Building and Operating a Low-Cost Elevated Carbon Dioxide Growth Chamber to Evaluate Microgreen Physiology under Spaceflight-Relevant CO2 Levels.

Carbon dioxide (CO2) levels aboard crewed spacecraft routinely exceed 3,000 ppm, significantly higher than atmospheric concentrations on Earth. Sustained exposure to extreme CO2 (exCO2) at levels above 3,000 ppm can alter plant growth, physiology, and nutritional composition. Yet, few laboratory systems can reliably reproduce these exCO2 concentrations. This protocol describes an accessible and low-cost method for constructing a benchtop growth chamber capable of maintaining stable CO2 levels above 3,000 ppm for plant experiments. The chamber is created by modifying a benchtop incubator to inc

Endophytic bacteria associated with turmeric, ginger and white radish exhibit dual roles as producers of antibacterial metabolites and carriers of multidrug resistance.

The increasing global burden of antimicrobial resistance (AMR) necessitates the exploration of unconventional microbial niches for novel bioactive compounds. Simultaneously, it is also necessary to evaluate their role in the dissemination of resistant bacteria through the food chain. Endophytic bacteria inhabiting edible plant tissues represent a largely underexplored interface between environmental microbiota and human exposure. In this exploratory study, endophytic bacteria were isolated from fresh rhizomes of Zingiber officinale (ginger), Curcuma longa (turmeric), and Raphanus sativus (whit

Health risk assessment of toxic elements and nitrate in tuberous vegetables: carcinogenic and non-carcinogenic perspectives, monte carlo simulation, and sensitivity analysis.

This study investigated the non-carcinogenic and carcinogenic risks associated with consuming carrots and radishes, focusing on heavy metal and nitrate contamination. Children had an overall hazard index (HI) for radish consumption of 23.77, in contrast to adult males (8.20) and adult females (9.49). The HI value for carrots was 38.81 in children; nevertheless, it was lower in adult males (10.46) and adult females (11.35). Zn had the lowest hazard quotient (HQ) values, ranging from 1.04 to 1.11; however, Cd and Pb showed the highest HQ values, especially in carrots (Cd: 13.05, Pb: 13.63 in chi

Liming effects on cadmium stabilization in upland soil affected by gold mining activity.

To reduce cadmium (Cd) uptake of plants cultivated in heavy metal-contaminated soil, the best liming material was selected in the incubation test. The effect of the selected material was evaluated in the field. In the incubation experimentation, CaCO(3), Ca(OH)(2), CaSO(4).2H(2)O, and oyster shell meal were mixed with soil at rates corresponding to 0, 400, 800, 1600, 3200 mg Ca kg(-1). The limed soil was moistened to 70% of field moisture capacity, and incubated at 25 degrees C for 4 weeks. Ca(OH)(2) was found to be more efficient on reducing soil NH(4)OAc extractable Cd concentration, due to

A green synthesis of Sm doped NiO nano catalyst as dual functional material for photocatalytic degradation and electrochemical sensing.

Samarium-doped nickel oxide (NiO-Sm) nanoparticles (NPs) were synthesized using Raphanus sativus leaf extract as a green reducing and capping agent. NiO nanoparticles showed a characteristic UV-Vis absorption peak at 220 nm with additional bands at 280-322 nm, while Sm doping induced a bathochromic shift to 230 nm and FTIR confirmed metal-oxygen bonding (Sm-O and Ni-O). XRD analysis revealed NiO as the dominant phase with minor cubic Sm2O3 features, and SEM images showed irregular, agglomerated nanoparticles with porous, loosely packed structures in the nano-to-micron range. The Photocatalytic

Brassicaceae-Based Biosolarization Reduces Lettuce Fusarium Wilt (FOLac) in Naturally Infested Soils.

Brassicaceae-based bio-disinfestation is a promising non-chemical option to manage lettuce Fusarium wilt caused by Fusarium oxysporum f. sp. lactucae (FOLac), but evidence integrating pathogen suppression, residue chemistry, and soil microbiome response remains limited. We conducted a greenhouse mesocosm assay with naturally infested soil to compare biofumigation (uncovered soil) and biosolarization (plastic-covered soil for 40 days) using five Brassicaceae residues (Brassica carinata, B. juncea, B. napus, Sinapis alba and Raphanus sativus) applied at two field-equivalent doses. Transcriptome-based gene profiling provides novel insights into the characteristics of radish root response to Cr stress with next-generation sequencing.

Radish (Raphanus sativus L.) is an important worldwide root vegetable crop with high nutrient values and is adversely affected by non-essential heavy metals including chromium (Cr). Little is known about the molecular mechanism underlying Cr stress response in radish. In this study, RNA-Seq technique was employed to identify differentially expressed genes (DEGs) under Cr stress. Based on de novo transcriptome assembly, there were 30,676 unigenes representing 60,881 transcripts isolated from radish root under Cr stress. Differential gene analysis revealed that 2985 uingenes were significantly d

Influence of nitisinone and its metabolites on l-tyrosine metabolism in a model system.

Nitisinone (NTBC) is currently used for the treatment of tyrosinemia type 1, a rare disease. It also exhibits potential in the treatment of other orphan diseases as well as nervous system disorders - this is however limited by its side effects. In all living organisms, NTBC inhibits 4-hydroxyphenylpyruvate dioxygenase activity, thereby affecting l-tyrosine (L-TYR) catabolism, which results in the therapeutic effect. The NTBC metabolites formed in patient's body is one of the causes of its side effects. The influence of NTBC and its metabolites; 2-amino-4-(trifluoromethyl)benzoic acid, 2-nitro-

Differential antifungal and calcium channel-blocking activity among structurally related plant defensins.

Plant defensins are a family of small Cys-rich antifungal proteins that play important roles in plant defense against invading fungi. Structures of several plant defensins share a Cys-stabilized alpha/beta-motif. Structural determinants in plant defensins that govern their antifungal activity and the mechanisms by which they inhibit fungal growth remain unclear. Alfalfa (Medicago sativa) seed defensin, MsDef1, strongly inhibits the growth of Fusarium graminearum in vitro, and its antifungal activity is markedly reduced in the presence of Ca(2+). By contrast, MtDef2 from Medicago truncatula, wh

BldDss Modulates the Functional Role of Streptomyces scabies through Directly Regulating the Biosynthesis of Phytotoxin Thaxtomin and Phytohormone Indole-3-Acetic Acid.

Streptomyces scabies is the first identified common scab-causing pathogen, producing thaxtomin A as a principal virulence factor. Interestingly, S. scabies also produces the phytohormone indole-3-acetic acid (IAA), but how it fine-tunes the production of phytotoxins and phytohormones remains poorly understood. Here, we found that BldDss from S. scabies acts as a dual-function regulator, activating thaxtomin biosynthesis while repressing IAA biosynthesis, both derived from l-tryptophan (Trp). BldDss directly bound to the txtR and txtE promoters to promote thaxtomin biosynthesis. bldDss deletion

Epidemiological drivers of turnip yellows virus and its vector Myzus persicae in Australian canola (Brassica napus) systems: a 3-year study of commercial crops and noncrop vegetation.

BACKGROUND: Turnip yellows virus (TuYV) is one of the most economically important viral pathogens of Brassica napus (canola) in Australia. However, the relative contributions of noncrop reservoirs, aphid vectors and virus genetic variation to TuYV epidemiology remain only partly resolved. Myzus persicae is the principal vector of TuYV and a major pest of B. napus production globally. In Australia, both M. persicae and TuYV persist between growing seasons in weeds and volunteer B. napus plants that can facilitate early-season crop infection. RESULTS: We conducted a 3-year study across southeas

Biotransformation of bisphenol A in vivo and in vitro by laccase-producing Trametes hirsuta La-7: Kinetics, products, and mechanisms.

Bisphenol A (BPA) is a ubiquitous endocrine disruptor that poses adverse human health risks. Herein, biotransformation kinetics, products, and mechanisms of BPA undergoing a laccase-producing Trametes hirsuta La-7 metabolism were for the first time reported. Strain La-7 could completely biotransform ≤0.5 mmol·L-1 BPA within 6 d in vivo. Notably, its extracellular crude laccase solution (ECLS) and intracellular homogenized mycelium (HM) only required 6 h to convert 85.71% and 84.24% of 0.5 mmol·L-1 BPA in vitro, respectively. The removal of BPA was noticeably hampered by adding a cytochrome P-4

Light intensity and production parameters of phytocenoses cultivated on soil-like substrate under controlled [correction of controled] environment conditions.

To increase the degree of closure of biological life support systems of a new generation, we used vermicomposting to involve inedible phytomass in the intra-system mass exchange. The resulting product was a soil-like substrate, which was quite suitable for growing plants (Manukovsky et al. 1996, 1997). However, the soil like substrate can be regarded as a candidate for inclusion in a system only after a comprehensive examination of its physical, chemical, and other characteristics. An important criterion is the ability of the soil-like substrate to supply the necessary mineral elements to the

Assessing the potential of turnip rape and radish cover crops as trap crops for Psylliodes chrysocephala in commercial field production conditions.

BACKGROUND: Cabbage stem flea beetle (CSFB; Psylliodes chrysocephala) is a major pest of winter oilseed rape (WOSR; Brassica napus), during autumn. Previous studies have shown some species of Brassicaceae, for example, turnip rape (Brassica rapa) and radish (Raphanus sativus var longipinnatus), to be attractive for this pest. Farmers' crop rotations regularly include the use of cover crops before spring sowing to avoid leaving the soil bare and unprotected. As these cover crops are generally destroyed in winter, incorporating these Brassicaceae into cover crops mixes could: (i) attract CSFB an
