MircoRNAs as regulators of regulated cell death in cancer progression: a focus on ferroptosis, anoikis and pyroptosis.

Cancer progression is shaped not only by genetic alterations but also by dynamic remodeling of regulated cell death. MicroRNAs (miRNAs) have emerged as central regulators of these processes by coordinating gene expression, metabolic balance, and stress responses. This review summarizes current understanding of how miRNAs modulate ferroptosis, anoikis, and pyroptosis across liver, lung, gastric, breast, and other cancers. They influence ferroptosis through lipid peroxidation and antioxidant systems, regulate anoikis via integrin signaling and cytoskeletal remodeling, and tune pyroptosis through

Copper-regulated cell death after spinal cord injury: evidence boundaries for non-coding RNA, epigenetics and cuproptosis.

BACKGROUND: Spinal cord injury (SCI) initiates a secondary-injury cascade involving mitochondrial dysfunction, metal dyshomeostasis, neuroinflammation, and regulated cell death. Cuproptosis, a copper-dependent form of regulated cell death driven by lipoylated mitochondrial protein aggregation, has reframed copper dyshomeostasis as a potential molecular vulnerability rather than a passive trace-metal disturbance. MAIN BODY: This targeted narrative review critically synthesizes the FDX1-lipoylation-DLAT/DLST cuproptosis pathway, copper trafficking and redistribution after SCI, and the evidence

RIPK1 in Alzheimer's Disease: A Context-Dependent Signaling Hub Linking Aβ, Tau, Neuroinflammation, and Regulated Cell Death.

Alzheimer's disease is a multifactorial neurodegenerative disorder in which amyloid-β accumulation, tau pathology, neuroinflammation, synaptic dysfunction, vascular injury, and neuronal loss interact across multiple cellular compartments. Although amyloid and tau remain central to disease definition and biomarker staging, growing evidence indicates that inflammatory and regulated cell-death pathways actively shape disease progression. Receptor-interacting protein kinase 1 (RIPK1) has emerged as a context-dependent signaling checkpoint that links inflammatory signaling to cell-fate decisions. I

Statins as Repurposed Anticancer Agents: Regulated Cell Death, the Tumor Immune Microenvironment, and the Translational Evidence Gap.

Statins inhibit 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR) and reduce flux through the mevalonate pathway, which supplies both cholesterol and the non-sterol isoprenoids required for prenylation of small GTPases. Because this pathway is frequently activated in cancer, statins have become one of the most intensively studied candidates for oncological drug repurposing. Preclinical work indicates that statins can lower the threshold for apoptosis, modulate autophagy in either a pro-death or a cytoprotective direction, induce pyroptosis, and sensitize cells to ferroptosis, while also

Identification and validation of prognostic genes associated with m6A-regulated programmed cell death in acute lymphoblastic leukemia.

This study investigated the prognostic significance of m6A-related programmed cell death (PCD) genes in acute lymphoblastic leukemia (ALL). Transcriptomic data from the TCGA-ALL and GSE48558 datasets were analyzed to identify m6A-related PCD genes (corgenes), which were then intersected with 3,063 differentially expressed genes (DEGs), resulting in 23 candidate genes. Univariate Cox regression and LASSO regression analyses identified CFLAR and CDK4 as key prognostic genes, facilitating the construction of a high-accuracy risk prediction model. Gene set enrichment analysis (GSEA) revealed signi

Immunogenic cell death after combined treatment with radiation and ATR inhibitors is dually regulated by apoptotic caspases.

INTRODUCTION: Inhibitors of the ATR kinase act as radiosensitizers through abrogating the G2 checkpoint and reducing DNA repair. Recent studies suggest that ATR inhibitors can also increase radiation-induced antitumor immunity, but the underlying immunomodulating mechanisms remain poorly understood. Moreover, it is poorly known how such immune effects relate to different death pathways such as caspase-dependent apoptosis. Here we address whether ATR inhibition in combination with irradiation may increase the presentation of hallmark factors of immunogenic cell death (ICD), and to what extent c

Mechanism and regulation of programmed cell death in aging-related diseases.

Aging and its associated diseases are characterized by a complex interplay of cellular dysfunction, chronic inflammation, and tissue degeneration. Dysregulated cell death and the resulting inflammatory responses are pivotal drivers of this pathological synergy. Among various cell demise pathways, programmed cell death (PCD) mechanisms, including apoptosis, necroptosis, and pyroptosis, are critical for maintaining homeostasis but can drive pathology when aberrantly activated. Crucially, they leave specific and reliable biomarkers, allowing for their distinct assessment in complex aging tissues.

Mechanisms regulating host cell death during Leishmania infection.

Parasites from the Leishmania genus are the causative agents of leishmaniasis and primarily reside within macrophages during mammalian infection. Their ability to establish intracellular infection provides a secure niche for proliferation while evading detection. However, successful multiplication within mammalian cells requires the orchestration of multiple mechanisms that control host cell viability. In contrast, innate immune cells, such as macrophages, can undergo different forms of cell death in response to pathogenic intracellular microbes. Thus, modulation of these different forms of ho

Multidimensional role of adapalene in regulating cell death in multiple myeloma.

AIMS: Multiple myeloma (MM) remains a challenging condition to cure, with persistent drug resistance negating the benefits of treatment advancements. The unraveling complexities in programmed cell death (PCD), inclusive of apoptosis, autophagy, and ferroptosis, have highlighted novel therapeutic avenues. Our study focuses on deciphering how adapalene (ADA), a small molecule compound, accelerates the demise of MM cells via targeting their compensatory survival mechanisms. METHODS: To assess the impact of ADA on MM, we employed flow cytometry and trypan blue exclusion assays to determine cell v

CaMKII in Regulation of Cell Death During Myocardial Reperfusion Injury.

Cardiovascular disease is the leading cause of death worldwide. In spite of the mature managements of myocardial infarction (MI), post-MI reperfusion (I/R) injury results in high morbidity and mortality. Cardiomyocyte Ca2+ overload is a major factor of I/R injury, initiating a cascade of events contributing to cardiomyocyte death and myocardial dysfunction. Ca2+/calmodulin-dependent protein kinase II (CaMKII) plays a critical role in cardiomyocyte death response to I/R injury, whose activation is a key feature of myocardial I/R in causing intracellular mitochondrial swelling, endoplasmic retic

NMDA receptor regulation of cell death in the rat olfactory bulb.

Cell death is widespread in the developing nervous system and is under complex regulation by numerous intra- and intercellular mechanisms. Blockade of the N-methyl-D-aspartate (NMDA) subtype of glutamate receptor has been shown to promote cell death in the developing brain (Ikonomidou et al., 1999), suggesting that afferent functional activation is an important regulator of cell survival. The olfactory bulb, the first central relay for olfactory information from the nose, is well suited for examining the role of afferent activity in neuronal development. Functional deprivation is easily perfor

CRISPR-Cas9 screen to identify genes regulating cell death.

Regulated cell death mediated by dedicated molecular machines, known as programmed cell death, plays important roles in health and disease. Understanding the mechanisms of cell death is crucial for elucidating the control of cellular homeostasis and developing therapies for related diseases. Despite extensive research efforts spanning decades, many aspects of cell death mechanisms remain elusive, highlighting the need for continued exploration. Here, we describe how to identify novel regulators involved in cell death pathways using a genome-wide screening approach.

Regulation of immunogenic cell death and potential applications in cancer therapy.

Immunogenic cell death (ICD), a type of regulatory cell death, plays an important role in activating the adaptive immune response. Activation of the tumor-specific immune response is accompanied by the cell surface exposure of calreticulin and heat-shock proteins, the secretion of adenosine triphosphate, and the release of high mobility group box-1. In this review, we summarize and classify the latest types of ICD inducers and their molecular mechanisms, and discuss the effects and potential applications of inducing ICD by chemotherapy drugs, targeted drugs, and oncolytic viruses in clinical r

NK cells promote cardiac cell death and regulate myelopoiesis in myocardial infarction.

Ischemic heart failure remains a major clinical challenge, underscoring the need to better understand post-infarction immune mechanisms and identify new therapeutic targets. Both innate and adaptive immunity contribute to adverse cardiac remodeling following myocardial infarction (MI), yet the role of cytotoxic cells such as natural killer (NK) cells remains poorly defined. Here, we show that after acute MI in mice, NK cells are recruited to the ischemic myocardium in a CCR2-dependent manner and become activated. Activated NK cells locally release granzyme B, promoting cardiomyocyte apoptosis,

Programmed cell death regulates hematopoietic cell homeostasis under radiation conditions.

BACKGROUND: It is well-known that hematopoietic cells are sensitive to irradiation exposure. Apoptosis, necroptosis, pyroptosis and ferroptosis might contribute to irradiation-induced hematopoietic injury. However, it is uncertain whether different hematopoietic cells apply specific cell death pathways under irradiation exposure. METHODS: We investigated the role of different programmed cell death pathways in irradiation-induced hematopoietic cell injury. In order to study the acute and long-term effects of ionizing radiation on hematopoietic system, we established injury models of mice at di

Modulating regulated cell death: mechanistic insights into traditional Chinese medicine metabolites for ischemia/reperfusion-induced acute kidney injury.

Acute kidney injury (AKI), characterized by a rapid decline in renal function, represents a significant global health burden due to its high mortality and frequent progression to chronic kidney disease (CKD). Ischemia/reperfusion (I/R) injury is a major cause of AKI, yet clinically effective pharmacotherapies remain elusive. The pathogenesis of I/R-AKI is critically driven by regulated cell death (RCD) pathways-including apoptosis, ferroptosis, pyroptosis, and necroptosis-which mediate tubular epithelial damage and inflammatory responses. Consequently, targeting these RCD pathways emerges as a

Unravelling the multi-faceted roles of microRNAs in non-apoptotic regulated cell death (RCD) to improve cancer therapy.

MicroRNAs (miRNAs) are short non-coding RNAs that orchestrate post-transcriptional gene regulation and govern critical cellular processes. Their dysregulation in cancer is intimately associated with tumor progression and metastasis. Concurrently, regulated cell death (RCD), particularly non-apoptotic forms, has emerged as a vital target for cancer therapy due to its ability to bypass apoptosis resistance. Growing evidence indicates that miRNAs modulate non-apoptotic RCD by functioning as tumor suppressors or oncogenes. Consequently, miRNA-based interventions may synergize with chemotherapy and
