase chain reaction following reverse transcription (RT-qPCR) was performed on a pool of five gastrocnemius tendons from ea

Connecting the Evolution and Spread of Turkey Reovirus Across the United States: A Genomic Perspective.

A major cause of lameness in turkeys is reoviral arthritis, driven by turkey reovirus (TRV) infection. In the U.S., TRV was first isolated in the 1980s but re-emerged as a significant pathogen causing arthritis in 2011. Since then, TRV outbreaks have spread nationwide across turkey-producing regions and have occasionally resulted in hepatitis-associated pathotypes. The absence of a consistently effective commercial vaccine continues to hinder disease control efforts. To better understand TRV's evolutionary trajectory and transmission dynamics, we analyzed 211 complete TRV genome sequences coll
