Glycolytic oscillations in yeast have been studied for many years simply by adding a glucose pulse to a suspension of cells and measuring the resulting transient oscillations of NADH. Here we show, using a suspension of yeast cells, that living cells can be kept in a well defined oscillating state indefinitely when starved cells, glucose and cyanide are pumped into a cuvette with outflow of surplus liquid. Our results show that the transitions between stationary and oscillatory behaviour are uniquely described mathematically by the Hopf bifurcation. This result characterizes the dynamical prop

Genesis and differentiation of so-called interdigitating cells (IDC) of secondary lymphatic organs are reported by electron microscopic investigation of human lymph nodes in different cutaneous disorders. The formation of these cells apparently take place by differentiation of monocytic cells, in which a cyto-morphological transition into typical IDC--showing characteristic tubulo-vesicular structures--has been observed. Therefore, the IDC apparently represent a special structural and functional form of the monocytic-macrophagic system in lymph nodes and spleen. A hypertrophic reaction of thes

Patients with systemic lupus erythematosus (SLE) experience cognitive abnormalities in multiple domains including processing speed, executive function, and memory. Here we show that SLE patients carrying antibodies that bind DNA and the GluN2A and GluN2B subunits of the N-methyl-d-aspartate receptor (NMDAR), termed DNRAbs, displayed a selective impairment in spatial recall. Neural recordings in a mouse model of SLE, in which circulating DNRAbs penetrate the hippocampus, revealed that CA1 place cells exhibited a significant expansion in place field size. Structural analysis showed that hippocam

During movement, there is a transition of activity across the population, such that place-field centers ahead of the rat are sequentially activated in the order that they will be encountered. Although the mechanisms responsible for this sequence updating are unknown, two classes of models can be considered. The first class involves head-direction information for activating neurons in the order that their place fields will be traversed. An alternative model contends that motion and turn-related information from the posterior parietal cortex shift the subset of active hippocampal cells across th

The in vivo effects of a single oral dose (50 mg/kg) of oxamniquine on the ultrastructure of Schistosoma mansoni were investigated. In male worms, severe disruption of the tegument and gastrodermis took place, and extensive extracellular spaces developed between the cells of the internal tissues. Elimination of the damaged worms was associated with complete tegumental breakdown and encapsulation by host cells. A small proportion of females showed similar drug-induced changes and were also eliminated. In the residual females, no drug-induced morphological damage was observed even after a second

It was found that the accumulation of 125I-bilignost by isolated hepatocytes as target cells is mediated by the saturated and unsaturated systems. In thymocytes as non-target cells no saturated system of 125I-bilignost absorption was detected. 125I-triombrast transport in isolated hepatocytes and thymocytes takes place by usual diffusion.

In acute rheumatic fever, various types of myocardial degeneration and subsequent transformation of the damaged muscle fibers into a variety of cells, classified and unclassified, are described from microscopic examination of autopsy specimens. This study suggests that in the origin of Aschoff bodies, the diagnostic feature of rheumatic fever, there are three different pathways (Types A, B and C) for cytogenesis of Aschoff cells from altered muscle fibers. Type A cytogenesis takes place through the stage of Anitschkow myocytes of cardiac muscle origin (McDONALD 1963), and the Type B pathway, t

Just as hippocampal lesions are principally responsible for "temporal lobe" amnesia, lesions affecting the anterior thalamic nuclei seem principally responsible for a similar loss of memory, "diencephalic" amnesia. Compared with the former, the causes of diencephalic amnesia have remained elusive. A potential clue comes from how the two sites are interconnected, as within the hippocampal formation, only the subiculum has direct, reciprocal connections with the anterior thalamic nuclei. We found that both permanent and reversible anterior thalamic nuclei lesions in male rats cause a cessation o

In the classical view of hippocampal function, the subiculum is assigned the role of the output layer. In spatial paradigms, some subiculum neurons manifest as so-called boundary vector cells (BVCs), firing in response to boundaries at specific allocentric directions and distances. More recently, it has been shown that some subiculum BVCs can be classified as vector trace cells (VTCs), which exhibit traces of activity after a boundary/object has been removed. Here, we propose a model of processing within subiculum that accounts for VTCs, taking into account proximodistal differences in subicul

Wolbachia is an intracellular endosymbiont present in most arthropod and filarial nematode species. Transmission between hosts is primarily vertical, taking place exclusively through the female germ line, although horizontal transmission has also been documented. The results of several studies indicate that Wolbachia spp. can undergo transfer between somatic and germ line cells during nematode development and in adult flies. However, the mechanisms underlying horizontal cell-to-cell transfer remain largely unexplored. Here, we establish a tractable system for probing horizontal transfer of Wol

For regenerative medicine applications, transplanted cells are difficult to retain in a specific place to maximize the localized release of cytokines, which can positively modulate immune responses, angiogenesis and tissue regeneration. Here, we developed a hollow electrospun (ESP) scaffold with one open and one closed end to enclose a hydrogel laden with human mesenchymal stem cells (hMSCs); we termed this combination the hydrocup. hMSCs remained viable for 28 days in vitro and released functional cytokines similar to hMSCs in transplanted but non-enclosed hydrogels. The hydrocups remained in

A luminescent-histochemical method was used in investigation of the enterochromaffin cells (EC-cells) of the ampulla in the major duodenal papilla of 3 control dogs and 6 dogs under experimental fasting. In the ampulla wall a large number of EC-cells containing serotonin and catecholamine were detected. The degree and shining colour of these endocrine cells depend upon their interrelationship. During fasting changes of secretory activity and fluorescence index of EC-cells in major duodenal papilla of dogs take place.

Memory research remains focused on just a few brain structures-in particular, the hippocampal formation (the hippocampus and entorhinal cortex). Three key discoveries promote this continued focus: the striking demonstrations of enduring anterograde amnesia after bilateral hippocampal damage; the realization that synapses in the hippocampal formation are plastic e.g., when responding to short bursts of patterned stimulation ("long-term potentiation" or LTP); and the discovery of a panoply of spatially-tuned cells, principally surveyed in the hippocampal formation (place cells coding for positio

The work is dedicated to the study of biofilms formation process by main pyelonephritis causative agents in children in vitro using methods of light, fluorescent and scanning microscopy. To study biofilms formation bacteria were cultivated in liquid substratum on glass in polystyrene Petri dishes d=40mm. The study demonstrated that all isolates formed biofilms. Adhesion of bacteria planktonic forms took place on the first stage, intracellular matrix formation took place on the second stage, and biofilms formation took place on the third stage. During the study of E. coli and Proteus sрр bacter

Studying the immunological processes taking place during the initial steps of acute hepatitis C virus (HCV) infection has been a challenge in patients. Shin et al. have recently reported that delayed induction, not impaired recruitment of specific CD8(+) T cells, causes the late onset of acute hepatitis C in chimpanzees (Gastroenterology, 2011). However, further elucidation of the underlying mechanisms is difficult in vivo. We made observations consistent with their conclusions in human liver slices inoculated ex vivo with HCV produced in cell culture (HCVcc). Autologous immune cells were puri

Dendritic cells are thought to regulate tolerance induction vs immunization by transferring Ags and peripheral signals to draining lymph nodes (LN). However, whether myelin Ag transfer and presentation in LN occurs during demyelinating brain disease is unknown. In this study, we demonstrate redistribution of autoantigens from brain lesions to cervical LN in monkey experimental autoimmune encephalomyelitis (EAE) and in multiple sclerosis (MS). Immunohistochemical analysis revealed significantly more cells containing myelin Ags in cervical LN of monkeys with EAE compared with those of healthy co

The carotid bodies are a small pair of highly vascularized and well perfused organs located at each carotid artery bifurcation, strategically situated to sense oxygen in arterial blood as it leaves the heart. Carotid body glomus cells are identified as the primary oxygen sensors, which respond to changes in blood P(O(2)) within milliseconds. Acute hypoxia causes a rapid increase in carotid sinus nerve (CSN) activity, providing afferent signals to the respiratory center in the brainstem. Glomus cells secrete numerous neurotransmitters that modulate CSN firing rates. This review will discuss maj

Silence of FMR1 causes loss of fragile X mental retardation protein (FMRP) and dysregulated translation at synapses, resulting in the intellectual disability and autistic symptoms of fragile X syndrome (FXS). Synaptic dysfunction hypotheses for how intellectual disabilities like cognitive inflexibility arise in FXS predict impaired neural coding in the absence of FMRP. We tested the prediction by comparing hippocampus place cells in wild-type and FXS-model mice. Experience-driven CA1 synaptic function and synaptic plasticity changes are excessive in Fmr1-null mice, but CA1 place fields are nor

Damage involving the anterior thalamic and adjacent rostral thalamic nuclei may result in a severe anterograde amnesia, similar to the amnesia resulting from damage to the hippocampal formation. Little is known, however, about the information represented in these nuclei. To redress this deficit, we recorded units in three rostral thalamic nuclei in freely-moving rats [the parataenial nucleus (PT), the anteromedial nucleus (AM) and nucleus reuniens NRe]. We found units in these nuclei possessing previously unsuspected spatial properties. The various cell types show clear similarities to place c

The histology of intestinal urinary conduits (8 ileal and 2 sigmoid) in place for 10 months to 10 years was studied in 10 patients between 20 and 65 years old. It is concluded that the ileum and the large intestine do not undergo adaptive metaplastic transformation. The only changes observed are related to a condition of chronic irritation to which the response is a chronic inflammatory reaction. Structures proving to be the most resistant were the cells of Paneth. The latter were not damaged in appearance and number as compared to normal, even when the conduit was in place for 10 years.
