Saliva NIR spectroscopy and Aquaphotomics: a novel diagnostic approach to Paratuberculosis in dairy cattle.

INTRODUCTION: Paratuberculosis is a granulomatous intestinal infection that affects ruminant animals worldwide. The disease is often detected when most animals are already infected due to the long incubation period and the high transmissibility of the infectious agent. The lack of a comprehensive method to diagnose Paratuberculosis is a global challenge. Therefore, a non-destructive, fast, and cost-effective diagnostic method for early detection of Paratuberculosis is crucial. METHODS: Near-infrared spectroscopy (NIRS) and Aquaphotomics have the potential to diagnose the disease by detecting

Mycobacterium avium subsp. paratuberculosis is not discerned in diabetes mellitus patients in Hyderabad, India.

Mycobacterium avium ssp. paratuberculosis (MAP) is an obligate intracellular pathogen. It causes chronic intestinal inflammation in ruminants known as Johne's disease and is associated with human Crohn's disease. Furthermore, association of MAP with other autoimmune diseases, such as type-1 diabetes, has been established in patients from Sardinia (Italy) which is a MAP endemic and genetically isolated region. Due to largest livestock population and consequently high MAP prevalence amidst a very high diabetes incidence in India, we sought to test this association on a limited number of patient

Experimental infection of a bovine model with human isolates of Mycobacterium avium subsp. paratuberculosis.

Mycobacterium avium subsp. paratuberculosis (Map), the etiologic agent of Johne's disease (JD) in ruminants, has been implicated in the pathogenesis of Crohn's disease (CD) in humans. We developed a bovine ileal cannulation model to facilitate comparison of the immune response to Map and the mechanisms of pathogenesis in cattle and humans. Initial studies showed a T cannula could be maintained for up to a year in calves without inducing inflammation or adversely affecting intestinal function. Map introduced through the cannula established a persistent low level of infection without inflammatio

Use of bioinformatics to predict a function for the GS element in Mycobacterium avium subspecies paratuberculosis.

Mycobacterium avium subsp. Paratuberculosis (MAP) is a member of the Mycobacterium avium complex (MAC) and causes the inflammatory bowel disease, Johne's disease, in livestock. MAP has also been implicated as the causative agent of a similar disease, Crohn's disease, in humans. One of three major genetic differences between MAP and non-pathogenic MAC is the 6496-bp GS element. Based on the output from freely available protein sequence and structural bioinformatics tools, and the close homology of GS genes with the SER2 region of the closely related Mycobacterium avium subsp. Avium (MAA), we pr

Lesions in subclinical paratuberculosis of goats are associated with persistent gut-associated lymphoid tissue.

The organized gut-associated lymphoid tissue (the Peyer's patches [PPs] of domestic ruminants) is an important site of lesions caused by Mycobacterium avium subsp. paratuberculosis. To investigate the association between PP morphology and the lesions of paratuberculosis in goats, two experiments were performed. Five healthy kids aged 4-5 weeks were examined and the morphology of organized lymphoid tissue in the small intestine was described. Morphological similarities were observed between the ileocaecal-valve PP (ICVPP) and the jejunal PPs (JPPs), with pear-shaped follicles, large submucosal

Specificity Evaluation of a Polyprotein-Based ELISA Designed for the Detection of Paratuberculosis in Multiple Species.

Paratuberculosis (PTB) is a disease affecting ruminant animals. The etiological agent, Mycobacterium avium subspecies PTB (Map), is a short, Gram-positive, acid-fast bacillus. Due to the nature of PTB, diagnosis often occurs at advanced stages of the disease, following the onset of clinical symptoms and prior dissemination of the agent. The specificity of current humoral diagnostic techniques, such as ELISA, is limited, mainly due to the presence of various species of mycobacteria in the environment and other closely related pathogenic mycobacteria that can interfere with the accurate diagnosi

Are there benefits of culture-based detection of Mycobacterium avium spp paratuberculosis over histopathology?

Paratuberculosis (Johne's disease) has devastating outcomes on ruminant health and impacts on national and international trade. The current work assessed the diagnostic value of the VersaTREK automated liquid culture system in isolating Mycobacterium avium subspecies paratuberculosis (MAP) from faecal and intestinal tissue samples from ovine under South African conditions and compared it with the current method of choice, histopathological examination. Intestinal tissue and faecal samples from 111 sheep (including complete set from 104 slaughter sheep from flocks with a history of MAP infectio

Economic viability of different surveillance strategies for the control of paratuberculosis in Swiss dairy cattle.

Paratuberculosis (PTB), or Johne's disease, is a globally prevalent disease caused by Mycobacterium avium subsp. paratuberculosis, leading to significant economic losses in the dairy industry. This study assessed the economic viability of potential PTB surveillance strategies in Switzerland using stochastic simulation models. Two non-targeted surveillance strategies (SS1 and SS2) were assessed under three participation scenarios over ten years. Additionally, a more targeted surveillance strategy (SS3) was evaluated, focusing on historically positive herds. In SS1, herds are screened using sock s

Bayesian Assessment of True Prevalence of Paratuberculosis Infection in Dairy Herds and Their Parity Subgroups.

Paratuberculosis is a widespread infectious disease in ruminants that leads to significant economic losses in livestock production. In this study, we developed a practical method for predicting the likelihood of the herd-level presence of the infection and estimating its prevalence in subgroups of a dairy herd-specifically, first-time calving cows (primiparous) and those that have calved more than once (multiparous). We fit a Bayesian hierarchical model to cow-level data, incorporating prior knowledge about regional prevalence of infection to improve the accuracy and reliability of the estimat

Herd-level true seroprevalence of caseous lymphadenitis and paratuberculosis in the goat population of Poland.

A large-scale study was carried out in the Polish goat population in 2014-2021 to determine the herd-level true seroprevalence (HTP) of caseous lymphadenitis (CLA) caused by Corynebacterium pseudotuberculosis (Cp) and paratuberculosis (PTB) caused by Mycobacterium avium ssp. paratuberculosis (Map). Two-stage cluster sampling was applied to herds counting at least 20 adult goats (aged >1 year) and in each herd all males and 10-13 females were tested. At least one seropositive goat regardless of its sex was necessary to consider the herd as infected. HTP was estimated using the Bayesian approach

First identification of Mycobacterium avium subsp. paratuberculosis in wild ruminants in a zoo in Mexico.

BACKGROUND AND AIM: Paratuberculosis (PTB) is an infectious disease that induces chronic enteritis in ruminants. It is caused by Mycobacterium avium subsp. paratuberculosis (MAP). In this study, we evaluated the presence of MAP using bacteriological, molecular, and anatomopathological studies, based on the clinical suspicion of PTB in a zoo, in an area housing 10 scimitar-horned oryx (Oryx dammah), five giraffes (Giraffa camelopardalis), and three blue wildebeests (Connochaetes taurinus). MATERIALS AND METHODS: From November 2016 to June 2017, fecal samples were collected from individuals of

Paratuberculosis on small ruminant dairy farms in Ontario, Canada: A survey of management practices.

A cross-sectional study was undertaken (October 2010 to August 2011) to determine the risk factors for dairy goat herds and dairy sheep flocks testing positive for paratuberculosis (PTB) in Ontario, Canada. A questionnaire was administered to 50 producers during a farm visit in which concurrently, 20 randomly selected, lactating animals over the age of 2 years underwent sampling for paratuberculosis testing. Only 1 of 50 farms (2.0%) was closed to animal movement, whereas 96.6% of dairy goat farms and 94.1% of sheep farms purchased livestock from other producers. Only 10.3% of dairy goat, and

Crohn's disease and Mycobacterium avium subsp. paratuberculosis: the need for a study is long overdue.

The initial suggestion that Mycobacterium avium subsp. paratuberculosis (Map) might be involved in the pathogenesis of Crohn's disease (CD) was based on the apparent similarity of lesions in the intestine of patients with CD with those present in cattle infected with Map, the etiological agent of Johne's disease (JD). Recent investigations have now revealed the presence of Map or Map DNA in blood or lesions from adults and children with CD. Of special interest, Map has also been found in patients with other diseases as well as healthy subjects. The latter observations indicate all humans are s

The thioamides methimazole and thiourea inhibit growth of M. avium Subspecies paratuberculosis in culture.

BACKGROUND: Thyrotoxicosis is conceptualized as an "autoimmune" disease with no accepted infectious etiology. There are increasingly compelling data that another "autoimmune" affliction, Crohn disease, may be caused by Mycobacterium avium subspecies paratuberculosis (MAP). Like M. tb, MAP is systemic. We hypothesized that some cases of thyrotoxicosis may be initiated by a MAP infection. Because other thioamides treat tuberculosis, leprosy and M. avium complex, we hypothesized that a mode of action of some thioamide anti-thyrotoxicosis medications may include MAP growth inhibition. METHODS: Th

Comparison of the resistance of C57BL/6 and C3H/He mice to infection with Mycobacterium paratuberculosis.

Susceptibility of C57BL/6 (Bcgs) and C3H/HeN (Bcgr) mice to an intraperitoneal infection with Mycobacterium paratuberculosis strain 19698 was compared (by histopathology and the number of mycobacteria isolated from the spleen). Mycobacterial counts from the spleen of Bcgr mice progressively decreased over the course of infection but remained unchanged in Bcgs mice. Granulomatous lesions and acid-fast bacteria were consistently present in the liver and lymph nodes of Bcgs mice, whereas lesions were transient or absent in Bcgr mice. These results indicate that Bcgr mice are inherently resistant

Stereotypic immune response in Mycobacterium avium ssp. paratuberculosis infection among different Swiss caprine genotypes.

Paratuberculosis is an infection with Mycobacterium avium ssp. paratuberculosis (MAP) causing chronic enteritis in domestic and wild ruminants worldwide. In goats, the infection is caused by C (cattle)-type and S (sheep)-type strains. In this study, the correlation between different MAP strains and histomorphological lesions in the small and large intestines, as well as the mesenteric lymph nodes, in Swiss goats (Caprae aegagrus hircus) was investigated. Ten Swiss caprine MAP isolates were characterized using polymerase chain reaction (PCR) and enzymatic restriction-based single nucleotide pol

Sheep vaccinated against paratuberculosis have increased levels of B cells infiltrating the intestinal tissue.

Systemic immunisation delivered subcutaneously is currently used to control paratuberculosis, a chronic enteritis of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP). These vaccines do not provide complete protection and a small cohort of animals still succumb to clinical disease. The aim of this study was to assess mycobacterial infection site-specific variations in immune cells in vaccinated sheep that did or did not develop the disease following controlled exposure to MAP. Immunohistochemical staining of terminal ileum demonstrated that vaccination increased infiltr

Association between calf rearing technology and farm-level paratuberculosis infection in Hungarian dairy farms.

The ruminant paratuberculosis (PTB) is a global problem. The disease causes significant economic loss, primarily in dairy farms, due to premature culling, reduced slaughter value, and production losses. Newborn and young calves are the most susceptible to being infected, and the chance of infection decreases with increasing age. Animals are most commonly infected orally. The study aimed to assess the factors that have a greater association with PTB prevalence in Hungarian cattle farms and could play a major role in the eradication process. We visited 26 large-scale Hungarian dairy herds (havin
