Throughout most of its range, the brown dog tick, Rhipicephalus sanguineus (Latreille), prefers dogs as a host, but human bites occasionally occur in the Mediterranean region and Central America; historically, this species has rarely bitten humans in the United States. A focus of 15 human bite cases by this tick is reported from four Air Force bases located within 200 miles of each other in north central Texas and southwestern Oklahoma. The sudden appearance of numerous documented bite cases indicates that either the species is becoming more anthropophilic in this area or than an introduction

The cattle tick Rhipicephalus microplus is a major livestock pest and disease vector in tropical and subtropical regions. Host species exhibit heritable variation in tick susceptibility, resulting in differences in tick burden. Building on our previous finding that R. microplus predominates on both Bos indicus and Bos frontalis across seasons and altitudes, with greater tick diversity in B. frontalis, this study examines host-parasite interactions between these bovine hosts. In Arunachal Pradesh, the semi-domesticated Bos frontalis and domesticated Bos indicus are significant bovine hosts with

Given the need to develop sustainable strategies for the management of ectoparasites, the inclusion of eco-friendly mycoacaricide and/or mycoinsecticide products has become increasingly imperative. Rhipicephalus microplus is an ectoparasitic arthropod of major economic and sanitary importance in livestock production, acting as a vector of pathogens of veterinary relevance and causing substantial production losses. Control of this tick species has become progressively more challenging due to the widespread development of resistance to synthetic chemical acaricides commonly used in cattle produc

This study was conducted to assess the progression of deltamethrin resistance in Rhipicephalus (B.) microplus populations over a 5-year period, from 2019 to 2023, on selected communal farms in the Eastern Cape Province, South Africa. Ticks were collected from three communal farms with a documented history of deltamethrin resistance in R. (B.) microplus populations in 2019. The sampled tick populations were tested for resistance using the adult immersion test (AIT). The data generated from the AIT indicated deltamethrin resistance, with oviposition inhibition percentages ≤ 95% at both the manuf

Ovine babesiosis, caused by Babesia ovis, is a significant tick-borne disease affecting sheep globally, with severe economic implications for sheep farming, particularly in Türkiye. Babesia ovis is transmitted exclusively by adult Rhipicephalus bursa ticks, but the potential role of infected larval stages in modulating disease severity has remained unclear. This study investigated whether infestation with B. ovis-infected R. bursa larvae reduces the severity of babesiosis following subsequent exposure to infected adult ticks. Three experimental sheep were infested with B. ovis-infected larvae,

The present study was conducted to evaluate the effects of carvacrol and thymol on the antioxidant and detoxifying enzymes of larvae from two populations of R. microplus: Jaguar (tick population resistant to six classes of acaricides) and Porto Alegre (susceptible tick population). Carvacrol and thymol were tested at concentrations ranging from 0.14 to 5.0 mg mL-1 in both populations to determine the LC50. In addition, the LC1, LC25, and LC75 were estimated using the LC50 and HillSlope of each compound. Larvae of both populations of R. microplus were then treated with the LC1, LC25, LC50, and

The resistance of Rhipicephalus microplus to acaricides is a serious control problem, so its early diagnosis by a molecular technique is important. This study aims to develop a multiplex allele-specific polymerase chain reaction (PCR) for single-nucleotide polymorphisms (SNPs) in the para-sodium channel gene and in the GABA-Cl gene, associated with pyrethroids (cypermethrin and flumethrin) and fipronil resistance, respectively. We used 22 tick field isolates from farms with tick control problems (sampling convenience). These farms are located in departments of northern Uruguay. Three mutations

Rhipicephalus microplus, better known as the Asiatic cattle tick, is a largely invasive ectoparasite of great economic importance due to the negative effect it has on agricultural livestock on a global scale, particularly cattle. Tick-borne diseases (babesiosis and anaplasmosis) transmitted by R. microplus are alarming as they decrease the quality of livestock health and production. In sub-Saharan Africa, cattle represent a major source of meat and milk, but this region of the world is severely affected by the Rhipicephalus microplus tick. The principal method for tick control is the use of ch

Proteins belonging to the serine protease inhibitor (serpin) superfamily play essential roles in many organisms. In arthropods these proteins are involved in innate immune system, morphogenesis and development. In mammals serpins regulate pathways that are essential to life such as blood coagulation, fibrinolysis, inflammation and complement activation, some of which are considered the host's first line of defense to hematophagous and/or blood dueling parasites. Thus, it is hypothesized that ticks use serpins to evade host defense, facilitating parasitism. This study describes eighteen full-le

Rickettsia conorii, the etiologic agent of Mediterranean spotted fever is widely distributed in Southern Europe, the Middle East, Africa, India and the Caspian region. In the Mediterranean region, the brown dog tick, Rhipicephalus sanguineus, is the recognized vector of R. conorii. To study tick-pathogen relationships and pathogenesis of infection caused in model animals by the bite of an infected tick, we attempted to establish a laboratory colony of Rh. sanguineus persistently infected with R. conorii. Rhipicephalus sanguineus ticks of North American and Mediterranean origin were exposed to

Rhipicephalus simus Koch was removed from a human upon his return to Connecticut, U.S.A. from Kenya, Africa. This person became ill in Africa, exhibited clinical manifestations of boutonneuse fever and had a significant rise (greater than or equal to 8-fold) in IgM titer against spotted fever-group rickettsiae. Rickettsia conorii was probably the infectious agent.

The cattle tick Rhipicephalus microplus is a vector for Anaplasma marginale in tropical and subtropical regions worldwide. The study of pathogen-microbiome-vector interactions at the tick organ scale is a promising area of development, unraveling control strategies for tick-borne diseases. Our study focused on how A. marginale infection affects the bacterial community within the salivary glands and ovaries of R. microplus. Engorged female ticks collected from cattle in a disease-endemic area of Argentina were further classified based on whether they were infected with A. marginale by targeting

The functional significance of a non-conserved cysteine residue (C86) proximal to the interfacial region of Rhipicephalus microplus triosephosphate isomerase (RmTIM) was investigated through systematic substitution with aspartic acid (C86D), lysine (C86K), and alanine (C86A) via site-directed mutagenesis. Kinetic analyses revealed substantial perturbations in enzymatic parameters for the C86D and C86K variants, with marked alterations in maximal velocity (Vmax), substrate affinity (Km), catalytic turnover (kcat), and catalytic efficiency (kcat/Km). Thermodynamic destabilization was universally

The tick Rhipicephalus sanguineus is one of the main ectoparasites that affects dogs, causing direct and indirect damage to parasitized animals. Currently, infestation control is mainly carried out by using synthetic acaricidal drugs. However, a decrease in efficacy and an increase in resistance to the main therapeutic protocols against tick infestations have been increasingly reported and confirmed, a factor that has driven research into the potential acaricide activity of natural compounds, including in association with synthetic molecules. The aim of this work was to evaluate whether the co

Sphingomyelinase D is a toxin present in venomous spiders and bacteria and is associated with infection symptoms in patients affected by spider bites. It was observed that in Ixodes scapularis ticks, sphingomyelinase-like protein secreted in saliva can modulate the host immune response, affecting the transmission of flavivirus to the host via exosomes. In this work, a sphingomyelinase D-like protein (RmSMase) from R. microplus, a tick responsible for economic losses and a vector of pathogens for cattle, was investigated. The amino acid sequence revealed the lack of important residues for enzym

The cattle tick Rhipicephalus microplus is one of the most important ectoparasites with great sanitary and economic impact for cattle rearing worldwide. Ivermectin is commonly used to control tick populations, but its use over the last 30 years has led to the development of resistant populations of R. microplus, and a concomitant loss of efficacy. In this context, we aimed to determine the metabolic mechanisms that contribute to ivermectin resistance in a resistant strain of this species. We performed lethal time bioassays with inhibitors of detoxifying enzymes and xenobiotic transporters (fou

Control of ticks on dogs is often done by application of repellents that contain permethrin as the active ingredient. In this research, we studied the role of a glutathione S-transferase (GST) gene in detoxification of permethrin by ticks using a gene silencing method RNA interference (RNAi). The brown dog tick, Rhipicephalus sanguineus, used in these studies, has a notable host preference for dogs, but also infests other mammals. In this research, R. sanguineus females were injected with gst double-stranded RNA (dsRNA) to effect gene silencing by RNAi and then exposed to sublethal doses of pe

The control of Rhipicephalus annulatus ticks in Egypt and other countries relies principally on the application of acaricides which have many drawbacks. Recently, cattle vaccination against ticks showed a potential unconventional approach to control ticks. As a target, salivary glands contain various proteins that may play specific roles during attachment, feeding and may modulate the immune system of the host. We have performed immunoscreening on expression normalized cDNA library to identify unique R. annulatus proteins from salivary gland (RaSal) that are particularly expressed during engor

Sixty-two questing adult Rhipicephalus sanguineus (Latreille) ticks were collected by direct removal from blades of turfgrass and adjacent concrete walkways at a suburban home in Riverside County, CA, and tested for the presence of Rickettsia, Bartonella, and Ehrlichia DNA. Polymerase chain reaction (PCR) was used to amplify fragments of the 17-kDa antigen gene and the rOmpA gene of the spotted fever group rickettsiae. One male tick contained R. rickettsii DNA; its genotype differed from R. rickettsii isolates found in Montana and Arizona that cause Rocky Mountain spotted fever and from Hlp#2

Twenty Rhipicephalus sanguineus ticks collected in eastern Arizona were tested by PCR assay to establish their infection rate with spotted fever group rickettsiae. With a nested PCR assay which detects a fragment of the Rickettsia genus-specific 17-kDa antigen gene (htrA), five ticks (25%) were found to contain rickettsial DNA. One rickettsial isolate was obtained from these ticks by inoculating a suspension of a triturated tick into monolayers of Vero E6 monkey kidney cells and XTC-2 clawed toad cells, and its cell culture and genotypic characteristics were determined. Fragments of the 16S rR
