IgY passive immunotherapy for pandemic preparedness: a One Health platform approach against pathogen X.

SUMMARYThe rising threat of emerging infectious diseases, especially zoonotic pathogens crossing species barriers, highlights the urgent global need for scalable, rapid-response passive immunotherapy platforms. Chicken egg yolk-derived immunoglobulin Y (IgY) antibodies offer unique conceptual and practical advantages. This review critically evaluates IgY antibodies (IgY-Abs) as a One Health immunotherapeutic strategy with strong potential for mitigating zoonotic spillover risks. Drawing on insights from SARS-CoV-2, we highlight the advantages of IgY-Abs over traditional approaches in specific

Strengthening rural pandemic preparedness: Lessons from Nova Scotia's COVID-19 response for pharmacy practice.

The COVID-19 pandemic exposed longstanding structural inequities in rural health care systems, including shortages of primary care providers, transportation barriers, geographic isolation, workforce limitations, and unequal access to digital health technologies. Rather than creating these challenges, the pandemic magnified existing weaknesses that limited equitable access to testing, vaccination, chronic disease management, and preventive health care. Nova Scotia provides an informative Public Health Mobile Units (PHMU) implementation of how innovative, community-based models of care can mitig

A Comprehensive Review of Vaccine Technologies for Pandemic Preparedness Against Highly Pathogenic Avian Influenza.

Highly Pathogenic Avian Influenza Virus (HPAIV) poses a serious threat to the poultry industry and public health due to its global spread to avian and non-avian hosts. To combat the spread of HPAIVs in poultry and reduce the risk of zoonotic transmission, the World Health Organization underscores the importance of strategic surveillance and advancements in vaccination strategies. For the prevention of avian influenza outbreaks in poultry and their transmission to humans and cattle, vaccination would be a critical tool. We critically review the limitations of current vaccine platforms and highl

Organising knowledge to prevent global health crises: a comparative analysis of pandemic preparedness indicators.

Preparing for the possibility of a global pandemic presents a transnational organisational challenge: to assemble and coordinate knowledge over institutionally diverse countries with high fidelity. The COVID-19 pandemic has laid these problems bare. This article examines the construction of the three main cross-national indicators of pandemic preparedness: a database with self-reported data by governments, external evaluations organised by the WHO and a global ranking known as the Global Health Security Index. Each of these presents a different model of collecting evidence and organising knowl

Development and Evaluation of Vero Cell-Derived Master Donor Viruses for Influenza Pandemic Preparedness.

The embryonated egg-based platform currently produces the majority of seasonal influenza vaccines by employing a well-developed master donor virus (MDV, A/PR/8/34 (PR8)) to generate high-growth reassortants (HGRs) for A/H1N1 and A/H3N2 subtypes. Although the egg-based platform can supply enough seasonal influenza vaccines, it cannot meet surging demands during influenza pandemics. Therefore, multi-purpose platforms are desirable for pandemic preparedness. The Vero cell-based production platform is widely used for human vaccines and could be a potential multi-purpose platform for pandemic influ

Nontraditional Data in Pandemic Preparedness and Response: Identifying and Addressing First- and Last-Mile Challenges.

The COVID-19 pandemic served as an important test case of complementing traditional public health data with nontraditional data, such as mobility traces, social media activity, and wearable data, to inform real-time decision-making. Drawing on an expert workshop and a targeted survey of epidemic modelers in Europe, this study assesses the promise and the persistent limitations of such data in pandemic preparedness and response. We distinguish between "first-mile" challenges (obstacles to accessing and harmonizing data) and "last-mile" challenges (difficulties in translating insights into actio

Community behavioural resilience and STIs: Predictors of pandemic preparedness in a low-resource setting in Cameroon.

BACKGROUND: Sexually transmitted infections (STIs) infect 374 million people annually, with sub-Saharan Africa bearing the highest burden. Community behavioural resilience remains poorly understood in low-income settings, creating a critical gap in STI pandemic preparedness. This study identifies predictors of willingness to adopt protective behaviours during a hypothetical STI outbreak in rural Cameroon. METHODS: We conducted a community-based survey in 2022 among residents of three riverine health areas in the Bafia Health District, Cameroon. Participants (N=372; 202 females [54.3%], 170 ma

From pandemics to preparedness: harnessing AI, CRISPR, and synthetic biology to counter biosecurity threats.

Biosecurity threats, which include natural outbreaks, laboratory accidents, and intentional bioterrorism, are a major issue for global health security. The impact of poor preparedness on the health, social, and economic effects of the 1918 influenza pandemic, the 2001 anthrax attacks, and the COVID-19 crisis is devastating. Standard methods, such as quarantine and serology, as well as traditional inoculations, offered basic defences but were often reactive, slow, and unfair. The recent scientific and technological progress has altered the concept of biosecurity preparedness by providing new in

A method for evaluating the cost-benefit of different preparedness planning policies against pandemic influenza.

•Our work presents a unifying method to calculate the net-benefit of different preparedness policies against different pandemic influeunza strains. Unlike previous methods, which have focused on evaluating specific strategies against specific pandemics, our method allows assessment of mass immunisation strategies in presence and absence of antiviral drugs for a large range of pandemic influenza strain characteristics and programme features. Overall, the model described here combines two parts to evaluate different preparedness planning policies against pandemic influenza.•The first part is ada

Preparedness as a technology of (in)security: Pandemic influenza planning and the global biopolitics of emerging infectious disease.

This article takes as its starting point the idea that re-emerging infectious disease has become a paradigmatic way of thinking about disease. The framing of infectious disease as a threat to global public health and economic security coincides with preemptive forms of control. A particular type of preemptive regulation is global pandemic influenza planning that entails the governing of an imminent, albeit uncertain, global health event. We examine the discourse of 'preparedness' within pandemic planning documents produced by the World Health Organization from 1999 to 2009. We present key find

Editorial: The 2025 World Health Assembly Pandemic Agreement and the 2024 Amendments to the International Health Regulations Combine for Pandemic Preparedness and Response.

The importance of pandemic preparedness is underscored by two recent and significant findings in the US, including outbreaks of measles in children and adults, as well as the demonstration of airborne transmission of the influenza A(H5N1) virus (bird flu). On June 1, 2024, the 77th World Health Assembly of the World Health Organization (WHO) reached a consensus on amendments to the 2005 International Health Regulations, representing a new universal legal framework for global health, pandemic preparedness, and response that will enter into force in September 2025. On May 20, 2025, the 78th Worl

The Nipah virus threat in India: Epidemiological trends, risk drivers, and lessons for pandemic preparedness.

Nipah virus (NiV), a WHO-priority zoonotic pathogen, poses a recurrent public health threat in India. Since its initial detection in 2001, India has encountered multiple NiV spillover events, including repeated outbreaks in Kerala and recent cases reported in West Bengal in 2026. Although the pig-amplified epidemic in Malaysia (1998-1999) and the seasonal outbreaks associated with NiV-contaminated date palm sap consumption in Bangladesh are extensively documented, India represents a unique and relatively understudied epidemiological setting. NiV outbreaks in India are primarily linked to the N

Biotechnological Preparedness for Novel Pandemics: Diagnostic Performance of IVDS Against SARS-CoV-2.

Although the COVID-19 pandemic has created many challenges and negative impacts around the world, some of which will persist into the future, its technological challenge has created a unique opportunity in a globalized world. It is a rare event that almost all of humanity to be directed towards a single goal and to try to produce solutions, but the necessity of a similar global action in the future has begun to enter the agenda again. The predictions made on the basis of countries and institutions against the possibility of a pandemic, which is defined as Disease X, are shaped by the experienc

Democratising Artificial Intelligence for Pandemic Preparedness and Global Governance in Latin American and Caribbean Countries.

Infectious diseases continue to pose a significant global health challenge, necessitating innovative approaches for predicting outbreaks, detecting variants, conducting contact tracing, discovering new drugs and managing misinformation. Artificial intelligence (AI) has significantly supported work in these areas, particularly during the COVID-19 pandemic. However, the benefits of AI must be equitably distributed, and its use must be responsible and inclusive. As various nations implement AI regulations, the global nature of AI necessitates international collaboration to establish ethical guide

Emergency Preparedness and Management of Mobile Cabin Hospitals in China During the COVID-19 Pandemic.

The healthcare systems in China and globally have faced serious challenges during the coronavirus disease (COVID-19) pandemic. The shortage of beds in traditional hospitals has exacerbated the threat of COVID-19. To increase the number of available beds, China implemented a special public health measure of opening mobile cabin hospitals. Mobile cabin hospitals, also called Fangcang shelter hospitals, refer to large-scale public venues such as indoor stadiums and exhibition centers converted to temporary hospitals. This study is a mini review of the practice of mobile cabin hospitals in China.

Lessons learned from the COVID-19 pandemic: opportunities and challenges to leverage investments in HIV, tuberculosis and malaria for pandemic preparedness and response.

INTRODUCTION: The COVID-19 pandemic strained public health infrastructure in low-income and middle-income countries (LMICs), leading them to leverage resources of existing public health programmes including HIV, tuberculosis (TB) and malaria. This manuscript systematically documents how these programmes were used in response to understand both the barriers and facilitators encountered in the process. METHODS: From March through August 2023, we conducted 72 structured interviews with staff in 13 countries who played central roles in the response. Countries were purposively selected from the po
