Paraoxonase1 192 (PON1 192) gene polymorphism and serum paraoxonase activity in panic disorder patients.

BACKGROUND/AIM: Reactive oxygen species (ROS) are involved in the development of certain neuropsychiatric disorders. Paraoxonase 1 (PON1) activity has been suggested to be adversely related to oxidative stress in plasma. The purpose of the present study was to demonstrate the relationship between serum PON1 activity and PON1 192 polymorphism in panic disorder (PD). MATERIALS AND METHODS: Fourty-two patients with PD and 46 healthy controls were included in this study. PON1 192 genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis. P

Novel selective and irreversible mosquito acetylcholinesterase inhibitors for controlling malaria and other mosquito-borne diseases.

We reported previously that insect acetylcholinesterases (AChEs) could be selectively and irreversibly inhibited by methanethiosulfonates presumably through conjugation to an insect-specific cysteine in these enzymes. However, no direct proof for the conjugation has been published to date, and doubts remain about whether such cysteine-targeting inhibitors have desirable kinetic properties for insecticide use. Here we report mass spectrometric proof of the conjugation and new chemicals that irreversibly inhibited African malaria mosquito AChE with bimolecular inhibition rate constants (k(inact)

Lack of inducibility of brain monooxygenase activities including parathion desulfuration.

The ability of phenobarbital and beta-naphthoflavone to induce parathion desulfuration, aminopyrine N-demethylation, and NADPH-cytochrome-c reductase activity in the brain and liver of male and female rats was investigated. Activities of all three enzymes were found in similar levels in both the mitochondrial and microsomal fractions of brain. There were no sex differences in brain activities. Liver activities were from 10- to 30-fold higher than brain activities when computed on a tissue-wet-weight-equivalent basis. Although exposure to both inducers increased all three enzyme activities and

A Nanozyme- and Ambient Light-Based Smartphone Platform for Simultaneous Detection of Dual Biomarkers from Exposure to Organophosphorus Pesticides.

A transparent, lateral-flow test strip coupled with a smartphone-based ambient light sensor was first proposed for detecting enzymatic inhibition and phosphorylation. The principle of the platform is based on the simultaneous measurement of the total amount of the enzyme and enzyme activity to biomonitor exposure to organophosphorus (OP) pesticides. In this study, butyrylcholinesterase (BChE) was adopted as the model enzyme and ethyl paraoxon was chosen as an analyte representing OP pesticides. The total amount of BChE was quantified by a sensitive colorimetric signal originating from a sandwi

Inhibition of human fetal brain acetylcholinesterase: marker effect of neurotoxicity.

Human fetal brains were obtained after medical termination of pregnancy at 8-10 wk from informed patients. A definite regionalization of AChE was found in the brain of the fetus, with cerebellum recording the highest and cerebral hemisphere the lowest activity. Optimum conditions for the cerebellar AChE activity were determined with respect to molarity of the buffer, pH, temperature, and concentrations of substrate (acetylthiocholine iodide), activators (NaCl, MgCl2), and thiol indicator (dithiobisnitrobenzoic acid). In vitro inhibition of cerebellar AChE with two commercial pesticides, Metaci

Acute and subacute organophosphate poisoning in the rat.

The intermediate syndrome in organophosphate poisoning is clinically characterized by weakness in the territory of cranial nerves, weakness of respiratory, neck and proximal limb muscles, and depressed deep tendon reflexes. It occurs between the acute cholinergic crisis and the usual onset of organophosphate-induced delayed neurotoxicity. The weakness has been ascribed to muscle fiber necrosis. Fenthion has been the most common cause. This study assesses the occurrence of the necrotizing myopathy in rats in relation to the clinical course and the acetylcholinesterase (AChE) inhibition after po

[The use of direct and indirect parasympathomimetics in poisonings with tricyclic antidepressive agents].

The use of cholinesterase inhibitors in poisonings with potentially lethal doses of tricyclic antidepressive drugs seems to be indicated on account of clinical experiences and own experimental investigations on animals. An uncritical use of these substances which have considerable side-effects must be warned of. A mixed intoxication with several drugs effecting on the central nervous system is only a relative indication for the use of indirect parasympathicomimetics of the cholinesterase type. They should be used only then, when the picture of poisoning is unequivocally determined by the thymo

Programmable Semi-Interpenetrating Living Materials With Robust Stability for Versatile Bioremediation and Biotherapeutics.

The practical application of engineered living materials (ELMs) is currently hindered by some critical challenges, such as streamlining fabrication processes and achieving long-term stability. Here, a semi-interpenetrating ELM was developed relying on thermosensitive self-assembly of hydroxybutyl chitosan (HBC) and spontaneous covalent protein interactions. This semi-interpenetrating network provided superior mechanical properties over HBC hydrogels. Furthermore, this material can be adapted for diverse scenarios based on engineered bacteria encapsulated, and its applications in biotherapy tre

Model acetylcholinesterase-Fc fusion glycoprotein biotechnology system for the manufacture of an organophosphorus toxicant bioscavenging countermeasure.

Organophosphate (OP) toxicants remain an active threat to public health and to warfighters in the military. Current countermeasures require near immediate administration following OP exposure and are reported to have controversial efficacies. Acetylcholinesterase (AChE) fused to the human immunoglobulin 1 (IgG1) Fc domain (AChE-Fc) is a potential bioscavenger for OP toxicants, but a reproducible AChE-Fc biomanufacturing strategy remains elusive. This report is the first to establish a comprehensive laboratory-scale bioprocessing strategy that can reproducibly produce AChE-Fc and AChE(W86A)-Fc

Molecular Modeling and Structural Analysis of Arylesterase of Ancylostoma Duodenale.

Parasitic worm infection of humans is one of the most commonly prevalent helminth infection that has imposed great impact on society and public health in the developing world. The two species of hookworm, namely Ancylostoma duodenale and Necator americanus may be primarily responsible for causing parasitic infections in human beings. The highly prevalent areas for Ancylostoma duodenale infections are mainly India, Middle East, Australia, northern Africa and other parts of the world. The serum arylesterases/paraoxonases are family of enzymes that is involved in the hydrolysis of a number of org

Cholinesterases, a target of pharmacology and toxicology.

BACKGROUND: Cholinesterases are a group of serine hydrolases that split the neurotransmitter acetylcholine (ACh) and terminate its action. Of the two types, butyrylcholinesterase and acetylcholinesterase (AChE), AChE plays the key role in ending cholinergic neurotransmission. Cholinesterase inhibitors are substances, either natural or man-made that interfere with the break-down of ACh and prolong its action. Hence their relevance to toxicology and pharmacology. METHODS AND RESULTS: The present review summarizes current knowledge of the cholinesterases and their inhibition. Particular attentio

Calcium-dependent human serum homocysteine thiolactone hydrolase. A protective mechanism against protein N-homocysteinylation.

Homocysteine thiolactone is formed in all cell types studied thus far as a result of editing reactions of some aminoacyl-tRNA synthetases. Because inadvertent reactions of thiolactone with proteins are potentially harmful, the ability to detoxify homocysteine thiolactone is essential for biological integrity. This work shows that a single specific enzyme, present in mammalian but not in avian sera, hydrolyzes thiolactone to homocysteine. Human serum thiolactonase, a 45-kDa protein component of high density lipoprotein, requires calcium for activity and stability and is inhibited by isoleucine

Exploring the mechanism of metabolic cell death-related genes AKR1C2 and MAP1LC3A as biomarkers in Parkinson's disease.

There is a strong relationship between metabolic cell death (MCD) and neurodegenerative diseases. However, the involvement of metabolic cell death (MCD)-related genes (MCDRGs) in Parkinson's disease (PD) pathogenesis remains poorly analyzed. Integrating PD-associated differentially expressed genes (DEGs) from GSE7621 with MCDRGs, we identified key biomarkers through protein-protein interaction networks and machine learning. Diagnostic performance was validated through nomogram analysis. Subsequent analyses included functional enrichment, immune profiling, drug prediction, and single-cell RNA s

Paroxysmal Slow Waves Mark Ictal Networks.

Epilepsy diagnosis and treatment monitoring are hindered by the episodic, heterogeneous expression of seizures and by normal-appearing scalp EEG in many patients. We previously described paroxysmal slow-wave events (PSWEs)-brief epochs of broadband slowing detectable on EEG. Here, using intracerebral and epidural recordings in a paraoxon rat model of temporal lobe epilepsy, we show that PSWEs arise preferentially in temporo-frontal networks, co-occur with global slowing, and increase during both spontaneous and pharmacologically induced seizures. Epidurally recorded PSWEs were temporally coupl

Effects of antiretroviral treatment on paraoxonase 1 (PON1) activity in rats.

Highly active antiretroviral therapy (HAART), especially protease inhibitors (PIs), commonly used in HIV-infected patients, effectively suppresses a viral replication. However, it is frequently associated with significant side effects, including fat redistribution, lipodystrophy, hyperlipidemia, insulin resistance and diabetes mellitus. Currently, metabolic complications and atherosclerosis resulting from them become the major cause of mortality in HIV-infected patients receiving HAART. Paraoxonase 1 (PON1) is the HDL-bound esterase, which inhibits development of atherosclerosis by decomposing

Effect of metal ions and calcium on purified PON1 and PON3 from rat liver.

The effect of several metal ions and calcium on purified paraoxonases (PON1 and PON3) from rat liver was studied. PON1 and PON3 were also inhibited by EDTA and both enzyme activities were restored by the addition of free calcium. The reactivation by calcium was a time-dependent effect for PON1; however, this was not the case for PON3. We also studied the response of PON1 and PON3 to several inhibitors: Co, Cu, Mn, Hg and p-hydroxymercurybenzoate (pOHMB), and determined the type of inhibition and the inhibition constants. Among all the compounds tested, mercurials (Hg and pOHMB) were the most p

Electrospray deposition as a smart technique for laccase immobilisation on carbon black-nanomodified screen-printed electrodes.

Enzymes immobilisation represents a critical issue in the design of biosensors to achieve standardization as well as suitable analytical performances in terms of sensitivity, selectivity, and stability. In this work electrospray deposition (ESD) has been exploited as a novel technique for the immobilisation of laccase enzyme on carbon black modified screen-printed electrodes. The aim is to fabricate an amperometric biosensor for phenolic compound detection. The electrodes produced by ESD have been analysed by scanning electron microscopy and characterised electrochemically to prove that this i

Surface assembly of nano-metal organic framework on amine functionalized indium tin oxide substrate for impedimetric sensing of parathion.

The present paper reports the assembly and pesticide sensing application of a nanometal organic framework [Cd(atc)(H2O)2]n ('atc'=2-aminoterephthalic acid). The assembly of the NMOF film has been achieved by sequential dipping of a 2-aminobenzylamine (2-ABA) modified indium tin oxide (ITO) slide in organic linker 'atc' and metal ion 'Cd(2+)' solutions. The different structural and morphological characteristics of the NMOF thin film have been characterized. The availability of pendent -COOH functional groups on the assembled NMOF film is exploited to synthesize a pesticide immunosensor by conju

Assessment of developmental delay in the zebrafish embryo teratogenicity assay.

In this study we analyzed some aspects of the assessment of developmental delay in the zebrafish embryotoxicity/teratogenicity test and explored the suitability of acetylcholinesterase (AChE) activity as a biochemical marker and as a higher throughput alternative to morphological endpoints such as head-trunk angle, tail length and morphological score. Embryos were exposed from 4 to 52 h post-fertilization (hpf) to a selection of known embryotoxic/teratogen compounds (valproic acid, retinoic acid, caffeine, sodium salicylate, glucose, hydroxyurea, methoxyacetic acid, boric acid and paraoxon-met
