Bioactive cytokines such as vascular endothelial growth factor (VEGF) and Activin A are critical for the differentiation of stem cells into vascular endothelial cells and cardiomyocytes. However, production of the cytokines using conventional Escherichia coli or mammalian cell expression systems carries risks of immunogenicity and viral contamination. In this study, we developed a VEGF and Activin A plant expression system and demonstrated that plant-expressed VEGF and Activin A are as active as their commercial counterparts. We also showed that plant-expressed VEGF and Activin A are as effici

The extracellular accumulation of aggregated amyloid-β (Aβ) in the brain leads to the early pathology of Alzheimer's disease (AD). The administration of exogenous plant-type ceramides into AD model mice can promote the release of neuronal exosomes, a subtype of extracellular vesicles, that can mediate Aβ clearance. In vitro studies showed that the length of fatty acids in mammalian-type ceramides is crucial for promoting neuronal exosome release. Therefore, investigating the structures of plant ceramides is important for evaluating the potential in releasing exosomes to remove Aβ. In this stud

Vismione H (VH) is a fluorescent prenylated anthranoid produced by plants from the Hypericaceae family, with antiprotozoal activities against malaria and leishmaniosis. Little is known about its biosynthesis and metabolism in plants or its mode of action against parasites. When VH is isolated from Psorospermum glaberrimum, it is rapidly converted into madagascine anthrone and anthraquinone, which are characterized by markedly different fluorescent properties. To locate the fluorescence of VH in living plant cells and discriminate it from that of the other metabolites, an original strategy comb

Stenodactylin, a highly toxic type 2 ribosome-inactivating protein purified from the caudex of Adenia stenodactyla Harms, is a potential anticancer drug candidate. Previous studies demonstrated that stenodactylin induces apoptosis and necroptosis in treated cells, involving the production of reactive oxygen species. We analyzed the effect of stenodactylin on Raji and Ramos (Human Burkitt's lymphoma cells) and MOLM-13 (acute myeloid leukemia cells). Moreover, we focused on the early events in MOLM-13 cells that characterize the cellular response to the toxin by whole-genome microarray analysis

Non-Hodgkin's B cell lymphomas (NHL) include a diverse set of neoplasms that constitute ∼90% of all lymphomas and the largest subset of blood cancers. While chemotherapy is the first line of treatment, the efficacy of contemporary chemotherapies is hampered by dose-limiting toxicities. Partly due to suboptimal dosing, ∼40% of patients exhibit relapsed or refractory disease. Therefore more efficacious drug delivery systems are urgently needed to improve survival of NHL patients. In this study we demonstrate a new drug delivery platform for NHL based on the plant virus Potato virus X (PVX). We o

Among plant-parasitic nematodes, the root-knot nematodes (RKNs) of the Meloidogyne spp. are the most economically important genus. RKN are root parasitic worms able to infect nearly all crop species and have a wide geographic distribution. During infection, RKNs establish and maintain an intimate relationship with the host plant. This includes the creation of a specialized nutritional structure composed of multinucleate and hypertrophied giant cells, which result from the redifferentiation of vascular root cells. Giant cells constitute the sole source of nutrients for the nematode and are esse

The cellular and intracellular localization of the non-proteogenic amino acid nicotianamine (NA) in leaves and root elongation zones was immunochemically investigated in pea (Pisum sativum L.) and tomato (Lycopersicon esculentum Mill.) plants grown under various iron regimes and in three mutants defective in the regulation of iron uptake. Strongest immunostaining was observed in the over-accumulating pea mutants brz and dgl, and in iron-loaded wild-type plants. Fe concentration and NA level paralleled staining intensity, indicating that NA synthesis is induced by high iron availability. While

Agrobacterium pathogenesis, which involves transferring T-DNA into plant cells, is the cornerstone of plant genetic engineering. As the applications that rely on Agrobacterium increase in sophistication, it becomes critical to achieve a quantitative and predictive understanding of T-DNA expression at the level of single plant cells. Here we examine if a classic Poisson model of interactions between pathogens and host cells holds true for Agrobacterium infecting Nicotiana benthamiana. Systematically challenging this model revealed antagonistic and synergistic density-dependent interactions betw

This study evaluated the antimicrobial and antibiofilm activities of ciprofloxacin (Cip) in combination with trans-cinnamaldehyde (Tc), geraniol (Ger), and eugenol (Eug) against multidrug-resistant Escherichia coli, Proteus mirabilis, and Klebsiella pneumoniae isolated from broiler feces. Disk diffusion and microdilution assays showed that Tc displayed the highest antibacterial activity in disk diffusion (IZD: 30 mm) and microdilution assays (MIC: 312.5 μg/mL), while Ger and Eug revealed smaller inhibition zones (∼9-12 mm) and higher MIC values (2500 μg/mL). Checkerboard assays indicated that

Colorectal cancer (CRC) is a complex multifactorial disease caused by genetic and epigenetic changes playing a vital role in its development and progression. Chemotherapy remains a major option in the treatment of CRC. However, due to its unintended effects on normal tissue, research on identifying plant-based therapeutic agents as an alternative treatment modality has gained attention. Fisetin, a plant-derived flavonoid, has shown promising effects as an anticancer agent against several human cancers, including colon cancer. However, there is limited research focusing on studying the mechanis

Oxidative stress has been linked to prostate carcinogenesis as human prostate tissue is vulnerable to oxidative DNA damage. Apigenin, a dietary plant flavone, possesses anti-proliferative and anticancer effects; however, its antioxidant properties have not been fully elucidated. We investigated sub-cellular distribution of apigenin, it's binding to DNA and protective effects against H2O2-induced DNA damage using transformed human prostate epithelial RWPE-1 cells and prostate cancer LNCaP, PC-3 and DU145 cells. Exposure of cells to apigenin exhibited higher accumulation in RWPE-1 and LNCaP cell

Alternative oxidase (AOX) plays a pivotal role in cyanide-resistance respiration in the mitochondria of plants, fungi and some protists. Here we show that AOX from thermogenic skunk cabbage successfully conferred cyanide resistance to human cells. In galactose medium, HeLa cells with mitochondria-targeted AOX proteins were found to have significantly less reactive oxygen species production in response to antimycin-A exposure, a specific inhibitor of respiratory complex III. These results suggest that skunk cabbage AOX can be used to create an alternative respiration pathway, which might be imp

Thirty broilers 8-10 weeks old were used to study the rate of food passage in chickens. Wheat bran and rice husks of three different sizes: more than 2 mm, between 1 and 1.5 mm, and less than 0.5 mm, mordanted with chromium, were used as markers. The suitability of these markers to study the rate of food passage in chickens and the possible influence of the size and hardness of the particle on the retention time was the objective of this study. Both T1, the time of first appearance of the marker, and Tm, the mean retention time, have been evaluated. Tm was a better parameter than T1 for studyi

Penicillium simplicissimum GP17-2 is a plant growth-promoting fungus (PGPF) and an inducer of systemic defense responses. The mechanisms underlying the effect of GP17-2 on the reduction of cucumber leaf damage caused by the anthracnose pathogen Colletotrichum orbiculare were investigated. Cucumber leaves treated with the culture filtrate (CF) of GP17-2 exhibited a clear systemic resistance against subsequent infection with C. orbiculare. The number and size of lesions caused by the disease were reduced in CF-treated plants, in comparison with that in the control plants. The results showed that

Intracellular delivery into walled cells is essential for advancing research in plant science, microbiology, and synthetic biology. Conventional enzymatic wall degradation methods are inherently destructive, leading to poor cell survival. Micro/nanoinjection technology, which enables the physical penetration of the cell wall for direct substance introduction, has emerged as a critical solution. This review article presents a systematic overview of this technology. First, we summarize the state-of-the-art in probe design and fabrication. Next, we analyze automation and force-feedback strategies

Higher plants are divided into three major photosynthetic groups known as C3, C4, and crassulacean acid metabolism (CAM) plants. It is considered that cell wall thickness (TCW) affects diffusion and leakiness of CO2 within leaves, but it is unclear whether TCW of photosynthetic cells differs among these groups. This study investigated TCW of photosynthetic cells in herbaceous C3, C4, and CAM species under an electron microscope. Among 75 species of monocots and eudicots grown in a growth chamber in the same environment, the TCW of mesophyll cells (MCs) was much higher in CAM species than in C3

In plants, the response to stress, such as salinity, pathogen attack, drought, high concentration of metals, hyperthermia, and hypothermia, is usually accompanied by potassium ion (K+) leakage from the cytosol to the cell wall, mediated by plasma membrane cation conductivity. Stress-induced electrolyte leakage co-occurs with accumulation of reactive oxygen species (ROS) and calcium ions (Ca2+) and often results in programmed cell death (PCD). The development of X-ray and mass spectrometry (MS) based imaging techniques has enabled insight into the spatial tissue and cell-specific redistribution

Pea leaf epidermis incubated with cyanide displayed ultrastructural changes in guard cells that are typical of apoptosis. Cycloheximide, an inhibitor of cytoplasmic protein synthesis, and lincomycin, an inhibitor of protein synthesis in chloroplasts and mitochondria, produced different effects on the dynamics of programmed death of guard cells. According to light microscopy data, cycloheximide reinforced and lincomycin suppressed the CN(-)-induced destruction of cell nuclei. Lincomycin lowered the effect of cycloheximide in the light and prevented it in the dark. According to electron microsco

Green nanotechnology offers a sustainable approach to improving the therapeutic efficacy of plant‑derived bioactive compounds while reducing environmental and biological toxicity. In this study, silver quantum dots (Ag QDs) were synthesized via an eco‑friendly green route using the aqueous bulb extract of Drimia maritima (L.) Stearn (D. maritima), which served as a natural reducing and stabilizing agent. Physicochemical characterization confirmed the successful formation of well‑defined nanoscale Ag QDs with appropriate structural features. The antioxidant potential of the biosynthesized Ag QD
