Plants in the field are exposed to varying light and moisture. Agronomic improvement requires knowledge of whole-plant phenotypes expressed in response to simultaneous variation in these essential resources. Most phenotypes, however, have been described from experiments where resources are varied singularly. To test the importance of varying shoot and root resources for phenotyping studies, sister pre-breeding lines of wheat were phenotyped in response to independent or simultaneous exposure to two light levels and soil moisture profiles. The distribution and architecture of the root systems d

Iron is an essential microelement for plant growth. After uptake from the soil, iron is chelated by ligands and translocated from roots to shoots for subsequent utilization. However, the number of ligands involved in iron chelation is unclear. In this study, we identified and demonstrated that GLU1, which encodes a ferredoxin-dependent glutamate synthase, was involved in iron homeostasis. First, the expression of GLU1 was strongly induced by iron deficiency condition. Second, lesion of GLU1 results in reduced transcription of many iron-deficiency-responsive genes in roots and shoots. The mutan

The aim of this study was to evaluate the effect of severe environmental conditions prevailing on metalliferous waste heaps and heavy metal-contaminated growth substrates on accumulation of selected secondary metabolites, antioxidant capacity, and heavy metal concentration in two metallicolous (MC, MZ) and one nonmetallicolous (NM) populations of Echium vulgare L. The shoots and the roots of the three studied populations were collected from their natural habitats. Additionally, the plants were cultivated on different growth substrates, i.e. a contaminated substrate obtained from the areas of g

Even though the potential toxicity and treatment methods for per- and polyfluoroalkyl substances (PFAS) have attracted extensive attention, the plant uptake and accumulation of PFAS in edible plant tissues as a potential pathway for human exposure received little attention. Our study in a hydroponic system demonstrated that perfluorooctanoic acid (PFOA) and its replacing compound, 2,3,3,3-tetrafluoro-2-(heptafluoropropoxy) propanoic acid (GenX) displayed markedly different patterns of plant uptake and accumulation. For example, the root concentration factor (RCF) of PFOA in lettuce is almost f

BACKGROUND: Switchgrass (Panicum virgatum L.) is an important bioenergy and forage crop. The outcrossing nature of switchgrass makes it infeasible to maintain a genotype through sexual propagation. Current asexual propagation protocols in switchgrass have various limitations. An easy and highly-efficient vegetative propagation method is needed to propagate large natural collections of switchgrass genotypes for genome-wide association studies (GWAS).
RESULTS: Micropropagation by node culture was found to be a rapid method for vegetative propagation of switchgrass. Bacterial and fungal contamin

Polybrominated diphenyl ethers (PBDEs) are potentially harmful and persistent environmental pollutants. Despite evidence that soils are a major sink for PBDEs, little is known regarding their behavior in this medium. An environmentally relevant level of a commercial penta-BDE mixture (75 microg kg(-1)) was added to topsoil, and the extractability of three congeners (BDE-47, -99, and -100) was monitored over 10 weeks in planted and unplanted treatments. The extractability of each congener decreased rapidly in the experimental soil due largely to abiotic sorption to soil particles, which was dem

Effects of 10 microM cadmium (supplied as Cd nitrate) on the utilization and allocation of iron (Fe) were investigated in poplar (Populus alba L.) plants grown in nutrient solution with Fe(III)-EDTA or Fe(III)-citrate as the Fe source. The effects of Cd were also compared with those of Fe deprivation. The accumulation of Fe in roots was 10-fold higher in plants grown with Fe-citrate than with Fe-EDTA. Cadmium decreased leaf chlorophyll concentrations and photosynthetic rates, and these decreases were more marked in plants grown with Fe-citrate than with Fe-EDTA. In both Fe treatments, addition

Plant genome editing is achieved by the expression of sequence-specific nucleases (SSNs). RNA virus vector-mediated expression of SSNs is a promising approach for transgene integration-free targeted mutagenesis in plants. However, the removal of virus vectors from infected plants is challenging because no antiviral drugs are available against plant viruses. Here, we developed a removable RNA virus vector that carries the target site of tobacco microRNA398 (miR398) whose expression is induced during shoot regeneration. In the inoculated leaves in which expression of miR398 is not induced, inser

In higher plants, gravitropism proceeds through three sequential steps in the responding organs: perception of gravity signals, signal transduction and asymmetric cell elongation. Light and temperature also influence the gravitropic orientation of plant organs. A series of Arabidopsis shoot gravitropism (sgr) mutants has been shown to exhibit disturbed shoot gravitropism. SGR5 is functionally distinct from other SGR members in that it mediates the early events of gravitropic responses in inflorescence stems. Here, we demonstrated that SGR5 alternative splicing produces two protein variants (SG

Pollution caused by residual antibiotics is a worldwide environmental issue. Antibiotic residues often occur in aquatic ecosystems, posing threats to the health of aquatic organisms. The effects of antibiotic residues on the growth of crop plants and on human health are reasonably well known. However, less is known about antibiotic effects on wetland plants. Therefore, we studied the response and tolerance of ten clonal wetland plants grown in soil spiked with sulfadiazine at 10 mg kg(-1) (an environmentally relevant concentration) and 100 mg kg(-1). At 10 mg kg(-1), ramet number was the least

Plants can be regenerated from various explants/tissues via de novo shoot meristem formation. Most of these regeneration pathways are indirect and involve callus formation. Besides plant hormones, the role of polyamines (PAs) has been implicated in these processes. Interestingly, the lateral root primordia (LRPs) of Arabidopsis can be directly converted to shoot meristems by exogenous cytokinin application. In this system, no callus formation takes place. We report that the level of PAs, especially that of spermidine (Spd), increased during meristem conversion and the application of exogenous

When and how much plants grow under environmental constraints are fundamental questions in biology and increasingly important for predicting biomass production and carbon sequestration under climate change. While temperature and water availability directly regulate plant growth, the timing and rate of growth are also shaped by internal developmental programming, though this is rarely considered in predictions of tree growth responses to future climates. Here, we revisit a concept central to this internal programming-(in)determinacy. Focusing on woody plants, we define it as the extent to which

Lead (Pb) is toxic to the development and growth of rice plants. Nanoparticles (NPs) have been considered one of the efficient remediation techniques to mitigate Pb stress in plants. Therefore, a study was carried out to examine the underlying mechanism of iron (Fe) and silicon (Si) nanoparticle-induced Pb toxicity alleviation in rice seedlings. Si-NPs (2.5 mM) and Fe-NPs (25 mg L-1) were applied alone and in combination to rice plants grown without (control; no Pb stress) and with (100 µM) Pb concentration. Our results revealed that Pb toxicity severely affected all rice growth-related traits

Shoot apical meristems (SAMs) harbor persistent stem cells and give rise to above-ground organs throughout life. In tissue culture-based shoot regeneration, a subpopulation of pluripotent callus cells is specified into SAMs. How callus cells decide whether or not to become SAMs stands as an important question in developmental biology. Furthermore, the developmental basis underlying the de novo construction of dome-shaped SAMs remained largely unknown. Recent high-resolution analyses have revealed the spatiotemporal dynamics of cell fate determination and meristem establishment during shoot reg

The wild nature monument "184-year-old oak tree in the village of Irinovka" (Leningrad oblast, Russia), better known in local historical literature as "Irinovsky Oak," was officially opened in 2013. It is represented by a separate pedunculate oak tree (Quercus robur), planted in 1829 and preserved in satisfactory condition at 194 years of age. This paper presents data from a survey of the shoot system of the tree investigated. A total of 12 fungal species were registered (Cladosporium herbarum, Colpoma quercinum, Coryneum depressum, Diatrypella quercina, Erysiphe alphitoides, Hyphoderma setige

Root-applied humic acids (HAs) promote plant development, yet the early signaling mechanisms underlying their biostimulant action remain incompletely understood. Here, we show that treatment of Arabidopsis thaliana roots with a sedimentary humic acid (SHA) triggers a transient eustress-like response, characterized by early inhibition of shoot and root growth and increased H2O2 accumulation in root hairs and apical roots. This initial oxidative phase was followed by enhanced shoot and root growth, supporting the view that SHA acts as an adaptive plant modulator through a controlled stress-relat

Aldrovanda vesiculosa is a rare and critically endangered carnivorous plant species. Its populations have declined worldwide, so there is a need to protect the species from extinction. The research was conducted to establish an effective method of in vitro propagation of the species in order to obtain plants for reintroduction in the wild. The procedures included disinfection, multiplication, and acclimatization of plants. Contamination-free in vitro cultures were established using shoots and turions, which were disinfected with 0.25% sodium hypochlorite. The shoots were first defoliated. The

A soil-column experiment was conducted to compare biochar (BC) and nanobiochar (NB) at 2.5, 5, and 10% (w/w) to investigate their potential for stabilizing cadmium (Cd) and lead (Pb) under bare-soil and maize-planted conditions, enabling separation of amendment effects from rhizosphere-mediated influences. Soil properties, metal leaching behavior, microbial biomass nitrogen, plant growth, tissue metal accumulation, and phytostabilization indices were assessed. In contaminated unamended soil, soil organic matter, cation exchange capacity, and microbial biomass nitrogen declined, with substantia
