Transcriptome Analysis of Populus tomentosa Reveals Molecular Mechanisms of Jasmonate Signaling Pathway on Arsenic Tolerance Induced by Arbuscular Mycorrhizal Fungi.

Arsenic (As) pollution poses serious threats to soil ecosystems and plant growth. Arbuscular mycorrhizal fungi (AMF) have been confirmed to enhance As tolerance in host plants, making mycorrhizal-assisted phytoremediation a promising and practical strategy for remediating As-contaminated soils. Populus tomentosa Carr. (Chinese white poplar) is a native fast-growing woody species suitable for phytoremediation in East Asia. However, comprehensive transcriptomic analyses focusing on the molecular mechanisms by which AMF improve As tolerance in this species remain limited. In this study, a pot-bas

Ameliorative potential of Populus alba leaf powder against hexaflumuron exposure in Nile tilapia: immune-antioxidant, biochemical, histological, and transcriptomic analysis.

Contamination of the aquatic bodies with pesticides is a serious issue that hinders the aquaculture industry worldwide. Preventing aquatic pollution is a challenge, and finding eco-friendly strategies could help to overcome such a problem. Herein, we studied the antagonistic potential of dietary fortification of white poplar (Populus alba; PA) leaf powder against chronic hexaflumuron (HX) toxicity in Nile tilapia (Oreochromis niloticus). Fish (n = 200; 36.20 ± 1.55 g) were eventually grouped into four groups with five replicates and kept for 60 days. The C (control) and PA groups were fed basa

Population Structure and Spatial Distribution Pattern of Populus euphratica Riparian Forest Under Environmental Heterogeneity Along the Tarim River, Northwest China.

Populus euphratica Oliv. (Euphrates poplar), as the dominant tree species of desert riparian forests along the Central Asian inland rivers, plays a critical role in protecting arid land ecosystems. In recent decades, climate change and excessive water resources utilization activities have led to the environmental degradation of desert riparian forests along the Tarim River in northwest China. Understanding the forest stand structure and spatial distribution pattern provide important guidance for monitoring forest dynamics in support of sustainable management. However, few studies have examined

Properties analysis of transcription factor gene TasMYB36 from Trichoderma asperellum CBS433.97 and its heterogeneous transfomation to improve antifungal ability of Populus.

The transcription of TasMYB36 in the biocontrol species T. asperellum was upregulated in four different pathogenic fermentation broths, suggesting that TasMYB36 plays an important role in the response to biotic stresses. Seventy-nine MYB transcription factors that were homologous to TasMYB36 from six sequenced Trichoderma genomes were analyzed. They were distributed in fourteen clades in the phylogenetic tree. The 79 MYBs contained 113 DNA binding domains, and their amino acid sequences were conserved and were different to those in plants. The promoters of 79 MYBs contained 1374 cis-regulators

Characterization, expression profiling, and functional analysis of a Populus trichocarpa defensin gene and its potential as an anti-Agrobacterium rooting medium additive.

The diverse antimicrobial properties of defensins have attracted wide scientific interest in recent years. Also, antimicrobial peptides (AMPs), including cecropins, histatins, defensins, and cathelicidins, have recently become an antimicrobial research hotspot for their broad-spectrum antibacterial and antifungal activities. In addition, defensins play important roles in plant growth, development, and physiological metabolism, and demonstrate tissue specificity and regulation in response to pathogen attack or abiotic stress. In this study, we performed molecular cloning, characterization, expr

Plant-herbivore interactions in a trispecific hybrid swarm of Populus: assessing support for hypotheses of hybrid bridges, evolutionary novelty and genetic similarity.

UNLABELLED: Natural systems of hybridizing plants are powerful tools with which to assess evolutionary processes between parental species and their associated arthropods. Here we report on these processes in a trispecific hybrid swarm of Populus trees. Using field observations, common garden experiments and genetic markers, we tested the hypothesis that genetic similarities among hosts underlie the distributions of 10 species of gall-forming arthropods and their ability to adapt to new host genotypes. KEY FINDINGS: the degree of genetic relatedness among parental species determines whether hy

Metal-based-oxide nanoparticles assisted the in vitro culture growth of Populus alba as micronutrients: essential metabolic processes and genetic stability.

The present study evaluates the in vitro culture growth rate of Populus alba upon using nano metal-based-oxides such as hematite (Fe2O3 NPs), zinc oxide (ZnO NPs), and manganese oxide (Mn2O3 NPs) nanoparticles as analogues of three primary micronutrients such as iron (Fe), zinc (Zn), and manganese (Mn), which exist in soil as micronutrients. Herein, the in vitro culture growth rate was investigated using three different concentrations (i.e., 20, 40, and 60 mg L-1) of as-prepared metal oxide nanoparticles compared to the control. In addition, the as-prepared nanoparticles have been prepared via

Light and the maintenance of photosynthetic competence in leaves of Populus balsamifera L. during short-term exposures to high concentrations of sulfur dioxide.

Leaves of Populus balsamifera grown under full natural sunlight were treated with 0, 1, or 2 μl SO2·1(-1) air under one of four different photon flux densities (PFD). When the SO2 exposures took place in darkness or at 300 μmol photons·m(-2)·s(-1), sulfate accumulated to the levels predicted by measurements of stomatal conductance during SO2 exposure. Under conditions of higher PFD (750 and 1550 μmol·m(-2)·s(-1)), however, the predicted levels of accumulated sulfate were substantially higher than those obtained from anion chromatography of the leaf extracts. Light-and CO2-saturated capacity as

Integrated Multi-Omics Exploration of the Molecular Mechanisms Underlying Differential Responses to Saline-Alkaline Stress in Leaves of Populus simonii × P. nigra.

Populus simonii × P. nigra is a valuable model for studying woody plant responses to saline-alkaline stress, however the molecular mechanisms underlying its adaptation remain poorly understood. Here, integrated transcriptomic and metabolomic analyses were used to examine the responses in leaves to low (50 mmol/L) and high (200 mmol/L) alkaline salt stress over 7 and 14 days. Physiological assessments revealed that high-concentration stress inhibited growth, induced oxidative damage, elevated malondialdehyde (MDA) content, enhanced antioxidant enzyme activity (SOD, POD, CAT), and promoted osmor

eYGFPuv-Assisted Transgenic Selection in Populus deltoides WV94 and Multiplex Genome Editing in Protoplasts of P. trichocarpa × P. deltoides Clone '52-225'.

Although CRISPR/Cas-based genome editing has been widely used for plant genetic engineering, its application in the genetic improvement of trees has been limited, partly because of challenges in Agrobacterium-mediated transformation. As an important model for poplar genomics and biotechnology research, eastern cottonwood (Populus deltoides) clone WV94 can be transformed by A. tumefaciens, but several challenges remain unresolved, including the relatively low transformation efficiency and the relatively high rate of false positives from antibiotic-based selection of transgenic events. Moreover,

Effects of overproduction of condensed tannins and elevated temperature on chemical and ecological traits of genetically modified hybrid aspens (Populus tremula × P. tremuloides).

Gene transfer techniques offer new possibilities to study regulation of phenolic pathways and the defensive role of phenolics. Hybrid aspen lines (Populus tremula × tremuloides) that overexpress the PtMYB134 transcription factor were used to study the effects of condensed tannin production on plant physiology and plant defenses. The MYB134 protein activates all the known genes of the biosynthetic pathway for condensed tannins (CTs), so overexpression of MYB134 was expected to increase CT concentration in all tissues of the plants. Two out of three MYB134 overexpression lines (46 and 54) accumu

Expression pattern of the poplar GSTU family members in response to Alternaria alternate and PdbGSTU10 confers A. alternate resistance to Populus davidiana × P. bolleana.

Plant tau glutathione S-transferase (GSTU) is a kind of multiple functions enzyme, but its specific roles in poplar disease resistance remain uncertain. In this study, 27 PdbGSTU-encoding genes from Populus davidiana × P. bollena were cloned and their protein architectures and phylogenetic relationships were subsequently analyzed. Expression analysis revealed that PdbGSTUs were differentially expressed under Alternaria alternate infection. Then, the PdbGSTU10 was further induced by phytohormones and H2O2, especially salicylic acid (SA), indicating its potential role in the pathogen defense of

Spatiotemporal analysis of copper homeostasis in Populus trichocarpa reveals an integrated molecular remodeling for a preferential allocation of copper to plastocyanin in the chloroplasts of developing leaves.

Plastocyanin, which requires copper (Cu) as a cofactor, is an electron carrier in the thylakoid lumen and essential for photoautotrophic growth of plants. The Cu microRNAs, which are expressed during Cu deprivation, down-regulate several transcripts that encode for Cu proteins. Since plastocyanin is not targeted by the Cu microRNAs, a cofactor economy model has been proposed in which plants prioritize Cu for use in photosynthetic electron transport. However, defects in photosynthesis are classic symptoms of Cu deprivation, and priorities in Cu cofactor delivery have not been determined experim

Heterologous expression of the Hsp24 from Trichoderma asperellum improves antifungal ability of Populus transformant Pdpap-Hsp24 s to Cytospora chrysosperma and Alternaria alternate.

The tolerance of plants to biotic and abiotic stresses could be improved by transforming with fungal resistance-related genes. In this study, the cDNA sequence (GenBank Acc. No. KP337939) of the resistance-related gene Hsp24 encoding the 24 kD heat shock protein was obtained from the biocontrol fungus Trichoderma asperellum ACCC30536. The promoter region of Hsp24 contained many cis-regulators related to stresses response, such as "GCN4" and "GCR1" etc. Hsp24 transcription in T. asperellum was up-regulated under six different environmental stresses, compared with the control. Metabolome and Lipidome Profiles of Populus × canescens Twig Tissues During Annual Growth Show Phospholipid-Linked Storage and Mobilization of C, N, and S.

The temperate climax tree species Fagus sylvatica and the floodplain tree species Populus × canescens possess contrasting phosphorus (P) nutrition strategies. While F. sylvatica has been documented to display P storage and mobilization (Netzer et al., 2017), this was not observed for Populus × canescens (Netzer et al., 2018b). Nevertheless, changes in the abundance of organic bound P in gray poplar trees indicated adaptation of the P nutrition to different needs during annual growth. The present study aimed at characterizing seasonal changes in metabolite and lipid abundances in gray poplar an
