Gas-Phase Integration of Trophically Distinct Microbial Cultures for Net-Reduced CO2 and Enhanced Metabolite Production.

The continued increase in atmospheric CO2 concentrations has intensified global efforts to develop sustainable biotechnologies that capture and reutilise carbon rather than releasing it. While photosynthetic microorganisms provide a renewable route for CO2 fixation into organic products, heterotrophic fermentation remains the industrial standard due to its high productivity, controllability and scalability. Consequently, integrating the carbon efficiency of autotrophic processes with the productivity of heterotrophic systems may represent a promising strategy toward circular biomanufacturing.

clpC operon regulates cell architecture and sporulation in Bacillus anthracis.

The clpC operon is known to regulate several processes such as genetic competence, protein degradation and stress survival in bacteria. Here, we describe the role of clpC operon in Bacillus anthracis. We generated knockout strains of the clpC operon genes to investigate the impact of CtsR, McsA, McsB and ClpC deletion on essential processes of B. anthracis. We observed that growth, cell division, sporulation and germination were severely affected in mcsB and clpC deleted strains, while none of deletions affected toxin secretion. Growth defect in these strains was pronounced at elevated tempera

Multifunctional hybrid films made from CoT3OTx4 and CoFeT2OT4 nanoparticles inside a poly 3-hydroxybutyrate matrix and study of their impact in methyl orange photodegradation.

This work aims to produce hybrid materials with potential applications in dye photodegradation. Therefore, hybrid films were obtained by incorporating cobalt (II, III) oxide (Co3O4) or cobalt ferrite (CoFe2O4) nanoparticles (NPs) with 18 ± 1.6 nm and 26 ± 1.3 nm, respectively, into a poly 3-hydroxybutyrate (P3HB) polymeric matrix. The Co3O4@P3HB and CoFe2O4@P3HB hybrid films were fabricated by solvent casting in a ratio of 85 mg to 15 mg (P3HB-NPs). Different spectroscopic and microscopy techniques characterized the Co3O4 and CoFe2O4 NPs and the P3HB, Co3O4@P3HB and CoFe2O4@P3HB films. The opt

From heterotrophic monoculture to light-driven coculture: A sustainable platform for PHA biosynthesis and insights into interspecies metabolic interactions.

Microbial consortia combining phototrophic and heterotrophic partners offer a sustainable and cost-effective strategy for producing polyhydroxyalkanoates, which have attracted considerable attention as promising alternatives to petroleum-derived plastics. Here, a synthetic cyanobacteria-heterotroph consortium composed of Synechococcus elongatus and Priestia aryabhattai was employed to produce polyhydroxybutyrate (PHB) directly from CO2. Transcriptomic analysis of the heterotrophic partner revealed the sucrose-to-PHB biosynthesis pathway and key gene cluster. Optimal coculture conditions were e

Producing and Characterizing Polyhydroxyalkanoates from Starch and Chickpea Waste Using Mixed Microbial Cultures in Solid-State Fermentation.

The environmental impact of plastic waste is a growing global challenge, primarily due to non-biodegradable plastics from fossil resources that accumulate in ecosystems. Biodegradable polymers like polyhydroxyalkanoates (PHAs) offer a sustainable alternative. PHAs are microbial biopolymers produced by microorganisms using renewable substrates, including agro-industrial byproducts, making them eco-friendly and cost-effective. This study focused on the isolation and characterization of PHA-producing microorganisms from agro-industrial waste, including chickpeas, chickpeas with bean residues, and

Copper stimulation on methane-supported perchlorate reduction in a membrane biofilm reactor.

The present study demonstrated that the perchlorate reduction rate in a methane-based membrane biofilm reactor was significantly enhanced from 14.4 to 25.6 mg-Cl/L/d by increasing copper concentration in the feeding medium from 1 to 10 μM, indicating a stimulatory effect of copper on the methane-supported perchlorate reduction process. Batch tests further confirmed that the increased copper concentration enhanced both methane oxidation and perchlorate reduction rates, which was supported by an increasing trend of functional genes (pmoA for methanotrophs and pcrA for specific perchlorate reduce

3-Hydroxybutyrate methyl ester as a potential drug against Alzheimer's disease via mitochondria protection mechanism.

Alzheimer's disease (AD) is induced by many reasons, including decreased cellular utilization of glucose and brain cell mitochondrial damages. Degradation product of microbially synthesized polyhydroxybutyrate (PHB), namely, 3-hydroxybutyrate (3HB), can be an alternative to glucose during sustained hypoglycemia. In this study, the derivative of 3HB, 3-hydroxybutyrate methyl ester (HBME), was used by cells as an alternative to glucose. HBME inhibited cell apoptosis under glucose deprivation, rescued activities of mitochondrial respiratory chain complexes that were impaired in AD patients and de

Metabolic versatility of Gram-positive microbial isolates from contaminated river sediments.

Gram-positive bacteria from river sediments affected by the proximity of a petrochemical industrial site were isolated and characterized with respect to their ability to degrade a wide range of aromatic compounds. In this study we identified metabolically diverse Gram-positive bacteria capable of growth on wide range aromatic compounds in the presence of heavy metals and with the ability to accumulate biopolymers. Thirty-four isolates that were able to use 9 or more common aromatic pollutants, such as benzene, biphenyl, naphthalene etc. as a sole source of carbon and energy included members of

Restructuring Biologically Assembled Binding Protein-Biopolymer Conjugates toward Advanced Materials.

Bacterial cell factories have been successfully engineered to efficiently assemble spherical polyhydroxybutyrate inclusions coated with functional proteins of interest. In these submicrometer-sized core-shell assemblies, proteins are bioconjugated to the polymer core, enabling bioengineering for uses as bioseparation resins, enzyme carriers, diagnostic reagents, and particulate vaccines. Here, we explore whether these functional protein-polymer assemblies could be restructured via dissolution and subsequent precipitation while retaining the functionality of the conjugated protein. Polymer core

Improvement of the Polyhydroxyalkanoates Recovery from Mixed Microbial Cultures Using Sodium Hypochlorite Pre-Treatment Coupled with Solvent Extraction.

The use of mixed microbial cultures (MMC) and organic wastes and wastewaters as feed sources is considered an appealing approach to reduce the current polyhydroxyalkanoates (PHAs) production costs. However, this method entails an additional hurdle to the PHAs downstream processing (recovery and purification). In the current work, the effect of a sodium hypochlorite (NaClO) pre-treatment coupled with dimethyl carbonate (DMC) or chloroform (CF) as extraction solvents on the PHAs recovery efficiency (RE) from MMC was evaluated. MMC were harvested from a sequencing batch reactor (SBR) fed with a s

Production of nitrous oxide from anaerobic digester centrate and its use as a co-oxidant of biogas to enhance energy recovery.

Coupled Aerobic-anoxic Nitrous Decomposition Operation (CANDO) is a new process for wastewater treatment that removes nitrogen from wastewater and recovers energy from the nitrogen in three steps: (1) NH4(+) oxidation to NO2(-); (2) NO2(-) reduction to N2O gas; and (3) N2O conversion to N2 with energy production. In this work, we optimize Steps 1 and 2 for anaerobic digester centrate, and we evaluate Step 3 for a full-scale biogas-fed internal combustion engine. Using a continuous stirred reactor coupled to a bench-scale sequencing batch reactor, we observed sustained partial oxidation of NH4(

Fast procedure for the analysis of poly(hydroxyalkanoates) in bacterial cells by off-line pyrolysis/gas-chromatography with flame ionization detector.

Poly(hydroxyalkanoates) (PHAs) are polyesters formed by saturated short chain hydroxyacids, among which 3-hydroxybutanoic (HB) and 3-hydroxypentanoic (3-hydroxyvalerate, HV) are the most common monomers of homopolymers (e.g. poly(3-hydroxybutyrate), PHB) and copolymers (e.g. poly(3-hydroxybutyrate-co-3-hydroxyhexanoate), PHB-HC). The most widely used approach for their determination is the polymer methanolysis followed by gas chromatography-mass spectrometry (GC-MS) analysis of the methylated monomers; this procedure generally requires the use of additional reagents (e.g. sulfuric acid) and is

Poly-3-hydroxybutyrate (PHB): a resorbable conduit for long-gap repair in peripheral nerves.

The repair of peripheral nerve injuries with an autologous nerve graft is still the treatment of choice, although it is widely recognised that the method is associated with donor-site morbidity and a suboptimal functional outcome. An alternative approach is to use a nerve conduit to provide a guidance channel for the regenerating nerve. This study investigates the potential of poly-3-hydroxybutyrate (PHB) nerve conduits to bridge long nerve gaps (up to 4 cm) in a rabbit common-peroneal-nerve injury model. Regeneration was assessed up to 63 days postoperatively, and compared with that achieved

Effect of nitrogen source on growth and trichloroethylene degradation by methane-oxidizing bacteria.

The effect of nitrogen source on methane-oxidizing bacteria with respect to cellular growth and trichloroethylene (TCE) degradation ability were examined. One mixed chemostat culture and two pure type II methane-oxidizing strains, Methylosinus trichosporium OB3b and strain CAC-2, which was isolated from the chemostat culture, were used in this study. All cultures were able to grow with each of three different nitrogen sources: ammonia, nitrate, and molecular nitrogen. Both M. trichosporium OB3b and strain CAC-2 showed slightly lower net cellular growth rates and cell yields but exhibited highe
