MRI derived brain atrophy in PSP and MSA-P. Determining sample size to detect treatment effects.

Progressive supranuclear palsy (PSP) and multiple system (MSA) atrophy are associated with progressive brain atrophy. Serial MRI can be applied in order to measure this change in brain volume and to calculate atrophy rates. We evaluated MRI derived whole brain and regional atrophy rates as potential markers of progression in PSP and the Parkinsonian variant of multiple system atrophy (MSA-P). 17 patients with PSP, 9 with MSA-P and 18 healthy controls underwent two MRI brain scans. MRI scans were registered, and brain and regional atrophy rates (midbrain, pons, cerebellum, third and lateral ven

Evaluation of fixed sample-size plans for Plutella xylostella (Lepidoptera: Plutellidae) on broccoli crops in Australia.

Fixed sample-size plans for monitoring Plutella xylostella (L.) (Lepidoptera: Plutellidae) on broccoli and other Brassica vegetable crops are popular in Australia for their simplicity and ease of application. But the sample sizes used are often small, approximately 10-25 plants per crop, and it may be that they fail to provide sufficient information upon which to base pest control decisions. We tested the performance of seven fixed sample-size plans (10, 15, 20, 30, 35, 40, and 45 plants) by resampling a large data set on P. xylostella in commercial broccoli crops. For each sample size, enumer

Impact of Sample Size and Deformation Measurement Techniques on Uniaxial Tensile Testing of Fiber-Based Materials.

The uniaxial tensile test is a common and fundamental test in materials science and engineering, in which a specimen is subjected to controlled tension until failure. From this, the stress-strain curve and many property parameters of the material can be calculated, such as tensile strength, ultimate strength, maximum elongation, Young's modulus, Poisson's ratio, and yield strength. As fibrous materials, such as paper and paperboard, become more popular, accurately measuring their mechanical properties becomes essential for developing and applying these materials, especially in packaging. Sample size estimates for biomarker-based outcome measures in clinical trials in autosomal dominant Alzheimer's disease.

INTRODUCTION: Alzheimer disease (AD)-modifying therapies are approved for treatment of early-symptomatic AD. Autosomal dominant AD (ADAD) provides a unique opportunity to test therapies in presymptomatic individuals. METHODS: Using data from the Dominantly Inherited Alzheimer Network (DIAN), sample sizes for clinical trials were estimated for various cognitive, imaging, and CSF outcomes. Sample sizes were computed for detecting a reduction of either absolute levels of AD-related pathology (amyloid, tau) or change over time in neurodegeneration (atrophy, hypometabolism, cognitive change). Characterisation of between-cluster heterogeneity in malaria cluster randomised trials to inform future sample size calculations.

Cluster randomised trials (CRTs) are important tools for evaluating the community-wide effect of malaria interventions. During the design stage, CRT sample sizes need to be inflated to account for the cluster heterogeneity in measured outcomes. The coefficient of variation (k), a measure of such heterogeneity, is typically used in malaria CRTs yet is often predicted without prior data. Underestimation of k decreases study power, thus increases the probability of generating null results. In this meta-analysis of cluster-summary data from 24 malaria CRTs, we calculate true prevalence and inciden

Topical pharmacokinetics of brinzolamide suspensions in rabbits and variability analysis for sample size and design considerations.

Identifying critical attributes for complex locally acting ophthalmic formulations and establishing in vitro-in vivo correlations can facilitate selection of appropriate thresholds for formulation changes that reflect lack of impact on in vivo performance. In this study the marketed antiglaucoma product Azopt® (1% brinzolamide suspension) and five other brinzolamide formulations varying in particle size distributions and apparent viscosities were topically administered in rabbits, and their ocular pharmacokinetics was determined in multiple ocular tissues. Banking Mesenchymal Stromal Cells from Umbilical Cord Tissue: Large Sample Size Analysis Reveals Consistency Between Donors.

Mesenchymal stromal cells (MSCs) have emerged as candidate cells with therapeutic potential to treat different pathologies. The underlying mechanism is paracrine signaling. The cells secrete proteins that can impact inflammation, apoptosis, angiogenesis, and cell proliferation. All are important in wound healing and tissue regeneration. Although the bone marrow has been the most widely used source of MSCs, umbilical cord tissue (CT) presents a source that is just starting to be used in the clinic, yet can be obtained with more ease and easily stored. Here, we characterize CT-MSCs obtained from

The unseen sample in cohort studies: estimation of its size and effect. Multicenter AIDS Cohort Study.

Recruitment of disease-free subjects into cohort studies and measurement of their time from exposure/infection to disease selectively excludes individuals (the unseen sample) who had earlier exposure and who have shorter times to disease. The unseen and observed samples may differ in other characteristics in addition to incubation period and exposure/infection time. For data with known truncation times, we develop non-parametric maximum likelihood estimates of the size, exposure/infection dates and distribution of incubation time in the unseen sample. We provide procedures to estimate and comp

Effect Size Deflation With Increasing Sample Size in Adjunctive Trials for Negative Symptoms in Schizophrenia: A Meta-Analysis and Meta-Regression Across Mechanisms of Action.

OBJECTIVE: Meta-analytic evidence supports the effectiveness of several mechanisms of action adjunctive to antipsychotics for the treatment of negative symptoms in schizophrenia. Nevertheless, the results have not been replicated consistently in multicenter randomized clinical trials. The authors' primary objective in this study was to assess whether differential scaling of response in the placebo arm by sample size could account for the discrepant results between smaller- and larger-scale trials. METHODS: The authors conducted a meta-analysis and meta-regression of the potential contribution

Can Synthetic Data Allow for Smaller Sample Sizes in Chronic Urticaria Research?

BACKGROUND: Robust data are essential for clinical and epidemiological research, yet in chronic spontaneous urticaria (CSU), certain patient groups, such as the elderly or comorbid patients, are often underrepresented. In clinical trials, strict inclusion and exclusion criteria frequently limit recruitment, making it difficult to achieve sufficient statistical power. Similarly, real-world observational studies may lack sufficient sample sizes for robust analysis. To address these limitations, we generated synthetic patient data that reflect these groups' clinical characteristics and variabilit

Strategy to Estimate Sample Sizes to Justify the Association between MMP1 SNP and Osteoarthritis.

Background: the impact of knee osteoarthritis (OA) poses a formidable challenge to older adults. Studies have reported that genetic factors, such as MMP1, are one of important risk factors for knee OA. Although the relationship between the genetic polymorphism of MMP1 rs1799750 and the risk of knee OA has been explored, conclusions have been nonunanimous and pending due to research sample sizes, one of determinants in studying genetic polymorphisms associated with disease. Objective: to establish a model to assess whether the genetic polymorphism of MMP1 rs1799750 is associated with knee OA ba

Estimating effective population size changes from preferentially sampled genetic sequences.

Coalescent theory combined with statistical modeling allows us to estimate effective population size fluctuations from molecular sequences of individuals sampled from a population of interest. When sequences are sampled serially through time and the distribution of the sampling times depends on the effective population size, explicit statistical modeling of sampling times improves population size estimation. Previous work assumed that the genealogy relating sampled sequences is known and modeled sampling times as an inhomogeneous Poisson process with log-intensity equal to a linear function of

Size-exclusion chromatography of biological samples which contain extremely alkaline proteins.

An improved size-exclusion chromatography (SEC) was developed to isolate extremely basic (alkaline) proteins, such as trypsin (pI=10.5), lysozyme (pI=11), and histone (pI=10.8). Develosil 300 Diol-5 (300 x 8 mm I.D., 30 nm average pore diameter) column was used with an eluent of 0.1 M sodium phosphate, 1.5 M sodium chloride, glycerol (40%, v/v), 2-propanol (10%, v/v), and Brij-58 (1%, v/v). Under these conditions, the final apparent pH becomes to 4.0, and pH adjustment is not necessary. Column temperature and flow rate were 15 degrees C and 0.2 ml/min, respectively. This elution system is stab

Fixed sized samples for type-specific surveillance of human papillomavirus in genital warts.

Surveillance data suggest that human papillomavirus (HPV) vaccination in Australia is reducing the incidence of genital warts. However, existing surveillance measures do not assess the proportion of the remaining cases of warts that are caused by HPV types other than 6 or 11, against which the vaccine has no demonstrated effectiveness. Using computer simulation rather than sample size formulae, we established that genotyping at least 60 warts can accurately test whether the proportion of warts due to HPV types not targeted by the vaccine has increased (Type I error probability ≤ 0.05, Type II

Coverage-based rarefaction and extrapolation: standardizing samples by completeness rather than size.

We propose an integrated sampling, rarefaction, and extrapolation methodology to compare species richness of a set of communities based on samples of equal completeness (as measured by sample coverage) instead of equal size. Traditional rarefaction or extrapolation to equal-sized samples can misrepresent the relationships between the richnesses of the communities being compared because a sample of a given size may be sufficient to fully characterize the lower diversity community, but insufficient to characterize the richer community. Thus, the traditional method systematically biases the degre

Substituting blood-based biomarkers for imaging measures in Alzheimer's disease studies: implications for sample size and bias.

BACKGROUND: Blood-based biomarkers for Alzheimer's disease (AD) pathology are appealing options in large population-based studies due to their low cost, minimal invasiveness, and feasibility of collection in non-clinical settings. Despite these benefits, blood-based biomarkers have lower test-retest reliability than neuroimaging measures like amyloid positron emission tomography (amyloid-PET) Centiloids; trade-offs in power and bias remain unexplored. METHODS: We use data from Alzheimer's Disease Neuroimaging Initiative (ADNI) and the Anti-Amyloid Treatment in Asymptomatic Alzheimer's Disease

A digital twin methodology using retrospective patient data for sample size reduction in Alzheimer's disease clinical trials.

BACKGROUND: Recruitment for Alzheimer’s disease randomized controlled trials (RCTs) is difficult and expensive. To reduce RCT sample sizes while maintaining high statistical power, our Digital Twin Trial (DTT) methodology combines an interpretable cognitive decline prediction model with prediction-powered inference. Unlike RCT sample size reduction techniques that maintain power by recruiting only patients likely to decline, prediction-powered inference is used within the data analysis stage of the trial and does not impose additional restrictions on participant eligibility. METHODS: For DTT p

Impact of sample size of birds processed per pen in research settings for performance and quality data.

Sampling procedures in scientific experiments have long been under scrutiny. The use of power testing and estimating replications has long been utilized as methods for designing experiments. In the same sense, simulation of reanalyzed data has become an approach for understanding on a more applied basis. In the current analysis, two recent trials were simulated 1000 times using a Monte Carlo design to represent performance and quality data of broilers reared to 52 and 56 days of age. Data consisted of male and female Ross 708 final live weights and then subsequently deboned into breast, tender
