Functional Insights in PLS3-Mediated Osteogenic Regulation.

Plastin-3 (PLS3) encodes T-plastin, an actin-bundling protein mediating the formation of actin filaments by which numerous cellular processes are regulated. Loss-of-function genetic defects in PLS3 are reported to cause X-linked osteoporosis and childhood-onset fractures. However, the molecular etiology of PLS3 remains elusive. Functional compensation by actin-bundling proteins ACTN1, ACTN4, and FSCN1 was investigated in zebrafish following morpholino-mediated pls3 knockdown. Primary dermal fibroblasts from six patients with a PLS3 variant were also used to examine expression of these proteins

A novel frameshift deletion in PLS3 causing severe primary osteoporosis.

Mutations in the gene encoding plastin-3, PLS3, have recently been associated to severe primary osteoporosis. The molecular function of plastin-3 is not fully understood. Since PLS3 is located on the X chromosome, males are usually more severely affected than females. PLS3 mutations have thus far been reported in approximately 20 young patients with low bone mineral density (BMD). We describe an 8-year-old Greek boy with severe primary osteoporosis with multiple vertebral compression fractures and one low-energy long bone fracture. His clinical manifestations were consistent with osteogenesis

Loss of cyclase-associated protein 2 alters actin cytoskeleton organization and biomechanical properties of the ocular lens.

Cyclase-associated protein 2 (CAP2) is a conserved actin-binding protein that promotes actin filament (F-actin) turnover by disassembling ADF/cofilin-decorated filaments, supporting F-actin remodeling in differentiating cells and tissues. In the ocular lens, the actin cytoskeleton is crucial for maintaining tissue biomechanical properties during fiber cell maturation. To assess the role of CAP2 in the lens, we examined lens-specific CAP2 knockout (CAP2cKO) mice at cellular and tissue levels. CAP2cKO lenses were normal in size, shape and transparency but exhibited increased stiffness under comp

Integrated chromatin accessibility and DNA methylation analysis to reveal the critical epigenetic modification and regulatory mechanism in gonadal differentiation of the sequentially hermaphroditic fish, Monopterus albus.

BACKGROUND: Monopterus albus is a hermaphroditic and economically farmed fish that undergoes sex reversal from ovary to testis via ovotestis during gonadal development. The epigenetic changes that are associated with gonadal development in this species remain unclear. METHODS: We produced DNA methylome, transcriptome, and chromatin accessibility maps of the key stages of gonad development: ovary, ovotestis, and testis. The expression of the key candidate genes was detected using qRT-PCR and in situ hybridization and the methylation levels were analysed using bisulphite sequencing PCR. Promote

Ossification of the calcaneus in the normal fetal foot and in clubfoot.

The ossification of the fetal calcaneus was studied in a series of plastinated sections of 44 normal feet and three clubfeet. The results documented a precise sequence of perichondral and endochondral ossification in the normal calcaneus characterized by ossification grooves and associated cartilage canals. Within the clubfoot calcaneus, the coordination of perichondral and endochondral ossification is disturbed, ossification grooves and associated cartilage canals are not found in regular positions, and the process of endochondral ossification obviously is disturbed. The findings in the clubf

Proteomic analysis of mononuclear cells of patients with minimal-change nephrotic syndrome of childhood.

UNLABELLED: Background/Aims. Recently, peripheral blood mononuclear cell transcriptome analysis has identified genes that are upregulated in relapsing minimal-change nephrotic syndrome (MCNS). In order to investigate protein expression in peripheral blood mononuclear cells (PBMC) from relapsing MCNS patients, we performed proteomic comparisons of PBMC from patients with MCNS in relapse and controls. METHODS: PBMC from a total of 20 patients were analysed. PBMC were taken from five patients with relapsing MCNS, four in remission, five patients with other glomerular diseases and six controls. T

[Disorders of vascularization following fracture of the ankle joint in children].

In the Department of Paediatric Surgery of the University of the Heidelberg, 64 children were admitted during five years to the hospital after an ankle fracture. 12 children showed a typical fracture of the growth plate (v. Laer). All 12 were not older than 12 years. The problem of this typical form of fracture is that even careful accurate treatment may be followed by growth problems of the tibial malleolus. A microangiographic postmortem study using plastinated shetts (v. Hagens) showed a normal distribution of vascularity of the ankle joint. The vascular damage after fracture and the damage

The Fimbrin TvFim1, an immunogenic protein involved in male trichomoniasis.

An active immunoproteome of Trichomonas vaginalis was obtained by 2D-Western blotting (2D-WB). Subsequent proteoform identification by mass spectrometry (MS) showed differential expression and specific immunoreactions of multiple proteins mediated by the presence of Zn2+. A total of 25 proteoforms were immunologically reactive, generally under Zn2+ conditions, and MS analysis revealed that the fimbrin (plastin) of T. vaginalis (TvFim1) was recognized by the sera of male patients with trichomoniasis but not by the sera of infected female patients. These findings suggest that the protein is immu

The different growth zones of the fetal foot.

Previous publications revealed no reliable data or models concerning the three-dimensional ontogenesis of the lower extremity. Using the method of plastination-histology in combination with 3D-computer-reconstructions we were able to produce exact, virtual 3D-specimens of 19 healthy fetal feet. The fetuses were aged between 9 to 38 weeks of gestation and age-dependently related to four defined age-groups. We compared these feet with the help of a new geometrical method. Thus, we obtained a kind of "slow-motion-picture" of the undisturbed three-dimensional development of the fetal foot. Our res

[Congenital or acquired disposition of the separate compartment of the extensor pollicis brevis tendon associated with stenosing tendovaginitis (de Quervain's disease)? Macroanatomical and fetal-plastinationhistological studies of the first compartment of the wrist].

Our macroanatomical studies with cadaver specimens and clinical findings in the operative treatment of tendovaginitis stenosans de Quervain confirm the high number of anatomical variations in the first dorsal compartment of the wrist. With the help of thick transparent transversal-sections in one of 14 fetal wrists manufactured by plastination histology we found the first compartment with two tendons of the abductor pollicis longus and dorsal of this compartment a connective tissue formation and a separate compartment for the extensor pollicis brevis tendon, proved as a primary anatomical disp

The investing layer of the deep cervical fascia does not exist between the sternocleidomastoid and trapezius muscles.

OBJECTIVE: We sought to describe the 3-dimensional organization of connective tissues in the suboccipital region. STUDY DESIGN AND SETTING: We conducted a sectional anatomic investigation with the use of E12 sheet plastination. SUBJECTS: Six human adult cadavers (2 male and 4 female; age range, 54 to 86 years) were used in this study. Five of them were sectioned as 2.5-mm-thick coronal (1 cadaver), transverse (2 cadavers), or sagittal (2 cadavers) sections. RESULTS: No aggregation of fibrous connective tissue was seen between the sternocleidomastoid and trapezius muscles. The intervening sp

X-linked Osteoporosis due to PLS3 Pathogenic Variant: Case Report on Zoledronic Acid Treatment in Siblings.

Osteoporosis in children is a rare condition, often associated with genetic factors. Monogenic forms of osteoporosis linked to the X chromosome are often related to mutations in the gene encoding plastin 3 (PLS3). PLS3 is a protein involved in actin bundle formation in the cytoskeleton. We present two brothers with recurrent peripheral fractures and vertebral compression fractures, both associated with low bone mineral density (BMD). The patients shared the same deletion (c.589_590) in PLS3 on Xq23, which was confirmed by next-generation sequencing. They were treated with zoledronic acid, calc

Experiencing Body Worlds: voyeurism, education, or enlightenment?

Until the advent of plastinated cadavers, few outside the medical professions have had firsthand experience with human corpses. Such opportunities are now available at the Body Worlds exhibits of Gunther von Hagens. After an overview of these exhibits, we explore visitor responses as revealed in comment books available upon exiting the exhibit. Cultural, philosophical, and religious issues raised in the comments serve as a microcosm of society at large. The conclusion considers the challenge of such exhibits in introducing the public to science education, notes the image of the body as machine

Compartment syndrome of the foot.

The hindfoot compartment syndrome occurs in 10% of cases after calcaneal fracture. We analyzed the pathological anatomical reasons for this syndrome using the 10 feet from cadavers plastinated and cut into 4-mm thick sequential sections. CT scans of patients with calcaneal fractures were then compared with the anatomical findings. The key component of this compartment syndrome is the quadratus plantae muscle. The sustentacular calcaneal fragment causes bleeding from the bone or the medial calcaneal arteries into this compartment. The medial and lateral plantar nerves and vessels are then compr

E12 technique: an aid to study sinuses of psittacine birds.

Infraorbital sinus infections, which form the bulk of upper respiratory tract infections in companion birds, are commonly encountered in clinical practice and often require medical and/or surgical management. The infraorbital sinus with its dorsal drainage into the nasal cavity makes it difficult to treat these infections. The sinus has various compartments throughout the skull, and subcutaneous tissue. As an aid in determining the location and extent of the sinuses of the parrot and macaw, a corrosion cast of the infraorbital sinus was made. Also, computed tomography (CT) images of cadaver he

PLS3 mutations in X-linked osteoporosis with fractures.

Plastin 3 (PLS3), a protein involved in the formation of filamentous actin (F-actin) bundles, appears to be important in human bone health, on the basis of pathogenic variants in PLS3 in five families with X-linked osteoporosis and osteoporotic fractures that we report here. The bone-regulatory properties of PLS3 were supported by in vivo analyses in zebrafish. Furthermore, in an additional five families (described in less detail) referred for diagnosis or ruling out of osteogenesis imperfecta type I, a rare variant (rs140121121) in PLS3 was found. This variant was also associated with a risk

[Disorders of vascularization in para-epiphyseal groove humeral fractures and their clinical significance for the growing skeleton].

We analysed retrospectively 50 humerus fractures in the vicinity of elbow joint and 40 fractures of the proximal humerus with respect to possible injuries of the vessel pattern of the epiphyseal plate. The normal vessel pattern of the epiphyseal plate was examined postmortem on 8 foetus and newborns up to 6 months. We plastinated the affected regions and could compare symmetry of the vessel pattern in the epiphyseal plates as well as therapeutic consequences, especially after injuries of the elbow joint. Vascular disturbances in the case of young children, i.e. by fractures of the lateral oder

Osteoporotic Burst Fracture in a Young Male Adult as First Presentation of a Rare PLS3 Mutation: A Case Report.

Low-impact spinal fractures in young patients are rare and should raise suspicion of an underlying condition, as these injuries are typically the result of high-energy trauma. We describe a case of a young male patient who sustained a burst fracture of the first lumbar vertebra (L1) following low-energy trauma. The patient underwent percutaneous posterior spinal instrumentation, yet the poor bone quality detected intraoperatively prompted further diagnostic evaluation. Subsequently, low bone mineral density (BMD) was detected, and a rare plastine-3 (PLS3) gene mutation was revealed in the gene

Borders and topographic relationships of the paraglottic space.

The precise topographic relationships of the paraglottic space (PGS) were investigated in serial plastinated or frozen whole-organ sections of 46 normal adult larynges. Laterally, the PGS was bordered by the thyroid cartilage. Superomedially, the PGS in some specimens was only separated from the periepiglottic space by a coherent collagenous fiber septum. The paraglottic adipose tissue extended between the caudal fibers of the thyroarytenoid muscle. Inferomedially, the PGS was bordered by the conus elasticus. The anteroinferior portion of the PGS extended beyond the larynx beneath the inferior

[Anatomy of the glottis and subglottis in the pediatric larynx].

BACKGROUND: The morphological development of the human larynx during the first years of life has previously not been studied in detail and has mainly been described on a qualitative basis. This study seeks to provide detailed morphometric data on the regular anatomy of the vocal cords, the subglottic airway and the tracheal airway dimensions gained from plastinated whole organ serial sections of 43 infant larynges and to determine morphological changes with age. Such information may be useful for the understanding of pediatric airway disease or for laryngeal surgery in children. MATERIAL AND
