[Cultivating human Schwann cells for tissue engineering of peripheral nerves].

Cultivation of human cells is well established. The cultivation of human Schwann cells may offer a new therapeutic approach for treatment of degenerative and traumatic lesions of the peripheral nervous system. Currently, Schwann cells in combination with other biological matrices are used as tissue-engineered biological nerve grafts in animal models. Cultivation of human Schwann cells, however, is more difficult than cultivation of rodent cells. A high cell yield is only achieved by pharmacological stimulation, which should not be used in clinical therapy. Thus, we aimed to establish an easy m

AAEM minimonograph #42: intraoperative monitoring of peripheral and cranial nerves.

Intraoperative studies of focal neuropathies have become increasingly common both as an aid to the localization of the neuropathy and a means of monitoring the integrity of the nerve in the course of surgery. Success very often depends on careful attention to the technical problems presented by working in the operating theater. Despite these difficulties, assessment of conduction directly from the exposed nerve offers opportunities for better understanding the pathophysiology of these neuropathies and correlating these findings with conventional electrodiagnostic studies.

Changes in the peripheral nerves in rats induced by vibration.

Effects of long-term low frequency vibrations on the structure of the sciatic and plantar nerves were examined in Wistar strain rats. Decrease in total number of nerve fibers was noted. This was associated with altered distribution of myelinated fiber diameters, disturbances in axon structure and with fiber demyelination. The extend of lesions depended upon duration of exposure and distance from the source of vibrations.

Damage and regeneration of peripheral nerves in advanced treated leprosy.

Despite the rapidly falling prevalence of leprosy, the disability and handicap resulting from loss of protective sensation, due to irreversible nerve damage, will remain a huge medical problem for many years. To elucidate the location and consequences of permanent nerve damage in treated leprosy, a prospective study involving nine patients who underwent leg amputation was conducted. Full-length nerves dissected from amputated legs were studied with histological and immunohistochemical methods. Our main findings were that: in both lepromatous and tuberculoid leprosy nerve damage increased dista

[Iatrogenic injuries of the peripheral nerves: analysis of 226 cases].

In 226 patients with iatrogenic peripheral nerve injuries, the causes were analyzed in detail. The iatrogenic nerve injuries were incurred through injection of therapeutic agents in 84 patients, inadvertent division by the surgeon in 44, traction during orthopaedic operation in 27, and undue compression from an ill-fitting splint or plaster cast in 26. The prophylaxis of such iatrogenic lesions is discussed. It cannot be over-stressed to exercise special care to prevent the nerve from being injured in the course of dissection or in any therapeutic maneuver, and to grasp a precise knowledge of

[Peripheral neurofibromatosis and involvement of lumbosacral nerves. Value of imaging].

We report a case of giant plexiform neurofibromatosis revealed by a sensori-motor, axo-myelinic polyneuropathy. Only the lumbar and sacral spinal nerves were involved in this peripheral form of Von Recklinghausen's disease. The polyneuropathy was due to compression of the nervous fibers by the neurofibromatous tumors. Ultrasonography DTPA 99 m Technetium scintigraphy, CT and MRI were the diagnostic procedures. MRI proved superior to CT in demonstrating the extent of the lesions. Biopsy confirmed the diagnosis. This form of neurofibromatosis did not involve the central nervous system, cranial n

The central-peripheral transitional regions of cranial nerves. Oculomotor nerve.

Oculomotor nerve rootlets varied more markedly in size and in transitional zone length and form than those of any other nerve studied to date. However, they could be classified into four main types, each of which was associated with a characteristic type of central-peripheral transitional zone. Type 1 rootlets emerged from the brainstem through a tongue-shaped elevation of the glia limitans. This type of central-peripheral transition is found elsewhere only in a minority of abducent rootlets (Fraher et al. 1988). Type 2 rootlets contained long segments made up entirely of central nervous tissu

[Late paralysis of the peripheral nerves from the neurologic viewpoint].

In a total of 791 cases of peripheral neuroparalysis, we found 77 cases of late paresis corresponding to approximately 10 per cent of the total number of patients. Almost half the number of late paresis were pareses of the ulnar nerve. The average latency period was 18 years. The causes were: 26 times fractures, 21 times open, sencondarily healed injuries of the soft parts, 14 times irradiation therapy and some closed injuries of the soft parts. Several late pareses occurred after industrial accidents or medical interventions, which may have legal consequences with respect to insurance. The ea

Familial amyloid polyneuropathy with marked hypertrophy of the peripheral nerves.

Autopsy findings in a 40-year-old male with heredofamilial amyloidosis and polyneuropathy are reported. He had been suffering from progressive autonomic as well as sensorimotor dysfunctions. Prominent amyloid deposit was found in the kidney, heart, thyroid, and testis, and less in the interstitium and small vessels of almost all organs. The peripheral nerves, some showing prominent hypertrophy, were most severely involved by amyloid deposit in a form of stellate mass, which ultrastructurally consisted of radially arranged amyloid filaments. In the hypertrophied nerves and ganglia, in addition

Clinical and experimental aspects of injection injuries of peripheral nerves.

Injury to peripheral nerves complicating deep intramuscular injections of antibiotic and other agents is well recognized and can result in significant permanent neurological deficit. The purpose of this paper is to review the subject of nerve injection injuries, and report on a series of recent experimental studies carried out in this laboratory designed to improve our understanding of the pathophysiology of this condition and help provide a rational basis for its treatment. A wide variety of chemotherapeutic, prophylactic, and local anaesthetic agents in common use were injected into the scia

Ultrastructural pathology of peripheral nerves in patients with diabetic neuropathy.

Sural nerve lesions in patients with clinically manifest diabetic neuropathy were investigated electron microscopically. Myelinated nerve fibers were reduced in all the diabetic patients. Axonal degeneration of both myelinated and unmyelinated nerve fibers was most conspicuous finding in the diabetic sural nerves. Structural changes of the axons were represented by axonal dwindling, depletion of axoplasmic organelles, vacuolarization and an increase in neurofilaments. Accumulation of glycogen-like particles and deposition of electron homogeneous amorphous materials were noted within a few axon

Targeted Polymersomes Enable Enhanced Delivery to Peripheral Nerves Post-Injury.

The gold standard therapy for peripheral nerve injuries involves surgical repair, which is invasive and leads to major variations in therapeutic outcomes. Because of this, smaller injuries often go untreated. However, alternative, noninvasive routes of administration are currently unviable due to the presence of the blood-nerve barrier (BNB), which prevents passage of small molecules from the blood into the endoneurium and the nerve. This paper demonstrates that ligands on the surface of nanoparticles, called polymersomes, can enable delivery to the nerve through noninvasive intramuscular inje

Neuroscience in peripheral cancers: tumors hijacking nerves and neuroimmune crosstalk.

Cancer neuroscience is an emerging field that investigates the intricate relationship between the nervous system and cancer, gaining increasing recognition for its importance. The central nervous system governs the development of the nervous system and directly affects brain tumors, and the peripheral nervous system (PNS) shapes the tumor microenvironment (TME) of peripheral tumors. Both systems are crucial in cancer initiation and progression, with recent studies revealing a more intricate role of the PNS within the TME. Tumors not only invade nerves but also persuade them through remodeling

Fibrin Glue Coating Limits Scar Tissue Formation around Peripheral Nerves.

Scar tissue formation presents a significant barrier to peripheral nerve recovery in clinical practice. While different experimental methods have been described, there is no clinically available gold standard for its prevention. This study aims to determine the potential of fibrin glue (FG) to limit scarring around peripheral nerves. Thirty rats were divided into three groups: glutaraldehyde-induced sciatic nerve injury treated with FG (GA + FG), sciatic nerve injury with no treatment (GA), and no sciatic nerve injury (Sham). Neural regeneration was assessed with weekly measurements of the vis

Optical Feedback Control and Electrical-Optical Costimulation of Peripheral Nerves.

BACKGROUND: Optogenetics is an emerging technology that enables the expression of light-activated ion channels in mammalian cells. Neurons expressing light-activated ion channels can be depolarized using the appropriate wavelength of light. Optical stimulation of neurons could have important implications for further understanding and managing peripheral nerve deficits leading to paresis or paralysis. This study examines the utility of this technology in a feedback-controlled system and the advantages of coupling this technology with conventional electrical stimulation. METHODS: The sciatic ne

[Decompression of Peripheral Nerves in the Treatment of Diabetic Polyneuropathy].

Polyneuropathy causing ulceration and amputation as a long-term consequence of diabetes mellitus is analysed for its pathophysiology, socioeconomic and medical relevance. Outcomes of decompression of peripheral nerves on the lower extremity regarding sensation, pain, development, and recurrence of ulceration, and amputation is evaluated by a systematic literature review in pubmed, medline and embase. Decompression of peripheral nerves in diabetic peripheral neuropathy seems to be a good treatment option for restoring sensation, decreasing pain, as well as for avoiding the development and recur

Variable spatial magnetic field influences peripheral nerves regeneration in rats.

Generator of spatial magnetic field is one of most recent achievements among the magnetostimulators. This apparatus allows to obtain the rotating magnetic field. This new method may be more effective than other widely used techniques of magnetostimulation and magnetotherapy. We investigated the influence of alternating, spatial magnetic field on the regeneration of the crushed rat sciatic nerves. Functional and morphological evaluations were used. After crush injury of the right sciatic nerve, Wistar C rats (n = 80) were randomly divided into four groups (control and three experimental). The e
