Identifying pyroptosis-related prognostic genes in the co-occurrence of lung adenocarcinoma and COPD via bioinformatics analysis.

Studies have indicated a complex association between chronic obstructive pulmonary disease (COPD) and lung adenocarcinoma (LUAD). However, the underlying mechanisms of their coexistence are still not fully understood. Thus, this study evaluated the possible mechanisms and biomarkers of COPD and LUAD by analyzing public RNA sequencing databases via bioinformatics analysis. This study obtained the LUAD datasets (TCGA-LUAD, GSE118370, and GSE30219) and the COPD dataset (GSE11784 and GSE39874) from TCGA and GEO databases, respectively. The differentially expressed genes (DEGs) were analyzed using

Screening of key genes related to disulfidptosis in psoriasis based on the analysis of WGCNA.

Psoriasis features keratinocyte hyperproliferation. This study aimed to identify disulfidptosis-related key genes in psoriasis for potential diagnostic biomarkers or therapeutic targets. Using the GSE30999 dataset, WGCNA was applied to identify key gene modules. GO, KEGG were used for analysis. IHC, Western blot, PCR and free thiol tests verified results. WGCNA clustered differentially expressed genes into 18 modules, with the blue module being significantly associated with psoriasis (Pearson's r = 0.82, P < 0.001). Six hub genes (FRK, GYS1, HECW2, MYH10, S100A12, SLC7A11) were identified. GYS

Noninvasive Biomarkers for Early Detection of Primary Sclerosing Cholangitis in Ulcerative Colitis.

Primary sclerosing cholangitis (PSC) is a progressive cholestatic liver disease strongly associated with ulcerative colitis (UC). Although uncommon, PSC-associated ulcerative colitis (PSCUC) represents a distinct subtype of inflammatory bowel diseases characterized by a higher risk of colorectal and hepatobiliary cancers. Early diagnosis remains a challenging issue because of the lack of specific biomarkers. Bioinformatic analysis of Gene Expression Omnibus (GEO) datasets identified potential, specific, and noninvasive biomarkers via differential gene expression and functional enrichment. Afte

Identification of pyroptosis-related gene S100A12 as a potential diagnostic biomarker for sepsis through bioinformatics analysis and machine learning.

Sepsis is a non-discriminatory inflammatory reaction that can result in a diverse array of organ dysfunctions, which can be fatal. Pyroptosis is a programmed mechanism of cell death that is distinguishable from apoptosis and other forms of cellular demise. However, the role of pyroptosis in sepsis remains to be further explored. In this study, by employing a combination of the difference analysis, WGCNA, Friends' analysis, and machine learning, the central gene S100A12 was successfully identified. S100A12 demonstrated superb diagnostic capabilities in both the integrated and external validatio

S100A12 as a key biomarker in a neutrophil-associated gene prediction model for sepsis diagnosis.

Sepsis is a life-threatening organ dysfunction syndrome caused by a dysregulated host response to infection. As a leading cause of mortality in intensive care units patients, it still lacks sensitive biomarkers. Therefore, this study aimed to develop a diagnostic model for sepsis and identify key driver biomarkers. Using single-cell RNA sequencing (scRNA-seq) data from the GEO database, we constructed a diagnostic model through 113 machine learning (ML) frameworks, supplemented with Shapley additive explanations (SHAP) analysis to identify pivotal genes. Results revealed a significant increase

Regulation mechanisms of disulfidptosis-related genes in ankylosing spondylitis and inflammatory bowel disease.

INTRODUCTION: Disulfidptosis is a recently identified form of cell death that contributes to maintaining the internal environment balance of an organism. However, the molecular basis of disulfidptosis in ulcerative colitis (UC), ankylosing spondylitis (AS), and Crohn's disease (CD) has not been thoroughly explored. METHODS: Firstly, the differentially expressed genes (DEGs) and disulfidptosis-associated genes (DAGs) were obtained through differential analysis between diseases (AS, CD, and UC) and control groups. After the disulfidptosis score was acquired using the single-sample gene set enri

Assessing the inflammatory response in horses undergoing gastric ulceration using salivary ADA and S100A12 as biomarkers.

The inflammatory status in horses with gastric ulcers was monitored by two inflammatory biomarkers, S100A12 and adenosine deaminase (ADA), using saliva samples from healthy and horses with gastric ulcers. Two trials were developed: an experimental trial, in which gastric ulceration was induced in 8 horses by feed deprivation, and a clinical trial, in which 20 horses without any symptomatology and 37 with symptoms compatible with gastric ulcers were used. S100A12 and ADA levels were measured in the saliva samples of all horses using previously validated assays. In both trials, horses with gastr

The alarmin S100A12 causes sterile inflammation of the human chorioamniotic membranes as well as preterm birth and neonatal mortality in mice†.

Sterile inflammation is triggered by danger signals, or alarmins, released upon cellular stress or necrosis. Sterile inflammation occurring in the amniotic cavity (i.e. sterile intra-amniotic inflammation) is frequently observed in women with spontaneous preterm labor resulting in preterm birth, the leading cause of neonatal morbidity and mortality worldwide; this condition is associated with increased amniotic fluid concentrations of alarmins. However, the mechanisms whereby alarmins induce sterile intra-amniotic inflammation are still under investigation. Herein, we investigated the mechanis

Plasma Protein Biomarkers to Detect Early Gastric Preneoplasia and Cancer: A Prospective Study.

Gastric cancer (GC) often presents a poor prognosis due to its asymptomatic phenotype at early stages. Upper endoscopy, which is the current gold standard to diagnose GC, is invasive with limited sensitivity for detecting gastric preneoplasia. Non-invasive biomarkers, such as blood circulating proteins, offer a promising alternative for the early detection of gastric lesions. In this prospective study, we identified plasma protein biomarkers for gastric preneoplasia and cancer using mass spectrometry-based proteomics in an exploratory cohort (n = 39). Fifteen promising protein candidates emerg

Single-cell transcriptome analysis reveals keratinocyte subpopulations contributing to psoriasis in corneum and granular layer.

BACKGROUND: Psoriasis is a chronic, inflammatory skin disease that is common and relapses easily. While the importance of keratinocyte proliferation in psoriasis development is well-documented, the specific functional subpopulations of epidermal keratinocytes associated with this disease remain enigmatic. MATERIALS AND METHODS: Therefore, in our analysis of single-cell transcriptome data from both normal and psoriatic skin tissues, we observed significant increases in certain keratinocytes in the stratum corneum (KC) and stratum granulosum (KG) within psoriatic skin. Furthermore, we identifie

Regulation of calcium homeostasis by S100A12 drives NETosis in chronic kidney disease.

Chronic kidney disease (CKD) is characterized by persistent renal injury and inflammation. Although S100A12 is associated with inflammatory states, its specific role in regulating neutrophil extracellular trap (NET) formation (NETosis) and contributing to renal pathology in CKD remains poorly understood. This study aimed to identify key diagnostic biomarkers and elucidate the mechanism by which S100A12 mediates NETosis in CKD. METHODS: Integrated bioinformatic analysis was performed on combined datasets (GSE37171, GSE66494, and GSE15072). Differentially expressed genes (DEGs) were screened us

S100 proteins: a new frontier in fibromyalgia research.

Fibromyalgia (FM) is a chronic condition that causes widespread pain, fatigue, and other symptoms that significantly affect quality of life. The underlying mechanisms of fibromyalgia involve both the immune system and the central nervous system. It has been proposed that changes in multiple ascending and descending pathways in the central nervous system may contribute to increased pain sensitivity in individuals with this condition. Recent research has identified S100 proteins as a new area of interest in fibromyalgia studies. These proteins are a group of small molecular weight proteins invol
