Effect of retinol status on retinol-binding protein biosynthesis rate and translatable messenger RNA level in rat liver.

Studies were conducted to explore the role of retinol in the control of the rate of synthesis of plasma retinol-binding protein (RBP) in the liver of the rat. Previous studies have shown that nutritional retinol status strongly influences RBP secretion from the liver cell. Both the in vivo relative rate of RBP synthesis and the in vitro translatable level of RBP-specific mRNA were examined in normal vitamin A, retinol-depleted, and retinol-repleted rats. The relative rate of RBP synthesis was estimated by measuring the extent of incorporation of [3H]leucine, [3H]lysine, and [3H]phenylalanine a

Structures and functions of enzymes involved in primary and secondary metabolite biosynthesis mediated by amino-group carrier protein.

Thermus thermophilus, an extremely thermophilic bacterium, synthesizes lysine through a novel type of 2-aminoadipate (AAA) pathway. The pathway starts from the synthesis of homocitrate using 2-oxoglutarate and acetyl-CoA. Homocitrate is converted to 2-oxoadipate by sequential reactions similar to the corresponding reactions in the tricarboxylic acid cycle. AAA formed by the transamination of 2-oxoadipate is loaded on amino-group carrier protein (AmCP) via isopeptide bond linkage. AAA is successively converted to lysine on AmCP by several reactions, and lysine is finally released from the carri

SbWRKY62 Coordinates With SbERF096 to Activate Pterostilbene Biosynthesis and Targets a Xylanase Inhibitor Protein for Anthracnose Resistance in Sorghum.

Sorghum anthracnose caused by Colletotrichum sublineola (C. sublineola) is a destructive disease leading to severe crop damages. Pterostilbene is a phytoalexin that contributes to defence responses in sorghum, but the transcriptional regulation of its biosynthesis remained poorly understood. Here, we characterised two transcription factors, SbERF096 and SbWRKY62, that are induced upon C. sublineola infection, with significantly higher expression levels in the resistant cultivar SC748-5 than the susceptible cultivar BTx623. SbERF096 directly activates SbSTS1, which encodes stilbene synthase, vi

Physiological copper exposure in Jurkat cells induces changes in the expression of genes encoding cholesterol biosynthesis proteins.

Copper is an essential micronutrient that functions as an enzymatic cofactor in a wide range of cellular processes. Although adequate Cu levels are essential for normal metabolism, excess Cu can be toxic to cells. Cellular responses to copper deficiency and overload involve changes in the expression of genes directly and indirectly involved in copper metabolism. However little is known on the effect of physiological copper concentration on gene expression changes. In the current study we aimed to establish whether the expression of genes encoding enzymes related to cholesterol (hmgcs1, hmgcr,

Structural basis of the interaction of MbtH-like proteins, putative regulators of nonribosomal peptide biosynthesis, with adenylating enzymes.

The biosynthesis of nonribosomally formed peptides (NRPs), which include important antibiotics such as vancomycin, requires the activation of amino acids through adenylate formation. The biosynthetic gene clusters of NRPs frequently contain genes for small, so-called MbtH-like proteins. Recently, it was discovered that these MbtH-like proteins are required for some of the adenylation reactions in NRP biosynthesis, but the mechanism of their interaction with the adenylating enzymes has remained unknown. In this study, we determined the structure of SlgN1, a 3-methylaspartate-adenylating enzyme

Lipid droplet-associated proteins are involved in the biosynthesis and hydrolysis of triacylglycerol in Mycobacterium bovis bacillus Calmette-Guerin.

Mycobacteria store triacylglycerols (TGs) in the form of intracellular lipid droplets (LDs) during hypoxia-induced nonreplicating persistence. These bacteria are phenotypically drug-resistant and therefore are believed to be the cause for prolonged tuberculosis treatment. LDs are also associated with bacilli in tuberculosis patient sputum and hypervirulent strains. Although proteins bound to LDs are well characterized in eukaryotes, the identities and functions of such proteins have not been described in mycobacteria. Here, we have identified five proteins: Tgs1 (BCG3153c), Tgs2 (BCG3794c), BC

Biosynthesis and differential processing of two pools of amyloid-beta precursor protein in a physiologically inducible neuroendocrine cell.

The amyloid-beta precursor protein (APP) is linked to Alzheimer's disease through its pathological proteolytic processing in the secretory pathway. Nevertheless, surprisingly little is known about the biosynthesis of endogenous APP. We therefore decided to investigate the intracellular fate of newly synthesized APP in a physiologically inducible neuroendocrine cell, the Xenopus intermediate pituitary melanotrope cell. We found that the level of both APP mRNA and protein was about threefold induced in the activated cells of black-adapted animals. Intriguingly, two pools of APP were found, only

The Ssc protein of enteric bacteria has significant homology to the acyltransferase Lpxa of lipid A biosynthesis, and to three acetyltransferases.

The Ssc protein, a novel essential protein affecting the function of the enterobacterial outer membrane, matched in a protein homology search best with LpxA (UDP-N-acetylglucosamine 3-hydroxymyristoyl transferase), the enzyme which catalyzes the first step of lipid A biosynthesis. The corresponding genes, located 0.56 kb apart, were 46.7% identical. The search also revealed homology to the bacterial acetyltransferases LacA and NodL, as well as to a hypothetical protein Yglm. The region of residues 109-149 Ssc displayed the highest homology and was also homologous with another bacterial acetylt

Transcriptome analysis reveals the promoting mechanism of corn steep liquor protein hydrolysate on cordycepin biosynthesis of Cordyceps militaris.

Cordycepin is an important nucleoside analogue and the principal active compound found in Cordyceps militaris. It exhibits anticancer, antiviral, immunomodulatory, and various other biological activities. However, the low cordycepin yields and high manufacturing costs limit its large-scale application. To develop a low-cost nutrient regulator to improve cordycepin production, we investigated the effect of corn steep liquor protein hydrolysate (CSLPH) on cordycepin biosynthesis during submerged fermentation of C. militaris, and the response mechanism of C. militaris to CSLPH utilization was rev

Redox/ROS regulation of lipopolysaccharide-induced mitogen-activated protein kinase (MAPK) activation and MAPK-mediated TNF-alpha biosynthesis.

Redox and ROS regulation of MAPK-mediated TNF-alpha biosynthesis is not well characterized. It was hypothesized that the involvement of the MAPK pathway in regulating LPS-mediated TNF-alpha secretion is redox-dependent, NF-kappaB-sensitive and attenuated by N-acetyl-L-cysteine (NAC) and other antioxidants. In alveolar epithelial cells, LPS induced a time- and dose-dependent phosphorylation of MAPK(p38). This was associated with the activation of MAPK-activated protein kinase, which phosphorylated the small heat-shock protein, Hsp27. MAPK(p38) inhibition (SB-203580) abrogated LPS-induced TNF-al

Valorization of Spirulina residue via black soldier fly larvae: Unraveling metabolic rewiring for optimized protein, lipid, and vitamin biosynthesis.

This study evaluated the potential of black soldier fly larvae (BSFL) for bioconverting Spirulina residue (SR) and the regulatory impacts of SR on BSFL quality. Multiscale analyses revealed that SR significantly improved the growth performance of BSFL, with a 9.8% increase in final larval weight and a 28.8% enhancement in bioconversion efficiency. Nutritionally, SR supplementation elevated the essential amino acid index by 28.0%, driven by a remarkable enrichment of branched-chain amino acids (e.g., valine increased by 1160%). The n-3 polyunsaturated fatty acid (PUFA) content increased by 23.9

The dual-function protein complex SnRK2.6-WRKY70 synergistically enhances the tanshinones biosynthesis and drought stress adaptation in Salvia miltiorrhiza.

Long-term domestication and cultivation have led to the differentiation of Salvia miltiorrhiza genetic resources in different regions of China, accompanied by differences in the accumulation of pharmacological components represented by tanshinones in the roots. Tanshinone, as a key biomarker with significant therapeutic effects on cardiovascular and cerebrovascular diseases, has become the primary target for improvement in S. miltiorrhiza breeding. It is particularly important to deeply analyze the differentiation of tanshinones biosynthetic pathways in S. miltiorrhiza of different ecotypes, s

Endothelin-1 depletion of cartilage oligomeric matrix protein modulates pulmonary artery superoxide and iron metabolism-associated mitochondrial heme biosynthesis.

This study examines if heme biosynthesis-associated iron metabolism is regulated in pulmonary arteries by endothelin-1 (ET1) potentially through modulating cartilage oligomeric matrix protein (COMP) availability. Our studies in organoid-cultured endothelium-rubbed bovine pulmonary arteries (BPAs) observed COMP depletion by siRNA or hypoxia increases NOX2 and superoxide and depletes mitochondrial SOD2. ET1 also increases superoxide in a manner that potentially impairs mitochondrial heme biosynthesis. In this study, organoid culture of BPA with ET1 (10 nM) increases superoxide in the mitochondri

Analysis of CFB, a cytokinin-responsive gene of Arabidopsis thaliana encoding a novel F-box protein regulating sterol biosynthesis.

Protein degradation by the ubiquitin-26S proteasome pathway is important for the regulation of cellular processes, but the function of most F-box proteins relevant to substrate recognition is unknown. We describe the analysis of the gene Cytokinin-induced F-box encoding (CFB, AT3G44326), identified in a meta-analysis of cytokinin-related transcriptome studies as one of the most robust cytokinin response genes. F-box domain-dependent interaction with the E3 ubiquitin ligase complex component ASK1 classifies CFB as a functional F-box protein. Apart from F-box and transmembrane domains, CFB conta

Recessiveness and dominance in barley mutants deficient in Mg-chelatase subunit D, an AAA protein involved in chlorophyll biosynthesis.

Mg-chelatase catalyzes the insertion of Mg2+ into protoporphyrin IX at the first committed step of the chlorophyll biosynthetic pathway. It consists of three subunits: I, D, and H. The I subunit belongs to the AAA protein superfamily (ATPases associated with various cellular activities) that is known to form hexameric ring structures in an ATP-dependant fashion. Dominant mutations in the I subunit revealed that it functions in a cooperative manner. We demonstrated that the D subunit forms ATP-independent oligomeric structures and should also be classified as an AAA protein. Furthermore, we add

CREB phosphorylation and melatonin biosynthesis in the rat pineal gland: involvement of cyclic AMP dependent protein kinase type II.

Phosphorylation of cyclic AMP response element binding protein (CREB) at amino acid serine 133 appears as an important link between the norepinephrine (NE)-induced activation of second messenger systems and the stimulation of melatonin biosynthesis. Here we investigated in the rat pineal gland: 1) the type of protein kinase that mediates CREB phosphorylation: and 2) its impact on melatonin biosynthesis. Immunochemical or immunocytochemical demonstration of serine133-phosphorylated cyclic AMP regulated element binding protein (pCREB) and radioimmunological detection of melatonin revealed that o

Biosynthesis of paf-acether. Activators of protein kinase C stimulate cultured mast cell acetyltransferase without stimulating paf-acether synthesis.

Antigen stimulation of cultured bone-marrow-derived mast cells sensitized with specific monoclonal IgE induced cell degranulation and paf-acether (paf; 1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine) biosynthesis via the deacylation/acetylation (remodelling) pathway. Phorbol myristate acetate (PMA; 20-100 ng/ml) triggered only acetyltransferase activation, without concomitant lyso-paf (1-O-alkyl-sn-glycero-3-phosphocholine) and paf formation. A low concentration of PMA (5 ng/ml) potentiated antigen-induced degranulation, acetyltransferase activation and paf formation by about 30% but did not c

Biosynthesis of Complex Phenazines Involves Dimerization and Amination Catalyzed by a Versatile Nuclear Transport Factor 2‑like Superfamily Protein.

Phenazine natural products represent a structurally diverse class of aromatic alkaloids with broad-spectrum biological activities. Among these compounds, phenazine dimers have attracted considerable attention owing to their intricate molecular architectures and potent antibacterial and anticancer properties. Although the biosynthesis of the core phenazine scaffold is well characterized, the enzymatic postmodifications, especially those governing dimerization processes that yield structurally varied diphenazines, remain poorly understood. In this study, we elucidate the complete biosynthesis of
