Effects of d-amphetamine and chlordiazepoxide on spaced responding in pigeons.

The effects of d-amphetamine and chlordiazepoxide were studied in pigeons on performance (1) under a schedule that reinforced responses on a key (food key) if they were more than 20 sec apart, (2) under the same schedule when responses also were required on a collateral key during the interresponse time on the food key, and (3) under the same schedule when responses were required on a collateral key during the interresponse time on the food key and collateral-key responses could produce a stimulus correlated with the availability of food. Under all three spaced-responding schedules, d-amphetam

Response inhibition is impaired by developmental methylmercury exposure: acquisition of low-rate lever-pressing.

Developmental methylmercury (MeHg) exposure produces response perseveration on discrimination reversal procedures, disrupts sensitivity to reinforcement, and enhances sensitivity to dopamine agonists - a profile suggesting a deficit in behavioral inhibition. To examine inhibition, we examined MeHg's effects on the acquisition and persistence of low-rate lever-pressing following a history of high-rate responding. Additionally, we examined whether chronic exposure to selenium protects against MeHg's developmental neurotoxicity. Female rats were exposed in utero via maternal exposure to drinking

Behavioral and neurochemical effects of cocaine and diphenhydramine combinations in rhesus monkeys.

RATIONALE: Diphenhydramine (DPH) is an over-the-counter medication used in the treatment of allergic symptoms. While DPH abuse is infrequent, recent preclinical evidence suggests that DPH and cocaine combinations may have enhanced reinforcing properties. OBJECTIVE: The aims were to assess the reinforcing effectiveness of cocaine and DPH alone or in combination under a second-order schedule of reinforcement and to examine the neurochemical basis of this interaction using in vivo microdialysis in awake rhesus monkeys. MATERIALS AND METHODS: Cocaine (0.03-0.3 mg/kg per injection), DPH (0.3-3.0

Effects of extended-access self-administration and deprivation on breakpoints maintained by cocaine in rats.

RATIONALE: Animal models that identify the effects of self-administration histories on subsequent patterns, levels of intake, and other aspects of reinforcement will help clarify the controlling variables of human drug use. OBJECTIVE: Identify the effects of extended-access to cocaine and 1 or 7 days of deprivation on cocaine-maintained breakpoints on a progressive ratio (PR) schedule of reinforcement. METHODS: Male, Sprague-Dawley rats were trained to self-administer intravenous cocaine (expt 1: 1.5 mg/kg per infusion; expt 2: 0.75 mg/kg per infusion), and then given various histories of se

Oral drug self-administration: an overview of laboratory animal studies.

Many abused drugs can be established as orally delivered reinforcers for rhesus monkeys and other animals. Benzodiazepines, barbiturates, opioids, psychomotor stimulants, dissociative anesthetics, and ethanol can come to serve as reinforcers when taken by mouth. The principal problems in establishing drugs as reinforcers by the oral route of administration are (1) aversive taste, (2) delay in onset of central nervous system effects, and (3) consumption of low volumes of drug solution. Strategies have been devised to successfully overcome these problems, and orally delivered drugs can be establ

Examination of clinical variables affecting resurgence: A reanalysis of 46 applications.

Despite the efficacy of behavioral interventions, resurgence of challenging behavior (e.g., aggression, self-injury) following successful treatment can still occur. Applied work has focused on identifying treatment-related variables thought to affect the occurrence and magnitude of resurgence. The current study describes the relation between several variables (i.e., phase duration, response rates in baseline and treatment, obtained rates of reinforcement, downshift in reinforcement step size) and resurgence in a retrospective consecutive controlled case series of 46 treatment applications for

Comparison of dehydroepiandrosterone (DHEA) and pregnanolone with existing pharmacotherapies for alcohol abuse on ethanol- and food-maintained responding in male rats.

The present study compared two putative pharmacotherapies for alcohol abuse and dependence, dehydroepiandrosterone (DHEA) and pregnanolone, with two Food and Drug Administration (FDA)-approved pharmacotherapies, naltrexone and acamprosate. Experiment 1 assessed the effects of different doses of DHEA, pregnanolone, naltrexone, and acamprosate on both ethanol- and food-maintained responding under a multiple fixed-ratio (FR)-10 FR-20 schedule, respectively. Experiment 2 assessed the effects of different mean intervals of food presentation on responding for ethanol under a FR-10 variable-interval

An optimized procedure for robust volitional cocaine intake in mice.

Substance use disorder (SUD) is a behavioral disorder characterized by volitional drug consumption. Mouse models of SUD allow for the use of molecular, genetic, and circuit-level tools, providing enormous potential for defining the underlying mechanisms of this disorder. However, the relevance of results depends on the validity of the mouse models used. Self-administration models have long been the preferred preclinical model for SUD as they allow for volitional drug consumption, thus providing strong face validity. While previous work has defined the parameters that influence intravenous coca

Prior commitment: Its effect on suboptimal choice in a gambling-like task.

Animals choose suboptimally when provided with cues that signal whether reinforcement is coming or not. For example, pigeons do not prefer an alternative that always provides them with a signal for reinforcement over an alternative that provides them with a signal for reinforcement only half of the time and a signal for the absence of reinforcement the rest of the time. In the present research, we tested the hypothesis that if the results of the choice are delayed, pigeons will choose less suboptimally. We tested this hypothesis by forcing pigeons to wait following their choice, requiring them

Noncontingent escape as treatment for self-injurious behavior maintained by negative reinforcement.

We extended research on the role of noncontingent positive reinforcement following a functional analysis of attention-maintained self-injurious behavior to self-injury maintained by negative reinforcement in 2 young males with developmental disabilities. During a pretreatment functional analysis, each participant's self-injury was shown to be differentially sensitive to escape from instructional activities as negative reinforcement. During noncontingent escape, escape from learning activities was provided on a fixed-time schedule that was not influenced by the participant's behavior. One parti

The phosphodiesterase-4 inhibitor rolipram attenuates heroin-seeking behavior induced by cues or heroin priming in rats.

Inhibition of phosphodiesterase-4 (PDE4), an enzyme that specifically hydrolyzes cyclic adenosine monophosphate (cAMP) increases intracellular cAMP/cAMP-response element binding protein (CREB) signaling. Activation of this signaling is considered as an important compensatory response that decreases motivational properties of drugs of abuse. However, it is not known whether PDE4 is involved in heroin seeking. Self-administration of heroin (50 μg/kg/infusion) was performed under the fixed ratio 1 (FR1) schedule for 14 d and then drug seeking was extinguished for 10 d. The progressive ratio sched

Differential reinforcement using matched examples and nonexamples expedites the practice needed to learn concepts.

Conceptual learning refers to correctly identifying novel examples and nonexamples of a concept. Concepts can be taught by presenting stimuli and differentially reinforcing responses to examples but not nonexamples. However, it is unclear which stimuli should be used and whether specifically designed stimuli can result in correct responding even in the absence of differential reinforcement. Stimulus selection can be informed by identifying features that a stimulus must have to be an example and can have without altering whether the stimulus is an example or a nonexample. Previous research sugg

Parametric evaluation of procedural fidelity level with combined errors during differential reinforcement of other behavior.

Differential reinforcement of other behavior (DRO) is an intervention used to address challenging behavior; however, its effectiveness may be diminished by procedural fidelity errors. Previous research has shown that DRO effects may be robust despite marginal errors of commission and omission, but the effects of combined errors (i.e., both commission and omission errors) on the efficacy of DRO have not been investigated. The purpose of this study was to parametrically evaluate procedural fidelity level (i.e., 100, 80, 60, 40, and 20% fidelity) in DRO when implemented with combined errors in a

[Near miss outcomes in gambling games].

BACKGROUND: Games of chance operate with an intermittent reinforcement schedule in which the number of games takes the player to win differ in each turn thus they can not predict when the next positive reinforcement arrives. The near miss outcome (close to winning but actually a losing outcome) can be interpreted as a secondary (built in) reinforcement within variable ratio reinforcement schedule that presumably contribute to the development and maintanance of gambling addiction. METHODS: The aim of this publication would be to introduce near miss outcomes and to summarize and critically anal

Effects of variable and fixed second-order schedules on schedule-induced polydipsia in the rat.

Schedule-induced polydipsia was studied in rats on a variable second-order schedule in Experiments 1 and 2. Drinking occurred only following presentations of the pellet reinforcer and was not induced by a stimulus that was associated with pellet delivery. Moreover, it was shown that lever pressing for food was not competing with the opportunity to drink following the stimulus during non-reinforced intervals. In Experiment 3 a fixed second-order schedule was used, and only one animal drank following the presentation of the stimulus. A positive contrast effect was obtained with schedule-induced

Selective suppression of schedule-induced ethanol drinking by antialcoholic drugs in rats.

Effects of disulfiram and calcium cyanamide, antialcoholic drugs, on schedule-induced ethanol drinking as well as on schedule-controlled response (lever-pressing) under a fixed interval 1 min schedule of food reinforcement were investigated in Wistar strain rats. When ethanol solution was available, the schedule-induced ethanol drinking decreased depending on the ethanol concentration (2-8%). However, the dose of ethanol intake during the 1 hr experimental session was at maximum (2.8 g/kg) when 4% ethanol solution was available. Thereafter, 4% ethanol solution was used in the experiment for st

Attenuation of cocaine and heroin seeking by μ-opioid receptor antagonism.

RATIONALE: Evidence has implicated the endogenous opioids, in particular μ-opioid receptors, in emotional behavior and regulation of reward circuits, especially in the context of heroin addiction and hedonic responses to ingestive rewards. The μ-opioid receptor antagonist naltrexone (NTX) has been reported to be effective in preventing relapse to alcoholism and in reducing alcohol and cocaine craving during abstinence. OBJECTIVES: The aim of the present experiments was to investigate the effects of a novel selective μ-opioid receptor antagonist GSK1521498 on cocaine and heroin seeking and the

mGluR5 antagonism attenuates methamphetamine reinforcement and prevents reinstatement of methamphetamine-seeking behavior in rats.

Addiction to methamphetamine is a significant public health problem, and there are currently no pharmacological agents that are approved for the treatment of addiction to this powerful psychostimulant. Chronic methamphetamine use leads to cognitive dysfunction as well as numerous psychiatric, neurological, and cardiovascular complications. There is a growing body of literature implicating an important role for glutamate neurotransmission in psychostimulant addiction. In the present study, we examined the effects of the selective type 5 metabotropic glutamate receptor (mGluR5) antagonist 3-((2-

Effects of diazepam on schedule-controlled and schedule-induced behavior under signaled and unsignaled shock.

Schedule-controlled lever pressing and schedule-induced licking were studied in rats under a multiple fixed-interval fixed-interval schedule of food reinforcement upon which was superimposed a multiple variable-time variable-time schedule of electric-shock delivery. Shocks were signaled in one component of the multiple schedule and unsignaled in the other. The effects of diazepam upon the suppression of behavior during the signal (conditioned suppression) and during signaled and unsignaled shock (differential suppression) were studied under several shock intensities (Experiment 1) and at incre
