Neurotoxicity of chemotherapeutic agents: mechanisms and mitigation strategies.

Chemotherapeutic drugs have neurotoxic consequences even if they are used to treat cancer. Peripheral neuropathy, cognitive impairment, neuropathic pain, seizures, encephalopathy, and other symptoms may be present. According to studies conducted worldwide, up to 75% of patients taking chemotherapeutic drugs may have detectable cognitive deficits, while 30-40% of patients acquire peripheral neuropathy. One of the most commonly reported neurological side effects of cancer treatment is chemotherapy-induced peripheral neuropathy, which raises morbidity and, in certain situations, death. This overv

An efficient cellular image-based platform for high-content screening of neuroprotective agents against chemotherapy-induced neuropathy.

Chemotherapy-induced peripheral neuropathy (CIPN) is a major dose-limiting side effect, with no approved therapy for prevention or treatment. Here, we aimed to establish a high-content image platform based on the neurite outgrowth of dorsal root ganglia (DRG)-derived neuron cells for the discovery of neuroprotective agents against paclitaxel-induced CIPN. ND7/23 cells, an immortalized hybrid DRG cell line, were maturely differentiated by induction with nerve growth factor and upregulation of intracellular cAMP levels. High-content image analyses of the neurofilament-stained neurite network sho

[Effectiveness of Angiotensin System Inhibitors Against Painful Peripheral Neuropathy in Type 2 Diabetic Old Patients: Validation by Analysis of Pharmacy Claims Data].

The angiotensin system participates in regulation of neuronal activity in the central and peripheral nervous systems, in addition to its role in the control of cardiovascular functions and fluid balance. A fundamental study employing laboratory animals and retrospective analysis of hospitals' medical records have recently suggested that treatment with angiotensin-converting enzyme inhibitors (ACEIs) or angiotensin II AT1 receptor blockers (ARBs) reduces the development of diabetic peripheral neuropathy (DPN) accompanying type 2 diabetes. To validate this novel notion, we conducted a retrospect

[Polypeptides and antagonists].

Polypeptides are endogenous agents, involved in the regulation of many physiologic functions and the pathogenesis of several diseases. Polypeptide antagonists form a group of new chemical entities which may provide valid therapeutic agents. Some polypeptides (angiotensin, kinins) are released through the action of proteolytic enzymes (renin, kallikreins) and act as hormones or autacoids; others (substance P, neurotensin) are synthetized by nervous cells to serve as neurotransmitters or neuromodulators. The main homeostatic role of the renin-angiotensin system is to uphold high systemic arteria

The neurotropic herpes viruses: herpes simplex and varicella-zoster.

Herpes simplex viruses types 1 and 2 (HSV1 and HSV2) and varicella-zoster virus (VZV) establish latent infection in dorsal root ganglia for the entire life of the host. From this reservoir they can reactivate to cause human morbidity and mortality. Although the viruses vary in the clinical disorders they cause and in their molecular structure, they share several features that affect the course of infection of the human nervous system. HSV1 is the causative agent of encephalitis, corneal blindness, and several disorders of the peripheral nervous system; HSV2 is responsible for meningoencephalit

Agent Orange Reviewed: Potential Role in Peripheral Neuropathy and Neurodegeneration.

Agent Orange, a dioxin-containing toxin, was used as an herbicide during the Vietnam War. Exposures to Agent Orange were initially linked to birth defects among Vietnamese civilians residing near aerially sprayed regions. Years later, returning South Korean and U.S. Veterans exposed to Agent Orange exhibited increased rates of malignancy, cardiovascular disease, diabetes and birth defects in their offspring. Growing evidence that herbicides and pesticides contribute to chronic diseases including neurodegeneration raises concern that Agent Orange exposures may have increased the risk for later

Pathogenesis of classical and atypical BSE in cattle.

It is known from earlier studies that the pathogenesis of BSE in cattle differs considerably from the TSE pathogenesis in sheep, where the lymphoreticular system (LRS) is majorly involved in the transport and propagation of the agent. In cattle, the BSE agent has only been detected in the Peyer's patches of the distal ileum and in the tonsils, which have both been identified as the portal of entry for the agent after oral uptake. It was shown that as opposed to most other animal species, in cattle the BSE agent amplifies almost exclusively in the central and peripheral nervous system. However,

[Intravital laboratory diagnosis of human subacute spongioid encephalopathies (Creutzfeldt-Jakob syndrome and amyotrophic leukospongiosis)].

Methods aimed at the detection of causative agents in the CSF and peripheral blood lymphocytes are recommended for the use in intravital laboratory diagnosis of slow infections of the central nervous system. The results obtained enable recommending the biotest on guinea-pigs or indication of the causative agent of amyotrophic leukospongiosis (AL) in cell culture coupled with the punctate immunoenzyme assay for the diagnosis of AL. As to the diagnosis of Creutzfeldt-Jacob disease, it is suggested that the biotest on guinea-pigs and the punctate immunoenzyme assay may be used.

An update on the involvement of inflammatory mediators in Parkinson's disease pathogenesis.

Several endogenous and exogenous mediators and pathogenic agents trigger inflammation. Despite the brain being less vulnerable to inflammation, many studies have underlined the contribution of inflammatory molecules in the pathogenesis of Parkinson's disease (PD). The onset of the features of PD is the result of an interaction of multiple contributors. These factors include inflammatory mediators, α-synuclein accumulation, oxidative stress, mitochondrial dysfunction, and neuronal cell death. Due to multifaceted interaction and cross-talk amongst a bunch of proteins and pathways, deciphering th

PrPTSE distribution in a primate model of variant, sporadic, and iatrogenic Creutzfeldt-Jakob disease.

Human prion diseases, such as Creutzfeldt-Jakob disease (CJD), are neurodegenerative and fatal. Sporadic CJD (sCJD) can be transmitted between humans through medical procedures involving highly infected organs, such as the central nervous system. However, in variant CJD (vCJD), which is due to human contamination with the bovine spongiform encephalopathy (BSE) agent, lymphoreticular tissue also harbors the transmissible spongiform encephalopathy-associated prion protein (PrP(TSE)), which poses a particularly acute risk for iatrogenic transmission. Two blood transfusion-related cases are alread

[Vasculitis of the nervous system in infectious diseases].

Vasculitis and vasculopathies of the central and peripheral nervous system can be caused by infectious diseases. Vasculitis can lead to stenosis, occlusion and aneurysm formation of blood vessels which may result in stroke or cerebral haemorrhage. In cases of peripheral nervous system involvement mononeuritis multiplex and symmetric peripheral neuropathy are possible. The diagnosis is based on clinical presentation, serology, cerebrospinal fluid analysis and neuroradiologic examinations. In cases of peripheral neuropathy neurophysiologic examinations and biopsy of the sural nerve can lead to t

Autoimmune neuromuscular disorders.

Autoimmune neuromuscular disorders affecting peripheral nerves, neuromuscular junction or muscle have a wide clinical spectrum with diverse pathogenetic mechanisms. Peripheral nervous system may be targeted in the context of complex immune reactions involving different cytokines, antigen-presenting cells, B cells and different types of T cells. Various immunomodulating and cytotoxic treatments block proliferation or activation of immune cells by different mechanisms attempting to control the response of the immune system and limit target organ injury. Most treatment protocols for autoimmune ne

Somatostatin as an Active Substance in the Mammalian Enteric Nervous System.

Somatostatin (SOM) is an active substance which most commonly occurs in endocrine cells, as well as in the central and peripheral nervous system. One of the parts of the nervous system where the presence of SOM has been confirmed is the enteric nervous system (ENS), located in the wall of the gastrointestinal (GI) tract. It regulates most of the functions of the stomach and intestine and it is characterized by complex organization and a high degree of independence from the central nervous system. SOM has been described in the ENS of numerous mammal species and its main functions in the GI trac

Activation of the AMPK Signaling and Brain Function: A Friend or Foe?

AMP-activated protein kinase (AMPK) is a central regulator of cellular energy balance, and its activation presents a therapeutic strategy for various metabolic diseases and neuronal disorders. Yet its effects on brain function remain unclear. This review evaluates evidence from animal and human studies examining AMPK activation through pharmacological agents (synthetic or natural compounds) and exercise. The effects of AMPK activation on synaptic and cognitive function seem highly context-dependent. Peripheral or acute AMPK activation often improves cognition, whereas chronic or central nervou

[The prevention of neurotoxic reactions during the treatment of pulmonary tuberculosis in middle-aged and elderly patients].

To prevent and control neurotoxic reactions in the treatment of pulmonary tuberculosis in elderly and old patients the author used dipromonium (derivative of pangamic acid). Use of this agent at the beginning of the treatment course reduced the incidence of side-effects of neurotoxic character. In case of development of reactions of the central and peripheral nervous system dipromonium treatment resulted in their weakening or liquidation.

[Mysteries of viral latency and reactivation].

Varicella-zoster virus is a Herpesvirus responsible for three distinct clinical features: chicken pox (varicella), shingles (herpes zoster) and post-zosterian pain (post-herpetic neuralgia). Neurological features of these diseases such as complications of chicken pox, viral latency in sensory ganglia and reactivation as shingles with concurrent and possibly subsequent prolonged pain, are the sequels of the invasion of the peripheral nervous system during primary infection. Prevention is achieved by vaccination with a live attenuated virus strain and therapy calls for specific antiviral agents.
