Deciphering the acute toxicity mechanisms of PFAS in algae: A molecular descriptor and binding energy hierarchy perspective.

Per- and polyfluoroalkyl substances (PFASs) are globally persistent pollutants, yet their molecular mechanisms of toxicity in aquatic organisms remain unclear. To elucidate the molecular mechanisms of perfluorocarboxylic acid (PFCA) toxicity in algae, we combined protein-protein interaction (PPI) network analysis, molecular docking, and molecular dynamics simulations (MDS). Three photosystem II (PSII) core proteins-D1, CP47, and CP43-were identified as primary PFCA-binding targets. Molecular docking revealed a chain-length-dependent enhancement of binding affinity (-5.2 to -9.7 kcal·mol⁻1) fol

Carbonic anhydrase activity in Arabidopsis thaliana thylakoid membrane and fragments enriched with PSI or PSII.

The procedure of isolating the thylakoids and the thylakoid membrane fragments enriched with either photosystem I or photosystem II (PSI- and PSII-membranes) from Arabidopsis thaliana leaves was developed. It differed from the one used with pea and spinach in durations of detergent treatment and centrifugation, and in concentrations of detergent and Mg(2+) in the media. Both the thylakoid and the fragments preserved carbonic anhydrase (CA) activities. Using nondenaturing electrophoresis followed by detection of CA activity in the gel stained with bromo thymol blue, one low molecular mass carri

Acclimation of tobacco leaves to high light intensity drives the plastoquinone oxidation system--relationship among the fraction of open PSII centers, non-photochemical quenching of Chl fluorescence and the maximum quantum yield of PSII in the dark.

Responses of the reduction-oxidation level of plastoquinone (PQ) in the photosynthetic electron transport (PET) system of chloroplasts to growth light intensity were evaluated in tobacco plants. Plants grown in low light (150 micromol photons m-2 s-1) (LL plants) were exposed to a high light intensity (1,100 micromol photons m-2 s-1) for 1 d. Subsequently, the plants exposed to high light (LH plants) were returned back again to the low light condition: these plants were designated as LHL plants. Both LH and LHL plants showed higher values of non-photochemical quenching of Chl fluorescence (NPQ

Comparative FTIR analysis of the microenvironment of in cyanide-treated, high pH-treated and iron-depleted photosystem II membrane fragments.

FTIR difference spectra due to light-induced formation were measured in control PS II membrane fragments and in samples where the magnetic interaction with the non-heme iron center in its high-spin Fe2+ state is eliminated by three different methods, i.e., extraction of the non-heme iron center, treatment with cyanide, and incubation at high pH (pH = 11). The results obtained reveal that (i) the most pronounced band at 1479 cm-1 reflecting the C../--O stretching mode of in the semiquinone anion radical remains invariant to "iron depletion" while it shifts by 4 and 2 cm-1 to lower frequencies u

Regulation and Functional Complexity of the Chlorophyll-Binding Protein IsiA.

As the oldest known lineage of oxygen-releasing photosynthetic organisms, cyanobacteria play the key roles in helping shaping the ecology of Earth. Iron is an ideal transition metal for redox reactions in biological systems. Cyanobacteria frequently encounter iron deficiency due to the environmental oxidation of ferrous ions to ferric ions, which are highly insoluble at physiological pH. A series of responses, including architectural changes to the photosynthetic membranes, allow cyanobacteria to withstand this condition and maintain photosynthesis. Iron-stress-induced protein A (IsiA) is homo

24-Epibrassinolide effectively alleviates UV-B stress-induced damage in the cyanobacterium Anabaena sp. PCC 7120 by employing nitric oxide: Improved PS II photochemistry, antioxidant system, and growth.

The alleviation of UV-B stress-induced damage by 24-epibrassinolide (EBL) in the cyanobacterium Anabaena sp. PCC 7120 and the role of nitric oxide (NO) signaling were investigated in this study. UV-B exposure caused a significant (P < 0.05) decline by 31% in the growth of test organism by negatively impacting photosynthetic pigments, whole cell O2 evolution, and PS II photochemistry [decrease in the values of Fv/F0, PIABS, and quantum efficiencies (Phi_Po, Phi_Eo, and Psi_0) and increase in specific energy fluxes (ABS, TR0, DI0, and ET0) per active RCs]. Stress enhanced the levels of oxidative

How do Mediterranean shrub species cope with shade? Ecophysiological response to different light intensities.

Under natural conditions, light exposure for Mediterranean shrubs can be highly variable, especially during cloudy days or under a canopy, and can interfere with other environmental factors such as temperature and water availability. With the aim of decoupling the effect of radiation and temperature from water availability, we conducted an experiment where two perennial and three summer semi-deciduous shrub species were subjected to different levels of irradiation. In order to follow plant responses to light exposure, we measured gas exchange, photosystem II photochemical efficiency, photosynt

Seasonal variation of leaf ecophysiological traits within the canopy of Quercus petraea (Matt.) Liebl. trees.

Facing contrasting light regimes during a vegetation season and depending on canopy position, physiological plasticity of leaves is vital for tree species to sustain the optimal ratio between the benefit of carbon assimilation and the costs of photoprotection in a given leaf. We tested the seasonal adjustment of sun and shade leaf photochemistry of sessile oak (Quercus petraea (Matt.) Liebl.) to changing light environments by parallel investigation of the meteorological conditions, photosynthetic pigment content, PSII quantum efficiency and excitation energy quenching. Sun and shade leaves got

Photosystem II proteins PsbL and PsbJ regulate electron flow to the plastoquinone pool.

The psbEFLJ operon of tobacco plastids encodes four bitopic low molecular mass transmembrane components of photosystem II. Here, we report the effect of inactivation of psbL on the directional forward electron flow of photosystem II as compared to that of the wild type and the psbJ deletion mutant, which is impaired in PSII electron flow to plastoquinone [Regel et al. (2001) J. Biol. Chem. 276, 41473-41478]. Exposure of Delta psbL plants to a saturating light pulse gives rise to the maximal fluorescence emission, Fm(L), which is followed within 4-6 s by a broader hitherto not observed second f

Light-induced changes in the chemical bond structure of light-harvesting complex II probed by FTIR spectroscopy.

Light-harvesting complex II (LHC-II) regulates the light energy distribution between photosystem I and II in plants. This process is mediated by phosphorylation of the LHC-II protein, which depends on the oxidation state of photosynthetic electron carriers. In addition to this regulatory mechanism, it has recently been proposed that light can directly induce a conformational change in isolated LHC-II. To provide biophysical evidence for such a conformational change in the protein, we studied infrared absorbance changes in isolated LHC-II upon exposure to light flashes. Compared to the signals

Chlorophyll fluorescence responses to CO2 availability reveal crassulacean acid metabolism in epiphytic orchids.

Crassulacean acid metabolism (CAM), a specialized mode of photosynthetic carbon assimilation characterized by nocturnal fixation of atmospheric CO2 and vacuolar malic acid storage, is found in a wide variety of vascular plant species, mainly those inhabiting water-limited environments. Identifying and characterizing diverse CAM species enhances our understanding of the physiological, ecological, and evolutionary significance of CAM photosynthesis. In this study, we examined the effect of CO2 elimination on chlorophyll fluorescence-based photosynthetic parameters in two constitutive CAM Kalanch

Modulation of Photosystem II Function in Celery via Foliar-Applied Salicylic Acid during Gradual Water Deficit Stress.

Water deficit is the major stress factor magnified by climate change that causes the most reductions in plant productivity. Knowledge of photosystem II (PSII) response mechanisms underlying crop vulnerability to drought is critical to better understanding the consequences of climate change on crop plants. Salicylic acid (SA) application under drought stress may stimulate PSII function, although the exact mechanism remains essentially unclear. To reveal the PSII response mechanism of celery plants sprayed with water (WA) or SA, we employed chlorophyll fluorescence imaging analysis at 48 h, 96 h

Herbicidal properties of antihypertensive drugs: calcium channel blockers.

Herbicide resistance is a worldwide problem in weed control. This prompts researchers to look for new modes of action to slow down the evolution of herbicide-resistant weeds. This research aims to determine the herbicidal action of thiazolo[3,2-a]pyrimidines derivatives, which are well known as antihypertensive drugs. The phytotoxic effects of ten compounds were investigated using leaf disc discoloration test and seed germination bioassay. At concentrations of 125 to 250 mg/L, the 5-(3-Fluoro-phenyl)-7-methyl-5H-thiazolo[3,2-a]pyrimidine-6-carboxylic acid ethyl ester (c) was highly active agai

Metallic nanoparticles influence the structure and function of the photosynthetic apparatus in plants.

The applications of nanoparticles continue to expand into areas as diverse as medicine, bioremediation, cosmetics, pharmacology and various industries, including agri-food production. The widespread use of nanoparticles has generated concerns given the impact these nanoparticles - mostly metal-based such as CuO, Ag, Au, CeO2, TiO2, ZnO, Co, and Pt - could be having on plants. Some of the most studied variables are plant growth, development, production of biomass, and ultimately oxidative stress and photosynthesis. A systematic appraisal of information about the impact of nanoparticles on these

Using the quantum yields of photosystem II and the rate of net photosynthesis to monitor high irradiance and temperature stress in chrysanthemum (Dendranthema grandiflora).

Under a dynamic greenhouse climate control regime, temperature is adjusted to optimise plant physiological responses to prevailing irradiance levels; thus, both temperature and irradiance are used by the plant to maximise the rate of photosynthesis, assuming other factors are not limiting. The control regime may be optimised by monitoring plant responses, and may be promptly adjusted when plant performance is affected by extreme microclimatic conditions, such as high irradiance or temperature. To determine the stress indicators of plants based on their physiological responses, net photosynthes

Involvement of molecular oxygen in the donor-side photoinhibition of Mn-depleted photosystem II membranes.

It has been shown by Khorobrykh et al. (Biochemistry (Moscow) 67:683-688, 2002); Yanykin et al. (Biochim Biophys Acta 1797:516-523, 2010); Khorobrykh et al. Experimental confirmation of this suggestion is presented in this work. The degree of photoinhibition was determined by the l

Supramolecular organization of photosystem II and its light-harvesting antenna in partially solubilized photosystem II membranes.

We present an extended analysis of the organization of green plant photosystem II and its associated light-harvesting antenna using electron microscopy and image analysis. The analysis is based on a large dataset of 16 600 projections of negatively stained PSII-LHCII supercomplexes and megacomplexes prepared by means of three different pretreatments. In addition to our previous work on this system [Boekema, E.J., van Roon, H., Calkoen, F., Bassi, R. and Dekker, J.P. (1999) Biochemistry 38, 2233-2239], the following results were obtained. Preferential damaging effects of limited magnesium bioavailability on photosystem I in Sulla carnosa plants.

Magnesium deficiency preferentially inhibits photosystem I rather than photosystem II in Sulla carnosa plants. The effects of magnesium (Mg(2+)) deficiency on growth, photosynthetic performance, pigment and polypeptide composition of chloroplast membranes were studied in the halophyte Sulla carnosa (Desf.), an annual legume endemic to Tunisia and Algeria. The results demonstrate a gradual decrease in biomass production with decreasing Mg(2+) availability in the growth medium. The increase of Mg(2+) deficiency was also associated with a decline of the net CO2 assimilation (Pn) in fully expanded
