Temporal Parasitemia Trends Predict Risk and Timing of Experimental Cerebral Malaria in Mice Infected by Plasmodium berghei ANKA.

BACKGROUND: Experimental models using Plasmodium berghei ANKA (PbA)-infected mice have been essential for uncovering cerebral malaria (CM) pathogenesis. However, variability in experimental CM (ECM) incidence, onset, and mortality introduce challenges when analyses rely solely on infection day, which may reflect different disease stages among animals. METHODS: We applied machine learning to predict ECM risk and onset in a cohort of 153 C57BL/6, 164 CBA, and 53 Swiss Webster mice. First, we fitted a logistic regression model to estimate the risk of ECM at any day using parasitemia data from da

Recurrent Plasmodium falciparum parasitemia and drug resistance mutations during intermittent preventive treatment of malaria in pregnancy in Uganda.

BACKGROUND: Intermittent preventive treatment with monthly sulfadoxine-pyrimethamine (IPTp-SP) is recommended during pregnancy in malaria-endemic countries. However, widespread resistance of Plasmodium falciparum to SP has compromised its efficacy, and the alternative dihydroartemisinin-piperaquine (DP) is under study. Potential selection of drug resistance is of interest. METHODS: We sequenced 1377 samples collected from pregnant women enrolled in a trial comparing monthly SP, DP, and DP+SP for IPTp in Busia, Uganda and with asymptomatic parasitemia at the time of IPTp administration. We cha

E3 ubiquitin ligase RNF123-deficient mice exhibit reduced parasitemia and mortality in rodent malaria (Plasmodium yoelii 17XL) infection.

Increased levels of several human ubiquitin ligases, including ring finger protein 123 (RNF123), in red blood cells with Plasmodium falciparum infection, have been reported. RNF123 is an E3 ubiquitin ligase that is highly expressed in erythroid cells. However, the function of the RNF123 gene and the relationship between the RNF123 gene and malarial parasite has not been clarified in vivo. In this study, we generated RNF123-deficient mice using the CRISPR/Cas9 system, and analyzed malaria susceptibility and erythrocyte morphology. The levels of parasitemia 5 days post-infection and mortality 21

Annual Versus Biannual Mass Azithromycin Distribution and Malaria Parasitemia During the Peak Transmission Season Among Children in Niger.

BACKGROUND: Azithromycin has modest efficacy against malaria, and previous cluster randomized trials have suggested that mass azithromycin distribution for trachoma control may play a role in malaria control. We evaluated the effect of annual versus biannual mass azithromycin distribution over a 3-year period on malaria prevalence during the peak transmission season in a region with seasonal malaria transmission in Niger. METHODS: Twenty-four communities in Matameye, Niger, were randomized to annual mass azithromycin distribution (3 distributions to the entire community during the peak transm

Trypanosoma acomys (Wenyon, 1909): reproductive forms and course of parasitemia in the natural host Acomys cahirinus (Desmarest, 1819).

Trypanosoma acomys (Stercoraria, Trypanosomatidae) was studied in the laboratory-bred spiny mouse Acomys cahirinus. Only trypomastigotes were found in peripheral blood, whereas the thymus of experimentally infected A. cahirinus showed amastigote syncytial reproductive forms whose nuclei of nuclei ranged from 2 to 128. These reproductive forms were detected 9-10 days after the i.p. injection of spiny mice with trypanosomes obtained from primary lung cultures. Transitional trypomastigote-like forms were observed in the thymus; no reproductive or diving forms were encountered in any other tissue,

Neutralizing Antibody Responses After mRNA COVID-19 Booster Vaccination are Unaffected by Parasitemia in a Malaria-Endemic Setting.

Subclinical malaria may reduce the immunogenicity of mRNA vaccines. We evaluated neutralizing antibody responses in adults with (n=87) and without (n=221) PCR-confirmed Plasmodium falciparum who received a COVID-19 booster. Similar boosted ID50 geometric mean titers >22,000 in parasitemic and non-parasitemic participants suggests that COVID-19 mRNA vaccine responses are not impaired.

Effect of moringa extract on parasitemia, monocyte activation and organomegaly among Mus musculus infected by Plasmodium berghei ANKA.

In Indonesia, malaria remains a problem, with 94,610 active cases in 2021 and its current therapy includes chloroquine and artemisinin; however, resistance has been commonly reported. To overcome this problem, studies about potential medicinal plants that can be used as antimalaria, such as moringa (Moringa oleifera) started to receive more attention. The aim of this study was to investigate the effects of moringa in parasitemia, monocyte activation, and organomegaly on animal model malaria. This experimental study used male Mus musculus, infected by Plasmodium berghei ANKA, as an animal malar

Persistence of Residual Submicroscopic P. falciparum Parasitemia following Treatment of Artemether-Lumefantrine in Ethio-Sudan Border, Western Ethiopia.

The emergence of artemisinin-resistant parasites in Africa has had a devastating impact, causing most malaria cases and related deaths reported on the continent. In Ethiopia, artemether-lumefantrine (AL) is the first-line drug for the treatment of uncomplicated falciparum malaria. This study is one of the earliest evaluations of artemether-lumefantrine (AL) efficacy in western Ethiopia, 17 years after the introduction of this drug in the study area. This study aimed at assessing PCR- corrected clinical and parasitological responses at 28 days following AL treatment. Sixty uncomplicated falcipa

Label-free imaging and classification of live P. falciparum enables high performance parasitemia quantification without fixation or staining.

Manual microscopic inspection of fixed and stained blood smears has remained the gold standard for Plasmodium parasitemia analysis for over a century. Unfortunately, smear preparation consumes time and reagents, while manual microscopy is skill-dependent and labor-intensive. Here, we demonstrate that deep learning enables both life stage classification and accurate parasitemia quantification of ordinary brightfield microscopy images of live, unstained red blood cells. We tested our method using both a standard light microscope equipped with visible and near-ultraviolet (UV) illumination, and a

Evaluation of antioxidant status, oxidative stress and serum trace mineral levels associated with Babesia ovis parasitemia in sheep.

Ovine babesiosis is a fatal disease characterized by severe progressive hemolytic anemia. In order to clarify the causal mechanisms implicated in anemia, this study was aimed to assess the antioxidant status and erythrocyte oxidative stress in sheep suffering from ovine babesiosis. Babesia infection was confirmed both with Giemsa's staining blood smears and semi-nested PCR amplified region of 18S rRNA gene. Thirty-eight Iranian sheep, naturally infected with Babesia spp., were considered as the infected group and divided into four subgroups according to parasitemia rates (<1%, 1-2%, 2-3% and >

The parasitemia of cloned Trypanoplasma borreli Laveran and Mesnil, 1901, in laboratory-infected common carp (Cyprinus carpio L.).

The course of parasitemia of cloned Trypanoplasma borreli in laboratory-infected common carp was investigated. In 25-42-g carp kept at 20 C, the prepatent period was 8 days; after a phase of exponential growth, the parasitemia peaked at day 39 postinjection (PI) at a level of about 10(3) T. borreli/microliters blood. This maximum was followed by a chronic phase of about 6 wk with large numbers of T. borreli. At 20 wk PI, T. borreli was absent in infected carp. In 2.2-g carp kept at 20 C, the prepatent period was 4 days only, and the parasitemia peaked at day 23 PI. At 30 C, T. borreli was pres

Prevalence and predictors of asymptomatic malaria parasitemia among pregnant women in the rural surroundings of Arbaminch Town, South Ethiopia.

BACKGROUND: In Sub-Saharan African countries, including Ethiopia, malaria in pregnancy is a major public health threat which results in significant morbidities and mortalities among pregnant women and their fetuses. In malaria endemic areas, Plasmodium infections tend to remain asymptomatic yet causing significant problems like maternal anemia, low birth weight, premature births, and still birth. This study was conducted to determine the prevalence and predictors of asymptomatic Plasmodium infection among pregnant women in the rural surroundings of Arba Minch Town, Southern Ethiopia. METHODS:

Reticulocytes in peripheral blood of mice infected with Trypanosoma brucei: dramatic decrease of reticulocytes during primary wave of parasitemia.

Infection of mice with rapidly proliferating trypanosomes that killed the mice in the first or second wave of parasitemia led to a dramatic decrease of immature erythrocytes in the peripheral blood during the primary wave of parasitemia. Trypanosome variants producing lower first waves of parasitemia, and/or less rapid deaths, caused a variable, on the average small, decrease of immature erythrocytes. Trypanosome variants causing rapidly lethal infections killed the mice before a decrease of immature erythrocytes became apparent.

Development and implementation of droplet digital PCR assays for accurate quantification of Plasmodium vivax parasitemia and G6PD viangchan genotyping.

Accurate quantification of Plasmodium vivax parasitemia and identification of glucose-6-phosphate dehydrogenase (G6PD) deficiency, particularly the Viangchan variant, the most prevalent in Southeast Asia, are crucial for effective malaria case management. This study developed, validated, and implemented droplet digital PCR (ddPCR) assays for precise quantification of P. vivax parasitemia and genotyping of the G6PD Viangchan variant. Duplex ddPCR assays targeting the P. vivax tubulin gene and the human RHCE gene were designed to accurately determine parasitemia, with precision and accuracy comp

The Association between Parasitemia and Liver Enzyme Alterations in Malaria Patients: ABO Blood Group as A Non-Contributory Factor.

BACKGROUND: Malaria, a life-threatening parasitic disease, exhibits diverse clinical manifestations influenced by parasite species, host immunity, and treatment access. Emerging evidence suggests that individual biological factors, such as ABO blood group and liver enzyme status, may also affect disease severity. This study investigates the relationship between blood type, liver enzyme levels, and malaria severity in affected patients. METHODS: A case-control study was conducted on malaria patients at the National Malaria Laboratory, Tehran University of Medical Sciences, from May 2022 to Oct

Effect of neem leaf extract (Azadirachta indica) in reducing the degree of parasitemia and apoptosis in C57BL mice with cerebral malaria.

BACKGROUND AND AIM: Brain malaria, which results from Plasmodium falciparum infection, is responsible for substantial fatalities and health issues. These processes, including cytoadherence, rosetting, and sequestration, induce an immune response, hypoxia, brain microvascular obstruction, disruption of the blood-brain barrier, and cell death. Parasitemia level can reveal the presence of infection and its association with apoptosis-related genes. Neem (Azadirachta indica) leaves with antimalarial properties could replace ineffective Indonesian malaria medications. This study was designed to eval

Impact of mass azithromycin distribution on malaria parasitemia during the low-transmission season in Niger: a cluster-randomized trial.

We assessed the effect of mass azithromycin treatment on malaria parasitemia in a trachoma trial in Niger. Twenty-four study communities received treatment during the wet, high-transmission season. Twelve of the 24 communities were randomized to receive an additional treatment during the dry, low-transmission season. Outcome measurements were conducted at the community-level in children < 1-72 months of age in May-June 2011. Parasitemia was higher in the 12 once-treated communities (29.8%, 95% confidence interval [CI] = 21.5-40.0%) than in the 12 twice-treated communities (19.5%, 95% CI = 13.0
