Preparation of a composite fibrous membrane loaded with mesalazine and metronidazole by interlaced electrospinning.

Novel composite fibrous membranes loaded with mesalazine and metronidazole were fabricated via interlaced electrospinning. The fibers were characterized by scanning electron microscopy and FTIR transmission spectra techniques. These characterizations were performed in the aim of optimizing the experimental conditions which allowed us to obtain good morphology of fibrous membrane loaded drugs. The in vitro release experiments revealed that mesalazine and metronidazole were released continuously from the loaded drug fibrous membrane. The fibrous membrane-loaded drugs also showed excellent stabil

Preparation and properties of PLGA microspheres containing hydrophilic drugs by the SPG (shirasu porous glass) membrane emulsification technique.

In the present paper, monodisperse poly (lactide-co-glycolide) (PLGA) microspheres containing the hydrophilic model drug, blue dextran (BLD), were manufactured by the solvent evaporation method and the shirasu porous glass (SPG) membrane emulsification technique. In order to prepare PLGA microspheres with a higher drug loading efficiency by the membrane emulsification technique, the test of stability and productivity of the primary emulsion (w(1)/o emulsion) was preliminary examined by change species or concentration of the oil-soluble surfactant and the ratio of water and organic solvent. Improved insulin loading in poly(lactic-co-glycolic) acid (PLGA) nanoparticles upon self-assembly with lipids.

Polymeric nanoparticles are widely investigated as drug delivery systems for oral administration. However, the hydrophobic nature of many polymers hampers effective loading of the particles with hydrophilic macromolecules such as insulin. Thus, the aim of this work was to improve the loading of insulin into poly(lactic-co-glycolic) acid (PLGA) nanoparticles by pre-assembly with amphiphilic lipids. Insulin was complexed with soybean phosphatidylcholine or sodium caprate by self-assembly and subsequently loaded into PLGA nanoparticles by using the double emulsion-solvent evaporation technique. Stimulated Drug Release to Human Melanoma Cell Line Using NIR-responsive PLGA Microparticles.

Stimulated on-demand drug release is critical in treating melanoma, enabling targeted therapeutic interventions with enhanced efficacy and minimized side effects. Poly(lactic-co- glycolic acid) (PLGA) microparticles (MPs) fabricated using the emulsion method provide a cost-effective biocompatible pathway towards developing a drug release system for such treatments. Near-infrared (NIR)-responsive particles such as indocyanine green (ICG) and silver nanoparticles (AgNPs) have been incorpo- rated for generating an on-demand release system. Characteriza- tion results showed that the MPs were aroun

Stem Cells Expansion Vector via Bioadhesive Porous Microspheres for Accelerating Articular Cartilage Regeneration.

Stem cell tissue engineering is a potential treatment for osteoarthritis. However, the number of stem cells that can be delivered, loss of stem cells during injection, and migration ability of stem cells limit applications of traditional stem cell tissue engineering. Herein, kartogenin (KGN)-loaded poly(lactic-co-glycolic acid) (PLGA) porous microspheres is first engineered via emulsification, and then anchored with chitosan through the amidation reaction to develop a new porous microsphere (PLGA-CS@KGN) as a stem cell expansion vector. Following 3D co-culture of the PLGA-CS@KGN carrier with m

Improved drug loading via spray drying of a chalcone implant for local treatment of cutaneous leishmaniasis.

Current chemotherapy of cutaneous leishmaniasis (CL), even the mildest forms, encompasses multiple and painful injections with toxic drugs that cause systemic adverse effects. Recently, we showed the promising use of poly(lactic-co-glycolic acid) (PLGA) microparticles loaded with an antileishmanial nitrosylated chalcone (CH8) for effective, safe, local, and single-dose treatment of CL. Here, we proposed to optimize the delivery system by increasing the CH8 loading in PLGA-microparticles using spray drying instead of emulsification-solvent evaporation. The effect of solvent composition and poly

Design of PLGA-functionalized quercetin nanoparticles for potential use in Alzheimer's disease.

Dysfunctional interaction of amyloid-β (Aβ) with excess metal ions is proved to be related to the etiology of Alzheimer's disease (AD). Hence, disruption of these metal-peptide interactions using nanoparticles (NPs) holds considerable promise as a therapeutic strategy to combat this incurable disease. Given that quercetin is a natural product, the biocompatibility and small size essential for permeating the blood-brain barrier make it a potential therapeutic drug candidate for treating AD. Nanocarriers formulated with the US Food and Drug Administration-approved biocompatible and biodegradable

Fabrication of a Micro-Needle Array Electrode by Thermal Drawing for Bio-Signals Monitoring.

A novel micro-needle array electrode (MAE) fabricated by thermal drawing and coated with Ti/Au film was proposed for bio-signals monitoring. A simple and effective setup was employed to form glassy-state poly (lactic-co-glycolic acid) (PLGA) into a micro-needle array (MA) by the thermal drawing method. The MA was composed of 6 × 6 micro-needles with an average height of about 500 μm. Electrode-skin interface impedance (EII) was recorded as the insertion force was applied on the MAE. The insertion process of the MAE was also simulated by the finite element method. Results showed that MAE could

pH-sensitive poly(lactide-co-glycolide) nanoparticle composite microcapsules for oral delivery of insulin.

This study proposes a new concept of pH-sensitive poly(lactide-co-glycolide) (PLGA) nanoparticle composite microcapsules for oral delivery of insulin. Firstly, insulin-sodium oleate complex was prepared by the hydrophobic ion pairing method and then encapsulated into PLGA nanoparticles by the emulsion solvent diffusion method. In order to reduce the burst release of insulin from PLGA nanoparticles and deliver insulin to specific gastrointestinal regions, hence to enhance bioavailability of insulin, the PLGA nanoparticles were further encapsulated into Eudragit(®) FS 30D to prepare PLGA nanopar

Towards clinical testing of a single-administration tetanus vaccine based on PLA/PLGA microspheres.

The availability of single-administration vaccines would assist in the control of global mortality caused by infectious diseases where protection can be achieved only upon repeated immunisations with appropriate vaccines. Biodegradable microspheres of poly(lactide-co-glycolide) have been studied pre-clinically for this purpose and shown to be promising for several protein and sub-unit antigens. In view of preparing a microsphere-based tetanus vaccine for clinical trials, final candidate vaccine-formulations were pre-clinically optimised here. Specifically, the importance of particular material

Refined Dual-Stage Release of Lidocaine by Erodible Microneedles for Postoperative Analgesia.

Effective postoperative pain management is crucial for major and minor surgeries, yet conventional approaches often fall short due to limitations in duration, dosing, or convenience. This study introduces an innovative microneedle-based drug delivery system highlighted by a refined dual-stage release profile to enhance surgical wound analgesia. The proposed erodible microneedle (EMN) contains two polymer networks, namely polyvinyl pyrrolidone (PVP) and poly(lactic-co-glycolic acid) (PLGA), and carries the effective local analgesic lidocaine hydrochloride (LIDH). The PVP network rapidly dissolv

Development and in vitro evaluation of a lignin-PLGA nanocarrier for florfenicol delivery.

Florfenicol (FF) is a widely used antimicrobial in veterinary medicine because of its broad antimicrobial activity, although it has certain limitations and raises concerns about the development of antimicrobial resistance genes. These limitations highlight the need to explore novel drug with controlled release systems to enhance the therapeutic efficacy of FF, while minimizing the potential for resistance development. This study introduces an innovative approach for the design, synthesis, and evaluation of lignin-poly(lactic-co-glycolic) acid (PLGA)-FF nanoparticles. By leveraging the properti

Characterization of Dye-Loaded Poly(lactic-co-glycolic acid) Nanoparticles by Comprehensive Two-Dimensional Liquid Chromatography Combining Hydrodynamic and Reversed-Phase Liquid Chromatography.

Analytical methods for the assessment of drug-delivery systems (DDSs) are commonly suitable for characterizing individual DDS properties, but do not allow determination of several properties simultaneously. A comprehensive online two-dimensional liquid chromatography (LC × LC) system was developed that is aimed to be capable of characterizing both nanoparticle size and encapsulated cargo over the particle size distribution of a DDS by using one integrated method. Polymeric nanoparticles (NPs) with encapsulated hydrophobic dyes were used as model DDSs. Hydrodynamic chromatography (HDC) was used

Development of uniform fenofibrate-loaded biodegradable microparticle by membrane emulsification.

Fenofibrate has shown therapeutic effects on diabetic retinopathy. However, fenofibrate can be rapidly cleared from the eye after a single intravitreal injection. Here, we aim to develop fenofibrate loaded PLGA microparticles (Feno-MP) with high drug loading and sustained in vitro release up to 6 months suitable for intravitreal injection. First, orthogonal array experimental design was applied for formulation optimization. The selected formulation parameters were used to formulate Feno-MP using homogenization method and direct membrane emulsification method. Both methods generated Feno-MP wit
