RECENT CHANGES IN INFECTIOUS DISEASES IN EUROPEAN WILDLIFE.

Many infectious diseases originating from, or carried by, wildlife affect wildlife conservation and biodiversity, livestock health, or human health. We provide an update on changes in the epidemiology of 25 selected infectious, wildlife-related diseases in Europe (from 2010-16) that had an impact, or may have a future impact, on the health of wildlife, livestock, and humans. These pathogens were selected based on their: 1) identification in recent Europe-wide projects as important surveillance targets, 2) inclusion in European Union legislation as pathogens requiring obligatory surveillance, 3

Grouper TRAF5 exerts negative regulation on antiviral immune response against iridovirus.

Tumor necrosis factor receptor-associated factor 5 (TRAF5) is an intracellular protein that binds to the cytoplasmic portion of tumor necrosis factor receptors and mediates the activation of downstream nuclear factor-kappa B (NF-κB), interferon regulatory factor 3, and mitogen activated protein kinase signaling pathways. Compared with other TRAF proteins, TRAF5 is largely unknown in teleosts. In the present study, a TRAF5 homologue (HgTRAF5) from the hybrid grouper (Epinephelus fuscoguttatus♂ × Epinephelus lanceolatus♀) was cloned and characterized. The open reading frame of HgTRAF5 consists o

SGIV envelope protein VP088 facilitated virus replication via interacting with other viral proteins and promoting p62-dependent autophagic degradation of TBK1.

Singapore grouper iridovirus (SGIV), a novel member of the genus Ranavirus, family Iridoviridae, frequently causes a severe disease with high mortality in grouper aquaculture. Although previous findings have demonstrated that SGIV envelope protein VP088 was crucial for its infectivity, the underlying mechanism still remained uncertain. Here, we screened the potential viral proteins that interacted with VP088 during SGIV infection using GFP pull-down assay. Co-immunoprecipitation (Co-IP) assays verified the interactions between VP088 and VP018, VP068, or VP156 in vitro. Moreover, confocal micro

Function analysis of fish Tollip gene in response to virus infection.

Toll-interacting protein (Tollip) is one of the important regulatory proteins of Toll-like receptor (TLR) signaling pathways. In previous studies, a Tollip sequence of grouper (Epinephelus coioides) was identified and the signal transduction functions of Tollip were studied. However, the response of Tollip to virus infection has not been characterized from grouper. In the present paper, the Tollip homolog (EtTollip) from grouper (Epinephelus tauvina) was cloned and its immune response to Singapore grouper iridovirus (SGIV) was investigated. EtTollip shares significant similarities to other mam

Analysis of antiviral functions, differences and apoptotic effects of two novel MHC-Iα genotypes in orange-spotted grouper (Epinephelus coioides).

The Major Histocompatibility Complex (MHC) is a group of tightly linked, highly polymorphic genes that play a critical role in vertebrate immune responses. MHC is closely associated with various diseases, including autoimmune disorders, antigen invasion, and inflammation. Notably, MHC class I (MHC-I) molecules exhibit extensive polymorphism and have the ability to specifically bind to a diverse array of viral antigens, which is crucial for antigen presentation and antiviral immunity. However, studies on the differential functions of different MHC-I genotypes in response to fish viral diseases

Grouper interleukin 15-like acts as a cytokine adjuvant in grouper iridovirus subunit vaccine.

Interleukin (IL) 15 and IL-15-like (IL-15L) are members of the cytokine receptor γ chain (γc) family proteins, which play critical roles in the growth, and differentiation of various cell lineages of the innate and adaptive immune systems. However, the functions of teleost IL-15L upon virus infection still remained unknown. In the current study, a homolog of IL-15L from grouper (Epinephelus coioides) was identified and characterized. Sequence analysis indicated that EcIL-15L contained a conserved IL-15 domain (aa 13-134). Quantitative Reverse Transcription PCR (qRT-PCR) analysis indicated that

Identification and functional characterization of interferon c (IFNc) in orange-spotted grouper (Epinephelus coioides) involved in antiviral response to SGIV and RGNNV infections.

The orange-spotted grouper (Epinephelus coioides) is highly susceptible to viral pathogens such as Singapore grouper iridovirus (SGIV) and red-spotted grouper nervous necrosis virus (RGNNV), resulting in significant economic losses in aquaculture. Type I interferons (IFNs) are pivotal antiviral cytokines in vertebrates, but their functions, particularly their antiviral effects against SGIV and RGNNV infections, remain incompletely understood. Here, we cloned and characterized the orange-spotted grouper ifnc gene (named Ec-ifnc), which encodes a 188 amino acids protein and clusters into a disti

PlCXCL12a differentially modulates SGIV and RGNNV replication during single and co-infection and regulates chemotaxis-associated immune responses.

Chemokines play important roles in coordinating innate immune defense in teleost fish. In this study, a CXCL12a homolog, designated PlCXCL12a, was cloned and characterized from leopard coral grouper (Plectropomus leopardus). PlCXCL12a encodes a conserved CXC chemokine containing a typical SCY domain and showed high sequence similarity to teleost CXCL12a homologs. PlCXCL12a was constitutively expressed in multiple tissues and was significantly induced by Singapore grouper iridovirus (SGIV), red-spotted grouper nervous necrosis virus (RGNNV), and pathogen-associated molecular pattern stimulation

Interferon regulatory factor 6 gene from Oreochromis niloticus is involved in resistance against tilapia lake virus.

Tilapia lake virus (TiLV) is an emerging virus that seriously threatens the tilapia industries worldwide. Interferon regulatory factors (IRFs) are the crucial mediators regulating the response of interferon (IFN) to combat invading viruses. Here, a IRF6 homologs from tilapia (Oreochromis niloticus) were characterized and analyzed. The full length of OnIRF6 mRNA contains a 249 bp 5'untranslated region (UTR), an open reading frame (ORF) of 1473 bp and a 3' UTR of 1209 bp. The OnIRF6 typically shared the conserved domains and phylogenetic relationship with IRF homologs of other species and showed

Modulatory role of tumor necrosis factor receptor type 1-associated death domain protein (TRADD) in innate antiviral immune responses and apoptosis in red-spotted grouper (Epinephelus akaara).

Tumor necrosis factor receptor type 1-associated death domain protein (TRADD) is a unique adaptor molecule involved in multiple receptor-mediated signaling pathways that regulate cell survival and apoptosis in a context-dependent manner. Despite extensive studies on TRADD in mammals, its functional role in teleosts remains poorly understood. To address this gap, the present study characterized a TRADD homolog from red-spotted grouper (Epinephelus akaara, EaTRADD) to determine its function in teleosts. The predicted EaTRADD encoded a putative protein of 291 amino acids, comprising an N-terminal

Grouper IFIT1 inhibits iridovirus and nodavirus infection by positively regulating interferon response.

Interferon-induced protein with tetratricopeptide repeats 1 (IFIT1), one of the interferon stimulated genes (ISGs), is strongly induced by type I interferon (IFN), double-stranded RNAs and virus infection. To investigate the actions of fish IFIT1 in response to virus infection, we cloned an IFIT1 homolog from orange spotted grouper (EcIFIT1) and clarified its function in this study. The full-length cDNA of EcIFIT1 is 1839 bp, which is composed of 436 amino acid (aa) residues, with 77.8% and 22.8% identity to IFIT1 homolog of yellow perch (Perca flavescens) and humans (homo sapiens), respective

PTPN2 modulates antiviral immunity in grouper by promoting TBK1 degradation and suppressing interferon signaling.

As a member of the class I non-receptor protein tyrosine phosphatase family, Tyrosine-protein phosphatase non-receptor type 2 (PTPN2) plays a pivotal role in the regulation of cellular signaling pathways. In this study, we successfully cloned and characterized a PTPN2 homolog from Epinephelus coioides, designated as EcPTPN2. The open reading frame (ORF) of EcPTPN2 spans 1186 nucleotides, encoding a protein of 395 amino acids (Accession number: PV289599). Sequence analysis revealed that EcPTPN2 contains the conserved PTPc domain and exhibits high sequence homology with Cottoperca gobio (93.40 %

TTRAP is a critical factor in grouper immune response to virus infection.

TTRAP (TRAF and TNF receptor-associated protein) is latest identified cytosolic protein that serves as a negative regulator for TNF signaling pathway. In this study, a member of TNF superfamily, TTRAP gene (designed as EcTTRAP) was cloned from grouper, Epinephelus coioides. There was an Exo_endo_phos type domain in EcTTRAP, and it was well conserved when compared with other TTRAPs, especially the endonuclease activity related motifs. EcTTRAP exhibited prominent endonuclease activity against the genome DNA from Escherichia coli, Vibrio vulnificus and E. coli JM109. Intracellular localization re

Nonclassical MHC-Restricted Invariant Vα6 T Cells Are Critical for Efficient Early Innate Antiviral Immunity in the Amphibian Xenopus laevis.

Nonclassical MHC class Ib-restricted invariant T (iT) cell subsets are attracting interest because of their potential to regulate immune responses against various pathogens. The biological relevance and evolutionary conservation of iT cells have recently been strengthened by the identification of iT cells (invariant Vα6 [iVα6]) restricted by the nonclassical MHC class Ib molecule XNC10 in the amphibian Xenopus laevis. These iVα6 T cells are functionally similar to mammalian CD1d-restricted invariant NKT cells. Using the amphibian pathogen frog virus 3 (FV3) in combination with XNC10 tetramers

The TLR9/NF-κB and cGAS/STING pathways mediate the assembly of the NLRP3 inflammasome induced by Singapore grouper iridovirus DNA.

Groupers are economically significant fish in southern China and Southeast Asia, where viral diseases severely hinder the sustainable development of aquaculture. Singapore grouper iridovirus (SGIV) is a highly pathogenic virus in groupers that can cause mass mortality upon infection. NOD-like receptor thermal protein domain associated protein 3 (NLRP3), a crucial component of innate immunity, plays vital roles in various viral infections. However, whether SGIV infection induces NLRP3 inflammasome activation and the precise mechanisms involved remain to be elucidated. In this study, we investig

Molecular cloning, characterization of one key molecule of teleost innate immunity from orange-spotted grouper (Epinephelus coioides): serum amyloid A.

The orange-spotted grouper (Epinephelus coioides), a favorite marine food fish, is widely cultured in China and Southeast Asian countries. However, little is known about its acute phase response (APR) caused by viral diseases. Serum amyloid A (SAA) is a major acute phase protein (APP). In this study, a new SAA homologous (EcSAA) gene was cloned from grouper, E. coioides, by rapid amplification of cDNA ends (RACE) PCR. The full-length cDNA sequence of SAA was 508 bp and contained a 363 bp open reading frame (ORF) coding for a protein of 121 aa. Similar to other fish known SAA genes, the EcSAA g
