Adolescent hyperactivity of offspring after maternal protein restriction during the second half of gestation and lactation periods in rats.

To clarify the effect of systemic growth retardation on behavior, pregnant rats were fed a synthetic diet with either a normal (20% casein) or low (10% casein) protein concentration from gestational day 10 to postnatal day (PND) 21 at weaning. Offspring were examined for sensory and reflex functions, detailed clinical observations, manipulative test, grip strength, motor activity and water-filled multiple T-maze test. Lowering trend in the air righting reflex index during lactation period and a decrease in grip strength on PND 72 were observed in the low protein diet group showing suppression

Faster immobilization and recovery with remimazolam compared to midazolam in medetomidine-butorphanol balanced anesthesia in male Sprague-Dawley rats: a randomized crossover study.

BACKGROUND: Balanced injectable anesthesia combining an α2-adrenoceptor agonist, a benzodiazepine, and an opioid is widely used in laboratory rodents because it provides sedation, muscle relaxation, and analgesia through complementary pharmacological mechanisms. In Japan, a mixture of medetomidine, midazolam, and butorphanol (MMB) is commonly used as an injectable anesthetic regimen in mice and rats. Remimazolam is an ultra-short-acting benzodiazepine that is rapidly metabolized by tissue esterases and produces no active metabolites. Because of its rapid metabolism and predictable pharmacokine

ACTIONS OF PROSTAGLANDINS E1, E2 AND E3 ON THE CENTRAL NERVOUS SYSTEM.

Prostaglandins E(1), E(2) and E(3), injected into the cerebral ventricles of unanaesthetized cats, produced sedation, stupor and signs of catatonia. The threshold dose was 3 mug/kg. Slight sedation was also observed following an intravenous injection, but a dose of 20 mug/kg was required. In chicks, intravenous injections of prostaglandins (10 to 400 mug/kg) caused respiratory depression, profound sedation, loss of normal posture and, with the higher doses, loss of the righting reflex.

Inhaled nonimmobilizers do not alter the middle latency auditory-evoked response of rats.

UNLABELLED: General anesthetics cause surgical immobility and oblivion (unconsciousness and amnesia). A class of compounds known as "nonimmobilizers" were predicted to be anesthetic, based on their physiochemical properties, but found to cause only amnesia. In humans, cerebrocortical electrical activity after auditory stimulation is depressed by concentrations of anesthetics which impair auditory recall. We sought to use these evoked responses to characterize the effects of the nonimmobilizer 1,2-dichlorohexafluorocyclobutane (2N) and conventional inhaled anesthetics on early sensory processin

Barbiturates inhibit stress-induced analgesia.

The effect of pentobarbitone and thiopentone on stress-induced analgesia was studied in 40 male Sprague-Dawley rats. Antinociception was determined by measuring motor reaction threshold to the noxious pressure on the tail with the use of an "Analgesymeter." Stress was induced by placement of a clamp on the hind paw. The stress procedure was found to cause an increase in reaction threshold, which was partially suppressed by naloxone 0.5 mg X kg-1. Pentobarbitone in a subanaesthetic dose of 25 mg X kg-1, SC, almost completely abolished the stress-induced increase in the reaction threshold (an in

Effect of caffeine on respiratory rate, recovery time, and brain wave activity during emergence from sevoflurane anaesthesia in rats.

OBJECTIVE: To determine whether caffeine affects the respiratory rate, recovery time, and brain slow-wave (theta and delta) activity during emergence from sevoflurane anaesthesia in rats. METHODS: Male Sprague-Dawley rats (n = 16) were randomly divided into two groups: control (saline) and caffeine (75 mg/kg). After tripolar electrode implantation, rats were placed in an anaesthesia induction chamber and brain electrocorticographic wave activity was recorded. Anaesthesia was induced with 4% sevoflurane (4 L/min O2) for 5 min, and once all the rats were asleep, sevoflurane concentration was re

Altered Behavioral Responses Show GABA Sensitivity in Muscleblind-Like 2-Deficient Mice: Implications for CNS Symptoms in Myotonic Dystrophy.

Considerable evidence from mouse models and human postmortem brain suggests loss of Muscleblind-like protein 2 (MBNL2) function in brain is a major driver of CNS symptoms in Myotonic dystrophy type 1 (DM1). Increased hypersomnia, fatigue, and surgical complications associated with general anesthesia suggest possible sensitivity to GABAergic inhibition in DM1. To test the hypothesis that MBNL2 depletion leads to behavioral sensitivity to GABAA receptor (GABAA-R) modulation, Mbnl2 knock-out (KO) and wild-type (WT) littermates were treated with the anesthetic sevoflurane, the benzodiazepine diaze

The differential effect of cyclosporine on hypnotic response and pain reaction in mice.

BACKGROUND: The calcineurin inhibitor, cyclosporine, is widely used for preventing allograft rejection in organ transplantation. Systemically administered cyclosporine is prevented from entering into the brain by the action of P-glycoprotein, encoded by the multidrug resistant 1 (mdr1) gene. However, in many transplant recipients, cyclosporine administration causes postoperative neuropsychological side effects, such as confusion, depression, and anxiety. Recently, calcineurin-inhibitor-induced pain syndrome, characterized by severe pain in the lower limbs, has also been recognized in both orga

Neurotensin levels in specific brain regions and hypnotic sensitivity to ethanol and pentobarbital as a function of time after haloperidol administration in selectively bred rat lines.

Evidence indicates that sensitivity to ethanol is a good predictor of the development of alcoholism. Thus, identification of neuronal processes that regulate ethanol sensitivity has been the subject of much recent research. The present studies were designed to further test the hypothesis that neurotensinergic processes mediate, in part, hypnotic sensitivity to ethanol. Single doses of haloperidol were administered to lines of rats [selectively bred for high and low sensitivity (HAS and LAS, respectively) to hypnotic effects of ethanol] to produce increases in neurotensin (NT) levels in brain r

The interaction of dopamine, cocaine, and cocaethylene with ethanol on central nervous system depression in mice.

The interactions between dopamine, cocaine, cocaethylene, and ethanol were studied in Swiss-Webster mice. The loss of the righting reflex (LORR) was used as a measure of CNS depression. Animals were injected intraperitoneally (IP) with ethanol (4.0 g/kg). which caused a LORR. Immediately upon regaining of the righting reflex, mice were injected intracerebroventricularly (ICV) with saline, dopamine (0.1, 0.5, or 1.0 mumol/kg), cocaine (1, 15, or 25 mumol/kg), or cocaethylene (1, 15, or 25 mumol/kg). In the presence of systemic ethanol, all three compounds produced CNS depression in a dose-depen

Pentobarbital administered intracerebroventricularly antagonizes morphine-induced antinociception in mice.

The purposes were to show that pentobarbital administered intracerebroventricularly (i.c.v.) antagonized the antinociceptive action of morphine given intrathecally in the tail-flick test in mice, determine whether this antagonistic action involved spinal release of dynorphin A (Dyn A) and demonstrate whether the mechanism involved gamma-aminobutyric acid (GABA) receptors. Previously, it was shown that midazolam given i.c.v. antagonizes morphine-induced antinociception by release of Dyn A in the spinal cord. This antagonism by midazolam is reversed by a variety of benzodiazepine/GABAA receptor

Kinetics of drug action in disease states. XXIX. Effect of experimental nephrotic syndrome on the pharmacodynamics of heptabarbital: implications of severe hypoalbuminemia.

The purpose of this investigation was to determine the effect of nephrotic syndrome (NS) on the pharmacodynamics of a barbiturate. NS was induced in male rats by puromycin aminonucleoside; it caused hypoproteinemia, increased liver and kidney weight and elevated serum creatinine and urea nitrogen concentrations. Serum albumin concentration decreased from 3.5% in controls to 0.90% in NS animals. The rats were infused i.v. with heptabarbital, 1 mg/min, until they lost their righting reflex. The total dose (mean +/- S.D.) required by rats with NS, 40.2 +/- 4.2 mg/kg, was substantially lower than

Evaluation of three medetomidine-based protocols for chemical restraint and sedation for non-painful procedures in companion rats (Rattus norvegicus).

Three medetomidine-based protocols were evaluated for sedation in companion rats undergoing diagnostic procedures. Group Me received medetomidine at 150 μg/kg intramuscularly (IM); group Me-Bu received medetomidine 100 μg/kg IM and butorphanol 2 mg/kg IM, and group Me-Bu-Mi received medetomidine 50 μg/kg IM, butorphanol 2 mg/kg IM and midazolam 1 mg/kg IM. The righting reflex disappeared more quickly in the Me-Bu-Mi group, but recovery after atipamezole was longer. In group Me, a palpebral reflex was present throughout sedation in more rats than in the other two groups. Pulse and respiratory r

Antagonism of anthracene-9-carboxylic acid as screening test for muscle relaxants.

Veratrinic response produced by anthracene-9-carboxylic acid in mice has been used as a model for studying effect of variety of drugs which induce muscle relaxation. None of the drugs listed as general depressants, central muscle relaxants, neuromuscular blocking agents and anti-convulsants were effective even at doses causing 100% loss of righting reflex. Local anaesthetics/direct acting muscle relaxants were effective. This simple model employed in conjunction with Straub's tail test can differentiate various categories of muscle relaxants and their mechanism of action.

n-3 PUFA ameliorate functional outcomes following repetitive mTBI in the fat-1 mouse model.

PURPOSE: Repeated mild traumatic brain injuries (mTBI) are a continuing healthcare concern worldwide, given its potential for enduring adverse neurodegenerative conditions. Past research suggests a potential protective effect of n-3 polyunsaturated fatty acids (PUFA) in experimental models of mTBI. The aim of this study was to investigate whether the neuroprotective benefits of n-3 PUFA persist following repetitive weight drop injury (WDI). METHODS: Male fat-1 mice (n = 12), able to endogenously convert n-6 PUFA to n-3 PUFA, and their wild type (WT) counterparts (n = 12) were maintained on a

N-methyl-d-aspartate receptors and glycinergic transmission, respectively, mediate muscle relaxation and immobility of pentobarbital in mice.

Pentobarbital-induced anesthesia is believed to be mediated by enhancement of the inhibitory action of γ-aminobutyric acid (GABA)ergic neurons in the central nervous system. However, it is unclear whether all components of anesthesia induced by pentobarbital, such as muscle relaxation, unconsciousness, and immobility in response to noxious stimuli, are mediated only through GABAergic neurons. Thus, we examined whether the indirect GABA and glycine receptor agonists gabaculine and sarcosine, respectively, the neuronal nicotinic acetylcholine receptor antagonist mecamylamine, or the N-methyl-d-a

The biologically active compound of Withania somnifera (L.) Dunal, docosanyl ferulate, is endowed with potent anxiolytic properties but devoid of typical benzodiazepine-like side effects.

BACKGROUND: Clinical and experimental studies support the therapeutic potential of Withania somnifera (WS) (L.) Dunal on anxiety disorders. This potential is attributable to components present in different plant extracts; however, the individual compound(s) endowed with specific anxiolytic effects and potential modulatory activity of the GABAA receptor complex (GABAAR) have remained unidentified until the recent isolation from a WS methanolic root extract of some GABAAR-active compounds, including the long alkyl-chain ferulic acid ester, docosanyl ferulate (DF). Lobsters with pre-existing damage to their mechanosensory statocyst organs do not incur further damage from exposure to seismic air gun signals.

Staotcysts, the mechanosensory organs common to many marine invertebrates, have shown sensitivity to aquatic noise. Previously, rock lobsters (Jasus edwardsii) from a remote site with little exposure to anthropogenic noise incurred persistent damage to the statocyst and righting reflex following exposure to seismic air gun signals. Here, J. edwardsii collected from a site subject to high levels of anthropogenic noise were exposed to an equivalent seismic air gun signal regime as the previous study of noise-naïve lobsters. Following exposure, both control and exposed treatments were found to ha

Hypoxic postconditioning improves behavioural deficits at 6 weeks following hypoxic-ischemic brain injury in neonatal rats.

Hypoxic-ischemic (HI) brain injury in newborns is associated with high morbidity and mortality, with many babies suffering neurological deficits. Recently, we showed that hypoxic postconditioning (PostC) immediately post injury can protect against HI up to one week in neonatal rats. Here, we aimed to examine whether long term functional deficits were also improved by PostC. Sprague-Dawley rats were assigned to control (C) or HI group on postnatal day 7 (P7). The HI group underwent unilateral carotid artery occlusion followed by hypoxia (7% oxygen, 3h). Half of each group were randomly assigned
