Recent studies have revealed that the ovary plays a crucial role in defending against pathogen infection in crustaceans, yet the underlying molecular mechanisms remain poorly understood. Our previous work identified numerous genes responsive to poly I:C challenge in the ovary of Macrobrachium nipponense, including several members of the fibrinogen-related protein (FREP) family. Here, we characterized a FREP member from M. nipponense and designated as Mnficolin-1. The Mnficolin-1' cDNA was 1086 bp, encoding 361 amino acids with a C-terminal fibrinogen-related domain (FReD). Mnficolin-1 mRNA was

A selenium dependent glutathione peroxidase (Se-GPx) cDNA was cloned from haemocyte by a reverse-transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA (RACE). The 913 bp cDNA contained an open reading frame (ORF) of 558 bp encoded a deduced amino acid sequence of 186 amino acids. The prawn Se-GPx sequence contains a selenocysteine (Sec) residue which is encoded by the unusual stop codon, (115)TGA(117). According to the molecular modeling analysis, the active site Sec residue, located in the loop between beta3 and alpha2 in a pocket on the protein surface, and hydroge

BACKGROUND: Allergy to specific shrimp species has not been studied systematically by oral challenges. A comparison of allergy to different shrimp species, especially seawater or freshwater varieties treatment, would be useful in testing shrimp-allergic subjects.
OBJECTIVE: The aim of the study was to identify cases of specific allergy to seawater shrimp, Penaeus monodon (Pm), or freshwater shrimp, Macrobrachium rosenbergii (Mr), among shrimp-allergic children. Comparisons of skin tests using commercial and crude shrimp extracts plus the prick-to-prick (PTP) method were investigated.

Diseases have remained the major issue for shrimp aquaculture industry for decades by which different shrimp species demonstrated alternative disease resistance or tolerance. However, there had been insufficient studies on the underlying host mechanisms of such phenomenon. Hence, in this study, the main objective involves gaining a deeper understanding into the functional importance of shrimp STAT gene from the aspects of expression, sequence, structure, and associated genes. STAT gene was selected primarily because of its vital signalling roles in stress, endocrine, and immune response. The d

Rel/nuclear factor (NF)-κB family of transcription factors paly vital roles in innate immunity response to bacterial and viral infection. Here, we cloned and identified a dorsal homologue (named as MnDorsal) from Macrobrachium nipponense. The full-length cDNA of MnDorsal is 2573 bp with a 1986 bp open reading frame that encodes 661 amino acids. Predicted MnDorsal protein contained a RHD (Rel homology domain), an IPT (Iglike, plexins, and transcriptions factors) domain, and two low complexity regions. Phylogenetic analysis showed that MnDorsal has a closer genetic distance with dorsal homologue

Toll-like receptors, which are a class of cell-surface proteins, have been regarded as the most important pattern recognition receptors in the innate immunity and play a vital role in multiple innate immune responses against pathogen invasion. The full-length cDNA of a novel Toll-like receptor (MrToll3) was identified from Macrobrachium rosenbergii in the current research. The nucleotide sequence of MrToll3 is 4481 bp long and contains a 3726-bp open reading frame encoding a putative protein of 1241 amino acids. MrToll3 was constitutively expressed in all the examined tissues, and high express

BACKGROUND: Seawater and freshwater shrimp are some of the most common causes of food allergy among children in Thailand. Tropomyosin has been reported as a major allergen for shrimp allergic populations around the world. Despite a high number of shrimp-allergic Thai children, however, it is unknown whether shrimp tropomyosin is a major cause of allergic reactions.
OBJECTIVES: To clone and characterize tropomyosin of giant freshwater shrimp Macrobrachium rosenbergii (Mr) and determine whether this tropomyosin is a major cross-reactive allergen for Thai children with shrimp allergy.

Phosphorylated sperm proteins are crucial for sperm maturation and capacitation as a priori to their fertilization with eggs. In the freshwater prawn, Macrobrachium rosenbergii, a male reproduction-related protein (Mar-Mrr) was known to be expressed only in the spermatic ducts as a protein with putative phosphorylation and may be involved in sperm capacitation in this species. We investigated further the temporal and spatial expression of the Mar-Mrr gene using RT-PCR and in situ hybridization and the characteristics and fate of the protein using immunblotting and immunocytochemistry. The Mar-

By voltage-clamp technique and simultaneous [Ca2+](i)measurements, we studied the modifications, induced by changes in membrane voltage, in the pattern of the [Ca2+](i)oscillation period, displayed by the Mg2+-stimulated oocyte of the prawn Palaemon serratus. When the Mg2+-stimulated oocytes were voltage clamped at 0mV, they developed a [Ca2+](i)signal with a more pronounced oscillatory pattern than that obtained on unclamped oocytes. Indeed, they displayed a first peak followed by a series of sharp [Ca2+](i)transients and a prominent [Ca2+](i)oscillatory plateau. By contrast, oocytes voltage

Protein kinase C (PKC), which is involved in cell signaling pathways, comprises a family of serine/threonine kinases ubiquitously present in animals and its members are grouped on the basis of structural and activation characteristics into novel, classical, and atypical PKC forms. In this study, an atypical PKC of Macrobrachium rosenbergii, designated MraPKC, was successfully cloned, and its protein comprised structural domains similar to those of atypical PKC homologues, including the Phox and Bem1 (PB1) domain, a zinc finger phorbol-ester/DAG-type signature, protein kinase signatures, and a

H9N2 and H5N2 avian influenza A viruses (IAVs) are two of the most widespread and economically damaging subtypes affecting the poultry industry. Controlling these viruses in poultry remains challenging, highlighting the need for a vaccine that can confer broad and effective protection across multiple avian IAV subtypes. Previously, we fused two copies of the matrix 2 ectodomain (M2e) of human and avian IAV (2xhuM2e-2xavM2e) at the C-terminus of Macrobrachium rosenbergii nodavirus capsid protein (NvC). This chimeric protein (NvC-2xhuM2e-2xavM2e) self-assembled into virus-like particles (VLPs),

X-box binding protein 1 (XBP1) and activating transcription factor 4 (ATF4) are transcription factors involved in the unfolded protein response (UPR) signaling pathway and have been reported to play important roles during viral infection in shrimp. To further investigate their functions during white spot syndrome virus (WSSV) infection in the prawn Macrobrachium nipponense, we cloned and characterized the full-length cDNA sequences of MnXBP1 and MnATF4. The MnXBP1 protein contains a basic leucine zipper (bZIP) domain and a low-complexity region, while MnATF4 possesses two internal repeats, a b

Macrophages are one of the important immune cells, which play important roles in innate and adaptive immune. However, the roles of macrophages in food allergy are not thoroughly understood. To investigate the roles of macrophages during food allergy, we focused on the relationship between macrophage polarization and allergic responses induced by tropomyosin (TM) in the present study. Arg 1 and CD206 expressions in the TM group were significantly higher than those of the PBS group, while iNOS and TNF-α expressions were no obvious difference, moreover, the morphology of macrophages stimulated by

We previously analyzed the central nervous system (CNS) transcriptome and found three isotypes of long neuropeptide F (MrNPF-I, -II, -III) and four isoforms of short NPF (sMrNPF) in the giant freshwater prawn, Macrobrachium rosenbergii. We now validate the complete sequences of the MrNPF-I and -II precursor proteins, which show high similarity (91-95 %) to NPFs of the penaeus shrimp (PsNPF). MrNPF-I and -II precursors share 71 % amino acid identity, whereas the mature 32-amino-acid MrNPF-I and 69-amino-acid MrNPF-II are identical, except for a 37-amino-acid insert within the middle part of the

Oncoprotein NM23, as a family of genes encoding the nucleoside diphosphate (NDP) kinase, plays important roles in bioenergetics, DNA replication, differentiation and tumor metastasis. In this study, a full-length cDNA of NM23 (designated EcNM23) was cloned from Exopalaemon carinicauda by using rapid amplification of cDNA ends (RACE) approaches. The full-length cDNA of EcNM23 was 755 bp, which contains an open reading frame (ORF) of 518 bp, encoding a 175 amino-acid polypeptide with the predicted molecular weight of 19.60 kDa and estimated isoelectric point of 7.67. The deduced amino acid seque
