Low wind, high impact: sustained low airflow alters water use efficiency and shapes plant acclimation.

Minor differences in airflow are often assumed to have negligible physiological effects, yet they directly modify heat and gas exchange by altering the leaf boundary layer. We investigated how sustained low airflow impacts mechanisms regulating leaf-microclimate exchange, and how these responses shape acclimation in Lactuca sativa and Solanum lycopersicum. Lettuce was grown under two low-velocity airflow regimes (0.22 m s-1 and 0.65 m s-1), and changes in the leaf surface microclimate, plant transpiration, stomatal regulation, leaf photosynthesis, and growth were quantified. Increased airflow

Leaf gas exchange and photosynthetic light-response curves of cassava genotypes under drought stress during the early growth stage and after recovery.

This study aimed to gain insights into the photosynthesis capacity and genotypic differences in response to drought imposed at an early growth stage of cassava. Leaf water potential (LWP), leaf gas exchange, and photosynthetic light-response curves were investigated in six field-grown cassava genotypes under full irrigation and drought imposed for 60 d during 3-5 months after planting. During the drought period, mean LWP at predawn (LWPpre) was significantly lower than that in the control plants, while the mean midday LWP (LWPmid) was similar. During the drought period, the mean stomatal condu

Drought-induced delays in stem hydraulic development shape gas exchange and growth recovery in Douglas fir.

The limiting factors of tree recovery from drought, particularly the coordination between carbon sources and sinks, remain poorly understood. In this study, juvenile Douglas fir (Pseudotsuga menziesii) were exposed to 28 d of mild or severe drought, followed by 35 d of recovery. We continuously monitored CO₂ and H₂O fluxes in shoots and roots to derive gas exchange and carbon accumulation, while measuring basal area to estimate stem growth and sapwood development. To identify underlying mechanisms of drought recovery, we periodically measured nonstructural carbohydrates (NSC), midday water pot

Advanced phenotyping features utilizing deep learning techniques for automated analysis of stomatal guard cell orientation.

Stomata are vital for controlling gas exchange and water vapor release, which significantly affect photosynthesis and transpiration. Characterizing stomatal traits such as size, density, and distribution is essential for adaptation to the environment. While microscopy is widely used for this purpose, manual analysis is labor-intensive and time-consuming that limit large scale studies. To overcome this, we introduce an automated, high-throughput method that leverages YOLOv8, an advanced deep learning model, for more accurate and efficient stomatal trait measurement. Our approach provides a comp

Loss of Stomatal Regulation Sensitivity to CO2 and Reduced Xylem Hydraulic Conductivity Contribute to Long-Term Tree Decline and Mortality.

Increasing aridity is a major threat to forests worldwide. Understanding tree functional constraints under drought and their impacts on resilience and mortality among species is crucial to assess the impacts of global change on forests. We analyzed the long-term drought and atmospheric CO2 responses in three Mediterranean co-occurring species with differing drought tolerances (Pinus pinaster < Pinus pinea < Juniperus oxycedrus). In this mixed forest, P. pinaster exhibited widespread mortality and mistletoe infection, P. pinea showed scattered mortality, and J. oxycedrus showed no decline. Usin

Can Oxygen Isotopes in Tree Rings Be Used to Detect Stomatal Responses to Global Change?

Stomatal conductance (gs) regulates CO2 and water fluxes of plants. Although experiments have shown that gs decreases with elevated CO2, it is unclear how gs is responding in situ to long-term exposures to rising CO2 and a changing climate. Tree ring isotope analysis provides a unique method to assess tree ecophysiological responses to long-term exposures of slowly changing environmental conditions. In particular, it has been suggested that changes in gs can potentially be inferred from tree ring stable oxygen isotope ratios (δ18Otrc). Several studies have indeed used δ18Otrc trends to conclud

Light prevents pathogen-induced aqueous microenvironments via potentiation of salicylic acid signaling.

Many plant pathogens induce water-soaked lesions in infected tissues. In the case of Pseudomonas syringae (Pst), water-soaking effectors stimulate abscisic acid (ABA) production and signaling, resulting in stomatal closure. This reduces transpiration, increases water accumulation, and induces an apoplastic microenvironment favorable for bacterial growth. Stomata are sensitive to environmental conditions, including light. Here, we show that a period of darkness is required for water-soaking, and that a constant light regime abrogates stomatal closure by Pst. We find that constant light induces

ALMT12 interacts with and inhibits SLAC1 to modulate stomatal movements and enhance plant biomass.

Stomata are pores that control carbon dioxide (CO2) and water exchange by modulating their aperture in response to different environmental and internal signals. The Slow-type SLOW ANION CHANNEL-ASSOCIATED 1 (SLAC1) and Rapid-type ALUMINUM-ACTIVATED MALATE TRANSPORTER 12 (ALMT12) anion channels mediate anion efflux in guard cells to promote stomatal closure. These channels were previously thought to function as 2 independent anion-permeable pores differing in their activation kinetics, voltage dependence, and anion selectivity. In this study, we found that ALMT12 interacts with and represses SL

High-resolution microCT reveals relationships between stomata and interior leaf anatomy in sorghum.

Stomata are pores in the leaf epidermis that regulate the trade-off between CO2 uptake for photosynthesis and water vapor loss to the atmosphere. Stomatal patterning therefore influences water use efficiency and is a target for engineering to avoid drought stress. However, there is limited understanding of how internal leaf anatomy is coordinated with stomatal development, in part due to the technical challenges of assessing three-dimensional anatomy with sufficient resolution. C4 grasses are understudied, and this is a significant knowledge gap, given their file-like stomatal distribution and

Wild Cicer species exhibit superior leaf photosynthetic phosphorus- and water-use efficiencies compared with cultivated chickpea under low-phosphorus conditions.

Domesticated chickpea cultivars exhibit limited genetic diversity. This study evaluated the effects of chickpea domestication on phosphorus (P)-use efficiency (PUE) under low-P conditions, using a diverse Cicer collection, including wild species. Two wild Cicer species - 54 C. reticulatum accessions and 15 C. echinospermum accessions, and seven domesticated C. arietinum accessions were grown in low-P soil. All three species exhibited significant variation in physiological PUE, leaf gas exchange characteristics, photosynthetic PUE (PPUE), and photosynthetic N-use efficiency (PNUE), with greater

Hydrogen sulfide acts downstream of jasmonic acid to inhibit stomatal development in Arabidopsis.

Main conclusion: Jasmonic acid (JA) negatively regulates stomatal development by promoting LCD expression and hydrogen sulfide (H2S) biosynthesis. H2S inhibits the initiation of stomata formation and acts upstream of SPEECHLESS. Abstract: Stomatal development is strictly regulated by endogenous signals and environmental cues. We recently revealed that jasmonic acid (JA) negatively regulates stomatal development in Arabidopsis thaliana cotyledons (Han et al., Plant Physiol 176:2871-2885, 2018), but the underlying molecular mechanism remains largely unknown. Here, we uncovered a role for H2S in

Physiological responses of the tropical tree Tibouchina pulchra Cogn under the influence of combustion of crude oil and natural gas at an oil refinery.

A refinery located on the slopes of a mountain range in the city of Cubatão (SE-Brazil) is the main source of sulfur dioxide (SO(2)) in the region. For this reason, the refinery replaced a system in which energy was produced from crude oil combustion in boilers with a system of energy and vapor co-generation in a thermoelectric power plant fueled by natural gas. The aim of this study was to investigate the responses of Tibouchina pulchra to the fuel switching. Saplings planted in pots were distributed throughout monitoring sites around the polluting source (sites I, II, III and IV) and in a si

Leaf nitrogen dioxide uptake coupling apoplastic chemistry, carbon/sulfur assimilation, and plant nitrogen status.

Emission and plant uptake of atmospheric nitrogen oxides (NO + NO(2)) significantly influence regional climate change by regulating the oxidative chemistry of the lower atmosphere, species composition and the recycling of carbon and nutrients, etc. Plant uptake of nitrogen dioxide (NO(2)) is concentration-dependent and species-specific, and covaries with environmental factors. An important factor determining NO(2) influx into leaves is the replenishment of the substomatal cavity. The apoplastic chemistry of the substomatal cavity plays crucial roles in NO(2) deposition rates and the tolerance

Understanding the possible cellular responses in plants under micro(nano)-plastic (MNPs): Balancing the structural harmony with functions.

The harmful impacts of micro(nano)-plastics (MNPs) on plants have gained significant attention in the last decades. Plants have a greater tendency to aggregate positively charged (+ve) MNPs on leaf surfaces and root tips, and it can be more challenging to enter the plant body than the negatively charged (-ve) MNPs. MNPs <20 nm can directly cross the cell wall and enter mainly via leaf stomata and root crack portion. Additionally, plants with aerenchyma tissue or higher water requirement might be more vulnerable to MNPs as well as environmental factors also affected MNPs uptake like porosity an

Induced tolerance to abiotic and biotic stresses of broccoli and Arabidopsis after treatment with elicitor molecules.

The plant hormones salicylic acid (SA) and jasmonic acid (JA) regulate defense mechanisms capable of overcoming different plant stress conditions and constitute distinct but interconnected signaling pathways. Interestingly, several other molecules are reported to trigger stress-specific defense responses to biotic and abiotic stresses. In this study, we investigated the effect of 14 elicitors against diverse but pivotal types of abiotic (drought) and biotic (the chewing insect Ascia monuste, the hemibiotrophic bacterium Pseudomonas syringae DC 3000 and the necrotrophic fungus Alternaria altern

Evolutionary trade-off between stomatal defense and gas exchange in Brassicaceae.

Stomatal opening is crucial for gas exchange, but it unavoidably offers invasion by pathogens. In response, plants close stomata to prevent pathogen entry, while the bacterial pathogen Pseudomonas syringae pv. tomato DC3000 (Pto) produces coronatine (COR), a jasmonate (JA) mimic, to counteract this plant response. Here, we demonstrate that by COR, Pto exploits CYP707A1 activation in Arabidopsis thaliana, encoding an enzyme that degrades abscisic acid, essential for stomatal closure. Notably, COR-induction of CYP707A1 is absent in other Brassicaceae species, such as Capsella rubella and Eutrema

2R,3R-butanediol, a bacterial volatile produced by Pseudomonas chlororaphis O6, is involved in induction of systemic tolerance to drought in Arabidopsis thaliana.

Root colonization of plants with certain rhizobacteria, such as Pseudomonas chlororaphis O6, induces tolerance to biotic and abiotic stresses. Tolerance to drought was correlated with reduced water loss in P. chlororaphis O6-colonized plants and with stomatal closure, indicated by size of stomatal aperture and percentage of closed stomata. Stomatal closure and drought resistance were mediated by production of 2R,3R-butanediol, a volatile metabolite of P. chlororaphis O6. Root colonization with bacteria deficient in 2R,3R-butanediol production showed no induction of drought tolerance. Studies w

SPEECHLESS duplication in grasses expands potential for environmental regulation of stomatal development.

Plants regulate stomatal development and function to acquire atmospheric carbon dioxide for photosynthesis while minimizing water loss. The ancestral basic helix-loop-helix transcription factor (TF) gene that drove stomata production in early land plants diversified to become paralogs SPEECHLESS (SPCH), MUTE, and FAMA. Extant grasses exhibit a particularly interesting set of duplications and losses of SPCH. Using phylogenetic methods, we tracked the history of SPCH duplications. Brachypodium distachyon and Oryza sativa plants bearing mutations in either SPCH1 or SPCH2, and B. distachyon plants

Growth and persistence of Listeria monocytogenes isolates on the plant model Arabidopsis thaliana.

While the majority of human listeriosis cases appear to be linked to consumption of processed ready-to-eat foods (e.g., deli meats), a few listeriosis outbreaks have been linked to consumption of contaminated vegetables. In this study, we assessed four isolates representing the major Listeria monocytogenes lineages for their abilities to attach to and grow on Arabidopsis thaliana, a well-characterized plant model. When plants were dipped for 5min into 3ml of water containing 8.8logCFU of L. monocytogenes and rinsed repeatedly, L. monocytogenes was recovered from the leaves at densities from 1.
