Strategies to Develop Regenerative Medicine Approaches for Olfactory Disorders.

Olfactory loss affects more than 12% of the population, with prevalence increasing in aging individuals. Multiple conditions can lead to a loss of smell (hyposmia or anosmia), including post-viral damage from coronavirus disease 2019 (COVID-19) or influenza, head injuries, sinusitis, or neurodegenerative conditions such as Alzheimer or Parkinson disease. Although treatments like surgery, anti-inflammatory medications, or olfactory training can be beneficial in certain cases, there remains an unmet need for effective therapies addressing many common causes of olfactory dysfunction. This is part

Reactive oxygen species-scavenging biomaterials for neural regenerative medicine.

Reactive oxygen species (ROS) are natural by-products of oxygen metabolism. As signaling molecules, ROS can regulate various physiological processes in the body. However excessive ROS may be a major cause of inflammatory diseases. In the field of neurological diseases, ROS cause neuronal apoptosis and neurodegeneration, which severely impede neuroregeneration. Currently, ROS-scavenging biomaterials are considered as a promising therapeutic strategy for neurological injuries due to their ability to scavenge excessive ROS at defects and modulate the oxidative stress microenvironment. Osteochondral Tissue Engineering Dilemma: Scaffolding Trends in Regenerative Medicine.

Orthopedic surgeons face a lot of difficulties in managing and repairing osteochondral defects. Damaged articular cartilage and the subchondral bone underneath are both present in osteochondral defects. The demands of the bone, cartilage, and the contact between the bone and the cartilage must be taken into consideration while repairing an osteochondral defect. Only palliative, not curative, therapeutic interventions are now available for the healing of osteochondral abnormalities. With its ability to successfully rebuild bone, cartilage, and the junction between bone and cartilage, tissue eng

Bioprinting of hair follicle germs for hair regenerative medicine.

Hair regenerative medicine is a promising approach to treat hair loss. The replication of in vivo tissue configurations and microenvironments, such as hair follicle germs, has been studied to prepare tissue grafts for hair regenerative medicine. However, such approaches should be scalable, because a single patient with alopecia requires thousands of tissue grafts. In this paper, we propose an approach for the scalable and automated preparation of highly hair-inductive tissue grafts using a bioprinter. Two collagen droplets (2 µL each) containing mesenchymal and epithelial cells were placed adj

Applications of Ultrasound-Mediated Gene Delivery in Regenerative Medicine.

Research on the capability of non-viral gene delivery systems to induce tissue regeneration is a continued effort as the current use of viral vectors can present with significant limitations. Despite initially showing lower gene transfection and gene expression efficiencies, non-viral delivery methods continue to be optimized to match that of their viral counterparts. Ultrasound-mediated gene transfer, referred to as sonoporation, occurs by the induction of transient membrane permeabilization and has been found to significantly increase the uptake and expression of DNA in cells across many org

CRISPR technologies for stem cell engineering and regenerative medicine.

CRISPR/Cas9 system exploits the concerted action of Cas9 nuclease and programmable single guide RNA (sgRNA), and has been widely used for genome editing. The Cas9 nuclease activity can be abolished by mutation to yield the catalytically deactivated Cas9 (dCas9). Coupling with the customizable sgRNA for targeting, dCas9 can be fused with transcription repressors to inhibit specific gene expression (CRISPR interference, CRISPRi) or fused with transcription activators to activate the expression of gene of interest (CRISPR activation, CRISPRa). Here we introduce the principles and recent advances

Regenerative medicine therapy for single ventricle congenital heart disease.

One of the most complex forms of congenital heart disease (CHD) involving single ventricle physiology is hypoplastic left heart syndrome (HLHS), characterized by underdevelopment of the left ventricle (LV), mitral and aortic valves, and narrowing of the ascending aorta. The underdeveloped LV is incapable of providing long-term systemic flow, and if left untreated, the condition is fatal. Current treatment for this condition consists of three consecutive staged palliative operations: the first is conducted within the first few weeks of birth, the second between 4 to 6 months, and the third and

Dual-acting biofunctionalised scaffolds for applications in regenerative medicine.

Off the shelf scaffolds for replacing ultra-small diameter vascular grafts are valuable for reconstruction of diseased or damaged vessels. The limitations for such grafts include optimal handling with ready availability of varied lengths of grafts, graft patency with the ability to replace the function of active cellular mechanisms and adequate mechanical properties to maintain physicochemical function. We used a well-established, solvent casting method for potential tissue replacement scaffold fabrication with incorporated bioactive molecules, which we have previously explored to confer haemo

Stem cells from amniotic fluid--Potential for regenerative medicine.

Regenerative medicine has recently been established as an emerging field focussing on repair, replacement or regeneration of cells, tissues and whole organs. The significant recent advances in the field have intensified the search for novel sources of stem cells with potential for therapy. Recently, researchers have identified the amniotic fluid as an untapped source of stem cells that are multipotent, possess immunomodulatory properties and do not have the ethical and legal limitations of embryonic stem cells. Stem cells from the amniotic fluid have been shown to differentiate into cell linea

Regenerative medicine in orthopedics using cells, scaffold, and microRNA.

Cells, scaffold, and growth factors are crucially important in regenerative medicine and tissue engineering. Progress in science and technology has enabled development of these three factors, with basic research being applied clinically. In the past decade, we have investigated tissue regeneration in animal models of musculoskeletal disorders by using cells, scaffold, and delivery systems which has been relatively easy to apply and develop in clinical settings. Moreover, microRNA (miRNA), which are important in biological processes and in the pathogenesis of human diseases, have been used in r

Regenerative medicine for congenital diaphragmatic hernia: regeneration for repair.

Regenerative medicine has developed recently as a new field of science aiming at restoring organ and tissue damage through the use of autologous constructs. Cellular therapies and relatively simple tissue engineering reconstructions have recently been successfully applied into patients. For babies born with congenital diaphragmatic hernia, regenerative medicine may play a role both in developing a myogenic patch capable of restoring muscle function and promoting regeneration of hypoplastic lungs that characterised those patients. The latter is particularly attractive because it may change the

Engineered tissue grafts: opportunities and challenges in regenerative medicine.

The human body has limited regenerative capacity in most of the major tissues and organs. This fact has spurred the field of regenerative medicine, promising to repair damage following traumatic injury or disease. Multiple therapeutic strategies are being explored including small molecules, gene delivery, and stem cells; however, tissue engineering remains a primary approach to achieving regeneration. Organ transplantation demonstrates that damaged tissues can be replaced, but technology to regenerate complex organs de novo is not yet available. Instead, tissue engineering can augment the body

Mesenchymal stem cells for tissue engineering and regenerative medicine.

Mesenchymal stem cells (MSCs) exist in bone marrow and other musculoskeletal tissues. These cells contribute to the homeostasis of musculoskeletal tissue as well as support for the growth and differentiation of primitive hemopoietic cells. Recent advancements in tissue engineering and regenerative medicine have highlighted MSCs as a potential source of cells which would differentiate to a variety of tissue tailored to individual needs. This paper briefly outlines the current status of MSCs, focusing on their biological characteristics and potential for clinical applications.

Regenerative medicine: Stem cell research turns to the spleen.

Extract: Though conventional medical wisdom has long considered the spleen a dispensable organ, it appears to be more than it seems. Newly identified stem cell populations have been found in the spleen of adult mice, challenging conventional wisdom and providing a potential source of stem cells for treating disease. These stem cells are relevant to future cellular therapies for diabetes and other diseases. At the interface of the circulatory and immune systems, the spleen normally has dual roles in maintenance and adaptation to stress or disease. The red pulp of the spleen holds macrophages (a

Chitosan in regenerative medicine: a translational perspective on biomaterial design.

Chitosan is an aminopolysaccharide platform with broad relevance to regenerative medicine because its charge density, solubility, degradation, processability, and biological interactions can be adjusted through source selection, deacetylation chemistry, molecular-weight control, derivatization, and formulation. However, this tunability is accompanied by substantial source- and process-dependent variability, which represents a central translational challenge: materials labelled as chitosan can differ markedly in degree of deacetylation, molecular weight, pattern of acetylation, polydispersity,

Designing of porous scaffolds for tissue engineering and regenerative medicine.

The potential of the porous scaffolds lies in its dynamic architectural features, such as porosity, pore size & shape, interconnectivity, and spatial distribution which are highly essential in tissue engineering applications. The porous structure of the scaffolds influences the biological and physicochemical activities in the tissue regeneration process. The porosity also allows better distribution of nutrients and diffusion of biological fluids thereby dispersing the seeding material uniformly. Generally, damaged tissues can be regenerated on their own but many times due to unavoidable factor
