Effect of host responses on the pathogenicity of strains of Porphyromonas gingivalis.

Porphyromonas gingivalis is implicated in the etiology of periodontitis. Strains of P. gingivalis have been classified as invasive or noninvasive based on their ability to form abscesses in a mouse model. The purpose of this study was to investigate the ability of P. gingivalis strains to cause abscesses and periodontal bone loss in an experimental rat model and the effect of serum and salivary responses on the pathogenicity of these strains. Subcutaneous injection of animals with P. gingivalis 33277, A7A1-28, W50 or 381 resulted in abscesses in a higher percentage of mice than rats. P. gingiv

Protective immunity to Porphyromonas gingivalis infection in a murine model.

The mouse abscess model has been used extensively to demonstrate protection after challenge with periodontopathic organisms. In the present study, an outer membrane (OM) preparation of P. gingivalis ATCC 33277 was used to immunize BALB/c mice prior to challenge with live P. gingivalis organisms. This OM preparation, particularly at the highest dose level of 100 micrograms/immunization, was able to induce high levels of specific antibody and subsequent protective immunity. Protection in all immunized mice was noted by the rapid healing of the primary lesions, a low incidence of secondary lesion

Porphyromonas gingivalis lipopolysaccharide increases blood-brain barrier permeability via ferroptosis.

OBJECTIVES: Ferroptosis, a newly characterized form of regulated cell death, has been associated with the dysfunction of the blood-brain barrier (BBB). Nevertheless, its precise role in BBB disruption caused by Porphyromonas gingivalis (P. gingivalis) lipopolysaccharide (LPS) remains unclear. DESIGN: Experimental periodontitis was induced by injecting P. gingivalis LPS into the left and right sides of the palatal gingiva between the maxillary first and second molars every two days for eight weeks, then changes in brain tissue in BBB permeability, and ferroptosis‑related factors were assessed.

Capsular K-antigen Coats Outer Membrane Vesicles of Porphyromonas gingivalis.

Periodontal disease is an inflammatory disorder that arises from dysbiosis of the subgingival microbiota, with Porphyromonas gingivalis acting as a keystone pathogen in the shift from health to disease. P. gingivalis employs multiple strategies to subvert host immune defenses, and its capsular K-antigen serves as a key virulence determinant. Here, a pre-adsorbed antiserum (pAds106) was generated by removing nonspecific antibodies using cells from a K-antigen-null mutant (W83ΔPG0106), resulting in exceptional specificity for the P. gingivalis K1-antigen. Immunofluorescence analysis revealed tha

Low DYNLL1 Resists Porphyromonas gingivalis-Induced Epithelial Jamming-Like State.

Incomplete regeneration of the junctional epithelium (JE) following periodontal therapy is associated with persistent alveolar bone resorption and disease recurrence. While pathogen clearance is essential, the host-intrinsic mechanisms dictating epithelial recovery remain obscure. Here, we reveal that Porphyromonas gingivalis challenge induces an aberrant jamming-like phase transition in the epithelium during the early recovery window, mechanically restricting the epithelial remodeling required for proper JE reconstruction. Through a genome-wide CRISPR-Cas9 screen, we identified Dynein Light C

Pervasive integrative and conjugative elements shape Porphyromonas gingivalis gene repertoires.

BACKGROUND: Porphyromonas gingivalis ( Pg ) is an oral pathobiont that contributes to periodontal disease and has been associated with systemic health conditions. Although Pg is recognized as exhibiting extensive strain-level genomic diversity and recombination, the extent to which mobile elements contribute to this variation, and their relevance to its fitness and virulence, remain incompletely understood. Our recent study of the Pg pangenome revealed diverse accessory defense-associated genes, raising the question of whether these are carried by unrecognized mobile genetic elements (MGEs). I

Association Between Porphyromonas gingivalis and Alzheimer's Disease: A Comprehensive Review.

BACKGROUND: Periodontitis has long been linked to various inflammatory, chronic, and immunological diseases, such as heart disease or diabetes. Recently, there has been increasing scientific interest in the bidirectional relationship that may exist between periodontitis and the presence and progression of Alzheimer's disease (AD), where it is hypothesized that the infiltration of oral microorganisms (mainly Porphyromonas gingivalis) into the bloodstream, which subsequently reaches the brain, causes inflammatory and neurodegenerative processes related to AD. PURPOSE: The purpose of this review

Porphyromonas gingivalis induces Zbp1-mediated macrophages PANoptosis in periodonitis pathophysiology.

Periodontitis is an oral immunoinflammatory disease, and macrophages play a crucial role in its pathophysiology. However, macrophage death during antibacterial activities will exacerbate inflammation and tissue damage. Porphyromonas gingivalis is a major constituent of subgingival biofilm plaques in periodontitis, but the effects and precise molecular mechanisms by which it triggers macrophage death remain unknown. Here we found that P. gingivalis infection notably activated multiple death pathways in bone-marrow-derived macrophages, including pyroptosis, apoptosis and necrosis. Furthermore, u

Splenic abscess caused by Porphyromonas gingivalis septicemia: a case report.

BACKGROUND: The spleen is a vital immune organ in the human body that is involved in both cellular and humoral immunity. It has unique physiological functions that contribute to fighting infections, and typically, infections do not lead to the formation of splenic abscesses, making them rare. CASE PRESENTATION: A 49-year-old healthy female presented with high fever, chills, and abdominal pain after tooth extraction. Abdominal enhanced computed tomography (CT) and enhanced magnetic resonance imaging (MRI) confirmed splenic infection and multiple abscesses. Porphyromonas gingivalis was identifi

Commensal Neisseria Inhibit Porphyromonas Gingivalis Invasion of Gingival Epithelial Cells.

PURPOSE: Periodontal disease is caused by periodontal invasion by pathogens such as Porphyromonas gingivalis. Although recent metagenomic analyses have shown that oral commensal bacteria are abundant in the mouth of healthy individuals, few studies have experimentally verified the benefits and functions of oral commensal bacteria in periodontal diseases. In this study, we focused on Neisseria among the oral commensal bacteria and aimed to experimentally verify its effects on P. gingivalis invasion. MATERIALS AND METHODS: We evaluated the inhibitory effect of Neisseria spp. on P. gingivalis in

Porphyromonas gingivalis-induced hematogenous disseminated severe pneumonia: a case report.

Porphyromonas gingivalis, a gram-negative oral anaerobe among more than more than 500 bacterial species that colonizing the oral cavity, is involved in the pathogenesis and prototypic polybacterial consortium of periodontitis. It is mainly found in oral infections and rarely present in other organ diseases. Here, we describe a 43-year-old man with underlying diabetes who developed hematogenous disseminated severe pneumonia after P. gingivalis had invaded the blood. Next-generation sequencing of early alveolar lavage fluid and blood samples confirmed the diagnosis. The patient's lung infection

Endotoxin tolerance induced by Porphyromonas gingivalis lipopolysaccharide alters macrophage polarization.

Endotoxin tolerance refers to a state refractory to subsequent lipopolysaccharide (LPS) stimulations following a primary LPS exposure. To study the relationship between endotoxin tolerance and macrophage polarization, endotoxin tolerance was induced by 1 μg/mL LPS from the periodontal pathogen, Porphyromonas gingivalis (P. gingivalis), in peritoneal macrophages (PMs) and bone marrow-derived macrophages (BMDMs). Repeated P. gingivalis LPS challenges increased the quantities of CD206+ PMs, while the number of CD86+CD206+ PMs was reduced compared with the non-tolerant group (p < 0.05). However, t

Computational Design of a Multi-Epitope Vaccine Against Porphyromonas gingivalis.

Porphyromonas gingivalis is a Gram-negative pathogenic bacterium associated with chronic periodontitis. The development of a chimeric peptide-based vaccine targeting this pathogen could be highly beneficial in preventing oral bone loss as well as other severe gum diseases. We applied a computational framework to design a multi-epitope-based vaccine candidate against P. gingivalis. The vaccine comprises epitopes from subunit proteins prioritized from the P. gingivalis reference strain (P. gingivalis ATCC 33277) using several reported vaccine properties. Protein-based subunit vaccines were prior

Porphyromonas gingivalis Infection Induces Amyloid-β Accumulation in Monocytes/Macrophages.

Abnormal accumulation of amyloid-β (Aβ) in the brain is the most significant pathological hallmark of Alzheimer's disease (AD). We have found that chronic systemic exposure to lipopolysaccharide of Porphyromonas gingivalis (P. gingivalis) induces the accumulation of Aβ in the brain of middle-aged mice. On the other hand, recent research has shown that circulating Aβ is transferred into the brain; however, the involvement of chronic systemic P. gingivalis infection in the peripheral Aβ metabolism is unknown. We hypothesized that chronic P. gingivalis infection expands Aβ pools in peripheral inf

Structural characterization of Porphyromonas gingivalis enoyl-ACP reductase II (FabK).

Enoyl-acyl carrier protein (ACP) reductase II (FabK) is a critical rate-limiting enzyme in the bacterial type II fatty-acid synthesis (FAS II) pathway. FAS II pathway enzymes are markedly disparate from their mammalian analogs in the FAS I pathway in both structure and mechanism. Enzymes involved in bacterial fatty-acid synthesis represent viable drug targets for Gram-negative pathogens, and historical precedent exists for targeting them in the treatment of diseases of the oral cavity. The Gram-negative organism Porphyromonas gingivalis represents a key causative agent of the costly and highly

Unprimed, M1 and M2 Macrophages Differentially Interact with Porphyromonas gingivalis.

Porphyromonas gingivalis is a keystone pathogen in the development of chronic periodontitis. Tissue macrophages are amongst the first immune cells to respond to bacteria and depending on the cytokine profile at the infection site, macrophages are primed to react to infection in different ways. Priming of naive macrophages with IFN-γ produces a classical pro-inflammatory, antibacterial M1 macrophage after TLR ligation, whereas priming with IL-4 induces an anti-inflammatory tissue-repair M2 phenotype. Previous work has shown that M1 are preferentially generated in gingival tissue following infec

Role of Porphyromonas gingivalis gingipains in multi-species biofilm formation.

BACKGROUND: Periodontal diseases are polymicrobial diseases that cause the inflammatory destruction of the tooth-supporting (periodontal) tissues. Their initiation is attributed to the formation of subgingival biofilms that stimulate a cascade of chronic inflammatory reactions by the affected tissue. The Gram-negative anaerobes Porphyromonas gingivalis, Tannerella forsythia and Treponema denticola are commonly found as part of the microbiota of subgingival biofilms, and they are associated with the occurrence and severity of the disease. P. gingivalis expresses several virulence factors that m

Translocation of Porphyromonas gingivalis infected monocytes and associated cellular responses.

Porphyromonas gingivalis (P. gingivalis), an important periodontal pathogen in adult chronic periodontitis, has been reported to colocalize in human atheromatous lesions. We have studied the phagocytosis and survival of P. gingivalis in human monocytes, together with the cellular responses of infected human monocytes. Human monocytes were cocultured with P. gingivalis and the external bacteria were killed with metronidazole and gentamycin. Localization of P. gingivalis in cells was studied by transmission electron microscopy (TEM). The survival of P. gingivalis was determined by lysing the mon

Histidine kinase-mediated production and autoassembly of Porphyromonas gingivalis fimbriae.

Porphyromonas gingivalis, a Gram-negative oral anaerobe, is strongly associated with chronic adult periodontitis, and it utilizes FimA fimbriae to persistently colonize and evade host defenses in the periodontal crevice. The FimA-related gene cluster (the fim gene cluster) is positively regulated by the FimS-FimR two-component system. In this study, comparative analyses between fimbriate type strain ATCC 33277 and fimbria-deficient strain W83 revealed differences in their fimS loci, which encode FimS histidine kinase. Using a reciprocal gene exchange system, we established that FimS from W83 i

Nuclear targeting of Porphyromonas gingivalis W50 protease in epithelial cells.

Porphyromonas gingivalis is an important pathogen associated with destructive periodontal disease and is able to invade the epithelial cell barrier. Its cysteine proteases are recognized as major virulence factors, and in this study, we examined the interaction of the arginine-specific protease with epithelial cells in culture. Three cell lines (KB, HeLa, and SCC4) were incubated with strain W50 culture supernatant; stained with monoclonal antibody 1A1, which recognizes an epitope on the adhesin (beta) component of the cysteine protease-adhesin (alpha/beta) heterodimer; and viewed using immuno
