Quadruple quorum-sensing inputs control Vibrio cholerae virulence and maintain system robustness.

Bacteria use quorum sensing (QS) for cell-cell communication to carry out group behaviors. This intercellular signaling process relies on cell density-dependent production and detection of chemical signals called autoinducers (AIs). Vibrio cholerae, the causative agent of cholera, detects two AIs, CAI-1 and AI-2, with two histidine kinases, CqsS and LuxQ, respectively, to control biofilm formation and virulence factor production. At low cell density, these two signal receptors function in parallel to activate the key regulator LuxO, which is essential for virulence of this pathogen. At high ce

Negative control of quorum sensing by RpoN (sigma54) in Pseudomonas aeruginosa PAO1.

In Pseudomonas aeruginosa PAO1, the expression of several virulence factors such as elastase, rhamnolipids, and hydrogen cyanide depends on quorum-sensing regulation, which involves the lasRI and rhlRI systems controlled by N-(3-oxododecanoyl)-L-homoserine lactone and N-butyryl-L-homoserine lactone, respectively, as signal molecules. In rpoN mutants lacking the transcription factor sigma(54), the expression of the lasR and lasI genes was elevated at low cell densities, whereas expression of the rhlR and rhlI genes was markedly enhanced throughout growth by comparison with the wild type and the

Chitinolytic activity in Chromobacterium violaceum: substrate analysis and regulation by quorum sensing.

Quorum sensing control mediated by N-acyl homoserine lactone (AHL) signaling molecules has been established as a key feature of the regulation of exoenzyme production in many gram-negative bacteria. In Chromobacterium violaceum ATCC 31532 a number of phenotypic characteristics, including production of the purple pigment violacein, hydrogen cyanide, antibiotics, and exoproteases are known to be regulated by the endogenous AHL N-hexanoyl-L-homoserine lactone (HHL). In this study we show that C. violaceum produces a set of chitinolytic enzymes whose production is regulated by HHL. The chitinolyti

Deciphering bacterial universal language by detecting the quorum sensing signal, autoinducer-2, with a whole-cell sensing system.

Bacteria communicate with neighboring bacteria of the same species or of other species by means of chemical signaling molecules. The concentration of such signaling molecules is proportional to the bacterial population size; upon reaching a threshold concentration, corresponding to a threshold cell density, certain specialized genes are expressed. This system of communication among bacteria is known as quorum sensing (QS). QS regulates diverse behaviors, such as formation of biofilms and production of pathogenic factors. Autoinducer-2 (AI-2) is a QS signaling molecule that is used for interspe

Characterization of a quorum-sensing communication based on gamma-butyrolactones in Rhodococcus erythropolis.

Quorum sensing (QS) enables bacteria to coordinate collective behaviors, including the production of secondary metabolites with potential biotechnological applications, through the synthesis and detection of small signaling molecules. In Streptomyces, QS is well-described and mainly mediated by 2,3-disubstituted γ-butyrolactones (GBLs), which play key roles in the regulation of secondary metabolism and spore production. Despite their importance, GBL-based QS systems remain poorly characterized in other actinomycetal genera. Here, we investigated the distribution, structure, and function of GBL

Identification and Characterization of Negative Regulators of Rgg1518 Quorum Sensing in Streptococcus pneumoniae.

Streptococcus pneumoniae is an agent of otitis media, septicemia, and meningitis and remains the leading cause of community-acquired pneumonia regardless of vaccine use. Of the various strategies that S. pneumoniae takes to enhance its potential to colonize the human host, quorum sensing (QS) is an intercellular communication process that provides coordination of gene expression at a community level. Numerous putative QS systems are identifiable in the S. pneumoniae genome, but their gene-regulatory activities and contributions to fitness have yet to be fully evaluated. To contribute to assess

A novel quorum sensing regulator LuxT contributes to the virulence of Vibrio cholerae.

Vibrio cholerae is a waterborne bacterium that primarily infects the human intestine and causes cholera fatality. Quorum sensing (QS) negatively regulates the expression of V. cholerae virulence gene. However, the primary associated mechanisms remain undetermined. This investigation identified a new QS regulator from the TetR family, LuxT, which increases V. cholerae virulence by directly inhibiting hapR expression. HapR is a master QS regulator that suppresses virulence cascade expression. The expression of luxT increased 4.8-fold in the small intestine of infant mice than in Luria-Bertani br

Andrographolide Inhibits Biofilm and Virulence in Listeria monocytogenes as a Quorum-Sensing Inhibitor.

Listeria monocytogenes is a major foodborne pathogen that can cause listeriosis in humans and animals. Andrographolide is known as a natural antibiotic and exhibits good antibacterial activity. We aimed to investigate the effect of andrographolide on two quorum-sensing (QS) systems, LuxS/AI-2 and Agr/AIP of L. monocytogenes, as well as QS-controlled phenotypes in this study. Our results showed that neither luxS expression nor AI-2 production was affected by andrographolide. Nevertheless, andrographolide significantly reduced the expression levels of the agr genes and the activity of the agr pr

Joint effects of antibiotics and quorum sensing inhibitors on resistance development in bacteria.

Quorum sensing inhibitors (QSIs) are promising alternatives to antibiotics. While QSIs have great application potential in a variety of fields, their joint effects with antibiotics on bacteria, especially on antibiotic resistance mutations, remain largely unexplored. Herein, we report the joint effects of four commonly used antibiotics and two QSIs on bacterial growth and resistance mutations in E. coli. It was found that QSIs presented antagonistic or additive effects with antibiotics on bacterial growth, and more importantly, QSIs exhibited an attenuating effect on antibiotic-induced resista

Synthesis and Evaluation of Indole-Based Autoinducers on Quorum Sensing in Vibrio cholerae.

Vibrio cholerae (V. cholerae) uses the autoinducer CAI-1 (cholera autoinducer 1) and several linked quorum sensing systems in order to efficiently sense its ever-changing environment and optimally coordinate population-wide gene expression. Indole has been reported as an important signal that is sensed by V. cholerae, and here, we report the synthesis and evaluation of a focused library of synthetic indole-CAI-1 derivatives as tools to probe quorum sensing (QS) in this human pathogen. Our results show interesting and diverging effects for several conjugates, as compared to CAI-1, on virulence

Effect of the quorum-sensing luxS gene on biofilm formation by Enterococcus faecalis.

Enterococcus faecalis is the species of bacterium most frequently isolated from the root canals of teeth that exhibit chronic apical periodontitis refractory to endodontic treatment. In this study, we evaluated the effect of the S-ribosylhomocysteine lyase (luxS) quorum-sensing gene on E. faecalis biofilm formation by constructing a knockout mutant. The biofilms formed by both E. faecalis and its luxS mutant strain were evaluated using the MTT method. Important parameters that influence biofilm formation, including cell-surface hydrophobicity and the nutrient content of the growth medium, were

Specific quorum sensing-disrupting activity (A QSI) of thiophenones and their therapeutic potential.

Disease caused by antibiotic resistant pathogens is becoming a serious problem, both in human and veterinary medicine. The inhibition of quorum sensing, bacterial cell-to-cell communication, is a promising alternative strategy to control disease. In this study, we determined the quorum sensing-disrupting activity of 20 thiophenones towards the quorum sensing model bacterium V. harveyi. In order to exclude false positives, we propose a new parameter (AQSI) to describe specific quorum sensing activity. AQSI is defined as the ratio between inhibition of quorum sensing-regulated activity in a repo

A Comprehensive Review on Various Mechanisms of Quorum Sensing Inhibition by Gold Nanoparticles.

Resistance to multiple drugs and biofilm-associated infections are major challenges to global health. The ability of gold nanoparticles (AuNPs) to disrupt bacterial quorum sensing (QS) mechanisms, reduce virulence, and inhibit biofilm formation without inducing resistance makes them a promising alternative for treating these infections. The potential of AuNPs as quorum-sensing inhibitors (QSIs) is reviewed, focusing on their ability to control bacterial communication pathways in pathogens, including Pseudomonas aeruginosa, Staphylococcus aureus, and Escherichia coli. AuNPs reduce QS by binding

Xanthoxylin as a quorum-sensing inhibitor of Aeromonas hydrophila with promising therapeutic effects.

Aeromonas hydrophila (A. hydrophila) is widely spread in different aquatic environments and still poses a serious threat to the health of aquatic animals. Nevertheless, with the emergence of drug-resistant A. hydrophila, antibiotics have shown limited efficacy as primary therapeutic agents, and novel treatment strategies are urgently needed. The increasing understanding of anti-virulence and anti-quorum-sensing (QS) strategies offers new hope for drug discovery against A. hydrophila infections, which aim not to inhibit or kill bacteria but to reduce pathogenicity by reducing the production or

Disruption of quorum sensing and biofilm formation by lawsone in gram-negative bacteria.

Antimicrobial resistance is a major global health threat, exacerbated by bacterial virulence mechanisms such as quorum sensing (QS) and biofilm formation, which enhance pathogenicity and reduce antibiotic susceptibility. Targeting QS offers a promising antivirulence strategy that may attenuate pathogenic traits without imposing selective pressure. In this study, lawsone was evaluated for anti-QS and antibiofilm activity against Chromobacterium violaceum, Pseudomonas aeruginosa, and Serratia marcescens. Lawsone significantly inhibited QS-regulated virulence factors, with reductions in violacein

Research Progress on the Quorum Sensing System of Acinetobacter baumannii and Its Inhibitors.

The escalating problem of bacterial drug resistance poses a severe threat to global public health, with Acinetobacter baumannii (A. baumannii) exhibiting particularly high resistance rates that are closely linked to its Quorum Sensing (QS) system. This narrative review synthesizes current knowledge on the A. baumannii QS system, focusing on its essential role in mediating antimicrobial resistance, as well as the latest progress in developing QS inhibitors. Key findings indicate that the A. baumannii QS system enhances bacterial survival by promoting biofilm formation, and regulating the expres

Malleilactone Is a Burkholderia pseudomallei Virulence Factor Regulated by Antibiotics and Quorum Sensing.

Burkholderia pseudomallei, the causative agent of melioidosis, encodes almost a dozen predicted polyketide (PK) biosynthetic gene clusters. Many of these are regulated by LuxR-I-type acyl-homoserine (AHL) quorum-sensing systems. One of the PK gene clusters, the mal gene cluster, is conserved in the close relative Burkholderia thailandensis The B. thailandensis mal genes code for the cytotoxin malleilactone and are regulated by a genetically linked LuxR-type transcription factor, MalR. Although AHLs typically interact with LuxR-type proteins to modulate gene transcription, the B. thailandensis

Quorum sensing in biofilms--how to destroy the bacterial citadels or their cohesion/power?

Biofilms or microbial communities formed by adherent and cohesive cells on cellular or inert substrata (like medical devices), are involved in ≈ 60% of all infections and characterized by moderate intensity symptoms, chronic evolution and resistance to antibiotics. Biofilms' pathogenicity, even of those formed by opportunistic microorganisms, is amplified by two major biofilm characteristics: 1) the increased resistance to antimicrobials; 2) the protection of cells against the host's defence mechanisms. The studies at the molecular level shown that the biofilms formation is controlled by cell-
