Cysteine cathepsin proteases in apicomplexan parasites.

Cysteine cathepsins are papain-like proteases which play key roles in a large range of organisms, including apicomplexan parasites. These obligate endoparasites are responsible for many devastating diseases in human and/or animal hosts, for which available treatments are limited or face the emergence of resistance. This review presents the cysteine cathepsins expressed by five major Apicomplexa (Plasmodium falciparum, Cryptosporidium parvum, Toxoplasma gondii, Eimeria tenella and Babesia bovis), highlighting available data on their structures, specific or common features, and biological functi

The Tubulin Superfamily in Apicomplexan Parasites.

Microtubules and specialized microtubule-containing structures are assembled from tubulins, an ancient superfamily of essential eukaryotic proteins. Here, we use bioinformatic approaches to analyze features of tubulins in organisms from the phylum Apicomplexa. Apicomplexans are protozoan parasites that cause a variety of human and animal infectious diseases. Individual species harbor one to four genes each for α- and β-tubulin isotypes. These may specify highly similar proteins, suggesting functional redundancy, or exhibit key differences, consistent with specialized roles. Some, but not all a

Can malaria parasites be spontaneously cleared?

A large body of evidence demonstrates that Plasmodium falciparum infections are chronic in malaria endemic areas; however, the notion of spontaneous clearance in the absence of antimalarial drug treatment is rarely discussed. In this opinion article, we review and reinterpret data to postulate that spontaneous clearance of P. falciparum infections occurs frequently, has been demonstrated in a range of transmission settings, and confirmed by the most sensitive malaria diagnostic techniques. We also discuss factors which may influence the likelihood, measurement, and conclusions of spontaneous c

Malaria parasites do respond to heat.

The capacity of malaria parasites to respond to changes in their environment at the transcriptional level has been the subject of debate, but recent evidence has unambiguously demonstrated that Plasmodium spp. can produce adaptive transcriptional responses when exposed to some specific types of stress. These include metabolic conditions and febrile temperature. The Plasmodium falciparum protective response to thermal stress is similar to the response in other organisms, but it is regulated by a transcription factor evolutionarily unrelated to the conserved transcription factor that drives the

Bioactive Peptides against Human Apicomplexan Parasites.

Apicomplexan parasites are the causal agents of different medically important diseases, such as toxoplasmosis, cryptosporidiosis, and malaria. Toxoplasmosis is considered a neglected parasitosis, even though it can cause severe cerebral complications and death in immunocompromised patients, including children and pregnant women. Drugs against Toxoplasma gondii, the etiological agent of toxoplasmosis, are highly toxic and lack efficacy in eradicating tissue cysts, promoting the establishment of latent infection and acute relapsing disease. Cryptosporidiosis has been recognized as the most frequ

Amino Acid Metabolism in Apicomplexan Parasites.

Obligate intracellular pathogens have coevolved with their host, leading to clever strategies to access nutrients, to combat the host's immune response, and to establish a safe niche for intracellular replication. The host, on the other hand, has also developed ways to restrict the replication of invaders by limiting access to nutrients required for pathogen survival. In this review, we describe the recent advancements in both computational methods and high-throughput -omics techniques that have been used to study and interrogate metabolic functions in the context of intracellular parasitism.

ApiAP2 Transcription Factors in Apicomplexan Parasites.

Apicomplexan parasites are protozoan organisms that are characterised by complex life cycles and they include medically important species, such as the malaria parasite Plasmodium and the causative agents of toxoplasmosis (Toxoplasma gondii) and cryptosporidiosis (Cryptosporidium spp.). Apicomplexan parasites can infect one or more hosts, in which they differentiate into several morphologically and metabolically distinct life cycle stages. These developmental transitions rely on changes in gene expression. In the last few years, the important roles of different members of the ApiAP2 transcripti

Protozoan Parasites and Type I IFNs.

For many years, the role of interferon (IFN)-I has been characterized primarily in the context of viral infections. However, regulatory functions mediated by IFN-I have also been described against bacterial infections and in tumor immunology. Only recently, the interest in understanding the immune functions mediated by IFN-I has dramatically increased in the field of protozoan infections. In this review, we discuss the discrete role of IFN-I in the immune response against major protozoan infections: Plasmodium, Leishmania, Trypanosoma, and Toxoplasma.

Recent advances in understanding apicomplexan parasites.

Intracellular single-celled parasites belonging to the large phylum Apicomplexa are amongst the most prevalent and morbidity-causing pathogens worldwide. In this review, we highlight a few of the many recent advances in the field that helped to clarify some important aspects of their fascinating biology and interaction with their hosts. Plasmodium falciparum causes malaria, and thus the recent emergence of resistance against the currently used drug combinations based on artemisinin has been of major interest for the scientific community. It resulted in great advances in understanding the resis

Translation in Organelles of Apicomplexan Parasites.

The protein translation machineries of the apicoplast and mitochondrion-the two actively translating organelles of apicomplexan parasites-have potential sites for drug intervention against diseases caused by these organisms. Work in the past few years, particularly on Plasmodium falciparum and Toxoplasma gondii, has shown that a reduced machinery of enzymes and factors is sufficient for organellar translation, which is also supported by components shared with the cytosolic translation system. This interplay between eukaryotic and prokaryotic-like components for mRNA translation in organelles i

Targeting Lysine Deacetylases (KDACs) in Parasites.

Due to an increasing problem of drug resistance among almost all parasites species ranging from protists to worms, there is an urgent need to explore new drug targets and their inhibitors to provide new and effective parasitic therapeutics. In this regard, there is growing interest in exploring known drug leads of human epigenetic enzymes as potential starting points to develop novel treatments for parasitic diseases. This approach of repurposing (starting with validated targets and inhibitors) is quite attractive since it has the potential to reduce the expense of drug development and acceler

Zoonotic foodborne parasites and their surveillance.

Humans suffer from several foodborne helminth zoonotic diseases, some of which can be deadly (e.g., trichinellosis, cerebral cysticercosis) while others are chronic and cause only mild illness (e.g., intestinal taeniosis). The route of infection is normally consumption of the parasite's natural host as a human food item (e.g., meat). The risk for infection with these parasites is highest wherever people have an inadequate knowledge of infection and hygiene, poor animal husbandry practices, and unsafe management and disposal of human and animal waste products. The design of surveillance and con

Interactions between helminth parasites and allergy.

PURPOSE OF REVIEW: To review the findings of recent human studies of the association between helminth parasite infections and allergy and discuss their potential relevance to public health. RECENT FINDINGS: Different helminth parasites may have different effects on allergy that may depend on the timing or intensity of the exposure or host genetic factors. Infections with Trichuris trichiura in early life are associated with a reduced prevalence of allergen skin test reactivity later in life and infants of helminth-infected mothers have been reported to have a reduced prevalence of eczema. Filarial parasites in the postgenomic era.

Filarial parasites are able to survive for many years in their host, with suppression of the host's immune response being one major survival strategy of the parasite. However, knowledge on molecules that induce these pathways is limited. Additionally, molecules that induce inflammation, thereby leading to severe pathology, such as elephantiasis, are also not fully identified. This article assesses the findings of a recently published analysis of stage-specific excretory-secretory proteins by sodium dodecylsulfate-polyacrylamide gel electrophoresis in combination with liquid chromatography-tand

Parasites in stranded cetaceans of Patagonia.

There is an increasing interest in parasites of marine mammals of Argentina. Here, we examined several poorly known cetaceans, i.e., 2 spectacled porpoises and 1 Burmeister's porpoise (Phocoenidae), and 1 Gray's beaked whale and 1 Cuvier's beaked whale (Ziphidae); we also updated the parasite information for 1 sperm whale (Physeteridae). These hosts strand only occasionally. We found Anisakis simplex s.l. in 2 spectacled porpoises and the Burmeister's porpoise, and recorded its distribution among the stomach chambers. Anisakis physeteris infected the sperm whale; Corynosoma cetaceum occurred i
