Comprehensive analysis of the tumor immune microenvironment in gastric cancer and peritoneal metastasis based on single-cell RNA sequencing analysis.

The development and metastasis of gastric cancer are complex processes involving the TME (Tumor microenvironment) and the interactions between various cell types. Here, we investigated the molecular mechanisms and biological processes underlying gastric cancer and its metastasis using single cell RNA-sequencing (scRNA-seq) with the aim of identifying new targets and approaches for clinical treatment. R version 4.4.1 and the SeuratV5 package were used to process 20 scRNA-seq samples sourced from the GEO database. Highly variable genes were selected for GO (Gene Ontology), KEGG (Kyoto Encycloped

GALNT6-glycosylated LRG1 mediates the interaction between M2 macrophages and pancreatic cancer cells by activating CCR1/PI3K/AKT axis.

Pancreatic cancer (PC) is a malignant tumor that often presents no obvious symptoms in its early stages, leading to late diagnosis and poor prognosis. Therefore, developing molecular markers for PC is essential for improving early diagnosis, optimizing treatment strategies, and enhancing patient outcomes. In this study, PC tissue and matched adjacent non-cancerous tissue were collected from patients. For in vitro analysis, the PC cell lines BxPC-3 and SW1990 were used. M2 macrophages were induced in THP-1 cells. Gene expression at both mRNA and protein levels was measured using real-time quant

Selective expression and cellular localization of pro-inflammatory chemokine ligand/receptor pairs in the sciatic nerves of a severe murine experimental autoimmune neuritis model of Guillain-Barré syndrome.

AIMS: To determine if specific pro-inflammatory chemokine ligand/receptor pairs expressed in the peripheral nerves of Guillain-Barré syndrome patients are expressed in a severe murine experimental autoimmune neuritis (sm-EAN) model and to determine their cellular localization as a prerequisite to designing potentially therapeutic interventions in vivo. METHODS: Sm-EAN was induced in 8-12-week-old female SJL/J mice using bovine peripheral nerve myelin emulsified in complete Freund adjuvant with pertussis toxin and recombinant mouse interleukin-12 acting as co-adjuvants, with appropriate contro

Expression of CXC and CC type of chemokines and its receptors in tuberculous and non-tuberculous effusions.

Chemokines mediate their biological functions by transmigration of various immune cells to the site of infection. Tuberculous pleurisy provides an effective model to study the role of chemokines in the recruitment of immune cells to the pleura. Our aim was to understand the cumulative effect of chemokines (IP-10, MIG, IL-8, MCP-1, MIP-1alpha and RANTES) and its receptors (CXCR2, CXCR3, CCR1, CCR2, CCR5 and CCR7) in the recruitment of CD4(+) T cells obtained from blood (BL) and pleural fluid (PF) of tuberculous (TB) and non-tuberculous (NTB) patients. We observed significant increase in CD4(+)

Expression of the chemokine receptor CCR7 in prostate cancer presenting with generalized lymphadenopathy: report of a case, review of the literature, and analysis of chemokine receptor expression.

PURPOSE: Generalized lymphadenopathy is a rare presentation of prostate cancer. We report a case and review reported cases in the literature. Because of the association of chemokine receptor expression with specific metastatic patterns, we tested for expression of chemokine receptors known to mediate migration to lymph nodes. METHODS: We performed a MEDLINE (National Library of Medicine, Bethesda, MD) database search for case reports during the last 32 years using "prostate cancer," "lymphadenopathy," "metastatic to lymph nodes," and "mimicking lymphoma" as keywords. Expression of the CXCR4 a

Increased responsiveness of murine eosinophils to MIP-1beta (CCL4) and TCA-3 (CCL1) is mediated by their specific receptors, CCR5 and CCR8.

In the present study, we investigated the regulation of chemokine-mediated responses and receptor expression on eosinophils from mice. MIP-1alpha (CCL3) and eotaxin (CCL11) induced a significant and only partially overlapping intracellular calcium flux in antigen-elicited and peripheral blood eosinophils, and MCP-1 (CCL2), MDC (CCL22), MIP-1beta (CCL4), and TCA-3 (CCL1) did not. To demonstrate functional use of the specific receptors, we examined chemotactic responses. Peripheral blood eosinophils migrated toward MIP-1alpha (CCL3) and eotaxin (CCL11) but not MCP-1 (CCL2), MDC (CCL22), MIP-1bet

Gene-expression profiling of laser-dissected islets and studies in deficient mice reveal chemokines as differential driving force of type 1 diabetes.

Although type 1 diabetes (T1D) results from the autoimmune destruction of the insulin-producing β-cells, its treatment is largely restricted to exogenous insulin administration. Only few therapies targeting the autoaggressive immune system have been introduced into clinical practice or are considered in clinical trials. Here, we provide a gene expression profile of the islet microenvironment obtained by laser-dissection microscopy in an inducible mouse model. Thereby, we have identified novel targets for immune intervention. Increased gene expression of most inflammatory proteins was apparent

The Effects of Pathogen-Associated Molecular Patterns on Peripheral Blood Monocytes in Patients with Non-alcoholic Fatty Liver Disease.

BACKGROUND: Innate immune responses to gut-derived pathogen-associated molecular patterns (PAMPs) have been implicated in the pathogenesis of nonalcoholic fatty liver disease (NAFLD). Whether NAFLD patients have increased sensitivity to PAMP exposure has yet to be reported. METHODS: Peripheral blood mononuclear cell (PBMC)/monocytes were exposed to lipopolysaccharide (LPS), Pam3CSK4, or BSA conjugated palmitate in vitro. Changes in toll-like receptors (TLR), cytokines, and chemokine receptors (CR) expressions were documented by flow cytometry and/or enzyme-linked immunoabsorbent assays (ELISA

Abnormal expression of chemokine receptors in Behçet's disease: relationship to intracellular Th1/Th2 cytokines and to clinical manifestations.

Dynamic interplay between cytokines and chemokines directs trafficking of peripheral blood mononuclear cells to tissues in autoimmune and/or viral diseases. The aim of the current study was to define the expression on CD3+ T cells of six chemokine receptors associated with inflammatory sites and the expression of intracellular cytokines, such as interferon-gamma (IFN-gamma) and interleukin-4 (IL-4), in Behcet's disease (BD). Flow cytometry was used to detect chemokine receptor and intracytoplasmic cytokines' expression. We observed that CD3+ T cells in the peripheral blood express a restricted

Role of CCL3/MIP-1alpha and CCL5/RANTES during acute Trypanosoma cruzi infection in rats.

Chagas' disease is caused by Trypanosoma cruzi infection and is characterized by chronic fibrogenic inflammation and heart dysfunction. Chemokines are produced during infection and drive tissue inflammation. In rats, acute infection is characterized by intense myocarditis and regression of inflammation after control of parasitism. We investigated the role of CCL3 and CCL5 during infection by using DNA vaccination encoding for each chemokine separately or simultaneously. MetRANTES treatment was used to evaluate the role of CCR1 and CCR5, the receptors for CCL3 and CCL5. Vaccination with CCL3 or

[Treatment strategy for rejection-free corneal transplantation--transition from full-thickness corneal transplantation to corneal endothelium transplantation].

The avoidance of allograft rejection is the most critical factor for favorable surgical outcome after corneal transplantation. We report experimental data including distribution of white blood cells in human corneas for rejection-free corneal transplantation. We focused on leukocyte trafficking based on the immunological mechanism leading to allograft rejection in a mouse full-thickness corneal transplantation model. We identified two chemokine-receptors, CCR1 and CCR7 which are functionally relevant to the occurrence of allograft rejection. These chemokine receptors can be new targets for the

Coreceptor usage of HIV-1 isolates representing different genetic subtypes obtained from pregnant Cameroonian women. European Network for In Utero Transmission of HIV-1.

In this study, coreceptor usage of HIV-1 other than subtype B in relation to HIV-1 transmission from mother to child was investigated. Repeated sampling of 42 HIV-1-seropositive, asymptomatic women in Cameroon during the second and third trimesters of pregnancy, at delivery, and 6 months postpartum were performed. Env subtyping was carried out from uncultured peripheral blood mononuclear cells (PBMCs) by heteroduplex mobility assay and, whenever necessary, by DNA sequencing. Virus isolates were tested for coreceptor usage on human cell lines-U87.CD4 and GHOST(3)-engineered to express stably CD

Hyperactivation of sympathetic nerves fuels basophil infiltration in atopic dermatitis.

Emotional stress exacerbates atopic dermatitis (AD) and contributes to overall disease burden. While sympathetic nervous system (SNS) activity broadly interacts with emotional states, its role in skin immunity remains underexplored. Using a murine model of AD-like disease, we demonstrated that skin inflammation triggered anxiety-like behaviors and hyperactivation of peripheral SNS innervating the affected skin. Disruption of peripheral SNS signaling, either by denervation or anxiety-relief interventions, markedly attenuated skin inflammation. Mechanistically, basophils were identified as key e

Obstructive nephropathy in the mouse: progressive fibrosis correlates with tubulointerstitial chemokine expression and accumulation of CC chemokine receptor 2- and 5-positive leukocytes.

The infiltration of leukocytes plays a major role in mediating tubulointerstitial inflammation and fibrosis in chronic renal disease. CC chemokines participate in leukocyte migration and infiltration into inflamed renal tissue. Because CC chemokine-directed leukocyte migration is mediated by target cell expression of a group of CC chemokine receptors, this study examined the expression of CC chemokines and their receptors during initiation of tubulointerstitial fibrosis after unilateral ureteral obstruction in C57BL/6 mice. Obstructed kidneys developed hydronephrosis, tubular cell damage, inte

Targeting CCR1-mediated macrophage phenotypic switching to alleviate bladder pain and urinary dysfunction in interstitial cystitis.

Interstitial cystitis/bladder pain syndrome (IC/BPS) is a chronic inflammatory condition with limited treatments. Although macrophages are implicated in its pathogenesis, the mechanisms driving their phenotypic switching remain unclear. This study identifies CCR1's role in IC/BPS and evaluates CCR1 inhibition as a therapeutic strategy. Integrated bulk and single-cell RNA sequencing reveal enrichment of pro-inflammatory CCR1⁺ macrophages in bladder tissue from patients with IC/BPS. In a lipopolysaccharide-induced rat model, pharmacological inhibition of CCR1 suppresses M1 polarization, promotes

Differential molecular mechanisms of heparin, BSA, and Ca2+-Induced boar sperm capacitation revealed by transcriptomics.

Sperm capacitation is a prerequisite for successful fertilization. In vitro, this process can be induced by various agents such as heparin, bovine serum albumin (BSA), and calcium ions (Ca2+), yet their underlying regulatory mechanisms remain incompletely understood. This study employed RNA sequencing (RNA-seq) to comparatively investigate the transcriptomic profiles and regulatory mechanisms during sperm capacitation induced by heparin, BSA, or Ca2+. Phenotypic analysis confirmed that all three inducers significantly promoted capacitation, albeit with differential effects on motility paramete

Hepatocyte-specific C-C motif chemokine ligand 9 signaling promotes liver fibrosis progression in mice.

BACKGROUND AND AIMS: Liver fibrosis involves the activation of HSCs and persistent inflammatory responses. Ccl9, a CC chemokine implicated in inflammation, has an undefined role in liver homeostasis. Our study investigates this murine homolog of human CCL15 to elucidate its role in the development of liver fibrosis. APPROACH AND RESULTS: We investigated the expression of Ccl9 and its upstream regulatory elements in liver fibrosis using mouse models induced by carbon tetrachloride (CCl 4 ), bile-duct ligation, and a high-fat, methionine-deficient and choline-deficient diet. A significant incre

CCR1 antagonist J-113863 corrects the imbalance of pro- and anti-inflammatory cytokines in a SJL/J mouse model of relapsing-remitting multiple sclerosis.

Multiple sclerosis (MS), an immune-mediated and neurodegenerative disorder of the central nervous system (CNS), is characterized by infiltrating myelin-reactive T lymphocytes and demyelinating lesions. Experimental autoimmune encephalomyelitis (EAE) is a well-established animal model used to study MS. To explore the impact of chemokine receptor CCR1 blockade in EAE and the underlying mechanisms, we used CCR1 antagonist J-113863 in PLP139-151-induced EAE in SJL/J mice. Following EAE induction, mice were treated with J-113863 (10 mg/kg) daily from day 14 until day 25. We investigated the effect
