Optical Control of Dopamine Receptors Using a Photoswitchable Tethered Inverse Agonist.

Family A G protein-coupled receptors (GPCRs) control diverse biological processes and are of great clinical relevance. Their archetype rhodopsin becomes naturally light sensitive by binding covalently to the photoswitchable tethered ligand (PTL) retinal. Other GPCRs, however, neither bind covalently to ligands nor are light sensitive. We sought to impart the logic of rhodopsin to light-insensitive Family A GPCRs in order to enable their remote control in a receptor-specific, cell-type-specific, and spatiotemporally precise manner. Dopamine receptors (DARs) are of particular interest for their

Blonanserin extensively occupies rat dopamine D3 receptors at antipsychotic dose range.

Antagonism of the dopamine D3 receptor has been hypothesized to be beneficial for schizophrenia cognitive deficits, negative symptoms and extrapyramidal symptoms. However, recent animal and human studies have shown that most antipsychotics do not occupy D3 receptors in vivo, despite their considerable binding affinity for this receptor in vitro. In the present study, we investigated the D3 receptor binding of blonanserin, a dopamine D2/D3 and serotonin 5-HT2A receptors antagonist, in vitro and in vivo. Blonanserin showed the most potent binding affinity for human D3 receptors among the tested

Pituitary and brain dopamine D2 receptors regulate liver gene sexual dimorphism.

Liver sexual gene dimorphism, which depends mainly on specific patterns of GH secretion, may underlie differential susceptibility to some liver diseases. Because GH and prolactin secretion are regulated by dopaminergic pathways, we studied the participation of brain and lactotrope dopamine 2 receptors (D2Rs) on liver gene sexual dimorphism, to explore a link between the brain and liver gene expression. We used global D2R knockout mice (Drd2(-/-)) and conducted a functional dissection strategy based on cell-specific Drd2 inactivation in neurons (neuroDrd2KO) or pituitary lactotropes. Disruption

Receptor reserve-dependent properties of antipsychotics at human dopamine D2 receptors.

Aripiprazole is the first dopamine D(2)/D(3) receptor partial agonist approved for use in the treatment of psychiatric disorders including schizophrenia, bipolar disorder, and unipolar depression in the US. Aripiprazole has demonstrated a relatively favorable side effect profile compared to other commonly prescribed antipsychotics, including a low propensity for treatment-limiting extrapyramidal symptoms, hyperprolactinemia, and body weight gain. In an effort to elucidate aripiprazole's pharmacological activity in relation to clinically relevant fluctuation of dopamine D(2) receptor reserves,

Characterization of aripiprazole partial agonist activity at human dopamine D3 receptors.

Aripiprazole is the first dopamine D2/D3 receptor partial agonist approved for use in the treatment of psychiatric disorders, including schizophrenia, bipolar disorder, and unipolar depression in the US. To explore the functional activity of aripiprazole at dopamine D3 receptors, we established Chinese hamster ovary (CHO) cell lines stably expressing high and low densities of Ser-9 and Gly-9 variants of human dopamine D3 receptors and compared aripiprazole's dopamine D3 pharmacological properties with other marketed and non-approved dopamine D3 receptor modulating agents on inhibition of forsk

Iron deficiency decreases dopamine D1 and D2 receptors in rat brain.

Iron deficiency (ID) in early life is known to alter neurological development and functioning, but data regarding specific effects on dopamine biology are lacking. The objective of this study was to determine the extent of functional alterations in dopamine receptors in two dopaminergic tracts in young, growing, iron-deficient rats. Forty male and 40 female weanling Sprague-Dawley rats were fed either an iron-deficient (ID) diet or control (CN) diet for 6 weeks. ID decreased densities of D(1) and D(2) receptors in the caudate-putamen and decreased D(2) receptor densities in the nucleus accumbe

Comparison of 12-chloroscoulerine enantiomers on animal behavior to dopamine receptors.

AIM: To compare the pharmacological characteristics of 12-chloroscoulerine (CSL) enantiomers to dopamine (DA) receptors. METHODS: Radioligand receptor binding assay with calf striatum and behavioral tests of mice or rats were used. RESULTS: In the competitive binding assay, the affinities (Ki) of l-CSL to D1 and D2 receptors were 5.7 nmol.L-1, while those of d-CSL for D1 and D2 receptors were 135 and 9150 nmol.L-1, respectively. The Ki of dl-CSL to D1 and D2 receptors were 8.9 and 9.6 nmol.L-1, respectively, which were slightly weaker than that of l-CSL. In the behavioral experiments, CSL en

Functional interaction between dopamine D1 and D2 receptors in 'MPTP' monkeys.

We have studied the motor response induced by independent administration of 4 different doses of a dopamine D2 [(+)-PHNO] and a dopamine D1 (CY 208-243) receptor agonist in 5 MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) monkeys. Both drugs had similar antiparkinsonian effects and both elicited choreic dyskinesias. Simultaneous administration of (+)-PHNO [(+)-4-propyl-9-hydroxynaphthoxazine] and CY 208-243 [(-)4,6,6a,7,8,12b-hexahydro-7-methylindolo[4,3a-b]phenan thyxidine] did not result in modification of the dose-response curve induced by each dopamine receptor agonist given alone. No evidence of vascular dopamine receptors in the rat portal vein.

Dopamine (5.3 X 10(-6) to 5.3 X 10(-5) M) decreased the force of spontaneous myogenic contractions of the rat portal vein in a concentration-dependent manner in the presence of the alpha-adrenoreceptor antagonist BE-2254. The DA1-selective agonist fenoldopam and the DA2-preferential agonist (-)-EOE were both inactive. Dopamine-induced relaxation was not inhibited by (+)-butaclamol (10(-8) M), but was reduced by a potent beta 2-selective adrenoreceptor antagonist ICI 118,551 (2 X 10(-8) M). Our results demonstrate that the rat portal vein is devoid of postsynaptic DA1 and DA2 dopamine receptors

[Role of dopamine receptors in the mechanism of action of neuroleptics].

The authors analyze the literature and their own data on the influence of neuroleptics on the cerebral dopamine and adrenoreceptors. It is shown that antipsychotic activity of neuroleptics correlates with their ability to block D2 dopamine receptor whereas their sedative properties are in a better agreement with the blockade of alpha 1-adrenoreceptor. Effects at the molecular level, other than the blockade of the above receptors, may be involved in forming the activity spectrum of each drug.

Further observations on vesicourethral dopamine receptors--a urodynamic study with haloperidol.

The effect of intravenous administration of 5 mg of haloperidol, a dopamine receptor antagonist upon cystometry, the urethral pressure profile, and the external urethral sphincter electromyography (EMG) was investigated in eight patients with neurogenic vesical dysfunction. There was no change in the detrusor reflex or in the sphincter EMG activities, but the peak urethral pressure decreased. The possible reasons for the difference in the above result as compared to our previous study using metoclopramide, another dopamine receptor antagonist, are discussed.

D2-like dopamine receptors blockade within the dentate gyrus shows a greater effect on stress-induced analgesia in the tail-flick test compared to D1-like dopamine receptors.

INTRODUCTION: Acute stress, as a protective mechanism to respond to an aversive stimulus, can often be accompanied by suppressing pain perception via promoting consistent burst firing of dopamine neurons. Besides, sensitive and advanced research techniques led to the recognition of the mesohippocampal dopaminergic terminals, particularly in the hippocampal dentate gyrus (DG). Moreover, previous studies have shown that dopamine receptors within the hippocampal DG play a critical role in induced antinociceptive responses by forced swim stress (FSS) in the presence of inflammatory pain. Aberrant behavioral effects of a dopamine D1 receptor antagonist and agonist in monkeys: evidence of uncharted dopamine D1 receptor actions.

BACKGROUND: Basic research indicates a role for dopamine (DA) D1 antagonism in the treatment of schizophrenia. Clinical trials have not confirmed any role. Besides the defining second messenger (adenylyl cyclase [AC]), DA D1 receptors are linked to other effectors (e.g., phospholipase C [PLC]). Differing actions of DA D1 antagonists upon differing effectors could explain conflicting results between the lab/clinic. METHODS: In a monkey model in which behavioral effects of DA D1 antagonists/agonists have been well characterized we examined: 1) SKF 83959, biochemically, a DA D1 antagonist, behav

D2 dopamine antisense RNA expression vector, unlike haloperidol, produces long-term inhibition of D2 dopamine-mediated behaviors without causing Up-regulation of D2 dopamine receptors.

Long-term inhibition of D2 dopamine receptors using classic D2 dopamine receptor antagonists such as haloperidol often causes a compensatory up-regulation of the D2 dopamine receptors. We investigated whether the long-term inhibition of D2 dopamine receptors using an eukaryotic expression vector housing a cDNA sequence encoding an antisense RNA directed to the D2 dopamine receptor transcript (D2 antisense vector) would also produce up-regulation of the D2 receptors. Single, bilateral injections of the D2 antisense vector into the corpora striata of mice inhibited the stereotypy induced by acut

The binding affinity and recognition specificity of dopamine to human dopamine receptors can be rationally altered by halogen doping.

Human hepta-helical transmembrane spanning G protein-coupled family of dopamine receptors (DRs) play key neurophysiological roles and are involved in the pathogenesis of Parkinson's disease due to dopamine deficiency. Previous studies found that halogen doping can be used as a chemical strategy to control the optical and spectroscopic properties of dopamine molecule and to shift the binding mode of various exogenous compounds to DRs. In this study, we attempted to investigate the halogen-doping effects by substituting the three positions (X1, X2 and X3) of dopamine's aromatic phenyl group with

Sex-specific behavioral and neurogenic responses to cocaine in mice lacking and blocking dopamine D1 or dopamine D2 receptors.

Adult neurogenesis in rodents is modulated by dopaminergic signaling and inhibited by cocaine. However, the sex-specific role of dopamine D1 and D2 receptors (D1R, D2R) in the deleterious effect of cocaine on adult neurogenesis has not been described yet. Here, we explored sex differences in (a) cell proliferation (5'-bromo-2'-deoxyuridine [BrdU]), (b) neural precursor (nestin), (c) neuronal phenotype (BrdU/β3-tubulin), and (d) neuronal maturity (NeuN) in the subventricular zone (SVZ) of the lateral ventricles and striatum of mice with genetic deletion (D1-/- , D2-/- ) or pharmacological block

Regulation by D2 dopamine receptors of in vivo dopamine synthesis in striata of rats and mice with experimental parkinsonism.

The effects of D2 dopamine (DA) receptor antagonism or stimulation by systemic haloperidol or quinpirole, respectively, on in vivo DA synthesis in 6-hydroxydopamine (6-OHDA)-lesioned rats and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated black mice were assessed by measuring the rate of dihydroxyphenylalanine (DOPA) accumulation following acute inhibition of L-aromatic amino acid decarboxylase with NSD-1015. 6-OHDA and MPTP caused partial lesions of nigrostriatal input to the striatum. Dopamine synthetic capacity was preserved relative to the severity of nigrostriatal lesion over

Contributions of the interaction between mu-opioid receptors and dopamine receptors in the regulation of risky decision making.

During decision making, the ability to appropriately weigh potential risks against their associated rewards is necessary to navigate daily life. Prior work has established a critical role for dopamine (DA) D2 receptors (D2Rs) in this process, with activation of these receptors decreasing choice of risky options. Much less is known, however, about how other neuromodulatory systems, such as the endogenous opioid system, contribute to risk-based decision making and whether these systems interact with D2R regulation of risky choice. To begin to address this gap in knowledge, the goals of the curre

Involvement of adrenoceptors, dopamine receptors and AMPA receptors in antidepressant-like action of 7-O-ethylfangchinoline in mice.

AIM: 7-O-ethylfangchinoline (YH-200) is a bisbenzylisoquinoline derivative. The aim of this study was to investigate the antidepressant-like action and underlying mechanisms of YH-200 in mice. METHODS: Mice were treated with YH-200 (15, 30, and 60 mg/kg, ig) or tetrandrine (30 and 60 mg/kg, ig) before conducting forced swimming test (FST), tail suspension test (TST), or open field test (OFT). RESULTS: YH-200 (60 mg/kg) significantly decreased the immobility time in both FST and TST, and prolonged the latency to immobility in FST. YH-200 (60 mg/kg) was more potent than the natural bisbenzylis

Activation of mu-opioid receptors slows pacemaking in hypothalamic A11 dopamine neurons.

Hypothalamic A11 dopamine neurons provide the only known source of spinal dopamine and critically modulate pain and motor systems. Yet, the electrophysiological properties of A11 neurons were unknown. Here, we characterized A11 dopamine neurons in mice using brain slice immunohistochemistry, and fluorescence-guided whole-cell patch-clamp and cell-attached electrophysiology. A11 dopamine neurons contained the enzymes necessary to synthesize dopamine, projected to the spinal cord, and were small, morphologically simple, and high resistance. Additionally, they received excitatory glutamatergic an
