LC-MS based metabolic fingerprinting of apricot pistils after self-compatible and self-incompatible pollinations.

LC-MS based metabolomics approach revealed that putative metabolites other than flavonoids may significantly contribute to the sexual compatibility reactions in Prunus armeniaca. Possible mechanisms on related microtubule-stabilizing effects are provided. Identification of metabolites playing crucial roles in sexual incompatibility reactions in apricot (Prunus armeniaca L.) was the aim of the study. Metabolic fingerprints of self-compatible and self-incompatible apricot pistils were created using liquid chromatography coupled to time-of-flight mass spectrometry followed by untargeted compound

Focusing on the focus: what else beyond the master switches for polar cell growth?

Cell polarity, often associated with polarized cell expansion/growth in plants, describes the uneven distribution of cellular components, such as proteins, nucleic acids, signaling molecules, vesicles, cytoskeletal elements, and organelles, which may ultimately modulate cell shape, structure, and function. Pollen tubes and root hairs are model cell systems for studying the molecular mechanisms underlying sustained tip growth. The formation of intercalated epidermal pavement cells requires excitatory and inhibitory pathways to coordinate cell expansion within single cells and between cells in c

Olfactory response of predatory mites to vegetative and reproductive parts of coconut palm infested by Aceria guerreronis.

The phytophagous mite Aceria guerreronis Keifer is an important pest of coconut worldwide. A promising method of control for this pest is the use of predatory mites. Neoseiulus baraki (Athias-Henriot) and Proctolaelaps bickleyi Bram are predatory mites found in association with A. guerreronis in the field. To understand how these predators respond to olfactory cues from A. guerreronis and its host plant, the foraging behavior of the predatory mites was investigated in a Y-tube olfactometer and on T-shaped arenas. The predators were subjected to choose in an olfactometer: (1) isolated parts (le

Analysis of the lipid body proteome of the oleaginous alga Lobosphaera incisa.

BACKGROUND: Lobosphaera incisa (L. incisa) is an oleaginous microalga that stores triacylglycerol (TAG) rich in arachidonic acid in lipid bodies (LBs). This organelle is gaining attention in algal research, since evidence is accumulating that proteins attached to its surface fulfill important functions in TAG storage and metabolism. RESULTS: Here, the composition of the LB proteome in L incisa was investigated by comparing different cell fractions in a semiquantitative proteomics approach. After applying stringent filters to the proteomics data in order to remove contaminating proteins from t

Three-dimensional ultrastructural analysis of fertilization-defective Arabidopsis mutants using STEM and array tomography.

Double fertilization in angiosperms is completed by the fusion of two sperm cells with the egg and central cells. While sperm membrane proteins such as GEX2, DMP8/9, and GCS1/HAP2 have been identified as key regulators of gamete attachment and fusion, their specific ultrastructural roles within the embryo sac have remained unclear owing to the technical limitations of live-cell imaging based on fluorescent markers. Here, we optimized high-pressure freezing and freeze-substitution protocols for Arabidopsis pistils and performed three-dimensional reconstructions using scanning transmission elect

Plant phospholipase D: novel structure, regulatory mechanism, and multifaceted functions with biotechnological application.

Phospholipases D (PLDs) are important membrane lipid-modifying enzymes in eukaryotes. Phosphatidic acid, the product of PLD activity, is a vital signaling molecule. PLD-mediated lipid signaling has been the subject of extensive research leading to discovery of its crystal structure. PLDs are involved in the pathophysiology of several human diseases, therefore, viewed as promising targets for drug design. The availability of a eukaryotic PLD crystal structure will encourage PLD targeted drug designing. PLDs have been implicated in plants response to biotic and abiotic stresses. However, the mol

A Chemically Driven Soft Growing Robot with Coupled Gas Generation and Growth Dynamics.

Soft growing robots achieve extension through material eversion and exhibit remarkable compliance and adaptability in confined environments. However, most existing soft growing robots still rely on tethered laboratory pneumatic supplies, onboard electric compressors, or pre-charged gas reservoirs for actuation. In contrast, chemical reaction--based actuation for soft growing robots remains largely unexplored. Inspired by the internally driven tip-growth process observed in biological systems such as pollen tubes, this work presents a chemically driven soft growing robot in which gas generated

[Basophil activation test--a practical approach to diagnosis of common respiratory allergy].

BACKGROUND: The diagnosis of immediate allergy is based on clinical data, skin prick tests (SPT), and measurements of allergen-specific IgE (sIgE). Basophil activation test (BAT) can supplement these methods and obviate their disadvantages, and possibly replace allergen challenge tests, such as a nasal provocative test (NPT). In this study, we assessed the influence of different storage times on BAT results. Futhermore, we compared the results of SPT, sIgE and NPT against BAT for common aeroallergens. METHODS: BAT was performed in twelve patients with allergic rhinitis sensitized to birch pol

[Biological profile of type I allergies at Mohamed V Hospital (Rabat- Morocco)].

Allergic diseases are ranked fourth considering the world health organization classification of diseases. The consequences linked to these ailments are huge for public health economics and the diagnosis is awkward due to clinical polymorphism and multifactorial aetiologies. The allergologic diagnosis is the result of weighing in clinical and biological findings. The biological assessment is made of qualitative specific and multiple-allergen serum IgE test, which once positive drives to skin test and each allergen-specific IgE level determination to conclude. Our study aims at displaying biolog

ROPs.

Rho-like GTPases from plants (ROPs) constitute a plant-specific subfamily of Rho-family small GTPases. The Rho family is conserved among all eukaryotic cells, and includes the Rho, Rac and Cdc42 subfamilies found in fungi and animals. Like other GTPases, ROPs act as molecular switches that cycle between the inactive GDP-bound and active GTP-bound states, and are typically involved in cell signaling. Functional studies have demonstrated that ROPs play key roles in regulating processes such as the establishment of cell polarity, polar growth of cells (e.g., pollen tubes and root hairs), cell sha

A next-generation sequencing approach for high-resolution S-locus genotyping in apricot.

Most temperate fruit crops exhibit a Gametophytic Self-Incompatibility (GSI) mechanism that prevents incompatible pollen tube growth and promotes outbreeding. In Prunus species, this system is governed by the multiallelic S-locus, which contains the S-haplotype-specific F-box (SFB) and S-RNase genes. Accurate determination of S-haplotypes is important for fruit breeding and orchard design and has traditionally relied on PCR-based analysis. However, PCR-based methods, combined with the partial sequencing of many S-alleles, may lead to ambiguous or incorrect allele identification. Next-generatio

Risk assessment for side-effects of neonicotinoids against bumblebees with and without impairing foraging behavior.

Bombus terrestris bumblebees are important pollinators of wild flowers, and in modern agriculture they are used to guarantee pollination of vegetables and fruits. In the field it is likely that worker bees are exposed to pesticides during foraging. To date, several tests exist to assess lethal and sublethal side-effects of pesticides on bee survival, growth/development and reproduction. Within the context of ecotoxicology and insect physiology, we report the development of a new bioassay to assess the impact of sublethal concentrations on the bumblebee foraging behavior under laboratory condit

Simple Whole-Mount Staining Protocol of F-Actin for Studies of the Female Gametophyte in Agavoideae and Other Crassinucellate Ovules.

During plant sexual reproduction, F-actin takes part in the elongation of the pollen tube and the movement of sperm cells along with it. Moreover, F-actin is involved in the transport of sperm cells throughout the embryo sac when double fertilization occurs. Different techniques for analysis of F-actin in plant cells have been developed: from classical actin-immunolocalization in fixed tissues to genetically tagged actin with fluorescent proteins for live imaging of cells. Despite the implementation of live cell imaging tools, fixed plant tissue methods for cytoskeletal studies remain an essen

Chromatin-remodeling factor OsINO80 is involved in regulation of gibberellin biosynthesis and is crucial for rice plant growth and development.

The phytohormone gibberellin (GA) plays essential roles in plant growth and development. Here, we report that OsINO80, a conserved ATP-dependent chromatin-remodeling factor in rice (Oryza sativa), functions in both GA biosynthesis and diverse biological processes. OsINO80-knockdown mutants, derived from either T-DNA insertion or RNA interference, display typical GA-deficient phenotypes, including dwarfism, reduced cell length, late flowering, retarded seed germination and impaired reproductive development. Consistently, transcriptome analyses reveal that OsINO80 knockdown results in downregula

A small multigene hydroxyproline-O-galactosyltransferase family functions in arabinogalactan-protein glycosylation, growth and development in Arabidopsis.

BACKGROUND: Arabinogalactan-proteins (AGPs) are ubiquitous components of cell walls throughout the plant kingdom and are extensively post translationally modified by conversion of proline to hydroxyproline (Hyp) and by addition of arabinogalactan polysaccharides (AG) to Hyp residues. AGPs are implicated to function in various aspects of plant growth and development, but the functional contributions of AGP glycans remain to be elucidated. Hyp glycosylation is initiated by the action of a set of Hyp-O-galactosyltransferase (Hyp-O-GALT) enzymes that remain to be fully characterized. RESULTS: Thr

Essential oils, asthma, thunderstorms, and plant gases: a prospective study of respiratory response to ambient biogenic volatile organic compounds (BVOCs).

Purpose: Prevailing opinion is that wind-pollinated plants affect asthma negatively and that insect- pollinated ones do not. "Thunderstorm" asthma, too, is attributed to bursting grass pollens. Additional biogenic volatile organic compounds (BVOCs) are identified here. Essential oils' BVOCs are inhaled from plants, oil diffusers, candles, room "fresheners", perfumes, and hygiene products. Claims of BVOC "safety" for sensitive respiratory systems are questioned. Methods: Fourteen volunteers, of mixed-age and gender, with seasonal asthma recorded peak expiratory flow (PEF) and 11 symptom scores.

Transcriptomic analysis reveals molecular phenological changes during the flower-to-fruit transition in Vanilla planifolia Andrews (Orchidaceae).

BACKGROUND: The transition from flower to fruit, encompassing flower formation to fruit maturation, has been extensively studied in model plants such as Arabidopsis thaliana. However, the Orchidaceae family, including Vanilla planifolia, exhibits a unique phenomenon known as post-pollination syndrome (PPS), where pollination initiates ovule development but often leads to premature ovary drop. This phenomenon significantly impacts the yield and stability of V. planifolia crops. Understanding the molecular mechanisms underlying PPS is essential for improving crop production. This study explores

Agaricus blazei-Based Mushroom Extract Supplementation to Birch Allergic Blood Donors: A Randomized Clinical Trial.

Since Agaricus blazei Murill (AbM) extract reduced specific IgE and ameliorated a skewed Th1/Th2 balance in a mouse allergy model, it was tested in blood donors with self-reported, IgE-positive, birch pollen allergy and/or asthma. Sixty recruited donors were randomized in a placebo-controlled, double-blinded study with pre-seasonal, 7-week, oral supplementation with the AbM-based extract AndosanTM. Before and after the pollen season, questionnaires were answered for allergic rhino-conjunctivitis, asthma, and medication; serum IgE was measured, and Bet v 1-induced basophil activation was determ
