Retinal Structure of Poecilia sphenops: Photoreceptor Mosaics, Synaptic Ribbon Patterns, and Glial Cell Expressions.

The specific arrangement and distribution of photoreceptors in the retina can vary among different fish species, with each species exhibiting adaptations related to its habitat, behavior, and visual requirements. Poecilia sphenops, a diurnal fish, was the focus of this study. The retinas of a total of eighteen Molly fish were investigated utilizing light and electron microscopy. The retina exhibited a square mosaic pattern of the inner segments of cones. This pattern comprised double cones positioned along the sides of a square, with two types of single cones situated at the center and corners

Electroretinographic and clinicopathologic correlations of retinal dysfunction in infantile neuronal ceroid lipofuscinosis (infantile Batten disease).

Infantile neuronal ceroid lipofuscinosis (INCL) is an autosomal recessive disease that results from deficiency of palmitoyl-protein thioesterase-1 (PPT1). INCL leads to retinal blindness, neurodegeneration, and early death. We studied the clinical features and electroretinogram (ERG) in three patients and histopathologic and immunofluorescence analyses of the retina in the third patient, who died at 3 years 2 months of age. The ERGs for the 2 youngest patients (ages 1.7 and 2.3 years) showed normal scotopic bright flash a-wave amplitudes with severe loss of b-wave (electronegative ERG), indica

The effects of zinc deficiency on developing photoreceptors in the rat retina: a scanning microscopic study.

A study was undertaken to observe the effects of dietary zinc deficiency on the topographical morphology of developing photoreceptor (rod) cells in the sensory retina of the rat. Observations were made by means of scanning electron microscopy (SEM). Pups were utilized from three groups including (1) zinc deficient (2) pair-fed controls and (3) ad libitum controls. Animals ranged in age from birth to twenty days. Femur zinc levels were measured by flame atomic absorption spectrophotometry. A statistically significant increase in the number of photoreceptor cells continued through the 6th day, h

A novel magnesium-dependent mechanism for the activation of transducin by fluoride.

Activation of transducin-GDP by NaF is mainly mediated by aluminofluorde or beryllofluoride complexes acting as GTP gamma-phosphate analogs. In millimolar magnesium, NaF at concentrations above 3 mM is active even in the absence of aluminium or beryllium. This activation has a Hill coefficient of 3 with respect to F-, and its rate is linear with respect to Mg2+ concentrations above 2 mM. Upon fluoride dilution, inactivation rate is hundreds of times faster than for aluminofluoride-activated T alpha GDP. We propose that at high NaF concentrations, 3 hydrogen-bonded fluorides in the gamma-phosph

Biochemical characterization of retinal protein and phospholipid synthesis in mice exposed transplacentally to N-methyl-N-nitrosourea.

Transplacental exposure to the DNA alkylating agent N-methyl-N-nitrosourea on day 16 of gestation in CD-1 albino mice induces a degeneration of the retina, the severity of which depends upon the dosage level of the drug. A 1 mg kg-1 dose provokes a progressive retinal degeneration in the offspring which begins at about 4-6 weeks of age and is characterized by gradual thinning of the retinal layers. A 15 mg kg-1 dosage of MNU provokes severe retinal dysplasia characterized morphologically by rosettes in the outer nuclear layer and loss of rod outer segments (ROS). In the present biochemical exp

Photoreceptor cell dysplasia in two Tippler pigeons.

Two 12-week-old Tippler pigeons were evaluated for ocular abnormalities associated with congenital blindness. The pigeons were emaciated and blind. Biomicroscopy and direct and indirect ophthalmoscopy findings of the Tippler pigeons were normal with the exception of partially dilated pupils at rest. Scotopic (blue stimuli) and photopic monocular electroretinograms were extinguished in the blind Tippler pigeons. Histological and electron microscopy studies revealed reduced numbers of rods and cones, and an absence of the double cone complex. The photoreceptor cells' outer segments were absent,

Vigabatrin-induced retinal toxicity is partially mediated by signaling in rod and cone photoreceptors.

Vigabatrin (VGB) is a commonly prescribed antiepileptic drug designed to inhibit GABA-transaminase, effectively halting seizures. Unfortunately, VGB treatment is also associated with the highest frequencies of peripheral visual field constriction of any of the antiepileptic drugs and the mechanisms that lead to these visual field defects are uncertain. Recent studies have demonstrated light exposure exacerbates vigabatrin-induced retinal toxicity. We further assessed this relationship by examining the effects of vigabatrin treatment on the retinal structures of mice with genetically altered ph

Measurement of sodium-calcium exchange in salamander rods.

1. Methods employing suction electrodes to measure the small inward currents associated with the exchange of internal Ca2+ for external Na+ in salamander rod outer segments are described. 2. The ratio of the integral of the exchange current to the integral of the Ca2+ current during the loading period averaged 0.37, which is consistent with 1 Ca2+ ion exchanging with 2.7 Na+ ions, in approximate agreement with Yau & Nakatani (1984b). 3. The transient pumping current observed when external Na+ was restored after a few seconds in isotonic Ca2+ with IBMX (3-isobutyl-1-methylxanthine) consisted of

The GTP-binding protein of rod outer segments. II. An essential role for Mg2+ in signal amplification.

The role of Mg2+ in the GTP hydrolytic cycle was investigated by using purified subunits (G alpha and G beta, gamma) of the GTP-binding protein isolated from Bufo marinus rod outer segments (ROS). Mg2+ markedly stimulated the rate of GTP and guanosine-5'-O-(3-thiotriphosphate) (GTP gamma-s) binding to G alpha. This effect was especially striking in the presence of very small quantities of illuminated ROS disc membranes. GTP hydrolysis could occur in the absence of Mg2+, and Mg2+ increased the rate of GTP hydrolysis only about 50%. These data indicate that Mg2+ plays a fundamental role in ampli

Development of the light response in neonatal mammalian rods.

The sensitivity to light is low in many neonatal mammals when compared with that in the adult. In human infants at one month of age, for example, the dark-adapted sensitivity for detection of large stimuli is 50 times lower than in the adult, and in rats the overall sensitivity of the neonatal retina is also low compared with the adult. This low sensitivity in the neonate has been attributed to a number of factors, but the possibility that the photoreceptors themselves might be an important limitation on the overall visual sensitivity has not so far been clearly established. Here we record the

Effect of light on the transfer of sugars from sugar nucleotides to rod outer segment membranes of control and dystrophic rats.

The transfer of N-acetyl-D-glucosamine (GlcNAc), D-mannose (Man), D-galactose (Gal) and L-fucose (Fuc) from their nucleotide complexes to isolated rod outer segment (ROS) membranes obtained from dark-adapted 21 +/- 2 days old dystrophic (RCS) and control (RCS-rdy+) rat retinas, was studied under light or dark conditions of incubation. It was found that all of these sugars were transferred to ROS membranes in the dark. Under these conditions there was significantly less (p less than 0.001) Gal transferred to dystrophic than to control membranes. Exposure to light affected the transfer of Gal an

Reactivating the phototransduction cascade with a mutation agnostic gene therapy preserves vision in rod-cone dystrophies.

Rod-cone dystrophy (RCD) comprises genetic conditions where rod photoreceptor degeneration leads to cone loss, causing progressive vision loss. We investigated the phototransduction cascade in degenerating cones using two RCD mouse models and found that opsin and arrestin expression continues in the cell body during outer segment degeneration. Based on this observation, we explored reactivating cones through G-protein-coupled inwardly rectifying K (GIRK) channel expression. Using adeno-associated viral delivery of GIRK channels, we achieved improved visual function in both mouse models. Additi

PIKfyve preserves endolysosomal function in photoreceptors and RPE cells to maintain retinal integrity.

Photoreceptors rely on efficient clearance of outer segment material and mislocalized proteins to maintain cellular health and visual function. While retinal pigment epithelial (RPE) cells remove shed outer segment tips through daily phagocytosis, the mechanisms by which photoreceptors eliminate misfolded or mistargeted proteins are not well understood. Here, we identify PIKfyve, a lipid kinase that synthesizes phosphatidylinositol 3,5-bisphosphate [PI(3,5)P₂], as a key regulator of degradative and metabolic pathways in the retina. PIKfyve is highly expressed in rod photoreceptors, and its sel

Neuronatin is a stress-responsive protein of rod photoreceptors.

Neuronatin (NNAT) is a small transmembrane proteolipid that is highly expressed in the embryonic developing brain and several other peripheral tissues. This study is the first to provide evidence that NNAT is detected in the adult retina of various adult rod-dominant mammals, including wild-type (WT) rodents, transgenic rodents expressing mutant S334ter, P23H, or T17M rhodopsin, non-human primates, humans, and cone-dominant tree shrews. Immunohistochemical and quantitative real time polymerase chain reaction (qRT-PCR) analyses were applied to detect NNAT. Confocal microscopy analysis revealed

Mettl14-mediated m6A modification is essential for visual function and retinal photoreceptor survival.

BACKGROUND: As the most abundant epigenetic modification of eukaryotic mRNA, N6-methyladenosine (m6A) modification has been shown to play a role in mammalian nervous system development and function by regulating mRNA synthesis and degeneration. However, the role of m6A modification in retinal photoreceptors remains unknown. RESULTS: We generated the first retina-specific Mettl14-knockout mouse models using the Rho-Cre and HRGP-Cre lines and investigated the functions of Mettl14 in retinal rod and cone photoreceptors. Our data showed that loss of Mettl14 in rod cells causes a weakened scotopic

Super-resolution microscopy reveals a Rab6a-dependent trafficking hub for rhodopsin at the mammalian rod photoreceptor Golgi.

Rod photoreceptor stability is critical for retinal health and lifelong vision. Rhodopsin (Rho) trafficking is essential for rod homeostasis, as its mislocalization precedes rod cell death in inherited retinal disorders such as retinitis pigmentosa. Despite its importance, the molecular mechanisms of Rho trafficking in mammalian rods remain largely undefined. We investigated Rho's subcellular organization in the mammalian rod Golgi complex. We utilized STORM and structured illumination microscopy super-resolution imaging to map Golgi proteins with Rho in mouse and macaque rods. Our analysis fo

Pigment epithelial and retinal phenotypes in the vitiligo mivit, mutant mouse.

PURPOSE: To describe the abnormal phenotype in retinal pigment epithelium (RPE) and neural retina of vitiligo mutant mice from embryonic stages to old age. METHODS: Eyes of wild-type controls and congenic vitiligo mutants were examined by light and electron microscopy from embryonic day (E) 12 to 2 years of age. The amount and distribution of pigment in the RPE was studied in wholemounts. RESULTS: Earliest phenotypic expression of mivit is seen in the RPE, which is abnormally multilayered dorsally at E12 to E13, and contains both hyperpigmented and hypopigmented patches. Cdhr1a and pcdh15b may link photoreceptor outer segments with calyceal processes revealing a potential mechanism for cone-rod dystrophy.

Cone-rod dystrophy (CRD) is a macular degeneration disorder characterized by initial cone cell degeneration. Mutations in CDHR1, a photoreceptor-specific cadherin, have been found to be associated with the incidence of CRD. While studying the function of CDHR1, we observed that the localization of the zebrafish homologue, cdhr1a, resembles that of calyceal process (CPs). When co-labeling CPs using pcdh15b, we observed that cdhr1a, in the outer segment (OS), juxtaposes with pcdh15b, found in the CP. Similar localization patterns were detected in human, macaque, xenopus, ducks, gerbil, and mouse

Distribution of macular xanthophylls between domains in a model of photoreceptor outer segment membranes.

A model of photoreceptor outer segment (POS) membranes has been proposed, consisting of an equimolar ternary mixture of 1-palmitoyl-2-docosahexaenoylphosphatidylcholine/distearoylphosphatidylcholine/cholesterol. It was shown that, as in membranes made from the raft-forming mixture, in the model of POS membranes, two domains are formed: the raft domain (detergent resistant membranes, DRM), and the bulk domain (detergent soluble membranes, DSM). Saturation-recovery EPR discrimination by oxygen transport method also demonstrated the presence of two domains in this model system in situ at a wide r
