Isolation and identification methods of Rothia species in oral cavities.

Rothia dentocariosa and Rothia mucilaginosa which are Gram-positive bacteria are part of the normal flora in the human oral cavity and pharynx. Furthermore, Rothia aeria, which was first isolated from air samples in the Russian space station Mir, is predicted to be an oral inhabitant. Immunocompromised patients are often infected by these organisms, leading to various systemic diseases. The involvement of these organisms in oral infections has attracted little attention, and their distribution in the oral cavity has not been fully clarified because of difficulties in accurately identifying the

Unified basis for the respirometric evaluation of inhibition for activated sludge.

This paper explores the merit of the oxygen uptake rate (OUR) profile obtained by means of respirometry as the basic mechanistic instrument for evaluating activated sludge inhibition. Experimental OUR data are generated using the synthetic peptone-based substrate and inhibition is tested with 60 mg/L hexavalent chromium and 33 mg/L nickel additions, corresponding to EC50 levels determined using the standard ISO 8192 procedure. Experimental results are evaluated by model calibration using ASM1 modified for dual hydrolysis and ASM3 modified for simultaneous growth. Model evaluations indicate tha

[Possible use of the indirect hemagglutination reaction for determining the sensitivity of the bacteria of glanders and melioidosis to antibiotics].

The use of IHAT for rapid assay of antibiotic sensitivity of bacteria causing malleus and melioidosis was shown to be possible. Pure bacterial cultures and suspensions of the spleen of infected animals were used as the investigation objects. The materials were additionally grown in meat-peptone broth with 5 per cent of glycerol at 37 degrees C with addition of an antibiotic or without it (control). Antibiotic efficacy was estimated by the difference in the titers in the main and control experiments. The test not requiring isolation of pure bacterial cultures provided a 2-3-fold decrease in the

Stable isotope ratios as a tool in microbial forensics--Part 2. Isotopic variation among different growth media as a tool for sourcing origins of bacterial cells or spores.

Since the anthrax attacks of 2001 the need for methods to trace the origins of microbial agents has become urgent. The stable isotope ratios of bacteria record information from both the nutrients and the water used to make their culture media and could potentially be used to provide information about their growth environment. We present a survey of carbon (C), nitrogen (N), and hydrogen (H) stable isotope ratios in 516 samples of bacteriological culture media. The observed variation was consistent with expected isotopic variation in the plant and animal products upon which the media are based.

Valorization of alkaline-enzymatic hydrolysates from sheep wool as bio-input-based technologies for Trichoderma harzianum cultivation and green forages.

The increase in sheep populations to meet the demand for meat generates large volumes of sheep wool, most of which is considered agro-industrial waste and poses a potential risk of contamination due to improper handling. Protein recovery is an attractive strategy for generating agricultural inputs, such as protein hydrolysates (PHs). This study aimed to evaluate the agrobiotechnological potential of sheep wool-derived PHs as (i) an alternative peptone source for Trichoderma harzianum and (ii) growth-promoting inputs in hydroponic green fodder (HGF) production. To this end, an alkaline-enzymati

Somatostatin inhibits intestinal mucosal mast cell degranulation in normal conditions and during mast cell hyperplasia.

Several studies demonstrate that intestinal mucosal mast cells (IMMC) are modulated by nervous reflexes as well as by intraluminal content. We recently demonstrated that peptones, such as ovalbumin hydrolysate (OVH), induce the release of rat mast cell protease II (RMCP II), indicating IMMC degranulation. The response is due to complex neuroendocrine reflexes. Somatostatin (SS) and its analogues have been used as potential treatments for inflammation in other body systems with contradictory results. The aim of this study was to evaluate if somatostatin could contribute to the reduction of inte

Optimization of Pyocyanin Production by Pseudomonas aeruginosa OG1 Using RSM: In Vitro Evaluation of Its Antibacterial and Anticandidal Efficacy Against Some Pathogens.

Background: The increasing antimicrobial resistance has led to a greater demand for alternative treatment options, which in turn has increased interest in naturally occurring biomolecules such as pyocyanin. Methods: In this study, a three-factor Box-Behnken Design (BBD)-based response surface methodology (RSM) was employed to optimize the effects of glycerol, peptone, and pH on pyocyanin production by Pseudomonas aeruginosa OG1. The antimicrobial efficacy of the optimized pyocyanin was subsequently evaluated in vitro against three Candida species and four clinically important bacterial pathoge

Characterization of Zalaria obscura Y1223 hydrolases: implications for lignocellulose conversion.

Overconsumption of fossil fuel reserves and its adverse effects has sparked interest in the production of second-generation biofuels due to the abundance of lignocellulosic waste and potential energy crops. However, processing costs associated with depolymerization of the cellulose crystalline structure have stalled advancement in cellulosic ethanol production. Current investigations range from identification of novel enzymes for lignocellulose hydrolysis to consolidation of enzyme production into a singular alcohol producing microorganism to potentially reduce cost for commercial processing.

The extremely high Al resistance of Penicillium janthineleum F-13 is not caused by internal or external sequestration of Al.

Penicillium janthinellum F-13 has been isolated in previous work as a fungus tolerating the presence of high concentrations of Al (as high as 100 mM AlCl3). Here its growth rate and yield in three acidic (pH 3.0) media of different composition with varying concentrations of Al are reported. The presence of Al did not affect these parameters. except that the growth yield was somewhat lower in GM (a glucose/peptone/yeast extract-containing medium) with the highest concentration tested (100 mM AlCl3). The amount of Al found in the mycelium was so low that it cannot lead to a significant decrease

Relation between cholecystokinin and antral innervation in the control of gastric emptying in the rat.

BACKGROUND: Antral motility and the hormone cholecystokinin (CCK) are major determinants of the rate of gastric emptying. The relation between CCK and antral neurons in regulating gastric emptying is uncertain. Benzalkonium chloride (BAC) causes selective lesions in gut myenteric neurons after serosal application. AIM: To develop a model of antral denervation using BAC to enable the study of the relation between CCK and antral neurons in regulating gastric emptying. METHODS: BAC, vehicle or the afferent neurotoxin capsaicin were applied to the serosal surface of the rat antrum or corpus; neu

[L-Glutamate oxidase from Streptomyces cremeus 510 MGU: effect of nitrogen sources on enzyme secretion].

Effect of nitrogen sources (organic complexes and mineral salts) on L-glutamate oxidase synthesis by Streptomyces cremeus 510 MGU was studied. Optimal enzyme production was not provided by any single nitrogen source. The most effective combination of nitrogen sources (soy flour, peptone, ammonium sulfate) was elaborated by the mathematical planning method. The results of experiment allowed to enhance biosynthesis of extracellular L-glutamate oxidase to 2.4-2.6 U/mL. It was shown that L-glutamate oxidase of Streptomyces cremeus 510 MGU is highly specific to substance and is stable during storag

EFFECTS OF CORN STEEP LIQUOR AND THIAMINE ON L-GLUTAMIC ACID FERMENTATION OF HYDROCARBONS.IV. UTILIZATION OF HYDROCARBONS BY MICROORGANISMS.

The effect of nutrients of natural source, such as corn steep liquor, peptone, and yeast extract, on the fermentative production of L-glutamic acid from hydrocarbons by a Corynebacterium was studied. Corn steep liquor and meat extract were found to be remarkably stimulatory to L-glutamic acid production; about 5 g per liter of L-glutamic acid were accumulated in a culture broth containing 3% n-paraffins, 0.01% corn steep liquor, and mineral salts. Among nutritional factors contained in corn steep liquor, biotin had very little effect on the accumulation of L-glutamic acid, but thiamine was hig

Can isoproteic diets with different amino acid profiles modulate the amino acid composition of the black soldier fly?

The nutritional composition of black soldier fly (Hermetia illucens; BSF) larvae is highly dependent on feed substrates, yet the independent effects of substrate protein levels and amino acid composition remain poorly understood due to the use of complex mixed feeds. This study employed a simplified isoproteic model using three purified peptones (meat, soybean, and casein) at two protein levels (124 g/kg and 46 g/kg DM) to investigate larval metabolic responses. The results reveal that BSF larvae employ a dual regulatory mechanism in response to varying nutrient inputs. The total amino acid (T

Mixed infection caused by V.cholerae and S.typhi was studied on guinea pig gall-bladder, used as an experimental model. These microorganisms coexisted in association in animals and exhibited no pronounced antagonistic properties in vitro. The cultures isolated from the organs of infected guinea pigs did not differ from initial ones. The study revealed that in nutrient broth containing 50% of dried bile salmonellae were preserved, but not V.cholerae.
