Choroidal innervation in primate eyes.

Arteries and arterioles of the choroid are surrounded by numerous nerve fibers staining for nitric oxide synthase (NOS) and vasoactive intestinal peptide (VIP). In most mammalian eyes these nerve fibers derive from the pterygopalatine ganglion via the facial nerve. Stimulation of the facial nerve causes vasodilation of the choroidal vasculature. In primates with a well developed fovea centralis there are ganglion cells in the choroidal stroma which in human eyes amount to around 2000. The postganglionic nerve fibers of these choroidal ganglion cells (CGC) join the perivascular nerve fiber plex

Oral transmission of primate lentiviruses.

Oral transmission of human immunodeficiency virus type 1 (HIV-1) is well documented in children who become infected postnatally through breast milk. In contrast, epidemiologic surveys have yielded conflicting data regarding oral HIV-1 transmission among adults, even though case reports have described seroconversion and the development of AIDS in adults whose only risk was oral-genital contact. To study oral virus transmission in primate models, we exposed rhesus macaques of various ages to cell-free simian immunodeficiency virus (SIV), including uncloned and molecularly cloned viruses. In neon

[Primates in studies on the safety of drugs].

Primates are more and more frequently used in the evaluation of the safety aspects of drugs. Because of their similar phylogenic profile, these animals appear to be more predictive than other species (rat, mouse, rabbit or dog) used in toxicological studies. This relationship is confirmed by anatomical, metabolic and physiological observations. Primates are rarely used in teratogenesis studies, because the methods and materials involved are too complex. However, even in these studies, primates appear to be the most predictive species.

Primate viral diseases in perspective.

The recent occurrence of fatal Herpesvirus simiae (B virus) infection in human subjects has again focused the attention of primatologists on this virus. B virus, however, is only one of a number of viral diseases that plays a role in primate colony management. This report is to emphasize to the primatologist a number of viruses other than H. simiae, with high morbidity and mortality rates, of importance for health management of nonhuman primate animal colonies. This concept is supported by the recent occurrence in colonies of nonhuman primates of simian hemorrhagic fever virus, SA8, herpesviru

Experimental cryptosporidiosis in a primate model.

Cryptosporidium causes a disease in infant macaques that is clinically, histologically, and microbiologically indistinguishable from that seen in young children. A reproducible experimental model of cryptosporidiosis has been developed in pigtailed macaques (Macaca nemestrina) and used to studied the infectious dose of oocysts and the effect of inoculum size on severity of disease. Inoculation with either 2 x 10(5) or 10 oocysts via nasogastric tube resulted in clinical enteritis and the fecal passage of large numbers of cryptosporidial oocysts in all four primates studied. The size of the ino

Primate model for onchocerciasis research.

A major impediment to progress in research in onchocerciasis has been the lack of a suitable animal model. However, chimpanzees can be reliably infected by injection of living third-stage larvae of Onchocerca volvulus, and develop a pattern of infection that closely resembles that seen in humans. This includes the formation of nodules by adult worms, the subcutaneous distribution of microfilariae and the apparent lack of development of resistance to infection after repeated larval challenge. Ocular lesions resembling those in humans have not been observed in animals studied so far, but this ma

Primate experiments on oral respiration.

Oral respiration associated with obstruction of the nasal airway is a common finding among patients seeking orthodontic treatment. The primate experiments reported here are part of a series designed to test some of the current hypotheses regarding the relationship between mouth breathing and dental malocclusions, that is, between deviations in orofacial muscle recruitment and jaw morphogenesis. Mouth-breathing was developed in the animals of this experiment by obstruction of the nasal passages with silicon nose plugs. The experiments showed that the monkeys adapted to nasal obstruction in diff

Myxoma of bone in a nonhuman primate.

A myxoma of the mandibles in an adult female Macaca mulatta is described. The left mandible was markedly enlarged by the tumor, which extended from the symphysis to the temporomandibular joint. The tumor had extended a short distance across the symphysis into the right mandible. It was composed of a glistening mucinous material. Radiographs of the tumor showed a large expansile radiolucent lesion of the left mandible without soft tissue involvement. Light microscopy demonstrated capillaries, stellate shaped cells with processes up to 15 mu long, very few other cell types, and an amorphous grou

Primate models of postural disorders.

Dorsal column lesions in the high cervical region of the monkey result in severe defects of movements projected into space and contactual orienting reactions of the forelimbs. The hindlimbs are less affected provided a pathway through the lateral columns, Morin's tract, remains intact. Interruption of this pathway results in a defect of hindlimb function similar to that of the forelimbs. Cerebellar ablations in monkeys result in postural and movement disorders, including hypotonia of limb extensor muscles. An important mechanism underlying the hypotonia is a depression of the responses to musc

Encephalomyocarditis virus (EMCV): An overlooked threat to primates.

Many emerging zoonotic viruses pose major risks to animal and human health, with most recent epidemics of viral origin. This review focuses on encephalomyocarditis virus (EMCV), a member of the Picornaviridae family and the Cardiovirus genus with significant but largely underrecognized zoonotic potential. While EMCV has not yet caused major public health crises, its broad host range and rodent reservoir suggest a wider ecological impact. It is typically detected only during high-fatality outbreaks, particularly in domestic animals and non-human primates (NHPs), which are among the most vulnera

Psychedelic studies in nonhuman primates: Past and future.

Studies of serotonergic or 'classic' psychedelics in nonhuman primates (NHPs) have provided valuable information about the drugs' effects on the brain and behavior in closely related species to humans. Psychedelics induce characteristic changes to both spontaneous and operant behaviors in NHPs, though variability exists in the different effects reported by different studies; this variability could be due to factors like differences across drugs, differences in dose ranges across studies, and inter-individual variability in drug responsiveness. Several effects of psychedelics in NHPs mirror tho

Testing pulmonary physiology in ventilated non-human primates.

BACKGROUND: Animal models of respiratory viral infections are essential for investigating disease pathogenesis and the efficacy of antivirals and vaccine candidates. A major limitation in the research of respiratory diseases in animal models is correlating clinically relevant changes in pulmonary physiology with cellular and molecular mechanistic studies. Few animal models have captured and correlated physiologic changes in lung function and immune response within same experiment, which is critical given the heterogeneous nature of lung disease due to viral infections. In ventilated human pati

Variation of thoracolumbar vertebral morphology in anthropoid primates.

OBJECTIVES: Morphological variation among extant primates in the lumbar vertebral column is well studied. However, knowledge concerning the thoracic spine, an important region responsible for supporting and facilitating movement in the upper trunk, remains relatively scarce. Consequently, our comprehension of the functional differentiation exhibited throughout the thoracolumbar vertebral column among various primate species remains constrained. In this study, we examined patterns of morphological variation in the thoracolumbar vertebral column of extant hominoids, cercopithecoids, and Ateles.

DT-109 ameliorates nonalcoholic steatohepatitis in nonhuman primates.

Nonalcoholic steatohepatitis (NASH) prevalence is rising with no pharmacotherapy approved. A major hurdle in NASH drug development is the poor translatability of preclinical studies to safe/effective clinical outcomes, and recent failures highlight a need to identify new targetable pathways. Dysregulated glycine metabolism has emerged as a causative factor and therapeutic target in NASH. Here, we report that the tripeptide DT-109 (Gly-Gly-Leu) dose-dependently attenuates steatohepatitis and fibrosis in mice. To enhance the probability of successful translation, we developed a nonhuman primate

Myocardial Changes in Diabetic and Nondiabetic Nonhuman Primates.

Diabetic human patients have increased risk of heart failure compared to healthy subjects. The underlying mechanisms for this are not fully understood, and to help develop improved treatment strategies, well-characterized animal models are essential. To investigate cardiac dysfunction in diabetes, this study evaluated myocardial changes in 10 aging rhesus monkeys with and without diabetes. Based on evaluation of plasma glycosylated hemoglobin and glucose, 7 of 10 rhesus macaques had diabetes for a minimum of 11 months, while 3 of 10 were categorized as nondiabetic. A detailed histological exam

BioSphincters to treat Fecal Incontinence in Nonhuman Primates.

Loss of anorectal resting pressure due to internal anal sphincter (IAS) dysfunctionality causes uncontrolled fecal soiling and leads to passive fecal incontinence (FI). The study is focused on immediate and long-term safety and potential efficacy of bioengineered IAS BioSphincters to treat passive FI in a clinically relevant large animal model of passive FI. Passive FI was successfully developed in Non-Human Primates (NHPs) model. The implantation of autologous intrinsically innervated functional constructs resolved the fecal soiling, restored the resting pressure and Recto Anal Inhibitory Ref

Sylvatic cycles of arboviruses in non-human primates.

Arboviruses infecting people primarily exist in urban transmission cycles involving urban mosquitoes in densely populated tropical regions. For dengue, chikungunya, Zika and yellow fever viruses, sylvatic (forest) transmission cycles also exist in some regions and involve non-human primates and forest-dwelling mosquitoes. Here we review the investigation methods and available data on sylvatic cycles involving non-human primates and dengue, chikungunya, Zika and yellow fever viruses in Africa, dengue viruses in Asia and yellow fever virus in the Americas. We also present current putative data t
