Non-parasite genome encoded virus-like RNAs reprogram the pathogenicity of human blood flukes.

Non-parasite genome encoded RNAs (ngRNAs) are often overlooked in their roles, especially their functions in shaping the biology of organisms. Here we show four non-parasite genome encoded virus-like RNAs that present in Schistosoma japonicum, a human blood fluke responsible for significant morbidity and mortality. These ngRNAs are predominantly localized in the germ cells that play crucial roles in oogenesis and egg production, pivotal processes contributing to host pathology. The ngRNA encode RNA-dependent RNA polymerase (RdRp) that synthesize complementary RNA strands, governing embryo viab

Staphylococcus aureus-Derived α-Hemolysin Evokes Generation of Specialized Pro-resolving Mediators Promoting Inflammation Resolution.

Underlying mechanisms of how infectious inflammation is resolved by the host are incompletely understood. One hallmark of inflammation resolution is the activation of specialized pro-resolving mediators (SPMs) that enhance bacterial clearance and promote tissue repair. Here, we reveal α-hemolysin (Hla) from Staphylococcus aureus as a potent elicitor of SPM biosynthesis in human M2-like macrophages and in the mouse peritoneum through selective activation of host 15-lipoxygenase-1 (15-LOX-1). S. aureus-induced SPM formation in M2 is abolished upon Hla depletion or 15-LOX-1 knockdown. Isolated Hl

A review of the effects of bromate on aquatic organisms and toxicity of bromate to oyster (Crassostrea gigas) embryos.

A critical literature review has been undertaken of the effects that bromate (BrO3-) has on aquatic organisms. Chronic (multigeneration) studies using four marine phytoplankton species indicate a stimulation of cell division at concentrations up to 13.6 mg BrO3- liter-1, such that the EC50 is > 13.6 mg BrO3- liter-1 (exposure period unspecified). No published data were available for any freshwater algal or phytoplankton species. For freshwater invertebrates, the available data are limited to acute studies using two species: the water flea, Daphnia magna, with a 48-h LC50 of 179 mg BrO3- liter-

Independent and parallel lateral transfer of DNA transposons in tetrapod genomes.

In animals, the mode of transmission of transposable elements is generally vertical. However, recent studies have suggested that lateral transfer has occurred repeatedly in several distantly related tetrapod lineages, including mammals. Using transposons extracted from the genome of the lizard Anolis carolinensis as probes, we identified four novel families of hAT transposons that share extremely high similarity with elements in other genomes including several mammalian lineages (primates, chiropters, marsupials), one amphibian and one flatworm, the planarian Schmidtea mediterranea. The discon

A Schmidtea nova (Benazzi, 1982)-like Population in SW Romania (Platyhelminthes, Tricladida, Dugesiidae), a Morphological Characterization.

This study has begun as exploratory research on freshwater planarians from southwestern Romania for faunal purposes. Specimens were sampled and processed for standard histology (paraffin wax embedding, horizontal and sagittal serial sections, and Haematoxylin-Eosin staining). The histological analysis provided evidence for a population belonging to the genus Schmidtea. The morphological and histological characteristics were compared with those of all other Schmidtea species. The histological slides reveal numerous diagnostic characters for Schmidtea nova: the presence of two nipples on the cou

Volume X-Ray Micro-Computed Tomography Analysis of the Early Cephalized Central Nervous System in a Marine Flatworm, Stylochoplana pusilla.

Platyhelminthes are a phylum of simple bilaterian invertebrates with prototypic body systems. Compared with non-bilaterians such as cnidarians, the bilaterians are likely to exhibit integrated free-moving behaviors, which require a concentrated nervous system "brain" rather than the distributed nervous system of radiatans. Marine flatworms have an early cephalized 'central' nervous system compared not only with non-bilaterians but also with parasitic flatworms or freshwater planarians. In this study, we used the marine flatworm Stylochoplana pusilla as an excellent model organism in Platyhelmi

Single-cell analysis of Schistosoma mansoni identifies a conserved genetic program controlling germline stem cell fate.

Schistosomes are parasitic flatworms causing one of the most prevalent infectious diseases from which millions of people are currently suffering. These parasites have high fecundity and their eggs are both the transmissible agents and the cause of the infection-associated pathology. Given its biomedical significance, the schistosome germline has been a research focus for more than a century. Nonetheless, molecular mechanisms that regulate its development are only now being understood. In particular, it is unknown what balances the fate of germline stem cells (GSCs) in producing daughter stem c

Disrupting quorum sensing alters social interactions in Chromobacterium violaceum.

Quorum sensing (QS) is a communication system used by bacteria to coordinate a wide panel of biological functions in a cell density-dependent manner. The Gram-negative Chromobacterium violaceum has previously been shown to use an acyl-homoserine lactone (AHL)-based QS to regulate various behaviors, including the production of proteases, hydrogen cyanide, or antimicrobial compounds such as violacein. By using combined metabolomic and proteomic approaches, we demonstrated that QS modulates the production of antimicrobial and toxic compounds in C. violaceum ATCC 12472. We provided the first evide

Identification of a Novel Allosteric Modulator of the Human Dopamine Transporter.

The dopamine transporter (DAT) serves a pivotal role in controlling dopamine (DA)-mediated neurotransmission by clearing DA from synaptic and perisynaptic spaces and controlling its action at postsynaptic DA receptors. Major drugs of abuse such as amphetamine and cocaine interact with DAT to mediate their effects by enhancing extracellular DA concentrations. We previously identified a novel allosteric site in the related human serotonin transporter that lies outside the central substrate and inhibitor binding pocket. We used the hybrid structure based (HSB) method to screen for allosteric modu

Schistosome sex matters: a deep view into gonad-specific and pairing-dependent transcriptomes reveals a complex gender interplay.

As a key event for maintaining life cycles, reproduction is a central part of platyhelminth biology. In case of parasitic platyhelminths, reproductive processes can also contribute to pathology. One representative example is the trematode Schistosoma, which causes schistosomiasis, an infectious disease, whose pathology is associated with egg production. Among the outstanding features of schistosomes is their dioecious lifestyle and the pairing-dependent differentiation of the female gonads which finally leads to egg synthesis. To analyze the reproductive biology of Schistosoma mansoni in-depth

Marr's ghost.

David Marr's tri-level analysis has organized cognitive neuroscience for four decades. We argue that it has also constrained it. The framework rests on three assumptions: that cognitive goals can be specified a priori, that neural processes implement algorithms, and that physical substrates are interchangeable realizers of abstract functions. Each assumption was defensible in 1982. Each is now demonstrably false. Slime molds, planarian flatworms, bioelectric networks in non-neural tissue, and degenerate neural circuits pose a dilemma the framework cannot resolve: either extend computational de

Divergent Axin and GSK-3 paralogs in the beta-catenin destruction complexes of tapeworms.

The Wnt/beta-catenin pathway has many key roles in the development of animals, including a conserved and central role in the specification of the primary (antero-posterior) body axis. The posterior expression of Wnt ligands and the anterior expression of secreted Wnt inhibitors are known to be conserved during the larval metamorphosis of tapeworms. However, their downstream signaling components for Wnt/beta-catenin signaling have not been characterized. In this work, we have studied the core components of the beta-catenin destruction complex of the human pathogen Echinococcus multilocularis, t

Zebrafish Ciliopathy Screen Plus Human Mutational Analysis Identifies C21orf59 and CCDC65 Defects as Causing Primary Ciliary Dyskinesia.

Primary ciliary dyskinesia (PCD) is caused when defects of motile cilia lead to chronic airway infections, male infertility, and situs abnormalities. Multiple causative PCD mutations account for only 65% of cases, suggesting that many genes essential for cilia function remain to be discovered. By using zebrafish morpholino knockdown of PCD candidate genes as an in vivo screening platform, we identified c21orf59, ccdc65, and c15orf26 as critical for cilia motility. c21orf59 and c15orf26 knockdown in zebrafish and planaria blocked outer dynein arm assembly, and ccdc65 knockdown altered cilia bea

The first Dugesia species (Platyhelminthes, Tricladida, Dugesiidae) documented for Saudi Arabia: an integrative description.

Exploration of various freshwater bodies in the Kingdom of Saudi Arabia led to the discovery of a new species of Dugesia, thus representing the first documented record of freshwater planarians in this country and the second fully documented record of a naturally sexual population of a Dugesia species in the Arabian Peninsula. Based on morphological, karyological, anatomical, and molecular data, this Dugesia population from Saudi Arabia is here identified as representing a distinct, new species, Dugesia bursagrossa Harrath & Sluys, sp. nov. The species is characterized by the following anatomic

A protocol for sequential isolation, purification, and collection of Luticola nematophagus eggs to obtain high-quality genomic DNA.

Conventional strategies for obtaining planarian tissue for genomic DNA extraction rely on diet-based rearing or amputation-induced regeneration. These approaches are inapplicable to soil-dwelling flatworms that predominantly reproduce by egg-laying and lack regenerative capacity, such as the recently discovered nematode predator Luticola nematophagus. Here, we developed a protocol for the sequential isolation, purification, and collection of L. nematophagus eggs, comprising three steps: (1) physical sieving through sintered meshes of graded pore sizes under recirculating water flow to exclude

Proposal of a new model with dopaminergic-cholinergic interactions for neuropharmacological investigations.

The motor system of Dugesia gonocephala shows a striking similarity with the extrapyramidal system of high vertebrates and of man with the evidence of correlations between dopaminergic and cholinergic neurons. The utilization of this model seems to be useful in testing drugs which presumably act on dopaminergic or cholinergic transmission. In this model, the quantification of animal behaviour seems considerably easier when compared with the difficulties met in other animal models commonly employed. Besides, it might be anticipated that this model, if correctly used, can display interesting per

A Stable Thoracic Hox Code and Epimorphosis Characterize Posterior Regeneration in Capitella teleta.

Regeneration, the ability to replace lost tissues and body parts following traumatic injury, occurs widely throughout the animal tree of life. Regeneration occurs either by remodeling of pre-existing tissues, through addition of new cells by cell division, or a combination of both. We describe a staging system for posterior regeneration in the annelid, Capitella teleta, and use the C. teleta Hox gene code as markers of regional identity for regenerating tissue along the anterior-posterior axis. Following amputation of different posterior regions of the animal, a blastema forms and by two days,

Identification and Complete Stereochemical Assignments of the New Resolvin Conjugates in Tissue Regeneration in Human Tissues that Stimulate Proresolving Phagocyte Functions and Tissue Regeneration.

Resolvin conjugates in tissue regeneration (RCTRs) are new chemical signals that accelerate resolution of inflammation, infection, and tissue regeneration. Herein, using liquid chromatography-tandem mass spectrometry-based metabololipidomics, we identified RCTRs in human spleen, lymph node, bone marrow, and brain. In human spleen incubated with Staphylococcus aureus, endogenous RCTRs were increased along with conversion of deuterium-labeled docosahexaenoic acid, conferring pathway activation. Physical and biological properties of endogenous RCTRs were matched with those prepared by total organ

Fraction of sex-inducing substances facilitates growth and body shape change in a Fasciola hepatica/gigantica hybrid: A novel in vitro research platform for studying liver-stage juveniles derived from mice.

The absence of an established in vitro platform is a major obstacle in research on parasitic flatworms, including Fasciola flukes. Fasciola flukes cause zoonotic infections that primarily affect the liver and the bile ducts. Infected juveniles can cause severe liver damage in animals, occasionally leading to sudden death. Although resistance to the only drug for the acute liver stage has been reported worldwide, the search for new drugs has been unsuccessful owing to the critical limitations of previous in vitro cultures. Previous studies have been unable to reproduce liver-stage development i

Systematic review of innate immune responses against Mycobacterium tuberculosis complex infection in animal models.

BACKGROUND: Mycobacterium tuberculosis (Mtb) complex (MTBC) includes ten species that affect mammals and pose a significant global health concern. Upon infection, Mtb induces various stages in the host, including early bacterial elimination, which may or may not involve memory responses. Deciphering the role of innate immune responses during MTBC infection is crucial for understanding disease progression or protection. Over the past decade, there has been growing interest in the innate immune response to Mtb, with new preclinical models emerging. METHODS: We conducted a systematic review foll
