Effects of polyinosinic-polycytidylic acid (Poly I:C) on naloxone-precipitated withdrawal in morphine-dependent mice.

Viral infections are frequently found in opioid addicts, subjecting them to immune challenge. However, the effects of immune challenge on opioid withdrawal are not fully understood. In the present study, mice were intraperitoneally injected with 2mg/kg polyinosinic-polycytidylic acid (Poly I:C, a viral mimetic) for 3 days to induce an immune challenge, followed by subcutaneous injection of morphine 3 times per day for 3 days to induce morphine dependence. Withdrawal was induced by an intraperitoneal injection of 5mg/kg naloxone, an opioid receptor antagonist. The results showed that Poly I:C p

[Smallpox vaccination with protection by interferon and an interferon inducer].

The vaccinal process was studied in monkeys vaccinated with smallpox vaccine under the protection of fibroblastic or leukocytic interferons and modified poly(I) . poly(C). Simultaneous vaccination and administration of one of the studied preparations induced intense immunity and at the same time prevented viremia and alleviated local inflammatory reactions caused by the vaccination. The protective effect of the fibroblastic interferon was more marked than that of the leukocytic one in the vaccinal process. When the monkeys received the preparations intramuscularly the leukocytic but not the fi

Type I interferon signaling in systemic immune cells from patients with alcoholic cirrhosis and its association with outcome.

BACKGROUND & AIMS: In immune cells, constitutively and acutely produced type I interferons (IFNs) engage autocrine/paracrine signaling pathways to induce IFN-stimulated genes (ISGs). Enhanced activity of IFN signaling pathways can cause excessive inflammation and tissue damage. We aimed to investigate ISG expression in systemic immune cells from patients with decompensated alcoholic cirrhosis, and its association with outcome. METHODS: Peripheral blood mononuclear cells (PBMCs) from patients and heathy subjects were stimulated or not with lipopolysaccharide (LPS, an IFN inducer) or increasing

The role of vitamin A in natural killer cell cytotoxicity, number and activation in the rat.

The cause of low natural killer (NK) cell activity in vitamin-A-deficient rats was investigated in the current study. Cytotoxicity correlated with the percentage of NK cells (3.2.3-positive cells) in peripheral blood mononuclear cells (PBMC; r = 0.85) and spleen (r = 0.70). Lytic activity per NK cell did not differ between vitamin-A-deficient rats and controls in PBMC, whereas it did in spleen. Vitamin A deficiency did not impair the ability to produce interferon (IFN), increase lytic efficiency and expand the NK cell pool upon poly(I:C) stimulation in vivo. Nor was the in vitro activation by

Tumor cell intrinsic dsRNA innate immune response triggered by PARP inhibitor is compromised in BRCA1-deficient breast cancer by repressing IRF3.

Poly(ADP-ribose) polymerase 1 (PARP1) inhibition represents a promising targeted therapy for BRCA-deficient cancer patients based on the synthetic lethality theory. Recent evidence shows that the efficacy of DNA damage drugs depends on two aspects: DNA repair signaling and immune response. Applying a functional proteomics approach, we find that the function of the spliceosome is perturbed by PARP inhibitors via enhancing interaction between PARP1 and SF3B1, a key factor of the spliceosome. We demonstrate that differential alternative spliced mRNA and accumulation of double-stranded RNA (dsRNA)

Protective effect of measles virus inoculation on subacute sclerosing panencephalitis virus-infected mice.

The Biken strain of subacute sclerosing panencephalitis (SSPE) virus caused a fatal neurologic disease in adult mice after intracerebral inoculation. However, the mice were completely protected from the disease when a high dose of measles virus was given intracerebrally after the SSPE virus infection. The measles virus inoculation induced interferon production and immune responses. An experiment with athymic nude mice showed that interferon and anti-measles antibody were able to prolong the incubation period of the disease but not to protect the SSPE virus-infected nude mice from death. Depression of oxidative metabolism of aspirin in mice via an interferon-associated mechanism in relation to Reye's syndrome.

Interferon synthesis was induced in inbred strains of mice with either polyriboinosinic-polyribocytidylic acid or Newcastle disease virus. The strains, BALB/cBy and C57BL/6By, respond to Newcastle disease virus by producing 'low' and 'high' levels of serum interferon, respectively. 24 h after interferon induction mice received [carboxyl-14C]-aspirin orally. The metabolic conjugation of aspirin was little changed after either treatment while the urinary excretion of gentisic acid, a minor product of metabolic oxidation, was significantly depressed after polyriboinosinic-polyribocytidylic acid i

The GABBR1/miR-19b-3p/WNT2B axis regulates insulin resistance and liver injury in diabetes with viral infection: mechanistic and therapeutic insights.

Insulin resistance (IR) is the main feature of type 2 diabetes mellitus. Furthermore, viral infection can aggravate the abnormal glucose metabolism in diabetic patients. GABBR1 can maintain normal glucose homeostasis, but its specific role in diabetes is not clear. We investigated the function of the GABBR1/miR-19b-3p/WNT2B axis in vitro and in vivo. miR-19b-3p and GABBR1 were overexpressed or knocked down in AML12 cells. Subsequently, these cells were treated with palmitic acid (PA) to induce damage or poly I : C to mimic viral infection. The degree of AML12 cell damage was assessed using the

Synthetic double-stranded RNA in antiviral immunity and vaccine adjuvant: insights into poly(I:C) and poly(A:U).

Synthetic double-stranded RNA (dsRNA) analogues function as viral mimetics that activate innate immune signaling pathways critical for antiviral defense and the induction of adaptive immunity. Among dsRNA analogues, polyinosinic:polycytidylic acid [poly(I:C)] and polyadenylic:polyuridylic acid [poly(A:U)] have been extensively studied for their immunostimulatory properties and potential as vaccine adjuvants. This review examines how host pattern-recognition receptors sense synthetic dsRNA and explains the rationale for using poly(I:C) and poly(A:U) as representative dsRNA analogues with distin

Fetal developmental trajectories following maternal immune activation are shaped by sex chromosome-gonadal interactions.

Maternal immune activation (MIA) contributes to neurodevelopmental disorders with male-biased prevalence. To disentangle the contributions of sex chromosomes (XX vs. XY) and gonads (ovaries vs. testes) in shaping fetal responses to MIA, we used the four-core genotypes (FCG) mouse model. Offspring representing all combinations of sex chromosome and gonadal status were generated and exposed to poly(I:C) or saline at embryonic day (E)12.5. We assessed maternal serum, fetal placentas, and fetal brain cytokines at E13.5; at E17.5, fetal development was evaluated via microultrasound and CT imaging.

Nuclear scaffold attachment factor A functions as a potential viral recognition receptor involved in the antiviral immunity of grass carp (Ctenopharyngodon idella).

As one of the most primitive vertebrates, fish have evolved a distinct repertoire of viral recognition receptors. However, the existence of nuclear viral recognition receptors in fish remains uncertain. In this study, we identified a scaffold attachment factor A (SAFA), CiSAFA, as a potential nuclear recognition receptor for grass carp reovirus (GCRV) in grass carp (Ctenopharyngodon idella). CiSAFA shares high amino acid sequence similarity (70.3-99.6 %) and identity (56.1-98.6 %) with its counterparts from other vertebrates and contains three conserved domains, namely, SAP, SPRY, and AAA_33.

Proteomics analysis of soluble secreted proteins of Lutzomyia longipalpis LL5 cells transfected with a dsRNA viral mimic: insights into cellular defense and repair signals.

Sand flies, which transmit diseases like leishmaniases, bartonellosis, and certain viruses, pose a significant public health threat. Our research focuses on the immune responses of Lutzomyia longipalpis, the primary vector for visceral leishmaniasis in the Americas. We use L. longipalpis LL5 cells as a model to study how sand flies respond to pathogens. These cells exhibit robust immune reactions, producing molecules mainly regulated by the Toll, IMD, Jak-STAT, and RNAi pathways. In previous studies, we detected a non-specific antiviral response in LL5 cells following double-stranded RNAs (dsR

Cysteine Peptidase Cathepsin X as a Therapeutic Target for Simultaneous TLR3/4-mediated Microglia Activation.

Microglia are resident macrophages in the central nervous system that are involved in immune responses driven by Toll-like receptors (TLRs). Microglia-mediated inflammation can lead to central nervous system disorders, and more than one TLR might be involved in these pathological processes. The cysteine peptidase cathepsin X has been recognized as a pathogenic factor for inflammation-induced neurodegeneration. Here, we hypothesized that simultaneous TLR3 and TLR4 activation induces synergized microglia responses and that these phenotype changes affect cathepsin X expression and activity. Murin

Genome-wide identification and characterization of Toll-like receptor genes in black rockfish (Sebastes schlegelii) and their response mechanisms following poly (I:C) injection.

Toll-like receptors (TLRs) are canonical transmembrane receptors that play an important role in defending against invading pathogens. In this study, we identified a total of 12 TLR genes in black rockfish (Sebastes schlegelii) with an analysis of their sequence characterizations. The phylogenetic analysis suggested that 12 distinct TLRs were grouped into five subfamilies (i.e., TLR1, TLR3, TLR5, TLR7, and TLR11 subfamilies), and each SsTLR gene respectively corresponded to the orthologs genes of other species. The protein domain analysis indicated that TLRs are type I transmembrane proteins, i

A preoptic neuronal population controls fever and appetite during sickness.

During infection, animals exhibit adaptive changes in physiology and behaviour aimed at increasing survival. Although many causes of infection exist, they trigger similar stereotyped symptoms such as fever, warmth-seeking, loss of appetite and fatigue1,2. Yet exactly how the nervous system alters body temperature and triggers sickness behaviours to coordinate responses to infection remains unknown. Here we identify a previously uncharacterized population of neurons in the ventral medial preoptic area (VMPO) of the hypothalamus that are activated after sickness induced by lipopolysaccharide (LP
