Preparation of high-purity 1,3-distearoyl-2-oleoylglycerol (StOSt) via a two-step enzymatic interesterification combined with two-step solvent fractionation.

High-purity 1,3-distearoyl-2-oleoylglycerol (StOSt) was produced from high-oleic sunflower oil and ethyl stearate via a two-step enzymatic interesterification combined with a two-step solvent fractionation. Lipozyme RM IM (Rhizomucor miehei) was employed as a biocatalyst and the reaction was conducted in a packed-bed reactor. Molecular distillation was used to remove fatty acid ethyl esters from the reaction mixtures after enzymatic reactions. The optimum conditions were a temperature of 55 °C, a water content of 300 ppm (based on the total weight of the substrate), and a molar ratio of 1:8 (t

Jianqu fermentation with the isolated fungi significantly improves the immune response in immunosuppressed mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Jianqu, a classical formula of traditional Chinese medicine, is used clinically to treat symptoms like chill and fever headache, diarrhea and loss of appetite and act on patients with low immunity. However, the quality control of Jianqu fermentation is not well established, and its function in regulating the body's immunity still remains unclear. AIM OF THE STUDY: The present study firstly assesses the structure and diversity of fungal community during Jianqu fermentation and then investigates the immune regulating function of Jianqu extract in mouse model. Sustainable Modification of Minimolide: Influence of Structure and Acylation Pattern on Anti-Trypanosoma cruzi Activity.

Minimolide, a sesquiterpene lactone isolated from Mikania minima, has displayed promising activity against Trypanosoma cruzi. The biocatalytic derivatization of minimolide and its analogs via selective acetylation and deacetylation is reported here using commercial lipases under mild and sustainable conditions. Reaction optimization identified Rhizomucor miehei lipase (RMIM) and Candida antarctica lipase B (CAL B) as the most efficient catalysts for acetylation and regioselective deacetylation, respectively. Structural transformations allow the synthesis of heliangolide and elemanolide analogs

Active Site Studies to Explain Kinetics of Lipas`s in Organic Solvents Using Molecular Dynamics Simulations.

This study investigates the intricate dynamics underlying lipase performance in organic solvents using comprehensive molecular dynamics (MD) simulations, supported by enzyme kinetics data. The study reveals that a single criterion can neither predict nor explain lipase activity in organic solvents, indicating the need for a comprehensive approach. Three lipases were included in this study: Candida antarctica lipase B (CALB), Rhizomucor miehei lipase (RML), and Thermomyces lanuginosus lipase (TLL). The lipases were investigated in acetonitrile, methyl tert-butyl ether, and hexane with increasin

Lipase-catalysed acidolysis for enrichment of medium-chain triacylglycerols in virgin coconut oil and its functional properties.

Lipase-catalysed acidolysis has emerged as a promising biocatalytic approach for producing structured lipids. In particular, the enrichment of medium-chain triacylglycerols (MCTs) in oils via lipase catalysis is attracting great attention due to their health benefits. In addition, MCTs are a known source of instant energy. In the current work, the immobilized lipase-catalysed enrichment of medium-chain triacylglycerols (MCTs) in a virgin coconut oil (VCO) matrix was achieved. Lipase -catalysed acidolysis of VCO with caprylic (C8:0) and capric acids (C10:0) was conducted to enhance the concentr

Pulmonary zygomycosis with Cunninghamella bertholletiae in a killer whale (Orcinus orca).

An adult female killer whale (Orcinus orca) was transported to the Port of Nagoya public aquarium in June 2010. While the animal was being maintained in the aquarium there was a gradual decrease in body weight. On October 1st, 2010 the whale exhibited signs of gastrointestinal disease and died on January 14th, 2011. At necropsy examination the gastric compartments were filled with a large number of variably-sized rocks (total weight 81.4 kg) and there was marked ulceration in the third compartment. There were multifocal tubercle-like nodules within the lungs and on sectioning there were numero

Burn wounds infected by contaminated water: case reports, review of the literature and recommendations for treatment.

First-aid education for the management of burns advocates cool running water over burnt skin to limit soft tissue damage. However, the water used may itself constitute a risk. We report three cases of severe invasive and necrotizing infection in patients who used or immersed themselves in contaminated water in an attempt to extinguish the fire following acute major burns. Wound cultures from all patients yielded Aeromonas hydrophila and two yielded Bacillus cereus. One patient had a complex polymicrobial infection, including zygomycosis with Rhizomucor variabilis. All patients were treated agg

Rhino-Orbito-Cerebral Mucormycosis: A Challenging Case.

Mucormycosis is a rare opportunistic fungal infection caused by Mucorales and associated with high mortality rates. Rhino-orbito-cerebral localization usually occurs in individuals with uncontrolled diabetes mellitus. We report the case of a 41-year-old male, with previously undiagnosed diabetes, who presented with unilateral facial extensive black eschar and signs of diabetic ketoacidosis. Cerebral magnetic resonance imaging showed left pansinusitis, left craniofacial edematous infiltrate, and left proptosis. A left internal temporal abscess was identified at an early pre-suppurative stage. M

Lactoferrin, a potential iron-chelator as an adjunct treatment for mucormycosis - A comprehensive review.

Mucormycosis is a deadly infection which is caused by fungi of the order Mucorales including species belonging to the genus Rhizopus, Mucor, Mycocladus, Rhizomucor, Cunninghamella, and Apophysomyces. Despite antifungal therapy and surgical procedures, the mortality rate of this disease is about 90-100% which is exceptionally high. The hypersensitivity of patients with raised available serum iron indicates that the Mucorales are able to use host iron as a critical factor of virulence. This is because iron happens to be a crucial element playing its role in the growth of cells and development. I

Enhancing the Stability of Fungal Lipases by Immobilization onto Accurel MP 1000 Support and Additional Glutaraldehyde Crosslinking.

Commercial fungal lipases from Rhizopus oryzae, Rhizopus niveus, Aspergillus niger, Rhizomucor miehei, and Candida rugosa were immobilized via physical adsorption onto Accurel MP 1000, a hydrophobic polypropylene support. The effects of enzyme concentration, pH, temperature, and glutaraldehyde post-treatment were systematically evaluated. Immobilization generally enhanced enzyme stability, which was further improved in several cases by glutaraldehyde crosslinking. The immobilized preparations retained over 50% of their initial activity for 3-6 cycles, and 7-10 cycles following glutaraldehyde t
