Transferrin receptor expression by non-Hodgkin's lymphomas. Correlation with morphologic grade and survival.

The monoclonal antibody OKT9 was applied to cryostat sections of 267 non-Hodgkin's lymphomas and related neoplasms. It was found that the transferrin receptor is expressed by a wide variety of B- and T-lineage non-Hodgkin's lymphomas. The OKT9 staining also was loosely correlated with the three morphologic grades of non-Hodgkin's lymphomas identified by the International Working Formulation. In general, higher grade lymphomas more often and more intensely expressed the T9 antigen. However, transferrin receptor expression by certain histologic subtypes of lymphoma did not correlate with their m

Identification of human transferrin receptor as an entry co-receptor for parvovirus B19 infection of human erythroid progenitor cells.

Parvovirus B19 (B19V), a member of the genus Erythroparvovirus within the Parvoviridae family, infects human erythroid progenitor cells (EPCs) of bone marrow and fetal liver and causes various hematological disorders. The minor capsid protein VP1 of B19V contains a unique N-terminal region (VP1u) that facilitates virus binding and internalization into EPCs via its receptor-binding domain (RBD). We previously identified tyrosine-protein kinase receptor UFO (AXL) as a proteinaceous receptor for B19V infection of EPCs. In this study, we employed an ascorbate peroxidase 2 (APEX2)-based proximity l

Determination of transferrin receptors on frozen sections of malignant B-cell lymphomas by immunofluorescence with a monoclonal antibody.

A series of 34 B-cell lymphomas was studied for the presence of transferrin receptors by immunofluorescence on frozen tissue sections with a monoclonal antibody. These lymphomas were classified by light microscopic study and furthermore investigated by immunohistochemical and morphometric methods. There were diffuse centrocytic, diffuse centroblastic, and follicular and diffuse centrocytic/centroblastic lymphomas. Significant differences in log mean nuclear area in these four histologic groups were found, as well as a significant correlation between log nuclear area and transferrin receptors a

Mechanistic insights into the interaction of emerging SARS-CoV-2 spike variants with the transferrin receptor.

Since the onset of the COVID-19 pandemic, SARS-CoV-2 has posed a substantial threat to global health and safety. While the virus's entry into human cells via the interaction of its spike protein with the angiotensin-converting enzyme 2 (ACE2) receptor is well-established, emerging studies suggest that the transferrin receptor protein (TfR) may also serve as an entry receptor. However, the precise binding mechanism between SARS-CoV-2 and TfR remains elusive, and how various SARS-CoV-2 variants interact with TfR warrants further exploration. In this study, we utilized molecular docking to model

E-selectin and transferrin receptor functionalized synthetic miRNA nanoshuttles with therapeutic potential in stroke.

Current acute ischemic stroke treatment is limited to reperfusion therapies within the first hours after the onset, underscoring the urgent need for novel therapeutic strategies to mitigate the progression of the brain lesion. Here, we describe the development of bio-inspired synthetic nanoshuttles-lipid nanoparticles (LNPs)-designed to deliver miRNAs and target post-stroke inflammation, a key contributor to secondary brain damage. miRNAs isolated from human induced pluripotent stem cells (hiPSCs) were encapsulated into LNPs prepared by a microfluidic technique. We produced small sized, unifor

A transferrin receptor-targeted liposome for iron metabolism regulation-augmented photodynamic therapy in glioblastoma.

The clinical standard treatments for glioblastoma (GBM) present a therapeutic challenge of high recurrence. 5-aminolevulinic acid (5-ALA)-based photodynamic therapy (PDT) offers a promising alternative owing to the selective biosynthesis of protoporphyrin IX (PpIX) in tumor mitochondria with favorable phototoxicity. However, the highly elevated iron metabolism leads to low biosynthetic efficiency and insufficient accumulation of PpIX, thus limiting the PDT efficacy. Here, we developed a transferrin receptor (TfR)-targeted liposome (AD@LST) that co-delivers 5-ALA and the iron chelator deferoxam

Transferrin receptor-targeted anti-amyloid antibody enhances brain delivery and mitigates ARIA.

Amyloid-related imaging abnormalities (ARIA), side effects of anti-amyloid drugs seen in magnetic resonance imaging of the brain, are a major safety concern in patients with Alzheimer's disease. We developed an antibody transport vehicle (ATV) targeting transferrin receptor (TfR) for brain delivery of anti-amyloid-β protein (anti-Aβ) using asymmetrical Fc mutations (ATVcisLALA) that mitigates TfR-related liabilities and retains effector function when bound to Aβ. Administration of ATVcisLALA:Aβ in mice exhibited broad brain distribution and enhanced parenchymal plaque target engagement. This b

Soluble transferrin receptor is associated with red blood cell transfusion in infant cardiac surgery.

BACKGROUND: Iron deficiency (ID) is linked to an increased need for perioperative red blood cell (RBC) transfusion in cardiac surgery. Traditional markers used to assess ID are often influenced by inflammation, whereas soluble transferrin receptor (sTfR) is less affected by inflammation. Therefore, the purpose of this study is to explore the relationship between sTfR levels and the need for high-volume RBC transfusion in pediatric cardiac surgery patients. METHODS: From August 2021 to July 2022, 236 low-weight infants (≤10 kg) who underwent cardiac surgery were included in this study. Preoper

Elevated transferrin receptor impairs T cell metabolism and function in systemic lupus erythematosus.

T cells in systemic lupus erythematosus (SLE) exhibit multiple metabolic abnormalities. Excess iron can impair mitochondria and may contribute to SLE. To gain insights into this potential role of iron in SLE, we performed a CRISPR screen of iron handling genes on T cells. Transferrin receptor (CD71) was identified as differentially critical for TH1 and inhibitory for induced regulatory T cells (iTregs). Activated T cells induced CD71 and iron uptake, which was exaggerated in SLE-prone T cells. Cell surface CD71 was enhanced in SLE-prone T cells by increased endosomal recycling. Blocking CD71 r

Transferrin Receptor-Mediated Uptake at the Blood-Brain Barrier Is Not Impaired by Alzheimer's Disease Neuropathology.

The transferrin receptor (TfR) is highly expressed by brain capillary endothelial cells (BCECs) forming the blood-brain barrier (BBB) and is therefore considered as a potential target for brain drug delivery. Monoclonal antibodies binding to the TfR, such as clone Ri7, have been shown to internalize into BCECs in vivo. However, since Alzheimer's disease (AD) is accompanied by a BBB dysfunction, it raises concerns about whether TfR-mediated transport becomes inefficient during the progression of the disease. Measurements of TfR levels using Western blot analysis in whole homogenates from human

Design, synthesis, and in vitro antitumor activity of a transferrin receptor-targeted peptide-doxorubicin conjugate.

In this study, a peptide-drug conjugate was designed and synthesized by connecting a transferrin receptor (TfR)-targeted binding peptide analog BP9a (CAHLHNRS) with doxorubicin (DOX) through N-succinimidyl-3-maleimidopropionate (SMP) as the cross-linker. Confocal laser scanning microscopy results indicated that free DOX mainly accumulated in the nuclei of both TfR overexpressed HepG2 hepatoma cells and L-O2 normal liver cells expressing low level of TfR; most of the BP9a-DOX conjugate displayed cytoplasmic location, and its cellular uptake by HepG2 cells was obviously reduced by TfR blockage t

Prevention of mantle lymphoma tumor establishment by routing transferrin receptor toward lysosomal compartments.

Mantle cell lymphoma (MCL) is one of the most frequent of the newly recognized non-Hodgkin's lymphomas. The major problem of MCL therapy is the occurrence of relapse and subsequent resistance to chemotherapy and immunotherapy in virtually all cases. Here, we show that one injection of anti-human transferrin receptor (TfR) monoclonal antibody A24 totally prevented xenografted MCL tumor establishment in nude mice. It also delayed and inhibited tumor progression of established tumors, prolonging mice survival. In vitro, A24 induced up to 85% reduction of MCL cell proliferation (IC(50) = 3.75 nmol

Double diagnostic meaning of serum transferrin receptor in hemodialysis patients: two case reports.

Hemodialysis patients on maintenance erythropoietin need an adequate supply of iron to optimize therapy and achieve and maintain target levels of hemoglobin. Evaluation of iron stores and early detection of iron deficiency are essential for management of erythropoiesis in chronic renal failure, but there is still no single biochemical or hematological parameter that is sensitive or specific enough to completely describe the distribution of iron in the body. Serum transferrin receptor (sTfR) is a marker of iron that is available for erythropoiesis. We selected 2 clinical cases in which hemodial

Immunohistochemical localization of intraneuronal transferrin receptor immunoreactivity in the adult mouse central nervous system.

Iron is essential for a variety of intracellular functions. Accordingly, the transfer of iron from blood to brain is vital for normal brain function. In the CNS, the receptor for iron-transferrin is generally accepted to be located in endothelial cells, whereas its occurrence in other cell types is less well established. I have investigated the distribution of the transferrin receptor in the adult mouse central nervous system by immunohistochemistry by using a monoclonal antibody raised against the transferrin receptor protein. Immunoreactive cell types comprised brain capillary endothelial ce

Plasmacytoid Dendritic Cells Exhibit High Transferrin Receptor Expression Without Iron Accumulation.

Plasmacytoid dendritic cells (pDCs) are specialized antiviral sentinels defined by rapid type I interferon (IFN‑I) production, yet their proteomic organization and metabolic requirements remain incompletely understood. We established the steady‑state proteome of murine splenic pDCs directly ex vivo using deep, absolute quantitative mass spectrometry and compared it with conventional dendritic cell subsets and human pDCs. pDCs exhibited a highly conserved proteomic architecture across species, with selective divergence in central carbon metabolism, amino‑acid utilization, and nutrient transport
