Coronavirus diversity and SARS-CoV-2 exposure at the wildlife-human interface in Northern Italy.

BACKGROUND: Members of the Coronaviridae family infect humans as well as domestic and wild animals. Over the past three decades, three members of this family, all with zoonotic origins, have caused significant epidemics or pandemics (SARS, MERS, and COVID-19). Despite the spread of SARS-CoV-2 being primarily driven by human-to-human transmission, various animal species are susceptible to infection and may contribute to viral circulation. Aim of this work was to monitor coronavirus (CoV) infections in wild mammals in the Emilia-Romagna region (RER), Italy, using a combined approach of molecular

Genetic Characterization of Sarcoptes scabiei from Black Bears (Ursus americanus) and Other Hosts in the Eastern United States.

Since the early 1990s there has been an increase in the number of cases and geographic expansion of severe mange in the black bear (Ursus americanus) population in Pennsylvania. Although there are 3 species of mites associated with mange in bears, Sarcoptes scabiei has been identified as the etiologic agent in these Pennsylvania cases. Historically, S. scabiei-associated mange in bears has been uncommon and sporadic, although it is widespread and relatively common in canid populations. To better understand this recent emergence of sarcoptic mange in bears in Pennsylvania and nearby states, we

Infrequency of gross skin lesions among Panamanian forest mammals with cutaneous leishmaniasis.

Natural leishmanial infections among feral mammals in the Republic of Panama were studied in relation to gross skin alterations. Three dermatotropic species of Leishmania, L. braziliensis, L. mexicana and L. hertigi, isolated from 230 sylvatic animals of 5 mammalian orders were involved. In 205 (89%) of the cases there were no gross skin alterations. Cryptic infections of all three leishmanial species among wild animals were confirmed by culture and hamster inoculation. This type of infection was always present in the sloths, Choloepus hoffmanni and Bradypus infusatcus, infected with L. brazil

Wnts are endothelial cell-derived PKD1/PKD2-dependent autocrine/paracrine vasodilators.

BACKGROUND: Wingless/Int-1 (Wnts) proteins are canonical Frizzled receptor ligands. Recent evidence indicates that some Wnts, including Wnt9b and Wnt5a, bind to polycystin 1 (PKD1), a transmembrane protein which can couple to polycystin 2 (PKD2) to form a non-selective cation channel. The functional significance of Wnts binding to PKD1 is unclear. Here, we tested the hypothesis that Wnts act through PKD1/PKD2 channels on endothelial cells (ECs) to regulate arterial contractility and blood pressure and investigated the cellular source and secretory regulation of vasoactive Wnt proteins. METHOD

Fatty acyl donor selectivity in membrane bound O-acyltransferases and communal cell fate decision-making.

The post-translational modification of proteins with lipid moieties confers spatial and temporal control of protein function by restricting their subcellular distribution or movement in the extracellular milieu. Yet, little is known about the significance of lipid selectivity to the activity of proteins targeted for such modifications. Membrane bound O-acyl transferases (MBOATs) are a superfamily of multipass enzymes that transfer fatty acids on to lipid or protein substrates. Three MBOATs constitute a subfamily with secreted signalling molecules for substrates, the Wnt, Hedgehog (Hh) and Ghre

The influence of fibre in the diet on growth rates and the digestibility of nutrients in the greater cane rat (Thryonomys swinderianus).

The greater cane rat Thryonomys swinderianus is a coprophagous rodent in which fermentation occurs in the large caecum. The extent to which a 45% increase in the fibre component of the diet influenced growth rates of cane rats and the digestibility of nutrients and energy was investigated in two feeding trials. Higher fibre levels in the diet reduced the digestibility of dry matter, protein and fat, while animals digested fibre components (neutral-detergent fibre, acid detergent fibre, hemicellulose and cellulose) with a comparable efficiency to those maintained on a low fibre diet. In one of

Cercopithifilaria spp. of dogs: little known but prevalent filarioids beneath the skin.

Filarioids of the genus Cercopithifilaria are little studied, yet widespread parasites, that are relatively unique in being one of the very few nematodes transmitted by hard ticks. These filarioids live in the subcutis while microfilariae are found in the dermis. Definitive hosts include domestic dogs as well as a wide range of vertebrates, such as ruminants, non-human primates, murids, marsupials, porcupines, viverrids, bears and lagomorphs. The genus Cercopithifilaria contains three taxa (i.e. C. bainae, C. grassii and a yet undescribed species, namely Cercopithifilaria sp. II) that are know

Traumatic, degenerative, and developmental lesions in wolves and coyotes from Saskatchewan.

A retrospective review was done of traumatic and osseous lesions in 241 wolves (Canis lupus) and 316 coyotes (Canis latrans) necropsied at the University of Saskatchewan between 1971 and 1990. Most lesions were the result of interspecific conflict. The most frequently occurring lesion in wolves was fracture of one or more bones, primarily ribs. Lesions were healed in most cases and appeared to be compatible with injuries caused by prey animals. One wolf, found dead, died as a result of thoracic trauma. Limb and skull fractures were less common. Fractures were uncommon in coyotes. The most freq

Exposure to selected pathogens in wild mammals from a rescue and rehabilitation center in southern Italy.

Wildlife is at the same time a reservoir and sentinel of numerous infections for humans and domestic animals. For this reason, wildlife rehabilitation centers represent an opportunity to carry out surveillance against the most varied infections. In this work, wild animals (canids, mustelids, erinaceids, and cervids) hospitalized at a first aid center in southern Italy were sampled and tested with multispecies ELISAs and rapid tests against a panel of pathogens, including viruses, bacteria, and parasites. Foxes and wolves were exposed to Brucella canis, Coxiella burnetii, canine coronavirus, an

An ER Assembly Line of AMPA-Receptors Controls Excitatory Neurotransmission and Its Plasticity.

Excitatory neurotransmission and its activity-dependent plasticity are largely determined by AMPA-receptors (AMPARs), ion channel complexes whose cell physiology is encoded by their interactome. Here, we delineate the assembly of AMPARs in the endoplasmic reticulum (ER) of native neurons as multi-state production line controlled by distinct interactome constituents: ABHD6 together with porcupine stabilizes pore-forming GluA monomers, and the intellectual-disability-related FRRS1l-CPT1c complexes promote GluA oligomerization and co-assembly of GluA tetramers with cornichon and transmembrane AMP

The epidemiology of blastomycosis in dogs: north central Wisconsin, USA.

The epidemiological features of 59 consecutive cases of blastomycosis in domestic dogs, from a single veterinary practice in Eagle River, Wisconsin over a 3-year period, were examined by owner interview. The control sample included: (i) all porcupine quill-injured dogs during this time period (outdoor exposed dogs); and (ii) every sixth dog receiving a rabies vaccination during the second year of study (representative of all dogs in the practice). The estimated mean annual incidence of blastomycosis in dogs in this region was 1420:100,000. Blastomycosis cases were more likely to reside within

Prediction and design optimization of mechanical properties for rubber fertilizer hose reinforced with helically wrapped nylon.

Predicting and optimizing the mechanical performance of the helically wound nylon-reinforced rubber fertilizer hose (HWNR hose) is crucial for enhancing the performance of hose pumps. This study aims to enhance the service life of HWNR hoses and the efficiency of liquid fertilizer transport. First, a finite element simulation model and a mathematical model were established to analyze the influence of fiber layer arrangement on the maximum shear strain on the coaxial surface (MSS) and the reaction force on the extrusion roller (RF). For the first time, the Crested Porcupine Optimizer algorithm

Effects of the pathological E200K mutation on human prion protein: A computational screening and molecular dynamics approach.

The human prion protein gene (PRNP) is mapped to the short arm of chromosome 20 (20pter-12). Prion disease is associated with mutations in the prion protein-encoding gene sequence. Earlier studies found that the mutation G127V in the PRNP increases protein stability. In contrast, the mutation E200K, which has the highest mutation rate in the prion protein, causes Creutzfeldt-Jakob disease (CJD) in humans and induces protein aggregation. We aimed to identify the structural mechanisms of E200k and G127V mutations causing CJD. We used a variety of bioinformatic algorithms, including SIFT, PolyPhe

Molecular survey of Ehrlichia canis and Coxiella burnetii infections in wild mammals of southern Italy.

Ehrlichiosis and Q fever caused by the intracellular bacteria Ehrlichia canis and Coxiella burnetii, respectively, are tick-borne diseases with zoonotic potential and widespread geographical distribution. This study investigated the prevalence of both infections in wild mammals in southern Italy. Tissue samples obtained from the red fox (Vulpes vulpes), European badger (Meles meles), gray wolf (Canis lupus), beech marten (Martes foina), and crested porcupine (Hystrix cristata) were processed for molecular detection of both pathogens. E. canis was detected in 55 out of 105 (52 %) red foxes and

Zygotic Porcn paternal allele deletion in mice to model human focal dermal hypoplasia.

In mouse and humans, the X-chromosomal Porcupine homolog (Porcn) gene is required for the acylation and secretion of all 19 Wnt ligands, thus representing a bottleneck in the secretion of Wnt ligands. In humans, mutations in PORCN cause the X-linked dominant syndrome Focal Dermal Hypoplasia (FDH, OMIM#305600). This disorder is characterized by ecto-mesodermal dysplasias and shows a highly variable phenotype, potentially due to individual X chromosome inactivation patterns. To improve the understanding of human FDH, we have established a mouse model by generation of Porcn heterozygous animals c

Unusual causes of fatal upper aerodigestive tract obstruction in wild Bottlenose Dolphins (Tursiops aduncus).

Necropsy examination of dolphins living in Gulf St Vincent, Australia is routinely undertaken to enable the evaluation of disease processes and to provide rapid medicolegal assessment of any inflicted and/or accidental injuries. Two Indo-Pacific Bottlenose Dolphins (Tursiops aduncus) are reported to demonstrate conditions that may result in unexpected death involving upper airway compromise by quite unusual mechanisms. In the first case an adult male was found with extensive soft tissue trauma suggesting human interaction. At necropsy, death was due instead to upper airway obstruction from an

Elucidating the role of DEAF1 in neurodevelopment and shared molecular pathways in high-risk autism genes using cortical organoids.

Neurodevelopmental disorders (NDDs), including autism spectrum disorder (ASD) and intellectual disability (ID), are genetically heterogeneous. DEAF1 has emerged as a key NDD risk gene, with pathogenic variants linked to DEAF1-associated neurodevelopmental disorder (DAND), but its role in human neurodevelopment remains unclear. Human cortical organoids (hCOs) provide a physiologically relevant model that recapitulates fetal brain development with an authentic human genetic background. Here, we show that a DEAF1 mutation in human embryonic stem cells disrupts chromatin accessibility at neuronal
