Optogenetic stimulation of inner retinal neurons restores retinal computations in a virtual degenerated human retina.

Recent advances in genetic engineering have enabled the creation of diverse novel opsins with significant potential for restoring vision. In this study, we introduce a computational model that simulates the human retina's response to optogenetic stimulation across various inner retinal cell types. Our model captures detailed retinal connectivity across multiple neural layers, incorporates conductance-based dynamics at both cellular and synaptic levels, and employs analytical formulations to represent optogenetic processes. Our findings suggest that it is possible to selectively modulate distin

Differential effects of experimental glaucoma on intrinsically photosensitive retinal ganglion cells in mice.

Glaucoma is a group of eye diseases characterized by retinal ganglion cell (RGC) loss and optic nerve damage. Studies, including this study, support that RGCs degenerate and die in a type-specific manner following the disease insult. Here we specifically examined one RGC type, the intrinsically photosensitive retinal ganglion cell (ipRGC), and its associated functional deficits in a mouse model of experimental glaucoma. We induced chronic ocular hypertension (OHT) by laser photocoagulation and then characterized the survival of ipRGC subtypes. We found that ipRGCs suffer significant loss, simi

The pineal complex: a morphological and immunohistochemical comparison between a tropical (Paracheirodon axelrodi) and a subtropical (Aphyocharax anisitsi) characid species.

Cardinal neon Paracheirodon axelrodi and bloodfin tetra Aphyocharax anisitsi are two species of characids with high trade value as ornamental fish in South America. Although both species inhabit middle water layers, cardinal neon exhibits a tropical distribution and bloodfin tetra a subtropical one. In this work, we carried out an anatomical, histological and immunohistochemical study of the pineal complex of P. axelrodi and A. anisitsi. In both species, the pineal complex consisted of three components, the pineal and parapineal organs and the dorsal sac (DS). The pineal organ was composed of

Immunoreactive pinopsin in pineal and retinal photoreceptors of various vertebrates.

Pinopsin is a pineal specific opsin newly identified in the pineal of birds which has an absorption maximum at 470 nm. As the opsin content of photoreceptors in the pineal complex of several species is not yet known, in the present work, we studied their pinopsin immunoreactivity in various vertebrates from cyclostomes to mammals. We also compared the immunoreactivity of pineal photoreceptors to that of retinal cones and rods of each animal. For the immunocytochemistry, we raised antibodies in rabbits against a 14 amino acids containing part of the chicken pinopsin molecule. The immunoreaction

The fate of immunoreactive opsin following phagocytosis by pigment epithelium in human and monkey retinas.

Polyclonal and monoclonal antibodies to human rhodopsin were used to identify and localize this principal glycoprotein of the photoreceptor outer segment discs on thin sections of human and monkey retinal pigment epithelium (RPE) and on immunoblots of RPE subcellular fractions following gel electrophoresis. Antiopsin was visualized with protein A-gold labeling by electron microscopy or peroxidase-linked second antibody on immunoblots. In immunocytochemical studies using polyclonal antibodies, the rod outer segments (ROS) were heavily labeled whereas cone outer segments labeling was variable an

Pinopsin is a chicken pineal photoreceptive molecule.

In avian pinealocytes, an environmental light signal resets the phase of the endogenous circadian pacemaker that controls the rhythmic production of melatonin. Investigation of the pineal phototransduction pathway should therefore reveal the molecular mechanism of the biological clock. The presence of rhodopsin-like photoreceptive pigment, transducin-like immunoreaction, and cyclic GMP-dependent cation-channel activity in the avian pinealocytes suggests that there is a similarity between retinal rod cells and pinealocytes in the phototransduction pathway. We have now cloned chicken pineal cDNA

The visual system of the alligator.

The eye tissues and liver of the alligator contain vitamin A(1) alone. The retina contains rhodopsin, typical in absorption spectrum (lambda(max) 500 mmicro); but synthesized in solution from neo-b retinene and opsin much more rapidly than are the frog, mammalian, or chicken rhodopsins previously examined. In this regard alligator rhodopsin resembles the rhodopsins and porphyropsins of fishes, all of which so far investigated are synthesized rapidly in solution. The rates of synthesis in vitro of frog and alligator rhodopsins are matched closely by the rates of rod dark adaptation in living fr

Syntaxin 3B Mediates Light-Dependent Interactions with STXBP1 and Arrestin 4: Distinct Roles in Rods and Cones.

Syntaxin 3 (STX3), a member of the soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) family, plays a central role in vesicle fusion. Beyond its synaptic localization, STX3 is also detected in the photoreceptor inner segment, where its function remains poorly understood. It is shown that STX3 interacts with rhodopsin, peripherin 2, and the rod outer segment protein 1. In rod-specific STX3 knockout retinas, these proteins are mislocalized, whereas cone opsins remain properly localized, suggesting a distinct STX3 function in cones. To further define its cone-specific d

Spectral characteristics of visual pigments in two mullet species (Pisces, Mugilidae).

The spectral properties of photoreceptors in two closely related fish species of the family Mugilidae, differing in their preferred salinity habitat, were subjected to a comparative study using microspectrophotometry and liquid chromatography. The results have shown that both species are tetrachromats and possess two classes of single cones and two classes of double cones, with paired elements containing visual pigments with the same absorption maximum (λmax). In the flathead grey mullet (Mugil cephalus), a predominantly marine species, the rods and the single (two classes) and double cones (t

Melanopsin hypersensitivity dominates interictal photophobia in migraine.

PURPOSE: To define the melanopsin and cone luminance retinogeniculate pathway contributions to photophobia in healthy controls and migraineurs. METHODS: Healthy controls and migraineurs were categorized according to the International Classification of Headache Disorders criteria. Photophobia was measured under full-field illumination using electromyography in response to narrowband lights spanning the melanopsin and cone luminance action spectra. Migraineurs were tested during their interictal headache-free period. Melanopsin-mediated post-illumination pupil responses quantified intrinsically

Immunocytochemical observations on pineal organ and retina of the Antarctic teleosts Pagothenia borchgrevinki and Trematomus bernacchii.

In spite of the unique conditions they have to operate under, the pineal organs of Antarctic fishes have not previously been examined. We determined immunohistochemically that in the end-vesicles and the pineal stalks of Pagothenia borchgrevinki (a species found directly beneath the sea-ice) as well as Trematomus bernacchii (a species preferring somewhat deeper water than the former) at least two populations of physiologically-different cells occurred that displayed reactions indicative of typical vertebrate photoreceptors. Lack of retinal degeneration in a Dram2 knockout mouse model.

Damage-regulated autophagy modulator 2 (DRAM2) is a homologue of the DRAM family protein, which can induce autophagy process. In the retina, DRAM2 is located to the inner segment of photoreceptors, the apical surface of retinal pigment epithelial (RPE) cells, and the lysosome. Pathogenic variants of DRAM2 lead to autosomal recessive Cone-rod dystrophy 21 (CORD21). Cone-rod dystrophy is characterised by primary cone involvement, or sometimes simultaneous cone and rod loss, thus leading to decreased visual acuity, colour vision deficits, photophobia, and decreased sensitivity of the central visu

Protective Effect of a Locked Retinal Chromophore Analog against Light-Induced Retinal Degeneration.

Continuous regeneration of the 11-cis-retinal visual chromophore from all-trans-retinal is critical for vision. Insufficiency of 11-cis-retinal arising from the dysfunction of key proteins involved in its regeneration can impair retinal health, ultimately leading to loss of human sight. Delayed recovery of visual sensitivity and night blindness caused by inadequate regeneration of the visual pigment rhodopsin are typical early signs of this condition. Excessive concentrations of unliganded, constitutively active opsin and increased levels of all-trans-retinal and its byproducts in photorecepto

Light, Sound, and Melatonin: Investigating Multisensory Pathways for Visual Restoration.

Multisensory integration is fundamental for coherent perception and interaction with the environment. While cortical mechanisms of multisensory convergence are well studied, emerging evidence implicates specialized retinal ganglion cells-particularly melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs)-in crossmodal processing. This review explores how hierarchical brain networks (e.g., superior colliculus, parietal cortex) and ipRGCs jointly shape perception and behavior, focusing on their convergence in multisensory plasticity. We highlight ipRGCs as gatekeepers

Direct retino-iridal projections and intrinsic iris contraction mediate the pupillary light reflex in early vertebrates.

The pupillary light reflex (PLR) adapts the amount of light reaching the retina, protecting it and improving image formation. Two PLR mechanisms have been described in vertebrates. First, the pretectum receives retinal inputs and projects to the Edinger-Westphal nucleus (EWN), which targets the ciliary ganglion through the oculomotor nerve (nIII). Postganglionic fibers enter the eye-globe, traveling to the iris sphincter muscle. Additionally, some vertebrates exhibit an iris-intrinsic PLR mechanism mediated by sphincter muscle cells that express melanopsin inducing muscle contraction. Given th

Insight from OPN1LW Gene Haplotypes into the Cause and Prevention of Myopia.

Nearsightedness (myopia) is a global health problem of staggering proportions that has driven the hunt for environmental and genetic risk factors in hopes of gaining insight into the underlying mechanism and providing new avenues of intervention. Myopia is the dominant risk factor for leading causes of blindness, including myopic maculopathy and retinal detachment. The fundamental defect in myopia-an excessively elongated eyeball-causes blurry distance vision that is correctable with lenses or surgery, but the risk of blindness remains. Haplotypes of the long-wavelength and middle-wavelength c

Activation of an Endogenous Opsin 3 Light Receptor Mediates Photo-Relaxation of Pre-Contracting Late Gestation Human Uterine Smooth Muscle Ex Vivo.

Spontaneous preterm birth (sPTB) remains a worldwide healthcare challenge. Preterm labor (PTL) is thought to be the largest reversible cause of sPTB, but current tocolytic therapies are ineffective and associated with systemic side effects from chronic use. Therefore, identifying novel mechanisms that promote human uterine smooth muscle (hUSM) relaxation is essential to improving clinical management of PTL. Here, we aimed to determine if an extraocular opsin receptor (OPN 3,4,5) system is expressed in pregnant hUSM and to characterize how photo-mediated relaxation of pre-contracting hUSM may b

Hemodynamic responses related to intrinsically photosensitive retinal ganglion cells in migraine.

To clarify whether photoreception of intrinsically photosensitive retinal ganglion cells (ipRGCs) is related to migraine, we investigated the relationship between hemodynamic responses related to neural activity and visual stimulation of ipRGCs. It has been established that photoreception in ipRGCs is associated with photophobia in migraine. However, the relationship between visual stimulation of ipRGCs and hemodynamic responses in the visual cortex has not been clarified. Hemodynamic responses in the visual cortex were measured using functional near-infrared spectroscopy (fNIRS) as signals re
