Circular RNA-mediated regulatory networks drive oxidative stress and metabolic dysregulation in broiler breast muscle under chronic heat stress.

Chronic heat stress (CHS) disrupts broiler breast muscle physiology homeostasis, yet the underlying molecular mechanisms remain unclear. In this study, 144 male Arbor Acres broilers (28 d old) were randomly allocated into 3 treatment groups: the normal control (NC) group (22 °C, ad libitum), the heat stress (HS) group (22 °C, ad libitum), the pair-fed (PF) group (22 °C, feed matched to HS). After two weeks of CHS, the HS group exhibited elevated ROS production, increased antioxidant enzyme activities (GSH-Px, CAT, SOD), and higher MDA and carbonyl levels, accompanied by pronounced ultrastructu

Integrative whole-transcriptome analysis of circRNAs, lncRNAs, miRNAs, and mRNAs reveals regulatory networks in mouse brain during Toxoplasma gondii infection.

Toxoplasmosis, caused by the protozoan Toxoplasma gondii, is a zoonotic parasite that infects humans and animals worldwide. Although RNAs contribute to many vital biological processes within the central nervous system, their involvement in T. gondii infection remains poorly understood. In this study, we investigated the differential expression of three types of noncoding RNAs and mRNAs in the brain tissues of mice orally infected with 20 cysts of T. gondii Pru strain per mouse during the acute and chronic phases of infection. We used RNA sequencing (RNA-seq) to comprehensively profile the diff

LINC01996 suppresses non-small cell lung cancer proliferation and metastasis by orchestrating the miR-12115/CNRIP1/Ras signaling axis.

Lung cancer is the most prevalent cancer and a major cause of cancer-related mortality worldwide. Long non-coding RNAs (lncRNAs) have been confirmed to be dysregulated in the progression of cancers. In this study, through RNA-next generation deep sequencing, we identified a new lncRNA (LINC01996), which was associated with prognosis of non-small cell lung cancer (NSCLC), downregulated in the NSCLC tissues and cell lines. We demonstrated that overexpressing LINC01996 inhibited NSCLC cell proliferation, EMT, and migration. Xenograft mouse models further validated the suppression role of LINC0199

Competitive virus and host RNAs: the interplay of a hidden virus and host interaction.

During virus infection, viral RNAs and mRNAs function as blueprints for viral protein synthesis and possibly as pathogen-associated molecular patterns (PAMPs) in innate immunity. Here, considering recent research progress in microRNAs (miRNAs) and competitive endogenous RNAs (ceRNAs), we speculate that viral RNAs act as sponges and can sequester endogenous miRNAs within infected cells, thus cross-regulating the stability and translational efficiency of host mRNAs with shared miRNA response elements. This cross-talk and these reciprocal interactions between viral RNAs and host mRNAs are termed

Deciphering testicular aging through whole transcriptome sequencing and ceRNA network analysis in cattle.

Male reproductive performance of livestock is widely recognized as a critical economic trait. The testes serve as the core organ of male reproduction, their aging impairs the semen quality and service life of breeding bulls. However, the impact of genes and non-coding RNAs (ncRNAs) on bovine testicular aging remains unclear. In this study, histomorphological and whole transcriptome sequencing analysis was performed on testicular tissues collected from mature (3-year-old) and aged (10-year-old) Wandong cattle. Histological results demonstrated age-related differences in the tubular compartment

Integrated Transcriptome Landscape of mRNAs, lncRNAs, circRNAs, and miRNAs Reveals Molecular Regulatory Networks of Sex Differentiation in the Zig-Zag Eel (Mastacembelus armatus).

The zig-zag eel (Mastacembelus armatus) exhibits sexual dimorphism in growth patterns. Identifying the genes involved in sex differentiation is a crucial step toward achieving single-sex breeding and serves as a vital foundation for elucidating the XY sex determination mechanism in M. armatus. This study measured the morphological characteristics of male and female M. armatus and found that males were significantly superior to females in body weight and nearly all morphological indices. Subsequently, whole-transcriptome sequencing was performed on the gonads of adult males and females, identif

Lesion-specific circular RNA and mRNA signatures in secondary progressive multiple sclerosis.

Circular RNAs (circRNAs) are single-stranded RNAs with a covalently closed loop structure which have been increasingly recognised as stable and functionally relevant regulators of gene expression via sponging miRNAs. Recently, studies have revealed their roles in human diseases, subsequently emerging as potential biomarkers. Multiple sclerosis (MS) is an inflammatory, neurodegenerative, and immune-related disease of the central nervous system characterised by distinct lesion types that reflect different stages of brain tissue injury and repair. Our study conducted a comprehensive transcriptomi

CircFN1 modulates IL-1β-induced osteoarthritis through the miR-19b-3p/PTEN signaling axis.

Osteoarthritis (OA) is a degenerative joint disease marked by cartilage breakdown, chondrocyte apoptosis, and extracellular matrix (ECM) degradation. Non-coding RNAs, particularly circular RNAs (circRNAs), are increasingly recognized as key regulators of OA pathogenesis. This study reveals that circFN1 (hsa_circ_0058152) may act as a competing endogenous RNA, sponging miR-19b-3p and suppressing PTEN expression. This mechanism promotes chondrocyte apoptosis, enhances inflammatory responses, and accelerates ECM degradation, driving OA progression. We investigated the circFN1/miR-19b-3p/PTEN axis

Recent advances in the role of long non-coding RNAs in ovarian cancer chemoresistance.

Ovarian cancer (OC) remains a leading cause of cancer-related mortality among women, with a persistently low five-year survival rate. Currently, the primary treatment modalities for OC include surgery and systemic chemotherapy. However, despite initial therapeutic efficacy, the majority of patients eventually experience relapse and drug resistance, severely limiting clinical outcomes. Consequently, there is an urgent need to elucidate the molecular mechanisms underlying OC and develop innovative strategies to overcome chemoresistance. Long non-coding RNAs (lncRNAs), emerging as novel transcrip

The Prospective Regulatory Functions of lncRNAs and Their ceRNA Networks in the Development of Motor Neurons and Associated Diseases.

Motor neurons form a highly specialized network composed of α-, β-, and γ-subtypes that coordinate skeletal muscle activity. Motor neuron diseases (MNDs), including amyotrophic lateral sclerosis (ALS) and spinal muscular atrophy (SMA), are characterized by the progressive degeneration of this network, resulting in motor dysfunction. Emerging evidence underscores the significant roles of long non-coding RNAs (lncRNAs) in motor neuron development and disease. However, only a few have been experimentally confirmed as true ceRNA regulators, highlighting the need to differentiate validated mechanis

Circular RNA profiling revealed an evolutionarily conserved circACACA promotes liver lipid metabolism, oxidative stress, and autophagy disorder in a ceRNA manner.

Non-alcoholic fatty liver disease (NAFLD) is a clinical syndrome characterized primarily by hepatocellular steatosis and lipid accumulation, which leads to hepatocyte apoptosis, autophagy, inflammation, and intracellular oxidative stress. NAFLD is recognized as one of the most prevalent and complex chronic liver diseases globally, with its occurrence and associated mortality rates rising swiftly each year. Due to the high similarity between chicken fatty liver syndrome (FLS) and NAFLD, as well as the easy availability of diseased chickens, the chicken is considered an ideal model for studying

MOV10 suppresses circRNA biogenesis by disrupting reverse complementary matches structure to drive hepatocarcinogenesis.

Circular RNAs (circRNAs) are frequently downregulated in hepatocellular carcinoma (HCC), yet the molecular underpinnings of this phenomenon remain poorly understood. Although reverse complementary matches (RCMs) are essential for circRNA biogenesis, how their integrity is regulated in cancer has not been explored. Here, we conducted a systematic analysis correlating the expression of RNA helicases with global circRNA levels in HCC patient cohorts. MOV10 was identified as the top candidate, and its mechanism of regulating circRNAs was verified using various molecular biology approaches. The imp

YTHDF2-regulated hsa_circ_0005882 functions as a sponge for miR-654-3p to suppress nasopharyngeal carcinoma proliferation.

Circular RNAs (circRNAs) are a subclass of non-coding RNAs, playing an important regulatory role in tumor progression. Accumulating evidence has revealed that circRNAs can be regulated by m6A machinery, influencing their expression and biological functions. However, the functions and mechanisms of m6A-mediated circRNAs in nasopharyngeal carcinoma (NPC) remain to be elucidated. In this study, the sequence of hsa_circ_0005882 (circ_0005882) was determined by Sanger sequencing, and its localization in both the cytoplasm and nucleus was confirmed by fluorescence in situ hybridization.
