Ebselen restores peri-implantitis-induced osteogenic inhibition via suppressing BMSCs ferroptosis.

It is hard to reconstruct bone defects in peri-implantitis due to osteogenesis inhibited by excessive reactive oxygen species (ROS). Ferroptosis, a recently identified regulated cell death characterized by iron- and ROS- dependent lipid peroxidation, provides us with a new explanation. Our study aims to explore whether ferroptosis is involved in peri-implantitis-inhibited osteogenesis and confirm ebselen, an antioxidant with glutathione peroxidase (GPx)-like activity, could inhibit ferroptosis and promote osteogenesis in peri-implantitis. In this study, we used LPS to mimic the microenvironmen

Antibiotic Resistance in Patients with Peri-Implantitis: A Systematic Scoping Review.

The implementation of adjunctive antibiotics has been recommended for the therapy of peri-implantitis (PI). In this review, antibiotic resistance patterns in PI patients were assessed. A systematic scoping review of observational studies and trials was established in conjunction with the PRISMA extension for scoping reviews. The SCOPUS, PubMed/MEDLINE, EMBASE, SCIELO, Web of Science, and LILACS databases were reviewed along with the gray literature. The primary electronic examination produced 139 investigations. Finally, four observational studies met the selection criteria. These studies eval

Current Concepts on the Pathogenesis of Peri-implantitis: A Narrative Review.

As implant treatment has been integrated in contemporary dental practice, complications with the forms of peri-implant mucositis and peri-implantitis have also increased in prevalence. Peri-implantitis is the more severe biological complication and is defined as an inflammatory disease affecting peri-implant tissues resulting in bone and eventually implant loss. In addition, the treatment of peri-implantitis has currently become a substantial global economic burden. In the current study, a search was conducted in several electronic databases using specific keywords relevant to the article's ma

Oral microbial profile variation during canine ligature-induced peri-implantitis development.

BACKGROUND: Dental implants have become well-established in oral rehabilitation for fully or partially edentulous patients. However, peri-implantitis often leads to the failure of dental implants. The aim of this study was to understand the core microbiome associated with peri-implantitis and evaluate potential peri-implantitis pathogens based on canine peri-implantitis model. RESULTS: In this study, three beagle dogs were used to build peri-implantitis models with ligature-induced strategy. The peri-implant sulcular fluids were collected at four different phases based on disease severity dur

Risk of Superinfection in Peri-implantitis After Systemic Broad Spectrum Antibiotics.

Peri-implant disease has developed over the last few years as a complication that is often difficult to resolve. The disease process is mainly attributed to bacterial infection. Proposed combined therapies use broad-spectrum antibiotics to halt its progression. A major associated risk is the undetected development of superinfections that are difficult to eradicate. A group of healthy individuals with advanced peri-implantitis (PI) were referred for evaluation due to severe, rapidly progressive bone loss. Previous nonsurgical and empiric antibiotic therapy had been rendered. Culture and polymer

Occlusion-activated autonomous piezoelectric implants for adaptive prevention of peri-implantitis.

More than one-third of the global population suffers from dental defects, with osseointegrated implants being the gold-standard intervention; yet, their long-term functional performance is hindered by peri-implantitis. Here, we present an autonomous piezoelectric implant that delivers adaptive antibacterial and anti-inflammatory functionalities through physiological occlusal activation without any external intervention. The implant demonstrates good functional durability, sustaining consistent bioelectric outputs through over 1,000,000 loading-unloading cycles and exhibiting stable electrical

Decoding host-microbial dynamics in peri-implantitis: Ecosystem-specific experimental models.

BACKGROUND: Peri-implantitis is an inflammatory disease arising within a unique host-microbial-biomaterial ecosystem. Unlike periodontitis, peri-implant tissues lack a periodontal ligament and epithelial junction integrity and interface with metal oxides on titanium surfaces. The inflammatory changes in peri-implant disease not only affect the host but also extend to the implant surfaces, which undergo corrosion and particle release. These specific ecological features must be accurately captured in preclinical experimental models to provide mechanistic insight and enable translational success.

Neutrophil Extracellular Traps Induce Pyroptosis of Gingival Fibroblasts in Peri-Implantitis.

OBJECTIVES: This study aimed to verify the release of neutrophil extracellular traps (NETs) in peri-implantitis and explore whether NETs induce pyroptosis and inflammatory responses in human gingival fibroblasts (HGFs). MATERIALS AND METHODS: Peri-implant soft tissue samples were collected from healthy individuals and patients with peri-implantitis. NETs and the expression of pyroptosis-related factors including NLRP3, Caspase1, GSDMD, and IL-1β were detected. In vitro, NETs induced from differentiated HL60 (dHL60) cells and human neutrophils were used to stimulate HGFs. Transcriptome sequenc

The impact of biocorrosion and titanium ions release on peri-implantitis.

OBJECTIVES: Biofilm accumulation is considered the primary cause of peri-implant inflammation. Still, metallosis caused by an increased concentration of titanium ions at the site of peri-implantitis site cannot be ignored. Whether titanium ions alone or in concert with bacterial biofilm trigger inflammation and bone destruction in peri-implant tissues remains unproven. MATERIALS AND METHODS: Articles were retrieved from PubMed/Medline, Web of Science. All studies focusing on titanium ions release in peri-implant reactions were included and evaluated. RESULTS: Titanium implants are considered

Cargo-less Nanoparticles Prevent Bone Loss in Periodontitis and Peri-implantitis.

Periodontal and peri-implant diseases are a significant public health problem worldwide, resulting in the destruction of the supporting bone. These bone defects can cause esthetic problems, increased relapse rate, and eventually tooth loss. The etiology of periodontal disease involves an influx of innate immune cells (neutrophils and monocytes) and upregulation of local inflammatory cytokines in the gingiva. Biodegradable polymeric nanoparticles are an inexpensive, safe, and effective means of preventing innate immune activation by bacterial biofilms. We therefore hypothesize that this technol

Biomaterials science and surface engineering strategies for dental peri-implantitis management.

Peri-implantitis is a bacterial infection that causes soft tissue inflammatory lesions and alveolar bone resorption, ultimately resulting in implant failure. Dental implants for clinical use barely have antibacterial properties, and bacterial colonization and biofilm formation on the dental implants are major causes of peri-implantitis. Treatment strategies such as mechanical debridement and antibiotic therapy have been used to remove dental plaque. However, it is particularly important to prevent the occurrence of peri-implantitis rather than treatment. Therefore, the current research spot ha

Microbial boundaries in peri-implantitis: a review of pathogen-related advances.

In the field of implantology, peri-implantitis is still a common complication of implant failure. Similar to periodontal disease, this kind of pathological condition is characterized by inflammation of the tissues surrounding dental implants or fillings. The sources of infection have been shown to be chronic periodontitis and poor maintenance of the communion. A thorough examination of the intricate components of peri-implantitis was sought in this review in order to identify common characteristics of the disease with regard to bacteria, biofilm formation, host immunological responses, diagnos
